Inhibition of contact sensitivity to oxazolone by the calmodulin inhibitor trifluoroperazine.

Topical application of trifluoroperazine (TFP), a calmodulin inhibitor, on the skin of CBA mice previously immunized with 2-phenyl-4-ethoximethylene-oxazolone (oxazolone) blocks the expression phase of contact sensitivity. More detailed analysis of the TFP-mediated inhibition of contact sensitivity shows that TFP significantly suppresses the passive transfer of contact sensitivity when added in vitro to oxazolone-immune cells at a molar concentration of 10(-4) -10(-5). Furthermore, the DNA synthesis of draining lymph node cells from immune mice challenged with oxazolone was suppressed when cul

Ca2+-calmodulin regulates the induction and expression of macrophage cytotoxicity.

Ca2+-activated calmodulin (CAM) is known to regulate cellular responses of diverse cell types to external stimuli. To examine the effects of Ca2+ influx and CAM on macrophage (MP) cytotoxicity, we used 51Cr-labeled cells as targets and in vivo or in vitro activated MPs from C57 BL/6 or BALB/c mice as effectors in a 16-h cytotoxicity assay. MPs activated in vivo by administration of vaccinia virus or in vitro with lymphokine (LK) were cytotoxic for a variety of tumor cell lines, including SV3T3, but not for normal 3T3 cells. Addition of verapamil (0.1 mM), a Ca2+ channel blocker, to MPs activat

A possible role for calmodulin in Ca2+-induced swelling of mitochondria.

Ca2+-induced mitochondrial swelling was inhibited by a low concentration of calmodulin antagonists. Two affinities of Ca2+ to mitochondrial swelling were observed: high (2-5 microM) and low (more than 100 microM) systems. The high-affinity change was inhibited by micromolar level of trifluoperazine and W-7, but not by W-5. The possible mechanism of this inhibition and the role of CaM in mitochondria are discussed.

Binding of trifluoperazine and fluorene-containing compounds to calmodulin and adducts.

Calmodulin can be specifically acylated with a fluorene-containing hydrophobic spin-labeling reagent at just Lys 75 or at Lys 75 and Lys 148. The binding of trifluoperazine to calmodulin and the two adducts was determined using a Hummel-Dreyer procedure, and binding of the phenothiazine was found to be characterized by apparent positive cooperativity and an apparent limiting stoichiometry of about seven binding sites per protein molecule. Two non-reactive fluorene-containing compounds were synthesized, and both reagents exhibited far less binding to calmodulin than did trifluoperazine. One of

[Effect of calmodulin inhibitors on the contraction and relaxation of the heart muscle].

The calmodulin inhibitor trifluoroperazine (5 microM) diminished the velocity of contraction and relaxation of the guinea-pig papillary muscle by 31 and 53%, respectively. Methophenazine showed approximately the same affinity to both calmodulin and trifluoroperazine and nearly 40-fold less affinity to troponin. Methophenazine (5 microM) did not change the contraction velocity and diminished the relaxation velocity ty 21%. It is suggested that calmodulin participates mainly in the control of myocardial relaxation.

Competitive binding assay for drugs that interfere with calmodulin function.

A direct 125I-calmodulin-binding system utilizing a particulate cytoskeletal preparation of the neuronal calmodulin-dependent protein kinase type II provides a simple, rapid filtration-based method for assessing anti-calmodulin drug activity. The binding assay avoids potential artifacts due to direct drug effects on the standard phosphodiesterase enzyme assay while obtaining comparable IC50 values for a spectrum of drugs. It also provides a tool to probe mechanisms of action of putative anticalmodulin agents which may help elucidate selective pharmacologic modifications of pathophysiological p

The nature of the trifluoperazine binding sites on calmodulin and troponin-C.

We have employed 1H-nuclear magnetic resonance spectroscopy to study the interaction of the drug trifluoperazine with calmodulin and troponin-C. Distinct trifluoperazine-binding sites exist in the N- and C-terminal halves of both proteins. Each site consists of a group of hydrophobic side-chains brought into proximity by the Ca2+-dependent juxtaposition of two alpha-helical segments of the protein, each, in turn, belonging to a different Ca2+-binding site in the protein half. The trifluoperazine-induced inhibition of the biological activating ability of calmodulin appears to result from confor

The role of calmodulin in human natural killer cell activity.

Calcium is known to play an essential role in the lytic mechanism of natural killer cells (NK), which form a subset of large granular lymphocytes. Many of the intracellular effects of calcium are mediated through the calcium-binding protein calmodulin. In this study we have demonstrated that the specific calmodulin inhibitors (naphthalene-sulphonamides) inhibit NK activity in humans at IC50's of 6.9 microM for W7 and 5.2 microM for W13. Comparison of the potency of these compounds with their less active counterparts suggests that NK activity is calmodulin-dependent.

Calmodulin-dependent stimulation of the NADPH oxidase of human neutrophils.

The NADPH oxidase of human neutrophils is highly sensitive to calcium concentration and is inhibited in intact cells and cell-free preparations by various phenothiazine drugs. Addition of calmodulin to preparations of NADPH oxidase stimulates enzymatic rates from 1.4-2.5-fold. Addition of calmodulin and calcium, but not calcium alone, to NADPH oxidase preparations which have been inactivated by EDTA results in the restoration of activity. No activation is observed when membrane preparations containing latent NADPH oxidase are exposed to calcium and calmodulin. These studies suggest a role for

Molecular recognition of myosin L by the Toxoplasma gondii calmodulin.

Calmodulin is a conserved EF-hand Ca2+ sensor that transduces intracellular Ca2+ signals through interactions with target proteins. Recent cryo-electron microscopy studies revealed the Toxoplasma gondii calmodulin (TgCaM) as a light chain associated with the neck of the unconventional myosin MyoL within the preconoidal P2 ring , a key component of the conoid involved in parasite motility and invasion. Here, we used NMR spectroscopy, isothermal titration calorimetry, and complementary biophysical approaches to characterize TgCaM and its interaction with a synthetic peptide spanning the predicte

Protective effects of interrupting the binding of calmodulin to mutant huntingtin.

There is evidence suggesting that transglutaminase (TG) 2 plays a role in stabilizing monomeric and aggregated huntingtin, thereby contributing to the pathophysiology of Huntington disease. Calmodulin (CaM) regulates TG2 cross-linking of N-terminal mutant huntingtin in cells and colocalizes with TG and huntingtin in inclusions in Huntington disease cortex. The current study examined the effects of small fragments of CaM in human embryonic kidney 293T cells expressing N-terminal mutant huntingtin and transglutaminase 2. Four CaM fragments were developed: first 76 amino acids, last 72 amino acid

Ca2+ and calmodulin regulate the binding of filamin A to actin filaments.

Filamin A (FLNa) cross-links actin filaments (F-actin) into three-dimensional gels in cells, attaches F-actin to membrane proteins, and is a scaffold that collects numerous and diverse proteins. We report that Ca(2+)-calmodulin binds the actin-binding domain (ABD) of FLNa and dissociates FLNa from F-actin, thereby dissolving FLNa.F-actin gels. The FLNa ABD has two calponin homology domains (CH1 and CH2) separated by a linker. Recombinant CH1 but neither FLNa nor its ABD binds Ca(2+)-calmodulin in the absence of F-actin. Extending recombinant CH1 to include the negatively charged region linker

Myosin V: regulation by calcium, calmodulin, and the tail domain.

Calcium activates the ATPase activity of tissue-purified myosin V, but not that of shorter expressed constructs. Here, we resolve this discrepancy by comparing an expressed full-length myosin V (dFull) to three shorter constructs. Only dFull has low ATPase activity in EGTA, and significantly higher activity in calcium. Based on hydrodynamic data and electron microscopic images, the inhibited state is due to a compact conformation that is possible only with the whole molecule. The paradoxical finding that dFull moved actin in EGTA suggests that binding of the molecule to the substratum turns it

[Expression of calmodulin and nNOS in the paraspinal muscles in idiopathic scoliosis].

OBJECTIVE: The aim of this study was to identify whether one side of the apex of the scoliotic curve showed greater muscular abnormalities than the other. This investigation sought to compare directly the expression and localization of Calmodulin (CaM) and nNOS of the thoracic spinal musculature on convex side with those on the concave side in the scoliosis patients attending for spinal surgery. METHODS: The patient group comprised 8 females and 2 males, scheduled for spinal surgery, in whom the apex of the scoliotic curve arose between T6 and T11. Muscle biopsies were taken bilaterally durin

Effect of calmodulin antagonists on the compound action potential of the cochlea.

This study aimed to evaluate the effect of calmodulin antagonists on the threshold of the compound action potential (CAP) and the functional recovery of the cochlea after transient ischemia. When trifluoperazine and W-7 were administered to albino guinea pigs with perilymphatic perfusion, these drugs did not significantly affect the CAP thresholds. Transient cochlear ischemia of 30-min duration was obtained via a skull base approach. Although trifluoperazine significantly ameliorated the post-ischemic CAP threshold shifts 4 h after the onset of reperfusion, 1 to 50 microM W-7 did not affect th

Effect of calmodulin-inhibitors and verapamil on the nephrotoxicity of cadmium in rat.

Recent reports indicate that calmodulin inhibitors (CIs) can modify cadmium (Cd) toxicity in rodents. Pretreatment with CIs prevents Cd-induced testicular damage in mice and reduces the severity of such damage in rats. On the other hand it has been suggested that the cellular transport of Cd can be partly inhibited by the calcium-channel inhibitor, verapamil. The aim of this study was to determine whether these inhibitors can prevent the toxic effects of Cd on the kidney which is the critical organ. For that purpose, we have examined the effects of two CIs (trifluoperazine and chlorpromazine)

Preliminary study on the effect of the selected calmodulin antagonists on the skin.

The aim of the study was to make a selection of the calmodulin antagonists, which could be useful in topical treatment of psoriasis. Four drugs were assessed: chlorpromazine, trifluoperazine, miconazole and ketoconazole. They were applied on the skin of guinea pigs, twice a day for the period of two weeks. Biopsy specimens from these sites were being taken for light microscopy, histoenzymatic examination and for evaluation of the proliferation activity of the epidermis (the AgNOR stain, after Ploton). Evidence for a functional interaction between calmodulin and the glucocorticoid receptor.

In the signaling cascade of membrane-bound receptors, calmodulin (CaM) plays an important role. However, little is known about the role of CaM in the activation of intracellular steroid receptors, which are known to act as ligand-regulated transcription factors. We report here that CaM can interact in a calcium-dependent manner with the untransformed glucocorticoid receptor (GR) complex containing hsp90. In addition, we demonstrate that four unrelated CaM antagonists (trifluoperazine, compound 48/80, W7, and phenoxybenzamine) can inhibit GR-mediated gene expression in mouse L929 cells stably-t

Effect of lead on the biological activity of calmodulin in rat brain.

In the present investigation, we observed that lead in vitro activates calmodulin at lower concentrations, and the maximum activation was observed at 30 microM concentration. In vivo lead exposure (50 mg/kg body weight, intragastrically) for a period of 8 weeks also stimulated the activity of calmodulin by 45%. The addition of trifluoperazine resulted in partially inhibiting the lead-stimulated calmodulin activity, whereas the calcium-stimulated calmodulin activity was completely inhibited by trifluoperazine. Increased covalent binding of acetaldehyde to calmodulin in the presence of calcium.

The regulatory protein, calmodulin, undergoes major conformational changes in response to changes in intracellular calcium concentration. Furthermore, calmodulin has been reported to have lysine residues which markedly increase their reactivity toward electrophilic substances in the calcium-loaded state. We found that calmodulin formed two to three times more stable adducts with acetaldehyde in the calcium-loaded state as compared to the calcium-free state. Competition-binding studies showed that calmodulin could preferentially compete with albumin for acetaldehyde in the presence, but not in
