Role of Isotocin in the Regulation of the Hypophysiotropic Dopamine Neurones in the Preoptic Area of the Catfish, Clarias batrachus.

Dopamine (DA) has emerged as a potent inhibitory neuromodulator of luteinsing hormone (LH) secretion and reproduction in teleosts. The DA neurones located in the anterior subdivision of nucleus preopticus periventricularis (NPPa) in the preoptic area (POA) innervate the pituitary gland and regulate LH cells. Although a reduction in the inhibitory DAergic tone is crucial for stimulatory action of gonadotrophin-releasing hormone (GnRH) on LH cells, the role of other hypothalamic factors is suggested but not fully understood. Nonapeptide, isotocin (IST) has emerged as a likely candidate that may

Diploid and triploid African catfish (Clarias gariepinus) differ in biomarker responses to the pesticide chlorpyrifos.

The impacts of environmental stressors on polyploid organisms are largely unknown. This study investigated changes in morphometric, molecular, and biochemical parameters in full-sibling diploid and triploid African catfish (Clarias gariepinus) in response to chlorpyrifos (CPF) exposures. Juvenile fish were exposed to three concentrations of CPF (mean measured μg/L (SD): 9.71 (2.27), 15.7 (3.69), 31.21 (5.04)) under a static-renewal condition for 21days. Diploid control groups had higher hepatosomatic index (HSI), plasma testosterone (T), and brain GnRH and cyp19a2 expression levels than triplo

Zebrafish (Danio rerio) bioassay for visceral toxicosis of catfish and botulinum neurotoxin serotype E.

Visceral toxicosis of catfish (VTC), a sporadic disease of cultured channel catfish (Ictalurus punctatus) often with high mortality, is caused by botulinum neurotoxin serotype E (BoNT/E). Presumptive diagnosis of VTC is based on characteristic clinical signs and lesions, and the production of these signs and mortality after sera from affected fish is administered to sentinel catfish. The diagnosis is confirmed if the toxicity is neutralized with BoNT/E antitoxin. Because small catfish are often unavailable, the utility of adult zebrafish (Danio rerio) was evaluated in BoNT/E and VTC bioassays.

Occurrence of Ichthyophthirius multifiliis within the peritoneal cavities of infected channel catfish Ictalurus punctatus.

Ichthyophthirius multifiliis is a ciliated protozoan parasite that infects the skin and gills of freshwater fish. This report describes the unusual finding of I. multifiliis within the peritoneal cavities of experimentally infected channel catfish Ictalurus punctatus. Twenty catfish fingerlings were exposed to I. multifiliis theronts using a standardized protocol. Five infected fish and 2 control fish were killed at various time points after infection and their tissues examined. Formalin-fixed, paraffin embedded sections were processed for light microscopy and immunohistochemical detection of

No detectable DNA excision repair in UV-exposed hepatocytes from two catfish species.

DNA repair is a critical process in protecting cellular genetic information from mutation. Nucleotide excision repair (NER) is a mechanism by which cells correct DNA damage caused by agents that form bulky covalent adducts and UV photoproducts such as thymine dimers and 6-4 photoproduct. NER, sometimes called dark repair, is generally accepted as being low in fish compared to mammals. This study was designed to quantitate NER in two related catfish species that have known differential sensitivities to liver carcinomas. The original hypothesis was that the more cancer resistant species, channel

Citrate enhances olfactory receptor responses and triggers oscillatory receptor activity in the channel catfish.

Citrate, a normal constituent of cellular metabolism, in a binary mixture with an amino acid enhanced asynchronous olfactory receptor responses in the channel catfish, Ictalurus punctatus. In addition, high concentrations of either citrate (> or =3 mM) alone or an amino acid (> or =0.1 mM) in a binary mixture with citrate (> or =1 mM) triggered synchronized voltage oscillations of olfactory receptor neurons (ORNs) known as "peripheral waves" (PWs). Binary mixtures containing lower concentrations of an amino acid also triggered PW activity if the concentration of citrate in the mixture was incr

GABA transporters and GABAC-like receptors on catfish cone- but not rod-driven horizontal cells.

The effects of GABA and related agents were studied in solitary rod- and cone-driven horizontal cells, acutely isolated from the catfish retina using enzymatic and mechanical treatment. Both types of horizontal cells, which normally receive glutamatergic input from photoreceptors, responded to pressure ejection of the glutamate analog kainate (50 microM) with an inward current of 300-800 pA when voltage-clamped at -70 mV using the whole-cell patch-clamp technique. But pressure ejection of GABA (500 microM) elicited an inward current only in cone-driven horizontal cells. This current, ranging b

Voltage-dependent and odorant-regulated currents in isolated olfactory receptor neurons of the channel catfish.

The electrical properties of olfactory receptor neurons, enzymatically dissociated from the channel catfish (Ictalurus punctatus), were studied using the whole-cell patch-clamp technique. Six voltage-dependent ionic currents were isolated. Transient inward currents (0.1-1.7 nA) were observed in response to depolarizing voltage steps from a holding potential of -80 mV in all neurons examined. They activated between -70 and -50 mV and were blocked by addition of 1 microM tetrodotoxin (TTX) to the bath or by replacing Na+ in the bath with N-methyl-D-glucamine and were classified as Na+ currents.

IP3- and cAMP-induced responses in isolated olfactory receptor neurons from the channel catfish.

Olfactory receptor neurons enzymatically dissociated from channel catfish olfactory epithelium were depolarized transiently following dialysis of IP3 or cAMP (added to the patch pipette) into the cytoplasm. Voltage and current responses to IP3 were blocked by ruthenium red, a blocker of an IP3-gated Ca(2+)-release channel in sarcoplasmic reticulum. In contrast, the responses to cAMP were not blocked by extracellularly applied ruthenium red, nor by L-cis-diltiazem or amiloride and two of its derivatives. The current elicited by cytoplasmic IP3 in neurons under voltage clamp displayed a voltage

Responses of single facial taste fibers in the sea catfish, Arius felis, to amino acids.

1. Taste buds in catfish are found not only within the oropharyngeal cavity, as in mammals, but are also located along the external body surface of the animal from the barbels and lips to the caudal fin. Because these taste buds are innervated by the facial (cranial VII) nerve, the extraoral taste system of catfish is analogous to the mammalian taste system of the anterior two-thirds of the tongue, which contains taste buds innervated by the chorda tympani nerve, and of the soft palate and nasoincisor ducts, which contain taste buds innervated by the greater superficial petrosal nerve. 2. The

Ultrastructural organization of seminal vesicles in the African catfish (Clarias gariepinus) during the spawning season.

The seminal vesicle is an important accessory reproductive organ in male teleosts, but detailed ultrastructural information on its organization in Clarias gariepinus remains limited. This study examined the seminal vesicle of sexually mature male African catfish during the spawning season using transmission electron microscopy (TEM). Forty wild-caught males from the Lake Chad region of Maiduguri, Nigeria were examined. Seminal vesicle tissues were fixed in phosphate buffered glutaraldehyde, post-fixed in osmium tetroxide, embedded in Epon 812, sectioned, and examined using TEM. The seminal ves

Evidence of genome-wide relaxed selection on mildly deleterious mutations in an ancient subterranean catfish.

About one hundred subterranean catfish species have been described, resulting from repeated colonization of cave environments by multiple surface lineages. Most cave-dwelling species are found in the Americas, in particular in South America, but a few species also live in Central and North America. Despite the availability of high-quality genome assemblies for two cave species, the Mexican blind catfish Prietella phreatophila and the Colombian blind catfish Trichomycterus rosablanca, genomic approaches to investigate genetic changes associated with subterranean life or to estimate cave coloniz

Adaptive Diversification of the Mstnb Locus Drives Genomic Architecture of Growth in Thai Clariid Catfish.

The production of clariid catfish in Thai aquaculture depends on culture of the native species (Clarias macrocephalus and C. batrachus) and introduced African catfish (C. gariepinus). Particularly, hybridization with C. gariepinus has improved productivity but raised significant conservation concerns. These lineages show distinct growth variations; however, the underlying genetic factors remain uncharacterized. We performed a comprehensive genomic assessment of the myostatin-b (Mstnb) gene in 535 individuals comprising 19 Thai populations. The study design involved a novel integration of high-

Drivers of the range expansion of the European catfish (Silurus glanis) within its native distribution.

The European catfish Silurus glanis L. continues to spread as an invasive species in Europe. Meanwhile, increasing abundance and range expansions are also suspected within its native distribution. The objective of this study was to characterize the feeding, growth and abundance of S. glanis in 12 lakes and 12 rivers within its European native range in relation to environmental changes that may explain population trends over time. The results suggest a shift in factors limiting the carrying capacity of catfish habitats within this range, leading to increases in growth and abundance. Statistical

Transcriptomic Divergence and Associated Markers Between Genomic Lineages of Silver Catfish (Rhamdia quelen).

Rhamdia quelen is a catfish widely distributed throughout South America, characterized by a complex taxonomic history. This species is a valuable resource for both fisheries and aquaculture. Due to its cultural and economic importance, it has been prioritized for conservation in the Neotropical region. Population genomics studies supported two main lineages latitudinally distributed (North and South) in the Neotropical basins Río de la Plata and Laguna Merín based on current genetic isolation and signals of local adaptation. In this study, we characterized the R. quelen transcriptome in brain,

Remediation of hypoxia induced brain alteration with pyrimidine treatment in the freshwater catfish, Heteropneustes fossilis.

Low dissolved oxygen (DO), or hypoxia, has a profound impact on the physiological and neurological health of aquatic organisms, particularly oxygen-sensitive organs such as the brain. The aim was to investigate the effects of pyrimidine and its derivative, arylvinylpyrimidine (AVP), on neuronal health in the brain of the lesser studied Heteropneustes fossilis under hypoxic conditions. Laboratory-acclimatized fish were exposed to critical threshold hypoxia (2 mg/L DO for 8 h) based on behavioural and brain histopathology observations. For remediation by pyrimidine and AVP in the hypoxic brain,
