Interaction of vitamin A deficiency and excess fluoride in calcification of rat bones and teeth.

Using a 2 X 2 factorial design, we evaluated the possible interaction of vitamin A deficiency and excess fluoride in rat dentin and bone. Simultaneous presence of excess fluoride plus vitamin A deficiency resulted in a significant decrease in bone fluoride concentration compared to the presence of excess fluoride alone. Vitamin A deficiency alone significantly reduced calcium concentration in dentin formed during the deficient period.

Oral fluoride reservoirs and the prevention of dental caries.

Current models for increasing the anti-caries effects of fluoride (F) agents emphasize the importance of maintaining a cariostatic concentration of F in oral fluids. The concentration of F in oral fluids is maintained by the release of this ion from bioavailable reservoirs on the teeth, oral mucosa and - most importantly, because of its association with the caries process - dental plaque. Oral F reservoirs appear to be of two types: (1) mineral reservoirs, in particular calcium fluoride or phosphate-contaminated 'calcium-fluoride-like' deposits; (2) biological reservoirs, in particular (with r

Otospongiosis and sodium fluoride. A blind experimental and clinical evaluation of the effect of sodium fluoride treatment in patients with otospongiosis.

The effect of sodium fluoride treatment in patients with otospongiosis has been evaluated blindly in a morphological and microchemical element analysis of otospongiotic specimens together with a prospective clinical double-blind, placebo-controlled study. The results show that using the calcium/phosphorus ratio as an indication for bone maturity, the sodium fluoride treatment can stabilize otospongiotic lesions in retaining calcium relative to phosphorus. The clinical double-blind, placebo-controlled study of 95 patients showed a statistically significant worse deterioration of the hearing los

Potential exposure and risk of fluoride intakes from tea drinks produced in Taiwan.

Tea is the second most commonly consumed drink in the world. Excess fluoride intakes from tea drinks may cause health effects. This work assesses infusible fluoride levels in popular tea sold in Taiwan and evaluates potential exposure factors. Lungjing, pouchong, tienguanyin, oolong, pureh, and black tea specimens were purchased from different counties in Taiwan. Fluoride levels were evaluated in one complete cycle of tea making as well as at different calcium carbonate contents in water, with glass or porcelain teapots, and with/without adding sugar. Oolong tea leaves in each manufacturing st

Oral decontamination with calcium or magnesium salts does not improve survival following hydrofluoric acid ingestion.

Hydrofluoric acid (HF) ingestion may result in rapid death from systemic fluoride poisoning. Because fluoride binds calcium and magnesium, oral administration of calcium or magnesium salts has been suggested as useful therapy for HF ingestion. This study evaluates oral administration of calcium and magnesium salts for the prevention or attenuation of acute HF toxicity following oral exposure in a mouse model. Mixed breed CF-1 mice were randomized to treatment with a lethal-dose HF premixed with calcium chloride, magnesium sulfate, or deionized water by gavage. Average survival time for the thr

Dental fluorosis: chemistry and biology.

This review aims at discussing the pathogenesis of enamel fluorosis in relation to a putative linkage among ameloblastic activities, secreted enamel matrix proteins and multiple proteases, growing enamel crystals, and fluid composition, including calcium and fluoride ions. Fluoride is the most important caries-preventive agent in dentistry. In the last two decades, increasing fluoride exposure in various forms and vehicles is most likely the explanation for an increase in the prevalence of mild-to-moderate forms of dental fluorosis in many communities, not the least in those in which controlle

Anabolic effects of fluoride on bone.

Fluoride exerts a biphasic action at the level of osteoblasts, on bone mineral, on bone structure and function, and in the treatment of osteoporosis. At low circulating concentrations, skeletal uptake of fluoride is limited and the effects are beneficial. At higher concentrations and greater skeletal uptake, fluoride may cause the formation of abnormally mineralized bone of impaired quality. A new treatment program entailing intermittent slow release sodium fluoride (SR-NaF) with continuous calcium citrate may capture desirable qualities of fluoride without toxic effects, and be therapeuticall

Correlations between the concentrations of fluoride and some other constituents in tea infusions and their possible dental caries-preventive effect.

Tannic acid, total, complexed and ionized fluoride, calcium, total phosphate, zinc, aluminium, copper, iron, mercury and manganese were determined in infusions prepared from some or all of 30 brands of tea. Mercury and copper were not detected. Significant positive correlations were observed between fluoride, aluminium and manganese. The low reactivity of fluoride in tea infusions with powdered hydroxyapatite compared with that in dilute sodium fluoride solutions may be in part explained by the presence of aluminium and manganese.

Environmental fluoride exposure and bone metabolism: Molecular pathways and health implications.

Fluoride, a common environmental pollutant in drinking water and air, poses a major public health risk in endemic regions. Fluorosis disrupts bone structure and density and impairs key physiological processes, including calcium-phosphorus homeostasis, hormone regulation, enzyme activity, and cytokine-mediated signaling-thereby compromising skeletal integrity. This review explores the molecular mechanisms through which chronic environmental fluoride exposure disrupts bone metabolism, leading to structural and functional skeletal damage. Chronic fluoride exposure alters bone mineral crystals due

Delayed fatal hyperkalemia in a patient with acute fluoride intoxication.

A 19-year-old man presented with acute fluoride poisoning. Initially his serum electrolytes were normal, but two hours later he developed ECG evidence of hyperkalemia followed by refractory ventricular fibrillation, suggesting that hyperkalemia may be important in the cardiotoxicity of acute fluoride intoxication. Treatment of fluoride-induced hyperkalemia consists of removal of fluoride from the body by dialysis, binding fluoride with aluminum or calcium, or enhancing fluoride excretion by inducing a metabolic alkalosis. Direct treatment of the hyperkalemia with glucose, insulin, and bicarbon

Formation of fluoride on enamel in vitro after exposure to fluoridated mouthrinses.

The aim of this study was to quantify the formation of alkali-soluble fluoride (loosely bound fluoride such as calcium fluoride-like material and absorbed fluoride) and alkali-insoluble fluoride (firmly bound fluoride or apatitically bound fluoride) when fluoride mouthrinsing solutions were applied on sound human enamel in vitro. Two commercial products containing 0.2% or 0.05% NaF were used during 30 sec, 60 sec, 5 min, and 60 min. The formation of loosely bound fluoride was determined by KOH extraction and visualized by scanning electron microscopy. The firmly bound fluoride was measured by

Contribution to the physicochemical rationale for the caries reducing effect of fluoride.

The effect of fluoride on the stability of apatites containing Na+ and CO3(2)- ions is described qualitatively. Also the boundaries of the apatite stability field in the quasi-septary system CaO - P2O5 - H2O - Na2O - CO2 - NaF - MX are presented. At these boundaries brushite (or monetite), octocalciumphosphate, calcite and calcium fluoride can become stable solid phases. As a consequence, certain mechanisms for the caries reducing effect of fluoride gain probability over other proposed mechanisms. First, it is likely that endemic fluoride yields more fully mineralized enamel which in turn resu

Effect of fluoride varnishes on oral bacteria of preschool children with cavitated and non-cavitated carious lesions: randomized clinical trial.

We compare the effect of calcium and phosphate-containing sodium fluoride (NaF) varnishes to conventional NaF varnish on S. mutans and L. fermentum counts. 3-4 years old children were grouped according to their caries status (n = 45 each): caries-free, with non-cavitated and with cavitated lesions. Each group was randomly subdivided (n = 15 each) into: Group 1- 5% NaF, Group 2- 5% NaF with tricalcium phosphate, Group 3- 5% NaF with casein phosphopeptide- amorphous calcium phosphate. Biofilm and saliva were collected to quantify microorganisms at baseline (T1) and 24-months (T2). Differences be
