Cervical cancer: cytological cervical screening in Iceland and implications of HPV vaccines.

This paper reviews the Icelandic experience regarding the age-specific effectiveness, optimal targeted age range and intervals in cervical cancer screening and the screening implications of the HPV16/18 vaccines. The background material is based on data from a screening programme with centralized records dating back to 1964, as well as from population-based studies on the distribution of oncogenic HPV types in cancer and histologically verified CIN2-3 lesions and from the Icelandic arm of the Future II trial with Gardasil. The findings confirm significant increased rates in the screened popula

Clinical role of cervical cancer vaccination: when and whom to vaccinate?

The recent development of two highly effective vaccines against persistent infection by the 2 most important types of human papillomavirus (HPV) (16 and 18) and against high grade premalignant lesions (CIN2+) has opened a new scenario for the primary prevention of cervical cancer. The optimum target population for vaccination should be individually defined taking the following into account: 1) the efficacy of the vaccine, 2) the epidemiological context and 3) the vaccination programs available in each country. To achieve the maximum preventive benefits, the vaccine should be administered befor

Lung cancer vaccines to enhance immune checkpoint inhibitor therapy: evidence and future perspectives.

Immune checkpoint inhibitors (ICIs) have transformed the treatment landscape of lung cancer over the past decade, markedly improving antitumor responses, overall survival, and quality of life. However, durable clinical benefit is achieved in only a subset of patients, and resistance to ICIs remains a major clinical challenge. Mechanistically, resistance arises from multiple, often overlapping processes, including inadequate tumor antigen presentation, dysfunctional T-cell priming and expansion, and the presence of physical and immunosuppressive barriers within the tumor microenvironment that l

mRNA vaccines targeting HPV E6/E7: A new frontier in cervical cancer immunotherapy.

Human papillomavirus (HPV) infections, particularly high-risk types 16 and 18, are responsible for approximately 70% of cervical cancer cases worldwide. Despite the availability of effective prophylactic vaccines, therapeutic interventions for established HPV-associated malignancies remain limited. This mini-review synthesizes recent advances (2020-2026) in mRNA-LNP vaccine development targeting the HPV E6 and E7 oncoproteins. We examine the mechanistic basis for targeting E6/E7, technological advantages of mRNA-LNP platforms, and key preclinical evidence demonstrating potent T cell-mediated a

mRNA-Based Cancer Vaccines: A Review of the Current Scenario and Future Prospects.

Messenger RNA (mRNA) has gained increasing attention as a valuable tool to cure various human diseases, particularly malignant tumors. Such growing interest has been triggered largely by the phenomenal clinical success of mRNA vaccines developed using lipid nanoparticle (LNP) technology against COVID-19. mRNA may be used to produce cancer immunotherapies in numerous different ways, including cancer vaccines to induce or enhance immunity to tumor-specific antigens (TSAs) or tumor-associated antigens (TAAs). mRNA can also be used to adoptively transfer T-cells for the expression of antigen recep

Reversing the "cold" tumor microenvironment: the role of neoantigen vaccines in prostate cancer.

BACKGROUND: Prostate cancer (PCa) is a common malignancy with a "cold" tumor microenvironment (TME) that limits immunotherapy efficacy. Neoantigens from non-synonymous mutations are potential immunotherapeutic targets. METHODS: We identified the neoantigens map in the RM-1 murine prostate cancer model by the whole exome sequencing. The immunogenicity of the identified neoantigens was subsequently assessed using enzyme-linked immunospot assays. In vivo neoantigen vaccination experiments were finally conducted in mice to assess the antitumoral effect on PCa, and comprehensively understand the i

Current "state of the art" on dendritic cell-based cancer vaccines in melanoma.

PURPOSE OF REVIEW: Dendritic cells (DCs) are the gatekeepers of our immune system and indispensable in the antitumor immune response. In recent years, their classification has been revised considerably using single-cell sequencing approaches. In this review, we focus on their unique role in cancer and how specific DC subsets can be manipulated to induce an effective and durable antitumor response. RECENT FINDINGS: Historically, due to the ease of their isolation in sufficient cell numbers from peripheral blood, the utility of monocyte-derived DCs as therapeutic cancer vaccines was explored in

Comprehensive mutanome analysis of Lewis lung cancer reveals immunogenic neoantigens for therapeutic vaccines.

Personalized neoantigen vaccines are capable of eliciting vigorous T-cell responses and have been demonstrated to achieve striking therapeutic effects against cancer. Here we performed comprehensive mutanome analysis of the mouse Lewis lung cancer cells to identify tumor neoantigens followed by prediction of their MHC affinity and immunogenicity. We adopted a strategy that enables us to select neoantigens that were predicted to have high affinity to both MHC I and MHC II. Ten neoantigens were selected to synthesize peptide vaccines and tested in vivo for immunogenicity. Four neoantigen peptide

Preclinical evaluation of mRNA trimannosylated lipopolyplexes as therapeutic cancer vaccines targeting dendritic cells.

Clinical trials with direct administration of synthetic mRNAs encoding tumor antigens demonstrated safety and induction of tumor-specific immune responses. Their proper delivery to dendritic cells (DCs) requires their protection against RNase degradation and more specificity for dose reduction. Lipid-Polymer-RNA lipopolyplexes (LPR) are attractive mRNA delivery systems and their equipment with mannose containing glycolipid, specific of endocytic receptors present on the membrane of DCs is a valuable strategy. In this present work, we evaluated the capacity of LPR functionalized with a tri-ante

EBV-transformed lymphoblastoid cell lines as vaccines against cancer testis antigen-positive tumors.

EBV-transformed lymphoblastoid cell lines (LCL) are potent antigen-presenting cells. To investigate their potential use as cancer testis antigen (CTA) vaccines, we studied the expression of 12 cancer testis (CT) genes in 20 LCL by RT-PCR. The most frequently expressed CT genes were SSX4 (50 %), followed by GAGE (45 %), SSX1 (40 %), MAGE-A3 and SSX2 (25 %), SCP1, HOM-TES-85, MAGE-C1, and MAGE-C2 (15 %). NY-ESO-1 and MAGE-A4 were found in 1/20 LCL and BORIS was not detected at all. Fifteen of 20 LCL expressed at least one antigen, 9 LCL expressed ≥2 CT genes, and 7 of the 20 LCL expressed ≥4 CT

Counterpoint. Cancer vaccines: single-epitope anti-idiotype vaccine versus multiple-epitope antigen vaccine.

Anti-idiotype (Id) vaccine therapy has been tested and shown to be effective, in several animal models, for triggering the immune system to induce specific and protective immunity against bacterial, viral and parasitic infections. The administration of anti-Id antibodies as surrogate tumor-associated antigens (TAA) also represents another potential application of the concept of the Id network. Limited experience in human trials using anti-Id to stimulate immunity against tumors has shown promising results. In this "counter-point" article, we discuss our own findings showing the potential of an

Programming regulated cell death to engineer whole-tumor-cell vaccines for cancer immunotherapy.

Regulated cell death (RCD) encompasses a variety of genetically controlled cell death programs that are essential for maintaining tissue homeostasis and have emerged as promising therapeutic targets in oncology. When used to prepare whole-tumor-cell (WTC) vaccines, dying cancer cells provide broad tumor-antigen repertoires together with damage-associated molecular patterns (DAMPs) that can function as endogenous adjuvants. This review examines how distinct RCD pathways shape antitumor immunity and how they can be exploited to engineer next-generation WTC vaccines. We summarize the molecular an

Engineering age-adaptive mRNA lipid nanoparticle cancer vaccines via reprogramming systemic gene expression.

Lipid nanoparticle (LNP)-based mRNA vaccines have transformed cancer immunotherapy, yet their efficacy in older individuals, who represent the majority of cancer patients, remains poorly understood. Here, we uncover a critical and previously underappreciated barrier to mRNA LNP cancer vaccine performance in aged hosts: impaired systemic transgene expression. Using the SM-102 mRNA LNPs as a benchmark formulation, we show that while local immune activation and antigen presentation at the injection site and draining lymph nodes remain largely intact with age, transgene expression in peripheral or

Tn Hitchhiker Vaccines for High-Affinity Antibody Production and Targeted Cancer Cell Binding.

The development of effective cancer vaccines relies on the ability to generate a strong and specific humoral immune response against tumor-associated antigens. The Tn antigen, a truncated O-glycan overexpressed by various carcinomas, is a potent immunotherapeutic target. In this study, we evaluate the antibody specificity elicited in mice by the Tn-DSPE-PEG-2000 amphiphilic vaccine, along with initial insights into its cellular processing using a lysosomal assay. The vaccine is found to undergo lysosomal escape, thereby facilitating cytoplasmic antigen delivery, which is a crucial step for effe

Mandatory preventive vaccines for infectious diseases expressing T-cell epitopes for cancer prevention.

We have previously reported that microorganism-associated antigens (MAAs) share high sequence and conformational homology with tumor-associated antigens (TAAs) as well as T cells cross-react with homologous MAA/TAA pairs. More recently, we have also shown that the SARS-CoV-2 preventive vaccine, besides the humoral response, is able to elicit also a T cell response which cross-react with homologous TAAs. In the present study we analyzed the mandatory pediatric vaccines, namely the hexavalent vaccine (Diphtheria, Tetanus, Pertussis (whooping cough), Polio, Haemophilus influenzae type b (Hib), He

Engineered Carrier-Free Nanosystem-Induced In Situ Therapeutic Vaccines for Potent Cancer Immunotherapy.

In situ vaccines that can stimulate tumor immune response have emerged as a breakthrough in antitumor therapy. However, the immunosuppressed tumor microenvironment and insufficient infiltration of immune cells lead to ineffective antitumor immunity. Hence, a biomimetic carrier-free nanosystem (BCC) to induce synergistic phototherapy/chemotherapy-driven in situ vaccines was designed. A carrier-free nanosystem was developed using phototherapeutic reagents CyI and celastrol as raw materials. In vitro and in vivo studies have shown that under NIR light irradiation, BCC-mediated photo/chemotherapy
