Madecassoside ameliorates cisplatin-induced nephrotoxicity by inhibiting activation of the mitogen activated protein kinase pathway.

Nephrotoxicity is a major side effect of cisplatin. Apoptosis, oxidative stress, inflammation, and the MAPK signaling pathway activation are concerned with the pathophysiology of cisplatin-induced acute kidney injury (AKI). Madecassoside (MA), an active constituent of Centella asiatica, has anti-oxidative and anti-inflammatory effects. The present research aim to investigate the underlying protective mechanisms of MA on cisplatin nephrotoxicity. Pretreatment of mice with MA markedly ameliorated cisplatin-induced renal tubular cell injury evidenced by the improvement of kidney function and kidn

Asiatic Acid Ameliorates Doxorubicin-Induced Cardiac and Hepato-Renal Toxicities with Nrf2 Transcriptional Factor Activation in Rats.

Asiatic acid (AA), a pentacyclic triterpenoid, is a key phytoconstituent of Centella asiatica. AA is a patented as a cytotoxic substance, and it exerts cytotoxicity against Hep3B, Hela and MCF-7 cell lines. However, pentacyclic triterpenoids also modulate the expression and transcriptional activities of Nrf2 and exert cytoprotective effects. In this study, we investigated the effects of AA on the doxorubicin (DXR)-induced organ toxicities and expression of the Nrf2. DXR toxicity was induced by a single intravenous injection of 65.75 mg/kg of DXR. Seven days pretreatment with AA at the doses of

MicroRNA-Enriched Plant-Derived Exosomes Alleviate Colitis by Modulating Systemic Immunity, Metabolic Homeostasis, and Gut Microbiota.

Ulcerative colitis (UC) is a condition with complex immune dysregulation and chronic intestinal inflammation. Currently, there are still a lack of natural and low-toxicity therapeutic approaches for UC. Emerging research has underscored the potential of plant-derived exosomes for UC treatment. In this study, it is found that Centella Asiatica-derived exosomes (CAEs) have anti-colitis effects. CAEs selectively accumulate in inflamed colons, enabling targeted delivery of RNA cargos. MicroRNA (miRNA)-enriched CAEs not only mitigate inflammation but also facilitate the reconstitution of a balanced

Madecassoside suppresses migration of fibroblasts from keloids: involvement of p38 kinase and PI3K signaling pathways.

Keloid is a specific skin scar that expands beyond the boundaries of the original injury as it heals. The invasive nature of keloid and notable migratory activity of fibroblasts are a hallmark, which distinguishes keloids from other common scars. Madecassoside, a triterpenoid saponin occurring in Centella asiatica herbs, possesses unique pharmacological properties to enhance wound-healing and diminish keloid formation. However, the effects of madecassoside on the formation of keloid scars have been poorly understood. Here, we focused on the potential of madecassoside on the migration of keloid

Healing the Sole: Combining Natural Plant Extracts and Conventional Drugs in Wound Delivery Systems for Diabetic Foot Ulcer Treatment.

Diabetic foot ulcers (DFUs) affect 19-34% of diabetic patients with a five-year mortality rate of 50-70%. It is characterized by a high infection risk, chronic inflammation, oxidative stress, and poor angiogenesis, rendering existing therapies inadequate, which causes antimicrobial resistance and amputations. Bioactive compounds, such as curcumin, flavonoids, tannins, polyphenols, and proteins, found in natural product extracts (NPEs) have antibacterial, anti-inflammatory, antioxidant, and regenerative properties but are prone to variability and instability. Conventional drugs can be applied f

The Zebrafish Model as a New Discovery Path for Medicinal Plants in the Treatment of Parkinson's Disease.

Parkinson's disease (PD) is one of the most frequent degenerative central nervous system disorders affecting older adults. Dopaminergic neuron failure in the substantia nigra is a pathological sign connected with the motor shortfall of PD. Due to their low teratogenic and adverse effect potential, medicinal herbs have emerged as a promising therapy option for preventing and curing PD and other neurodegenerative disorders. However, the mechanism through which natural compounds provide neuroprotection against PD remains unknown. While testing compounds in vertebrates such as mice is prohibitivel

Intestinal interleukin-10 mobilization as a contributor to the anti-arthritis effect of orally administered madecassoside: a unique action mode of saponin compounds with poor bioavailability.

Madecassoside, a triterpenoid saponin present in Centella asiatica herbs with extremely low bioavailability, possesses excellent anti-rheumatoid arthritis property after oral administration. Such a disconnection between poor pharmacokinetic property and undoubted bioactivity also exists in many other herbal medicines. However, there is no reasonable explanation for this phenomenon to date. Here we showed that orally administered madecassoside displayed marked therapeutic effect on collagen-induced arthritis (CIA) in rats, which was accompanied by a systemic downregulation of inflammatory cytok
