Reassembly of the anchoring structures of the corneal epithelium during wound repair in the rabbit.

Reappearance of the structures involved in adhesion of the corneal epithelium to the stroma was studied in healing 7 mm keratectomy wounds in rabbit corneas. Corneas were taken at 48 and 66 hr, 1, 2, 3, 4, 6 and 8 weeks, and 4, 6 and 12 months post-wounding. Immunolocalization of bullous pemphigoid antigen (BPA), laminin and type VII collagen was used to determine time and sequence of appearance of hemidesmosomes, basement membrane and anchoring fibrils, respectively. Electron micrographs from three regions in the wound were used to correlate the immunohistochemical data and to quantitate the

The 'ins' and 'outs' of podosomes and invadopodia: characteristics, formation and function.

Podosomes and invadopodia are actin-based dynamic protrusions of the plasma membrane of metazoan cells that represent sites of attachment to - and degradation of - the extracellular matrix. The key proteins in these structures include the actin regulators cortactin and neural Wiskott-Aldrich syndrome protein (N-WASP), the adaptor proteins Tyr kinase substrate with four SH3 domains (TKS4) and Tyr kinase substrate with five SH3 domains (TKS5), and the metalloprotease membrane type 1 matrix metalloprotease (MT1MMP; also known as MMP14). Many cell types can produce these structures, including inva

New morphological observations on the initial events of Toxoplasma gondii entry into host cells.

Toxoplasma gondii is an obligate intracellular protozoan of worldwide distribution. It is effective in the infection of various homoeothermic animals of economic importance. The process of T. gondii invasion of host cells occurs in less than 20 s by the active mechanism of penetration. First, a mobile junction is formed due to the association between the apical end of the parasite and the host cell surface. Then, the secretion of invasive and docking proteins allows the formation of the mobile junction before the complete internalization of the parasite. Here, using high-resolution microscopy,

Nanoscale Curvature-Facilitated Membrane Intercalation of Conjugated Oligoelectrolytes Revealed by Nanobar-Supported Lipid Bilayers.

Conjugated oligoelectrolytes (COEs) constitute a powerful toolbox for detecting and modulating cell membrane properties. The versatility in their molecular structural design enables fine-tuning of their membrane intercalating behaviors, ranging from membrane disruption for antimicrobial applications to membrane stabilization for cell labeling and biosensing. However, a detailed description of the intercalation mechanism is absent, despite efforts to understand the impact of charge density and hydrophobic core length on the membrane intercalation efficiency of COEs. A critical yet overlooked fa

Membrane permeabilization by Islet Amyloid Polypeptide.

Membrane permeabilization by Islet Amyloid Polypeptide (IAPP) is suggested to be the main mechanism for IAPP-induced cytotoxicity and death of insulin-producing beta-cells in type 2 diabetes mellitus (T2DM). The insoluble fibrillar IAPP deposits (amyloid) present in the pancreas of most T2DM patients are not the primary suspects responsible for permeabilization of beta-cell membranes. Instead, soluble IAPP oligomers are thought to be cytotoxic by forming membrane channels or by inducing bilayer disorder. In addition, the elongation of IAPP fibrils at the membrane, but not the fibrils themselve

mDia1 Assembles a Linear F-Actin Coat at Membrane Invaginations To Drive Listeria monocytogenes Cell-to-Cell Spreading.

Direct cell-to-cell spreading of Listeria monocytogenes requires the bacteria to induce actin-based finger-like membrane protrusions in donor host cells that are endocytosed through caveolin-rich membrane invaginations by adjacent receiving cells. An actin shell surrounds these endocytic sites; however, its structure, composition, and functional significance remain elusive. Here, we show that the formin mDia1, but surprisingly not the Arp2/3 complex, is enriched at the membrane invaginations generated by L. monocytogenes during HeLa and Jeg-3 cell infections. Electron microscopy reveals a band

The cell envelope of the Legionnaires' disease bacterium. Morphologic and biochemical characteristics.

We studied the cell-envelope structure of the Legionnaires' disease (LD) bacterium by electron microscopy and biochemical assays. There were apparent differences in cell structure by electron microscopy using two different prefixation methods. Organisms prefixed with gluteraldehyde had a single surrounding membrane. The typical two-membrane structure of gram-negative bacteria, however, was observed after prefixation with a combination of gluteraldehyde, formalin, and creosol. The cell wall (peptidoglycan) was seen in electron micrographs of plasmolyzed bacteria and in papain-digested cells. Bo

Ultrastructure of odontogenic jaw cysts.

The epithelial ultrastructure of six radicular cysts, four follicular cysts, and five keratocysts was studied with special attention to the epithelium-connective tissue junction. Inflammation was found to cause widened interepithelial cell spaces which often harbored inflammatory cells in the radicular and follicular cysts. The characteristic structures at the epithelium-connective tissue junction (plasma membrane, lamina lucida and basal lamina) were not affected by inflammation. Fibrous structures were seen to connect the basal lamina to the underlining collagenous connective tissue. The ker

Recent Progress on Genetically Modified Animal Models for Membrane Skeletal Proteins: The 4.1 and MPP Families.

The protein 4.1 and membrane palmitoylated protein (MPP) families were originally found as components in the erythrocyte membrane skeletal protein complex, which helps maintain the stability of erythrocyte membranes by linking intramembranous proteins and meshwork structures composed of actin and spectrin under the membranes. Recently, it has been recognized that cells and tissues ubiquitously use this membrane skeletal system. Various intramembranous proteins, including adhesion molecules, ion channels, and receptors, have been shown to interact with the 4.1 and MPP families, regulating cellu

Structural basis for regulating lipopolysaccharide transmembrane transport.

Gram-negative bacteria are surrounded by a multilayered cell envelope with a mostly impermeable outer membrane that provides intrinsic resistance to many antibiotics1-5. The outer membrane is an asymmetrical bilayer with phospholipids in the inner leaflet and lipopolysaccharide (LPS) in the outer leaflet6,7. An LPS transport (Lpt) machine called LptB2FGCADE moves LPS across a protein bridge from the inner membrane to the outer membrane8,9. LPS biosynthesis is regulated to prevent toxic accumulation of LPS molecules in the inner membrane during growth10-12. Whether LPS transport across the Lpt

Amyloid peptide pores and the beta sheet conformation.

Over 20 clinical syndromes have been described as amyloid diseases. Pathologically, these illnesses are characterized by the deposition in various tissues of amorphous, Congo red stainingdeposits, referred to as amyloid. Under polarizing light microscopy, these deposits exhibit characteristic green birefringence. X-ray diffraction reveals cross-beta structure of extended amyloid fibrils. Although there is always a major protein in amyloid deposits, the predominant protein differs in each ofthe clinical syndromes. All the proteins exhibit the characteristic nonnative beta-sheet state. These pro

[Fuchs' endothelial dystrophy--the appearance of the corneal endothelium under the electron microscope (author's transl)].

Studying the various stages of Fuchs' endothelial dystrophy with transmission and surface electronmicroscopy makes the dystrophic and degenerative changes in the endothelial and Descemet's membrane visible: dystrophic changes by the formation of tuberosities, whose structure is very similar to that Descemet's membrane, and degenerative changes by reduction in the number of endothelial cells through destruction of the intercellular connections and through cell necrosis. The second stage is the reformation of the structure by fibroblastic cells. The dystrophic cells lead to the formation of a fi

Biomolecular Condensates, Crowding, and Membranes: The Role of Water Revealed with Advanced Fluorescence Microscopy and Phasor Analysis.

The intracellular milieu is densely packed with macromolecules, creating a confined space for water molecules. The influence of macromolecular crowding on water activity impacts cell physiology and homeostasis, as well as the behavior of biomolecules and their supramolecular structures. In this context, understanding how water dynamics can be regulated could have profound implications for the tuning of the emergent properties of membrane-bound and membraneless organelles at the mesoscale. In this chapter, we discuss how the application of hyperspectral imaging (HSI) and fluorescence lifetime mi

PM100117 and PM100118, new antitumor macrolides produced by a marine Streptomyces caniferus GUA-06-05-006A.

Two new bioactive polyhydroxyl macrolide lactones PM100117 (1) and PM100118 (2) were isolated from the culture broth of the marine-derived Streptomyces caniferus GUA-06-05-006A. Their structures were elucidated by a combination of spectroscopic methods, mainly one-dimensional and 2D NMR and HRESI-MS. They consist of 36-membered macrolides with a side chain containing three deoxy sugars and a 1,4-naphthoquinone chromophore. Compounds 1 and 2 displayed potent cytotoxicity against three human tumor cell lines with GI50 values in the micromolar range, as well as slight antifungal activity against

Sperm head membrane reorganisation during capacitation.

The sperm cell has a characteristic polarized morphology and its surface is also highly differentiated into different membrane domains. Junctional protein ring structures seal the surface of the mid-piece from the head and the tail respectively and probably prevent random diffusion of membrane molecules over the protein rings. Despite the absence of such lateral diffusion-preventing structures, the sperm head surface is also highly heterogeneous. Furthermore, lipid and membrane protein ordering is subjected to changes when sperm become capacitated. The forces that maintain the lateral polarity

Mechanism of membrane perforation in rotavirus cell entry.

Cell entry of nonenveloped animal viruses requires translocation of a macromolecular assembly across a cellular membrane. Double-stranded RNA viruses introduce into the target cell an inner capsid particle that does not uncoat further. Instead, it extrudes capped viral mRNA by virtue of polymerase and capping activities within it. As described here, we used cryogenic electron tomography to visualize the full course of rhesus rotavirus entry, from cell attachment and virion uptake to release of the subviral particle. The cryo-tomograms and subtomogram averaging of classified subparticles link h

Localization of host endocytic and actin-associated proteins during Shigella flexneri intracellular motility and intercellular spreading.

Shigella flexneri (S. flexneri), the causative agent of bacillary dysentery, uses an effector-mediated strategy to hijack host cells and cause disease. To propagate and spread within human tissues, S. flexneri bacteria commandeer the host actin cytoskeleton to generate slender actin-rich comet tails to move intracellularly, and later, plasma membrane actin-based protrusions to move directly between adjacent host cells. To facilitate intercellular bacterial spreading, large micron-sized endocytic-like membrane invaginations form at the periphery of neighboring host cells that come into contact

Stabilization and structural analysis of a membrane-associated hIAPP aggregation intermediate.

Membrane-assisted amyloid formation is implicated in human diseases, and many of the aggregating species accelerate amyloid formation and induce cell death. While structures of membrane-associated intermediates would provide tremendous insights into the pathology and aid in the design of compounds to potentially treat the diseases, it has not been feasible to overcome the challenges posed by the cell membrane. Here, we use NMR experimental constraints to solve the structure of a type-2 diabetes related human islet amyloid polypeptide intermediate stabilized in nanodiscs. ROSETTA and MD simulat

The fine structural localization of endogenous and exogenous peroxidase activity in human bone marrow mast cells under pathological conditions.

We have examined the ultrastructural characteristics of peroxidase activity in human bone marrow mast cells. These studies were performed in three patients with systemic mast cell disease, and in another six patients showing bone marrow mast cell hyperplasia. Endogenous peroxidase activity was localized in the perinuclear cisternae and strands of endoplasmic reticulum, but never in the granules. We have also demonstrated the "in vivo" existence of exogenous peroxidase activity in two of the three cases of systemic mast cell disease. The peroxidase internalization involved its binding to the pl

Detailed Protocol for Segmentation and Quantification of Overlapping Prospore Membranes using DeMemSeg.

Quantitative analysis of biological membrane morphology is essential for understanding fundamental cellular processes such as organelle biogenesis and remodeling. While manual annotation has been the standard for complex structures, it is laborious and subjective, and conventional automated methods often fail to accurately delineate overlapping objects in 2D projected microscopy images. This protocol provides a complete, step-by-step workflow for the quantitative analysis of overlapping prospore membranes (PSMs) in sporulating yeast. The procedure details the synchronous induction of sporulati
