Understanding the possible cellular responses in plants under micro(nano)-plastic (MNPs): Balancing the structural harmony with functions.

The harmful impacts of micro(nano)-plastics (MNPs) on plants have gained significant attention in the last decades. Plants have a greater tendency to aggregate positively charged (+ve) MNPs on leaf surfaces and root tips, and it can be more challenging to enter the plant body than the negatively charged (-ve) MNPs. MNPs <20 nm can directly cross the cell wall and enter mainly via leaf stomata and root crack portion. Additionally, plants with aerenchyma tissue or higher water requirement might be more vulnerable to MNPs as well as environmental factors also affected MNPs uptake like porosity an

Joubert syndrome causing mutation in C2 domain of CC2D2A affects structural integrity of cilia and cellular signaling molecules.

Cilia are organelles extend from cells to sense external signals for tuning intracellular signaling for optimal cellular functioning. They have evolved sensory and motor roles in various cells for tissue organization and homeostasis in development and post-development. More than a thousand genes are required for cilia function. Mutations in them cause multisystem disorders termed ciliopathies. The null mutations in CC2D2A result in Meckel syndrome (MKS), which is embryonic lethal, whereas patients who have missense mutations in the C2 domain of CC2D2A display Joubert syndrome (JBTS). They surv

Functional Loss of Terminal Complement Complex Protects Rabbits from Injury-Induced Osteoarthritis on Structural and Cellular Level.

The terminal complement complex (TCC) has been described as a potential driver in the pathogenesis of posttraumatic osteoarthritis (PTOA). However, sublytic TCC deposition might also play a crucial role in bone development and regeneration. Therefore, we elucidated the effects of TCC on joint-related tissues using a rabbit PTOA model. In brief, a C6-deficient rabbit breed was characterized on genetic, protein, and functional levels. Anterior cruciate ligament transection (ACLT) was performed in C6-deficient (C6-/-) and C6-sufficient (C6+/-) rabbits. After eight weeks, the progression of PTOA w

Structural Manipulation of Ruthenium(II) Polypyridine Nitrone Complexes to Generate Phosphorogenic Bioorthogonal Reagents for Selective Cellular Labeling.

We report a new class of ruthenium(II) polypyridine complexes functionalized with a nitrone group as phosphorogenic bioorthogonal probes. These complexes were very weakly emissive owing to rapid C=N isomerization of the nitrone moiety, but exhibited significant emission enhancement upon strain-promoted alkyne-nitrone cycloaddition (SPANC) reaction with bicyclo[6.1.0]nonyne (BCN)-modified substrates. The modification of nitrone with a dicationic ruthenium(II) polypyridine unit at the α-C-position and a phenyl ring at the N-position led to remarkably accelerated reaction kinetics, which are subs

Single-Cell and Spatial Transcriptomic Analysis Reveals Shared and Cancer-Type-Specific Cellular Interactions and Chemokine Signaling Associated With Tertiary Lymphoid Structures in Colorectal and Gastric Cancers.

Tertiary lymphoid structures (TLS) demonstrate prognostic significance and associations with immunotherapy response in gastrointestinal malignancies, though their regulatory mechanisms remain incompletely defined. The current understanding of TLS at single-cell resolution is limited. Here, we integrated single-cell and spatial transcriptomics with TLS-specific signatures to map spatial distributions and chemokine signaling within colorectal (CRC) and gastric cancer (GC) microenvironments. We identified significant enrichment of characteristic T cell and macrophage subsets in the TLS regions. S

Actin filament and microtubule-centered cytoskeletal abnormalities are associated with intracellular structural disorder and cellular softening in keratoconus stromal fibroblasts.

Keratoconus (KC) is characterized by progressive corneal thinning and biomechanical weakening, however, the cellular mechanobiological basis underlying stromal softening remains poorly understood. In this study, we investigated actin filament and microtubule-centered cytoskeletal abnormalities and their associations with intracellular structural disorder and cellular mechanics in primary stromal fibroblasts derived from corneas with KC and normal corneas. Confocal microscopy revealed disrupted actin filament orientation and altered microtubule coherency in KC stromal fibroblasts. Quantitative

Biodegradable antistatic starch foams via two-step extrusion: Linking additive effects, melt rheology, and cellular structure.

Developing biodegradable starch foams with antistatic functionality remains challenging because antistatic additives can simultaneously influence charge dissipation, melt viscoelasticity, and foam-structure formation. Herein, a hierarchical film-to-foam strategy was developed to separate additive-specific effects from changes introduced during two-step extrusion foaming of starch-based materials. Homogeneous hydroxypropyl starch (HPS) films were first used to screen additives with strong antistatic potential and favorable interactions with the starch matrix, while pellet rheology was further a

Structural insights into molecular and cellular level FXR binding potentials of GW4064 and LY2562175 hybrids by multi in silico modelling analyses.

CONTEXT: Non-alcoholic fatty liver disease (NAFLD) has become a significant health concern. Existing farnesoid X receptor (FXR) agonists like GW4064 and LYS2562175 show poor pharmacokinetics, prompting researchers to develop alternative molecules. This study aims to pinpoint the structural features responsible for exhibiting FXR agonism of a series of hybrid structures of GW4064 and LYS2562175 with improved pharmacokinetic properties which supersede the existing parent ligands. Electronegative components were found to critically influence biological activity on the molecular level, supported b

Co-modification of amylase by acrolein with formaldehyde or methylglyoxal: Impact on enzyme function, structure, and cellular stress in two cell lines.

Reactive carbonyl species (RCS), such as acrolein (Acr), formaldehyde (FA), and methylglyoxal (MGO) are highly reactive aldehydes co-existing in processed foods and food materials. RCS may modify the digestive enzymes individually while whether they act together on the enzymes remains unknown. In this study, the co-action of RCS on α-amylase was studied. The results show that when Acr is present together with MGO or FA, they simultaneously modify amylase to form carbonylated proteins and inhibit the activity of amylase. The co-action of RCS increases the modification sites of amylase, enhances

Interaction between Sars-CoV-2 structural proteins and host cellular receptors: From basic mechanisms to clinical perspectives.

Severe acute respiratory syndrome coronavirus 2 (Sars-CoV-2) has caused a global pandemic that has affected the lives of billions of individuals. Sars-CoV-2 primarily infects human cells by binding of the viral spike protein to angiotensin-converting enzyme 2 (ACE2). In addition, novel means of viral entry are currently being investigated, including Neuropillin 1, toll-like receptors (TLRs), cluster of differentiation 147 (CD147), and integrin α5β1. Enriched expression of these proteins across metabolic regulatory organs/tissues, including the circulatory system, liver, pancreas, and intestine

Synthesis of conjugates of 6-aminophenanthridine and guanabenz, two structurally unrelated prion inhibitors, for the determination of their cellular targets by affinity chromatography.

The synthesis of affinity matrices for 6-aminophenanthridine (6AP) and 2,6-dichlorobenzylidenaminoguanidine (Guanabenz, GA), two unrelated prion inhibitors, is described. In both cases, the same simple spacer, epsilon-aminocaproylaminopentanol, was introduced by a Mitsunobu reaction and the choice of the anchoring position of the linker was determined by the study of the residual antiprion activity of the corresponding 6AP or GA conjugates. Very recently, these two affinity matrices were used for chromatography assays leading to the identification of ribosome (via the rRNA) as a common target

Gene expression changes in thalamus and inferior colliculus associated with inflammation, cellular stress, metabolism and structural damage in thiamine deficiency.

Identification of gene expression changes that promote focal neuronal death and neurological dysfunction can further our understanding of the pathophysiology of these disease states and could lead to new pharmacological and molecular therapies. Impairment of oxidative metabolism is a pathogenetic mechanism underlying neuronal death in many chronic neurodegenerative diseases as well as in Wernicke's encephalopathy (WE), a disorder induced by thiamine deficiency (TD). To identify functional pathways that lead to neuronal damage in this disorder, we have examined gene expression changes in the vu

Transcriptome analysis of Deinagkistrodon acutus venomous gland focusing on cellular structure and functional aspects using expressed sequence tags.

BACKGROUND: The snake venom gland is a specialized organ, which synthesizes and secretes the complex and abundant toxin proteins. Though gene expression in the snake venom gland has been extensively studied, the focus has been on the components of the venom. As far as the molecular mechanism of toxin secretion and metabolism is concerned, we still knew a little. Therefore, a fundamental question being arisen is what genes are expressed in the snake venom glands besides many toxin components? RESULTS: To examine extensively the transcripts expressed in the venom gland of Deinagkistrodon acutus

3-Amino-5-phenoxythiophenes: syntheses and structure-function studies of a novel class of inhibitors of cellular L-triiodothyronine uptake.

A series of substituted 3-amino-5-phenoxythiophenes was synthesized starting from malodinitrile and carbon disulfide. The resulting dicyanoketenedithiolate reacts via Thorpe-Dieckmann cyclization with halogen methanes bearing electron-withdrawing groups to give thiophene-2-thiolates, which can be transformed into 3-amino-5-(methylsulfonyl)thiophene-4-carbonitriles. Replacement of the methylsulfonyl groups by substituted phenolates provides the substituted 3-amino-5-phenoxythiophenes. Some of the derivatives show a considerable inhibitory potency for the L-T3 uptake in inhibition studies on hum

A study of the cellular immune response to enteroviruses in humans: identification of cross-reactive T cell epitopes on the structural proteins of enteroviruses.

We have attempted to extend our understanding of the enteroviral cross-reactive T cell response in humans. Peripheral blood mononuclear cells (PBMC) from healthy donors were stimulated in vitro with six different serotypes of enterovirus and 15 synthetic peptides representing conserved regions in the four structural proteins of these viruses. Upon challenge with different antigens, PBMC from donors responded specifically with proliferation and production of interferon-gamma (IFN-gamma). In contrast, synthesis of interleukin-4 (IL-4) or IL-10 was not detected. A T cell response to each enterovi

Congenital genetic murine (ch) hydrocephalus. A structural model of cellular dysplasia and disorganization with the molecular locus of deficient proteoglycan synthesis.

The recessively inherited ch hydrocephalic mouse has been sporadically investigated over the past 40 years as a genetic murine model for congenital hydrocephalus. Since central nervous system anomalies are commonly associated with congenital anomalies in the musculoskeletal and urogenital systems, and since congenital hydrocephalus is also associated with other developmental abnormalities, an understanding of the pathophysiology at the cellular and molecular levels of the genetic defects of this hydrocephalic murine model has application to related human disorders associated with hydrocephalus

SbHsp70 overexpression enhances drought and salinity tolerance in wheat through improved cellular stability and stress-associated structural adaptations.

INTRODUCTION: Drought and salinity are major constraints to wheat productivity worldwide. Heat shock protein 70 (Hsp70) is a conserved molecular chaperone implicated in plant stress responses, but its role in cellular stability and structural adaptation in wheat remains poorly understood. METHODS: To investigate its function, the sorghum-derived SbHsp70 gene was constitutively overexpressed in durum wheat (cv. Kofa) and bread wheat (cv. Bobwhite) via particle bombardment. Transgenic lines were evaluated under controlled drought and salinity stress conditions using physiological, cellular, mol

Effects of Layered Nanoclays on the Cellular Structure, Dynamic-Mechanical-Thermal Properties and Fire Behavior of Flame-Retardant ABS Foams.

The present work deals with the preparation and characterization of fire-retardant acrylonitrile-butadiene-styrene (ABS) foams incorporating 25 wt% of a phosphorus flame-retardant (PFR) system formed by 50% of ammonium polyphosphate (APP) and 50% of aluminum diethylphosphinate (AlPi). To further enhance performance, 5 wt% of the PFR was replaced by either montmorillonite (MMT) or layered double hydroxide (LDH) nanoparticles, maintaining the overall FR content constant. The formulations were prepared by melt blending, and foams were produced using a one-step supercritical carbon dioxide (sCO2)

Longitudinal structural, functional, and cellular myocardial alterations with chronic centrifugal continuous-flow left ventricular assist device support.

BACKGROUND: Left ventricular assist device (LVAD) support triggers adaptations within failing hearts. The HeartWare (HeartWare International, Inc., Framingham, MA) LVAD exhibits different flow profiles and afterload dependence compared with previous-generation devices, which may alter remodelling patterns. We sought to characterize myocardial adaptation to third-generation centrifugal-flow LVADs at a functional, hemodynamic, and structural level in addition to profiling transcriptomal changes using next-generation sequencing platforms. METHODS: We studied 37 patients supported with the HeartW

In vivo structural and cellular remodeling of engineered bone-ligament-bone constructs used for anterior cruciate ligament reconstruction in sheep.

Anterior cruciate ligament (ACL) ruptures rank among the most prevalent and costly sports-related injuries. Current tendon grafts used for ACL reconstruction are limited by suboptimal biomechanical properties. We have addressed these issues by engineering multiphasic bone-ligament-bone (BLB) constructs that develop structural and mechanical properties similar to native ACL. The purpose of this study was to examine the acute remodeling process that occurs as the BLB grafts advance toward the adult ligament phenotype in vivo. Thus, we implanted BLB constructs fabricated from male cells into fema
