Structural and functional aspects of porcine endometrial capillaries on days 13 and 15 after oestrus or mating.

The permeability of subepithelial capillaries in porcine endometrium was studied during midcycle and early pregnancy. Gilts were slaughtered on Day 13 or Day 15 of the oestrous cycle or pregnancy, 15 min after injection through the ear vein of 11.1 MBq of 125I-labelled human albumin in phosphate-buffered saline. The radioactivity of endometrial strips taken along the mesometrial and antimesometrial aspects of the uterine horn varied on average from 270 to 701 c.p.m./g and no difference (P greater than 0.05) was found between reproductive status, days of slaughter or sampling sites. The majorit

Vascular permeability in the peripheral autonomic and somatic nervous systems: controversial aspects and comparisons with the blood-brain barrier.

Endothelium, choroidal epithelium, and arachnoid exclude plasma proteins from most parts of the mammalian central nervous system (CNS). Nerve roots, in contrast, have permeable capillaries and permeable pia-arachnoid sheaths. Diffusion of plasma proteins into the cerebrospinal fluid is probably prevented by slow bulk flow along a pressure gradient from the subarachnoid space into the veins of the roots. In nerves, the perineurium prevents diffusion of proteins from the epineurium into the endoneurium. Capillaries within fascicles are permeable to macromolecules, though less so than the microve

Vascular damage in acute experimental leptospirosis of the guinea-pig.

The pathogenesis of the haemorrhagic diathesis in experimental leptospirosis of the guinea-pig was investigated in the lung, diaphragm and kidney. The vascular damage was found to be focal and mainly capillary. Swollen endothelium with dilated endoplasmic reticulum, enlarged mitochondriae and open junctions seemed to be the initial lesions and endothelial necrosis the final picture in all tissues. The lung capillaries showed endothelial and epithelial blebs and desquamation with many myelin figures. Capillary thrombosis was observed in the pulmonary microcirculation, probably acting as an aggr

Dextran fractional clearance studies in acute dengue infection.

BACKGROUND: Although increased capillary permeability is the major clinical feature associated with severe dengue infections the mechanisms underlying this phenomenon remain unclear. Dextran clearance methodology has been used to investigate the molecular sieving properties of the microvasculature in clinical situations associated with altered permeability, including during pregnancy and in various renal disorders. In order to better understand the characteristics of the vascular leak associated with dengue we undertook formal dextran clearance studies in Vietnamese dengue patients and healthy

Endothelial permeability in uremia.

The risk of cardiovascular disease is significantly higher in patients with long-term uremia than in otherwise healthy adults. In addition, the risk is greater than what would be predicted from classic vascular risk factors. This is true even before patients proceed to dialysis. Although the reasons for this accelerated vascular disease are unknown, transendothelial transport of lipids and other macromolecules in both large vessels and the microcirculation has been implicated in the generation of cardiovascular disease. Furthermore, endothelial cell dysfunction is widely accepted as a cardinal

[Edematous syndromes caused by capillary hyperpermeability. Diffuse angioedema].

Edema due to increased capillary permeability (ICP) may be diffuse or localized. Local edemas (Quincke edema, angioneurotic edema) are most often allergic or very rarely due to a defect in C1-inhibitor. Generalized edemas due to ICP share the following clinical features: Fluid retention (subcutaneous edema and diffused swelling) is predominant in lower limbs; it is worsened by orthostatism and warmth and alleviated by decubitus and cold, with important weight variations between morning and evening. It is associated with enhanced thirst, hypotension, oliguria, headaches and blood volume reducti

An analysis of the effect of synovial capillary distribution upon trans-synovial concentration profiles and exchange.

Recent morphometric studies of synovium lining the rabbit knee showed that capillaries extend to greater than 200 microns below the synovial surface; that a proportion of the superficial capillaries are fenestrated; and that the numerical density of the capillaries varies with depth ( Knight & Levick , 1983, 1984). Such observations raised the question 'Down to what depth do capillaries contribute significantly to the diffusional flux of a small extracellular solute (e.g. sulphate) into the joint cavity?' This problem was addressed by a mathematical model of trans-synovial diffusion. The peria

Effect of octanoate on blood-brain barrier permeability using L-dopa as a marker.

Short-chain fatty acid injection into animals produces coma, seizures, and hyperventilation. One mechanism of coma production may be through alterations in membrane permeability characteristics. The SPG histofluorescence technique was used to evaluate changes in blood-brain barrier permeability to injected L-dopa in rats after intraperitoneal injections of the short-chain fatty acid, sodium octanoate. Diffusion of intravascular fluorescence was observed around brain capillaries in the octanoate-injected rats suggesting an alteration in capillary permeability to L-dopa. Diffusion of fluorescenc

Assessment of pulmonary vascular injury.

The assessment of pulmonary injury in experimental and clinical situations remains a challenge. A method of quantifying pulmonary vascular aberrations in animal models and human clinical situations is described. A multiple-indicator dilution technique is employed to assess changes in lung capillary permeability--surface area product, extravascular lung water, and cardiac output. A mixture of 51Cr-RBC, 125I-albumin, tritiated water, and [14C]urea is injected into a central vein. Samples of the subject's blood are collected at timed intervals and counted for beta and gamma activity. Time concent

Edema and lymphatic clearance: molecular mechanisms and ongoing challenges.

Resolution of edema remains a significant clinical challenge. Conditions such as traumatic shock, sepsis, or diabetes often involve microvascular hyperpermeability, which leads to tissue and organ dysfunction. Lymphatic insufficiency due to genetic causes, surgical removal of lymph nodes, or infections, leads to varying degrees of tissue swelling that impair mobility and immune defenses. Treatment options are limited to management of edema as there are no specific therapeutics that have demonstrated significant success for ameliorating microvascular leakage or impaired lymphatic function. This

Connexins in endothelial barrier function - novel therapeutic targets countering vascular hyperpermeability.

Prolonged vascular hyperpermeability is a common feature of many diseases. Vascular hyperpermeability is typically associated with changes in the expression patterns of adherens and tight junction proteins. Here, we focus on the less-appreciated contribution of gap junction proteins (connexins) to basal vascular permeability and endothelial dysfunction. First, we assess the association of connexins with endothelial barrier integrity by introducing tools used in connexin biology and relating the findings to customary readouts in vascular biology. Second, we explore potential mechanistic ties be

Interleukin-2 toxicity.

Because of the ability of interleukin-2 (IL-2) to support the proliferation and activation of numerous types of immunocompetent cells and to support the survival of adoptively transferred lymphocytes, there has been considerable interest in its use in the immunotherapy of malignancies. While studies to date have indicated that IL-2 has activity against some malignancies, considerable toxicity has also been observed. Careful prescreening and selection of patients and appropriate management of toxicity can minimize adverse outcomes. Studies of IL-2 effects have provided intriguing evidence of in

Do circulating factors play a role in the pathogenesis of minimal change nephrotic syndrome?

This review examines the studies which have been undertaken to test the hypothesis that minimal change nephrotic syndrome of childhood (MCNS) is a primary immune disorder and that there is altered T-cell function which results in release of a circulating factor. This factor alters glomerular permeability, perhaps by modifying charge sites in the glomerular capillary bed, and results in selective proteinuria. The abnormalities in immune function observed in MCNS are summarized, as are the studies of circulating factors which have been identified. Although some agents have been shown to alter ca

A novel assay provides sensitive measurement of physiologically relevant changes in albumin permeability in isolated human and rodent glomeruli.

Increased urinary albumin excretion is a key feature of glomerular disease but has limitations as a measure of glomerular permeability. Here we describe a novel assay to measure the apparent albumin permeability of single capillaries in glomeruli, isolated from perfused kidneys cleared of red blood cells. The rate of decline of the albumin concentration within the capillary lumen was quantified using confocal microscopy and used to calculate apparent permeability. The assay was extensively validated and provided robust, reproducible estimates of glomerular albumin permeability. These values we

Therapy for microcirculatory disorders in severe acute pancreatitis: effectiveness of platelet-activating factor receptor blockade vs. endothelin receptor blockade.

Many of the complications of severe acute pancreatitis are the result of the amplifying effects of microcirculatory disruption. The factors causing microcirculatory disorders in acute pancreatitis involve vasoactive mediators such as platelet-activating factor (PAF) and endothelin-1 (ET) activated during the inflammatory response to pancreatic injury. To further evaluate the potential therapeutic role of specific receptor antagonists (RA) to these mediators, the present study compares the effect of PAF and ET receptor blockade on microcirculation and organ function in a well-established rodent

Trained hypertensive rats exhibit decreased transcellular vesicle trafficking, increased tight junctions' density, restored blood-brain barrier permeability and normalized autonomic control of the circulation.

Introduction: Chronic hypertension is accompanied by either blood-brain barrier (BBB) leakage and autonomic dysfunction. There is no consensus on the mechanism determining increased BBB permeability within autonomic areas. While some reports suggested tight junction's breakdown, others indicated the involvement of transcytosis rather than paracellular transport changes. Interestingly, exercise training was able to restore both BBB permeability and autonomic control of the circulation. We sought now to clarify the mechanism(s) governing hypertension- and exercise-induced BBB permeability. Metho

Concurrent measurements of blood flow and transcapillary transport in avian sarcoma virus-induced experimental brain tumors: implications for chemotherapy.

A blood-to-tissue transfer constant, K, and tissue blood flow, F, were measured concurrently in seven rats with a total of 19 separate brain tumors induced by intracerebral inoculation of avian sarcoma virus. Regional and local measurements of K and F were obtained using double-label quantitative autoradiography with alpha-[14C]aminoisobutyric acid and [131I]iodoantipyrine, computerized microdensitometry and image analysis. Apparent tissue extraction fractions and capillary permeability-surface area products were calculated for different tumor regions, brain adjacent to tumor and tumor-free br
