Phalangeal bone anomalies in the European common toad Bufo bufo from polluted environments.

Every spring, many of amphibians are killed by motor vehicles on roads. These road-killed animals can be used as valuable material for non-invasive studies showing the effect of environmental pollution on amphibian populations. The aims of our research were to check whether the phalanges of road-killed toads may be useful as material for histological analysis, and whether various degrees of human impact influence the level in bone abnormalities in the common toad. We also examined whether the sex and age structure of toads can differ significantly depending in the different sites. We chose thr

Differential toxicity and uptake of Diazinon on embryo-larval development of Rhinella arenarum.

Diazinon, an anti-cholinesterase organophosphate, is an extensively used pesticide. The main objective of this work was to assess the lethal and sublethal effects of Diazinon and its comparison with the uptake by embryos and larvae of the common South American toad Rhinella arenarum by means of standardized bioassays during acute (96 h), short-term chronic (168 h) and chronic (504 h) exposures. Toxicity resulted time- and stage-dependent, thus the lethal concentration 50 for 96 h, 168 h and 504 h were 27.2; 20.1 and 6.8 mg Diazinon L(-1) for embryos and 8, 6.7 and 1.9 mg Diazinon L(-1) for lar

Cyanide inhibition of chloride conductance across toad skin.

The effect of cyanide (CN(-)) on voltage-activated or cAMP-induced passive chloride conductance (G(Cl)) was analyzed in isolated toad skin. Comparatively low concentrations of CN(-) inhibited G(Cl) almost completely and fully reversibly, regardless of whether it was applied from the mucosal or serosal side. The IC(50) was 180 +/- 12 microm for voltage-activated G(Cl) and 305 +/- 30 microm for the cAMP-inducted conductance. At [CN] <100 microm, the initial inhibition frequently declined partly in the continuous presence of CN(-). Inhibition was independent of the presence of Ca(2+). Inhibition

An Efficient Strategy Based on Liquid-Liquid Extraction with Three-Phase Solvent System and High Speed Counter-Current Chromatography for Rapid Enrichment and Separation of Epimers of Minor Bufadienolide from Toad Meat.

This study presents an efficient strategy based on liquid-liquid extraction with three-phase solvent system and high speed counter-current chromatography for rapid enrichment and separation of epimers of minor bufadienolide from toad meat. The reflux extraction conditions were optimized by response surface methodology first, and a novel three-phase solvent system composed of n-hexane/methyl acetate/acetonitrile/water (3:6:5:5, v/v) was developed for liquid-liquid extraction of the crude extract. This integrative extraction process could enrich minor bufadienolide from complex matrix efficientl

Bioaccumulation and maternal transfer of mercury and selenium in amphibians.

Amphibian population declines have been documented worldwide and environmental contaminants are believed to contribute to some declines. Maternal transfer of bioaccumulated contaminants to offspring may be an important and overlooked mechanism of impaired reproductive success that affects amphibian populations. Mercury (Hg) is of particular concern due to its ubiquity in the environment, known toxicity to other wildlife, and complex relationships with other elements, such as selenium (Se). The objectives of the present study were to describe the relationships between total Hg (THg), methlymerc

Temperature and central chemoreceptor drive to ventilation in toad (Bufo paracnemis).

The central chemoreceptor drive to ventilation was assessed in unanesthetized toads, Bufo paracnemis, exposed to three different temperatures: 15, 25 and 35 degrees C. The acid-base status of the fourth ventricle was manipulated by mock CSF perfusion. In additional experiments, arterial pH was varied by inspiration of hypercapnic gas mixtures. Ventilation was measured directly by pneumotachography and arterial blood samples were analyzed using electrodes for pH and PO2. Regardless of temperature, the ventilatory control of acid-base status was predominantly central. Moreover, an increase in te

Life-threatening pediatric poisoning due to ingestion of Bufo bufo toad eggs: A case report.

Bufo parotid glands and eggs contain cardiac glycosides also known as bufadienolides. This class of molecules can cause digoxin-like cardiac toxicity, as they can block the sodium potassium-adenosine triphosphatase (Na/K-ATPase) pump. Poisoning with these toxins is rare but carries a high mortality risk. There are only a few cases of toad poisoning that have been reported worldwide, mainly in the southern hemisphere. We will describe the case of a child on the autistic spectrum disorder who developed an acute and severe cardiac bradyarrhythmia soon after being in a mountain creek. The child in

Naproxen and Its Phototransformation Products: Persistence and Ecotoxicity to Toad Tadpoles (Anaxyrus terrestris), Individually and in Mixtures.

Although pharmaceutical pollution is a global environmental concern, much remains unknown about the transformation of pharmaceuticals in the wild and their effects on wildlife. In the environment, pharmaceuticals typically transform to some extent into different, structurally related compounds. Pharmaceutical transformation products resulting from exposure to sunlight (i.e., ultraviolet radiation) in surface waters are of particular concern; these products can be more hydrophobic, persistent, and toxic than their parent compounds. In the present study, naproxen, a widely used nonsteroidal anti

Effect-based environmental monitoring for thyroid disruption in Swedish amphibian tadpoles.

It is well-known that the metamorphosis process in amphibians is dependent on thyroid hormones. Laboratory studies have shown that several environmental contaminants can affect the function of thyroid hormones leading to alterations in the amphibian metamorphosis. The basic idea of the present study was to elucidate if the amphibian metamorphosis might be a useful tool as biomarker for effect-based environmental monitoring, examining wild tadpoles for potential thyroid hormone disruption. A laboratory test was performed to identify the responses from exposure to 6-propylthiouracil (PTU), which

Immediate and lag effects of pesticide exposure on parasite resistance in larval amphibians.

Across host-parasite systems, there is evidence that pesticide exposure increases parasite loads and mortality following infection. However, whether these effects are driven by reductions in host resistance to infection or slower rates of parasite clearance is often unclear. Using controlled laboratory experiments, we examined the ability of larval northern leopard frogs (Lithobates pipiens) and American toads (Anaxyrus americanus) to resist and clear trematode (Echinoparyphium sp.) infections following exposure to the insecticide carbaryl. Northern leopard frogs exposed to 1 mg L-1 of carbary

Temperature effects on baroreflex control of heart rate in the toad, Rhinella schneideri.

For an adequate blood supply to support metabolic demands, vertebrates regulate blood pressure to maintain sufficient perfusion to avoid ischemia and other tissue damage like edema. Using a pharmacological approach (phenylephrine and sodium nitroprusside) we investigated baroreflex sensitivity at 15, 25, and 30°C in toads Rhinella schneideri. Baroreflex sensitivity presented a high thermal dependence (Q10=1.9-4.1), and the HR-baroreflex curve was shifted up and to the right as temperature increased from 15 to 30°C. Baroreflex variables, namely, HR range, gain50 (maximal gain) and normalized ga

Berberine-induced pigment dispersion in Bufo melanostictus melanophores by stimulation of beta-2 adrenergic receptors.

Reduced production of melanin by decreased or the absence of melanocytes leads to various hypopigmentation disorders, and the development of melanogenetic agents for photoprotection and hypopigmentation disorders is one of the top priority areas of research. Hence, the present study was carried out to elucidate the ability of berberine, a principal active ingredient present in the roots of the herb Berberis vulgaris to stimulate pigment dispersion in the isolated skin melanophores of the toad Bufo melanostictus. In the present study, mean melanophore size index of the isolated skin melanophore

A study of the relict fish fauna of northern Chad, with the first records of a polypterid and a poeciliid in the Sahara desert.

Seventeen species and sub-species of fishes belonging to four families (Cyprinidae, Clariidae, Aplocheilidae, Cichlidae) were known to occur in perennial bodies of water in the Sahara desert. The study of fishes collected in Lake Boukou near Ounianga Serir (Borkou, northern Chad) shows, for the first time, the occurrence in the Sahara desert of relict populations of Polypterus senegalus (Polypteridae) and Poropanchax normani (Poeciliidae). The Cichlidae Tilapia zilli was also collected in this lake. With these new records, the relict fish fauna currently known in lakes and gueltas of the Borko

The Heterogametic Transition in European Bufo Toads Switches the Sex Linkage of Key Vertebrate Sex Determination Genes and Associates with a Large Sex Chromosome Effect.

Characterizing the diversity and lability of the amphibian sex chromosomes holds key to understand what drives sex chromosome turnovers and assess the role of sex-linked genes in reproductive isolation and speciation. Here, we show that the heterogametic transition previously reported between the hybridizing toads Bufo bufo (ZW) and Bufo spinosus (XY) is nonhomologous, potentially implicates key genes of the vertebrate sex determination cascade (SOX9, DMRT1, and AMH), and is characterized by a much shorter ZW than XY segment. Integrating this information with published hybrid zone data suggest
