IL-27 shapes NK cell heterogeneity and function in colorectal cancer.

BACKGROUND: Colorectal cancer (CRC) is a leading cause of cancer-related mortality worldwide and is characterized by an immunosuppressive tumor microenvironment (TME). While adaptive immunity contributes to tumor control, growing evidence underscores the role of innate lymphocytes, particularly natural killer (NK) cells, in early antitumor surveillance. However, tumor-infiltrating NK cells often exhibit defective maturation and impaired effector functions, whereas the signals regulating NK cell differentiation and activity in CRC remain poorly defined. Interleukin-27 (IL-27) has emerged as a r

A minimal mechanically consistent model of smoothly dividing disk-shaped cells.

Replication through cell division is one of the fundamental processes of life and a major driver of dynamics in systems ranging from bacterial colonies to embryogenesis, tissues and tumors. While regulation also shapes self-organization, many biologically relevant behaviors arise from a limited number of physical ingredients, and particle-based models have become a popular platform to investigate these emergent dynamics. However, incorporating division into such models often produces aberrant mechanical fluctuations that hinder meaningful analysis. Here, we introduce a minimal model ensuring m

Manufacturing as biology: How T-cell processing shapes therapeutic outcomes.

T-cell therapies have transformed the treatment landscape for hematologic malignancies and show growing promise for solid tumors and non-oncologic diseases. Unlike conventional drugs, these living therapeutics are shaped not only by genetic engineering but also by the ex vivo manufacturing process itself, which functions as a powerful biological determinant of cell fate and function. Here, we present a framework that conceptualizes manufacturing as a form of biological programming that governs therapeutic outcomes through its effects on T-cell phenotype, fitness, and functional durability. We

Cell division timing shapes the morphology and size of nascent multicellular organisms.

Upon making the transition from unicellularity to multicellularity, many previously optimized cellular traits experience the renewed scrutiny of natural selection due to their novel effects on emergent multicellular phenotypes. Yet we lack a comprehensive understanding of how and why specific cellular traits influence multicellular phenotypes and fitness. The snowflake yeast model system provides a tractable entry point for such investigations. The effects of several cell-level traits (cellular aspect ratio, cell volume, bud neck strength) on multicellular cluster size have been characterized,

Hypoxia shapes immune cell organization and bacterial control in tuberculosis granulomas.

Spatial profiling of tuberculosis granulomas from non-human primates reveals that the myeloid core is partitioned into two distinct metabolic zones, one of which is hypoxic. Hypoxia is associated with dysfunctional immune cell states and altered spatial organization, including lymphocyte exclusion, and may be a driver of impaired immunity in tuberculosis.

Spatial metabolic heterogeneity shapes CD8+ T cell function in cancer.

Tumors are spatially heterogeneous ecosystems in which malignant, stromal, vascular, and immune cells interact within metabolically distinct niches. These localized microenvironments are shaped by factors such as nutrient availability, hypoxia, acidosis, and immunomodulatory metabolites, all of which strongly influence CD8⁺ T cell infiltration, migration, persistence, and effector function. Growing evidence indicates that spatial metabolic heterogeneity contributes to immune exclusion, T cell dysfunction, and resistance to immunotherapy. This has been accompanied by major advances in spatial m

NK cell allorecognition shapes reprogramming of neutrophils infiltrating heart allografts.

BACKGROUND: Recent developments in neutrophil biology have demonstrated that neutrophils are phenotypically and functionally heterogeneous. Tissue microenvironments dictate changes in neutrophil cell states in infection and cancer, but little is known about how alloimmune responses or solid organ transplantation influence neutrophil heterogeneity and plasticity. METHODS: Here, we used the murine heterotopic heart transplant model in conjunction with high dimensional flow cytometry and transcriptome analysis to interrogate how the alloimmune response and microenvironment of a transplanted orga

Satellite glial cells: Shaping peripheral input into the brain-body axis?

Satellite glial cells (SGCs) are peripheral nervous system glial cells enveloping sensory and sympathetic ganglion neuronal soma. Traditionally viewed as mere supportive cells, recent studies reveal SGCs' dynamic role in regulating sensory and autonomic processing, positioning them to shape peripheral neural signaling. This role has the potential to impact the healthy function of numerous biological processes and contribute to disease progression. Studies now implicate peripheral sensory and autonomic deficits in the etiology of many disorders, including cognitive decline in aging, neurodevelo

Unmasking hidden shapes: unusual red cell morphologies in urine sediment.

Schistocytes are fragmented red blood cells (RBCs) classically restricted to peripheral blood smears, where they serve as a hallmark of microangiopathic haemolytic anemia (MAHA) and thrombotic microangiopathy (TMA). Their presence in urine has been rarely documented. We report a 30-year-old postpartum woman who developed severe anemia, thrombocytopenia, acute kidney injury, and biochemical evidence of intravascular haemolysis following massive transfusion after caesarean delivery. Urinalysis revealed haematuria and proteinuria, and manual phase-contrast microscopy demonstrated atypical RBC mor

How Morphology Shapes Survival in Invasive Squamous Cell Carcinoma of the Lung.

BACKGROUND AND OBJECTIVES: Squamous cell carcinoma (SQCC) represents a significant proportion of human malignancies affecting various anatomical sites, including the lung. Understanding the prognostic factors is crucial for establishing effective risk stratification in these patients, as multiple critical aspects significantly impact overall survival. MATERIALS AND METHODS: A retrospective study was conducted on 99 patients with operable lung SQCC treated at a tertiary center. The exclusion criteria included patients under 18, those with in situ or metastatic SQCC, and those who received neoa

The Effect of the Acid-Base Imbalance on the Shape and Structure of Red Blood Cells.

Red blood cells respond to fluctuations in blood plasma pH by changing the rate of biochemical and physical processes that affect the specific functions of individual cells. This study aimed to analyze the effect of pH changes on red blood cell morphology and structure. The findings revealed that an increase or decrease in pH above or below the physiological level of pH 7.4 results in the transformation of discocytes into echinocytes and causes significant alterations in the membrane, including its roughness, cytoskeleton structure, and the cell's elastic modulus. Furthermore, the study shown

Reconstruction of Squamous Cell Carcinoma on Oral Commissure With Hatchet-Shaped Flap.

The mouth is a unique and prominent element of the lower face. Given the complex anatomy, aesthetic appearance, and function of the oral commissure, its reconstruction due to various causes presents a significant challenge for surgeons. Squamous cell carcinoma (SCC) of the lip is the most common type of oral cancer, accounting for approximately 25% to 30% of all oral cancers. Wide excision is the treatment of choice, and the prognosis is generally favorable. We encountered a case of SCC of the right oral commissure in a 69-year-old man. We designed a hatchet-shaped flap to minimize anatomical

Lactate exposure shapes the metabolic and transcriptomic profile of CD8+ T cells.

INTRODUCTION: CD8+ T cells infiltrate virtually every tissue to find and destroy infected or mutated cells. They often traverse varying oxygen levels and nutrient-deprived microenvironments. High glycolytic activity in local tissues can result in significant exposure of cytotoxic T cells to the lactate metabolite. Lactate has been known to act as an immunosuppressor, at least in part due to its association with tissue acidosis. METHODS: To dissect the role of the lactate anion, independently of pH, we performed phenotypical and metabolic assays, high-throughput RNA sequencing, and mass spectr

Tsc2 shapes olfactory bulb granule cell molecular and morphological characteristics.

Tuberous Sclerosis Complex (TSC) is a neurodevelopmental disorder caused by mutations that inactivate TSC1 or TSC2. Hamartin and tuberin are encoded by TSC1 and TSC2 which form a GTPase activating protein heteromer that inhibits the Rheb GTPase from activating a growth promoting protein kinase called mammalian target of rapamycin (mTOR). Growths and lesions occur in the ventricular-subventricular zone (V-SVZ), cortex, olfactory tract, and olfactory bulbs (OB) in TSC. A leading hypothesis is that mutations in inhibitory neural progenitor cells cause brain growths in TSC. OB granule cells (GCs)

Tonic NMDA receptor signalling shapes endosomal organisation in mammalian cells.

Calcium signalling through NMDA-type glutamate receptors (NMDARs) plays a key role in synaptic plasticity in the central nervous system (CNS). NMDAR expression has also been detected in other tissues and aberrant glutamate signalling has been linked to cancer; however, the significance of NMDAR function outside of the CNS remains unclear. Here, I show that removal of extracellular calcium rapidly decreases the size of early endosomes in primary human fibroblasts. This effect can be mimicked by blockade of NMDA-type glutamate receptors but not voltage-gated calcium channels (VGCCs), and can als

Schwann cells shape the neuro-immune environs and control cancer progression.

At present, significant experimental and clinical data confirm the active involvement of the peripheral nervous system (PNS) in different phases of cancer development and progression. Most of the research effort focuses on the impact of distinct neuronal types, e.g., adrenergic, cholinergic, dopaminergic, etc. in carcinogenesis, generally ignoring neuroglia. The very fact that these cells far outnumber the other cellular types may also play an important role worthy of study in this context. The most prevalent neuroglia within the PNS consists of Schwann cells (SCs). These cells play a substant

Hypoxia in the glioblastoma microenvironment: shaping the phenotype of cancer stem-like cells.

Glioblastoma is the most common and aggressive malignant primary brain tumor. Cellular heterogeneity is a characteristic feature of the disease and contributes to the difficulty in formulating effective therapies. Glioma stem-like cells (GSCs) have been identified as a subpopulation of tumor cells that are thought to be largely responsible for resistance to treatment. Intratumoral hypoxia contributes to maintenance of the GSCs by supporting the critical stem cell traits of multipotency, self-renewal, and tumorigenicity. This review highlights the interaction of GSCs with the hypoxic tumor micr

Shape of large bound polysomes in cultured fibroblasts and thyroid epithelial cells.

Large bound polysomes were observed by conventional electron microscopy in surface or en face views of the rough endoplasmic reticulum (RER) in two cultured cell types. Cultured thyroid follicular epithelial cells and dermal fibroblasts, both from rats, were prepared for electron microscopy. Ultrathin sections were cut in the plane of the flattened cells to maximize the incidence of RER surface views. Some observations were also made on tissue sections of rat thyroid. Most of the large, RER-bound polysomes in both cell types appeared as two parallel rows of ribosome, thus resembling the shape

Anaesthetics may change the shape of isolated type I hair cells.

Type I hair cells isolated from animals anaesthetised with barbiturates or ether were found to be shorter and to lack a prominent 'neck' region when compared to cells isolated from non-anaesthetised animals. Ketamine did not have this effect. The changes observed could have important implications for the physiology of inner ear receptors. These findings infer that care should be taken in the choice of anaesthetics used in studies on cells from the inner ear.
