Biochemical evaluation of triploid progenies of diploid × tetraploid breeding populations of Camellia for genotypes rich in catechin and caffeine.

To verify the quality of triploid varieties of Camellia tea species at the secondary metabolite level, we tested caffeine and catechin profiles of 97 F(1) segregating progenies in two breeding populations with a common tetraploid parent and diploid parents of two geographic and varietal origins. Catechin and caffeine levels of the triploid progenies were quantified and compared against their diploid parent. Some of the progenies showed better performance than their diploid parent. Most of the progenies of the diploid C. sinensis × tetraploid cross showed heterosis for caffeine and EGCG. Progen

The molecular mechanism of CsWRKY24-4 and CsWRKY24-5 in response to cold stress in tea plants (Camellia sinensis).

Cold stress has emerged as a major limiting factor for tea yield. In this study, we identified two low-temperature responsive WRKY transcription factors (CsWRKY24-4 and CsWRKY24-5) from tea plants (Camellia sinensis). Their expression was significantly upregulated under cold induction. Heterologous overexpression of CsWRKY24-4 and CsWRKY24-5 in Arabidopsis thaliana markedly enhanced cold tolerance in transgenic lines. Tea seedlings overexpressing CsWRKY24-4 or CsWRKY24-5 showed significantly stronger cold resistance under low-temperature stress, confirming their roles in enhancing cold toleran

Genomics insights into flowering and floral pattern formation: regional duplication and seasonal pattern of gene expression in Camellia.

BACKGROUND: The formation and domestication of ornamental traits are influenced by various aspects, such as the recognition of esthetic values and cultural traditions. Camellia japonica is widely appreciated and domesticated around the world mainly due to its rich variations in ornamental traits. Ornamental camellias have a diverse range of resources, including different bud variations from Camellia spp. as well as inter- and intra- specific hybridization. Despite research on the formation of ornamental traits, a basic understanding of their genetics and genomics is still lacking. RESULTS: He

Exploring the Effects of Magnesium Deficiency on the Quality Constituents of Hydroponic-Cultivated Tea (Camellia sinensis L.) Leaves.

Magnesium (Mg) plays important roles in photosynthesis, sucrose partitioning, and biomass allocation in plants. However, the specific mechanisms of tea plant response to Mg deficiency remain unclear. In this study, we investigated the effects of Mg deficiency on the quality constituents of tea leaves. Our results showed that the short-term (7 days) Mg deficiency partially elevated the concentrations of polyphenols, free amino acids, and caffeine but decreased the contents of chlorophyll and Mg. However, long-term (30 days) Mg-deficient tea displayed decreased contents of these constituents. Pa

Effects of Nigella sativa, Camellia sinensis, and Allium sativum as Food Additives on Metabolic Disorders, a Literature Review.

UNLABELLED: Both acquired mutations and germline genetic variation are known risk factors for chronic lymphocytic leukemia (CLL). The joint characterization of germline, acquired, and clinical risk has the potential to improve CLL risk prediction. In this study, we investigated whether the inclusion of a CLL-associated polygenic score (PGS) and two common types of clonal hematopoiesis (CH), autosomal mosaic chromosomal alterations (mCA) and CH of indeterminate potential (CHIP), could improve CLL risk stratification in 436,784 participants in the UK Biobank and a replication set of 35,382 parti

Identification of MTP gene family in tea plant (Camellia sinensis L.) and characterization of CsMTP8.2 in manganese toxicity.

Cation diffusion facilitators (CDFs) play central roles in metal homeostasis and tolerance in plants, but the specific functions of Camellia sinensis CDF-encoding genes and the underlying mechanisms remain unknown. Previously, transcriptome sequencing results in our lab indicated that the expression of CsMTP8.2 in tea plant shoots was down-regulated exposed to excessive amount of Mn2+ conditions. To elucidate the possible mechanisms involved, we systematically identified 13 C. sinensis CsMTP genes from three subfamilies and characterized their phylogeny, structures, and the features of the enc

CsBPC6-CsNACX2-CsTCS1 Module Positively Regulates Caffeine Biosynthesis Under PEG-Induced Drought Stress in Tea Plants (Camellia sinensis).

Caffeine biosynthesis in tea plants (Camellia sinensis) is dynamically regulated by environmental cues; however, the mechanisms linking early drought signals to caffeine accumulation remain unclear. Here, we show that acute PEG-induced osmotic stress induces rapid caffeine accumulation, accompanied by transcriptional activation of the key biosynthetic gene CsTCS1. Through integrative analyses, including GWAS and co-expression network modeling, we identified a CsBPC6-CsNACX2-CsTCS1 module as a central regulatory hub. Mechanistically, CsNACX2 directly binds the promoter of the caffeine synthase

Integrated metabolomic and transcriptomic analysis of Camellia sinensis var. pubilimba 'Rucheng Baimaocha' reveals distinct flavonoid and strictinin biosynthesis.

BACKGROUND: 'Rucheng Baimaocha' (RCBMC) is a traditional tea landrace found in Hunan Province, China. Its morphological characteristics differ substantially from those of the other cultivars, featuring larger mature leaves with thicker cuticle layers. Both young buds and leaf undersides are densely covered in silvery-white trichomes, indicating that RCBMC has a distinct metabolite composition compared to other cultivars. To elucidate the metabolic profile differences and their underlying molecular mechanisms, we conducted an integrated metabolomic and transcriptomic analysis of RCBMC and repre

Camellia nitidissima Flower Extract Alleviates Stress-Induced Sebaceous Dysfunction by Targeting the 11β-HSD1/PI3K/Akt/mTOR Axis.

Stress-induced sebaceous hyperactivity is a key driver of acne and seborrheic dermatitis; however, research investigating the pathway-specific mechanisms through which stress exerts its effects and the suppression of sebum production via targeting stress signaling remains relatively limited. In this study, we investigated whether Camellia nitidissima flower extract (CNF) could counteract stress-induced sebaceous dysfunction and its underlying mechanisms. Using a cortisone-stimulated SZ95 human sebocyte model, we evaluated lipid accumulation, cortisol production, signaling pathway activation, a

The transcription factor CsPAT1 from tea plant (Camellia sinensis) is involved in drought tolerance by modulating phenylpropanoid biosynthesis.

Tea plants, in particular, leafy cash crops, prefer warm and humid climates. Our previous work identified CsPAT1 as a facilitator of lignin biosynthesis in tea plants. The specific role of CsPAT1 in tea plants' abiotic stress response remains unclear. In this study, we found that the expression of CsPAT1 in tea plants was induced under drought, cold, heat, and ABA treatments. CsPAT1 transgenic Arabidopsis lines displayed enhanced drought tolerance compared with wild-type (WT) controls. The SOD and POD activities, proline content, and expression levels of drought-responsive genes were significa
