The ahpC c-54t compensatory mutation is not always a valid surrogate for isoniazid resistance in Mycobacterium tuberculosis.

Thirteen commercial genotypic antimicrobial susceptibility assays interrogate mutations upstream of ahpC to infer isoniazid resistance for Mycobacterium tuberculosis. We demonstrate that relying on one of these compensatory mutations (i.e., ahpC c-54t)-rather than causative resistance mutations in katG that ahpC compensates for-can result in systematic false-resistant results for isoniazid with the Cepheid Xpert MTB/XDR and suboptimal treatment. The WHO mutation catalog should be refined to address this scenario.

The practical labor lawyer: an employers' guide to strike contingency planning.

An employer's capacity successfully to survive a strike will depend largely on the extent to which it has anticipated the strike and taken the steps necessary to defend against it. It is the purpose of this article to catalog the factors employers should consider in approaching an imminent strike and devising contingency plans. It attempts to focus on the types of problems, legal and practical, that frequently arise in strike situations, and suggests certain measures that employers may adopt in preparing for, and defending against, the economic impact of a strike.

Oxidative stress in diabetic retinopathy.

Diabetic retinopathy (DR) remains a leading cause of blindness, with oxidative stress as a central pathogenic driver. However, the translation of this mechanistic knowledge into clinical diagnostics and targeted therapies has been hampered by inconsistent biomarkers and a simplistic view of redox balance. This critical review moves beyond cataloging oxidative markers to evaluate their biological specificity, analytical robustness, and compartmentalization between systemic circulation and the ocular microenvironment. We dissect the technical challenges of direct reactive species measurement and

Astrobiology in a mesophilic tropical country: A database and a review of analog environments in Brazil.

Astrobiological analog environments comprise Earth-based environments that share similarities with a target setting (another planetary body or Precambrian Earth conditions) and whose defining features influence extant or fossil biological components, enabling astrobiological interpretation. Although not synonymous with extreme environments, they often impose selective pressures that favor extremophiles and extremotolerant organisms, making them suitable for exploring habitability, the limits of life, and its evolutionary history. Global cataloging initiatives have identified such sites worldwi

From functional annotation to functional meaning in microbiome research.

Microbiome research has moved from cataloging community composition to asking what these communities do, but "function" is often used to describe fundamentally different levels of evidence. Functional claims may refer to functional capacity (what is encoded), functional realization (what molecular functions are actively engaged under specific conditions), or functional impact (the resulting consequences for hosts, microbial communities, or ecosystems). In this Perspective, we examine how functional annotations in microbiome research generate biological meaning and why current approaches suppor

Non-Resolving Repair in Idiopathic Pulmonary Fibrosis: From Failed Cellular Transitions to Architectural Lock-In.

Repair programs are pervasive in idiopathic pulmonary fibrosis (IPF), yet they fail to reach completion. While single-cell and spatial omics have identified cellular states associated with injury-repair programs, a central paradox remains: why do these programs persist without reconstituting functional lung architecture? This review integrates evidence from multiscale omics, spatial analyses, and translational studies to propose a lesion-centered "non-resolving repair" framework that explains IPF progression. We argue that disease progression is driven by the persistence of cellular repair pro

Exploring the global mosaic of medicinal plant databases: unveiling nature's pharmacopoeia.

Medicinal plants have long served as an important asset in the treatment of diseases. Recent developments in computer science have enabled the rise of specialized databases cataloging medicinal plant knowledge. However, a systematic comparison of available region-specific medicinal plant databases is lacking. This review summarizes globally available medicinal plant databases that focus on specific geographical regions, aiming to inspire and guide people from specialists to the general public toward fostering innovation and making informed decisions. Through a systematic search of literature a

The spatial and multi-omic landscape of Down syndrome leukemogenesis: moving beyond cellular heterogeneity.

Down syndrome (DS) confers a developmentally rooted predisposition to both myeloid and lymphoid leukemias, particularly myeloid leukemia associated with DS (ML-DS) and acute lymphoblastic leukemia associated with DS (ALL-DS). While trisomy 21-driven gene dosage imbalance is central to this risk, DS leukemogenesis cannot be fully explained by recurrent mutations alone; it reflects a dynamic interplay between altered hematopoietic development, cell-intrinsic programs, and tissue microenvironmental cues. In this perspective, we argue that the field should move beyond cataloging cellular heterogen

MicroRNAs in Colorectal Cancer Immunotherapy: Biomarkers, Resistance Mechanisms, and Strategies to Convert "Cold" Tumors to "Hot".

Colorectal cancer (CRC) remains a major cause of cancer morbidity and mortality. Immune checkpoint inhibitors (ICIs) have transformed the treatment of microsatellite instability-high/mismatch repair-deficient (MSI-H/dMMR) CRC, yet most CRCs are microsatellite-stable/mismatch repair-proficient (MSS/pMMR) and remain poorly responsive to immunotherapy. MicroRNAs (miRNAs) are well positioned to influence this biology because individual miRNAs can coordinate multiple tumor-intrinsic and microenvironmental programs that shape antitumor immunity. Rather than cataloging miRNAs one by one, this review

Disparities in women's health and clinical considerations from a translational science perspective: A narrative review and framework for future directions.

BACKGROUND: Sex and gender disparities persist across biomedical research, clinical care, and health policy, despite increasing recognition of sex-specific disease mechanisms and treatment responses. Underrepresentation of women in clinical trials continues to produce male-centric treatment protocols and insufficient data to support precision care. METHODS: This narrative review applies a translational science framework to examine recurring and cross-cutting gaps in sex and gender health science. Rather than cataloging all disease areas, the review uses three exemplar domains-mental and neuro

Metabolomics and proteomics: synergistic tools for understanding snake venom inhibition.

Snake envenomation presents a significant global health challenge, especially in rural areas of tropical and subtropical regions. Traditional antivenom therapies face limitations related to efficacy, availability, and specificity, prompting a need for novel approaches. Recent advancements in omics technologies, particularly metabolomics and proteomics, have enhanced our understanding of snake venom composition, toxicity, and potential therapeutic strategies. Metabolomics allows for the study of metabolic changes induced by venom, providing insights into disrupted pathways and possible inhibito

The application of ceragenins to orthopedic surgery and medicine.

Osteomyelitis and infections associated with orthopedic implants represent a significant burden of disease worldwide. Ceragenins (CSAs) are a relatively new class of small-molecule antimicrobials that target a broad range of Gram-positive and Gram-negative bacteria as well as fungi, viruses, and parasites. This review sets the context of the need for new antimicrobial strategies by cataloging the common pathogens associated with orthopedic infection and highlighting the increasing challenges of managing antibiotic-resistant bacterial strains. It then comparatively describes the antimicrobial p

Expanded skin virome in DOCK8-deficient patients.

Human microbiome studies have revealed the intricate interplay of host immunity and bacterial communities to achieve homeostatic balance. Healthy skin microbial communities are dominated by bacteria with low viral representation1-3, mainly bacteriophage. Specific eukaryotic viruses have been implicated in both common and rare skin diseases, but cataloging skin viral communities has been limited. Alterations in host immunity provide an opportunity to expand our understanding of microbial-host interactions. Primary immunodeficient patients manifest with various viral, bacterial, fungal, and para

New challenges for public health care: biological and chemical weapons awareness, surveillance, and response.

Recent events in the United States have demonstrated a critical need for recognizing nurses and emergency health care providers as important elements of the nation's first line of defense and response against terrorist attacks involving biological, chemical, or radiological weapons. The anthrax letter attacks of September/October 2001 demonstrate the importance of vigilance and attention to detail while interviewing and attending patients and when entering, reviewing, and cataloging patient records. Nursing professionals, emergency care responders, and physicians can perform a crucial role in

Inventory of the proteins in Neisseria meningitidis serogroup B strain MC58.

A protein inventory of Neisseria meningitidis strain MC58, a meningococcal strain belonging to the serogroup B, was performed by proteomics. A differential extraction procedure was employed and 238 protein species were identified by 2D mini-maps and MALDI-ToF analyses. In this catalog, we detected protein products from 33 genes, which were not yet annotated in previous N. meningitidis proteomic studies. This approach is suitable for high-throughput studies on differential expression of N. meningitidis genomes.

Genomic diversity of human-associated Mycobacterium bovis in Canada.

Human infections caused by Mycobacterium bovis in Canada are not well documented. The following study analyzed M. bovis (excluding BCG) isolates identified in public health laboratories across Canadian provinces between 2000 and 2025. Whole-genome sequencing was used to catalog mutations linked to antimicrobial resistance and evaluate the genomic relatedness of isolates. The majority of samples in this study were non-respiratory (60%). Of 74 study isolates, 69 (93%) were phenotypically sensitive to first-line drugs isoniazid, rifampin, and ethambutol. The most frequently encountered M. bovis c

Genomic and biosynthetic landscape of high-temperature Daqu microbiome.

As the core starter for Chinese Baijiu, high-temperature Daqu is produced through open solid-state fermentation with recurrent inoculation by mature Daqu, forming a rich yet largely untapped reservoir of genomes and bioactive compounds. This study constructs the High-temperature Daqu Fermentation Microbiome catalog using 463 metagenomes spanning the full fermentation cycle. The catalog comprises 4,264 metagenome-assembled genomes that are dereplicated into 252 representative genome-based species, 82 % of which are absent from current global food microbiome databases. It further contains 14.3 m

Antibiotic resistance gene reservoir in live poultry markets.

OBJECTIVES: The heavy use of antibiotics in farm animals contributes to the enrichment and spread of antibiotic resistance genes (ARGs) in "one-health" settings. Numerous ARGs have been identified in livestock-associated environments but not in Chinese live poultry markets (LPMs). METHODS: We collected 753 poultry fecal samples from LPMs of 18 provinces and municipalities in China and sequenced the metagenomes of 130 samples. Bioinformatic tools were used to construct the gene catalog and analyze the ARG content. PCR amplification and Sanger sequencing were used to survey the distribution of

Identifying a polymorphic 'switch' that influences miRNAs' regulation of a myasthenia gravis risk pathway.

The significant roles of genetic variants in myasthenia gravis (MG) pathogenesis have been demonstrated in many studies, and recently it has been revealed that aberrant level/regulation of microRNAs (miRNAs) might contribute to the initiation and progression of MG. However, the dysfunction of miRNA associated with single nucleotide polymorphisms (miRSNPs) has not been well investigated in MG. In this study, we created a contemporary catalog of 89 MG risk genes via manual literature-mining. Based on this risk gene catalog, we obtained 18 MG risk pathways. Furthermore, we identified 93 miRNAs th
