We describe a novel preparation of the isolated brain stem and spinal cord from prometamorphic tadpole stages of the South African clawed frog ( Xenopus laevis) that permits whole cell patch-clamp recordings from neurons in the ventral spinal cord. Previous research on earlier stages of the same species has provided one of the most detailed understandings of the design and operation of a central pattern generator circuit. Here we have addressed how development sculpts complexity from this more basic circuit. The preparation generates bouts of fictive swimming activity either spontaneously or i

The pituitary gland releases hormones in a pulsatile fashion guaranteeing signalling efficiency. The determinants of pulsatility are poorly circumscribed. Here we show in magnocellular hypothalamo-neurohypophyseal oxytocin (OT) neurons that the bursting activity underlying the neurohormonal pulses necessary for parturition and the milk-ejection reflex is entirely driven by a female-specific central pattern generator (CPG). Surprisingly, this CPG is active in both male and female neonates, but is inactivated in males after the first week of life. CPG activity can be restored in males by orchide

Periodic behavioral motor patterns are normally controlled by neural circuits, such as central pattern generators. We here report a novel mechanism of motor pattern generation by non-neural cells. The defecation motor program in Caenorhabditis elegans consists of three stereotyped motor steps with precise timing and this behavior has been studied as a model system of a ultradian biological clock [J.H. Thomas, Genetic analysis of defecation in C. elegans, Genetics 124 (1990) 855-872; D.W. Liu, J.H. Thomas, Regulation of a periodic motor program in C. elegans, J. Neurosci. 14 (1994) 1953-1962; K

BACKGROUND: The 2006 international consensus defines cerebral palsy as a permanent disorder of movement and posture caused by non-progressive brain injury in the developing fetus or infant, yet it sets no upper age limit for the injury; in practice, countries adopt arbitrary chronological-age cut-offs, most commonly 24 months, without any neurobiological basis, and the 2025-2026 updated descriptions, although shifting toward a broader "description," retain the same temporal framework and provide no operational diagnostic boundary.
CORE HYPOTHESIS: The diagnostic essence of cerebral palsy lies

Constant drive provided to the human lumbar spinal cord by epidural electrical stimulation can cause local neural circuits to generate rhythmic motor outputs to lower limb muscles in people paralysed by spinal cord injury. Epidural spinal cord stimulation thus allows the study of spinal rhythm and pattern generating circuits without their configuration by volitional motor tasks or task-specific peripheral feedback. To reveal spinal locomotor control principles, we studied the repertoire of rhythmic patterns that can be generated by the functionally isolated human lumbar spinal cord, detected a

Current source generators (dipoles) of the human visual evoked potentials to pattern-onset stimuli were investigated with the dipole tracing method, using a realistic four-layer head model of scalp-skull-fluid-brain, which can equate the surface potential distributions on a scalp to one or two corresponding equivalent dipoles. Three healthy adult human subjects were used, and 29 electrodes were set on a scalp of each subject. Visual stimulus of a checkerboard pattern was presented for 250 ms in each of eight different visual fields (central and peripheral parts of each of four quadrant fields)

Fitting models to experimental intracellular data is challenging. While detailed conductance-based models are difficult to train, phenomenological statistical models often fail to capture the rich intrinsic dynamics of circuits such as central pattern generators (CPGs). A recent trend has been to employ tools from deep learning to obtain data-driven models that can quantitatively learn intracellular dynamics from experimental data. This paper addresses the general questions of modeling, training, and interpreting a large class of such models in the context of estimating the dynamics of a neura

Motor behavior requires the balanced production and integration of a variety of neural cell types. Motor neurons are positioned in discrete locations in the spinal cord, targeting specific muscles to drive locomotive contractions. Specialized spinal interneurons modulate and synchronize motor neuron activity to achieve coordinated motor output. Changes in the ratios and connectivity of spinal interneurons could drastically alter motor output by tipping the balance of inhibition and excitation onto target motor neurons. Importantly, individuals with Fragile X syndrome (FXS) and associated autis

A partly synchronized pulsatile secretion of luteinizing hormone (LH) and prolactin has previously been suggested as an indication of the coupling of the respective pulse generators under certain conditions. In women with hyperandrogenemic chronic anovulation, episodic LH secretion is disturbed. It was, therefore, the aim of the present study to evaluate possible changes in episodic prolactin secretion pattern and in LH/prolactin co-pulsatility, and to relate the results to the accelerated LH pulse frequencies often seen in patients with hyperandrogenemic chronic anovulation. Blood samples of

The simpler nervous systems of certain invertebrates provide opportunities to examine colocalized classical neurotransmitters in the context of identified neurons and well defined neural circuits. This study examined the distribution of γ-aminobutyric acid-like immunoreactivity (GABAli) in the nervous system of the panpulmonates Biomphalaria glabrata and Biomphalaria alexandrina, major intermediate hosts for intestinal schistosomiasis. GABAli neurons were localized in the cerebral, pedal, and buccal ganglia of each species. With the exception of a projection to the base of the tentacle, GABAli

Mechanically stimulating the dorsal lumbar region inhibits locomotion and reduces weight support during standing in rabbits and cats. However, how this inhibitory effect from the lumbar skin is mediated is poorly understood. Here we evaluated the effect of mechanically stimulating (vibration or pinch) the dorsal lumbar region on short-latency (8- to 13-ms onset) cutaneous reflex responses, evoked by electrically stimulating the superficial peroneal or distal tibial nerves, in seven adult cats with a low thoracic spinal transection (spinal cats). Cutaneous reflexes were evoked before, during, a

μ-Opioid receptors are distributed widely in the brain stem respiratory network, and opioids with selectivity for μ-type receptors slow in vivo respiratory rhythm in lowest effective doses. Several studies have reported μ-opioid receptor effects on the three-phase rhythm of respiratory neurons, but there are until now no reports of opioid effects on oscillatory activity within respiratory discharges. In this study, effects of the μ-opioid receptor agonist fentanyl on spike train discharge properties of several different types of rhythm-modulating medullary respiratory neuron discharges were an

Neurons in cold-blooded animals remarkably maintain their function over a wide range of temperatures, even though the rates of many cellular processes increase twofold, threefold, or many-fold for each 10°C increase in temperature. Moreover, the kinetics of ion channels, maximal conductances, and Ca(2+) buffering each have independent temperature sensitivities, suggesting that the balance of biological parameters can be disturbed by even modest temperature changes. In stomatogastric ganglia of the crab Cancer borealis, the duty cycle of the bursting pacemaker kernel is highly robust between 7

Neurons in the central pattern-generating circuits in the crustacean stomatogastric ganglion (STG) release neurotransmitter both as a graded function of presynaptic membrane potential that persists in TTX and in response to action potentials. In the STG of the male crab Cancer borealis, the modulators oxotremorine, C. borealis tachykinin-related peptide Ia (CabTRP1a), red pigment concentrating hormone (RPCH), proctolin, TNRNFLRFamide, and crustacean cardioactive peptide (CCAP) produce and sustain robust pyloric rhythms by activating the same modulatory current (IMI), albeit on different subset

The unique, hypnotic pulsation behavior of certain soft corals of the Xeniidae family, the result of rhythmic opening and closing of their tentacles, has fascinated scientists since the 18th century. Repetitive motion is regulated by autonomous neural circuits known as pacemakers or central pattern generators. However, little is known about such circuits in nonbilaterian organisms like corals. In this report, Xenia umbellata, a fast-growing octocoral, served as a model organism to study the muscular and neural mechanisms of pulsation. Leveraging this coral's rapid regeneration ability, we test

INTRODUCTION: Epidural electrical stimulation of the conus medullaris has helped facilitate native motor recovery in individuals with complete cervicothoracic spinal cord injuries (SCI). A theorized mechanism of clinical improvement includes supporting central pattern generators intrinsic to the conus medullaris. Because spinal cord stimulators (SCS) are approved for the treatment of neuropathic pain, we were able to test this experimental therapy in a subject with complete L1 paraplegia and neuropathic genital pain due to a traumatic conus injury.

BACKGROUND: Rats exhibit extremely limited motor function recovery after total transection of the spinal cord (SCT). We previously reported that SM-216289, a semaphorin3A inhibitor, enhanced axon regeneration and motor function recovery in SCT adult rats. However, these effects were limited because most regenerated axons likely do not connect to the right targets. Thus, rebuilding the appropriate connections for regenerated axons may enhance recovery. In this study, we combined semaphorin3A inhibitor treatment with extensive treadmill training to determine whether combined treatment would furt

Locomotion in vertebrates is controlled by central pattern generators in the spinal cord. The roles of specific network architecture and neuronal properties in rhythm generation by such spinal networks are not fully understood. We have used multisite recording from dissociated cultures of embryonic rat spinal cord grown on multielectrode arrays to investigate the patterns of spontaneous activity in randomised spinal networks. We were able to induce similar patterns of rhythmic activity in dissociated cultures as in slice cultures, although not with the same reliability and not always with the

INTRODUCTION: Abdominal pain is an occasionally observed non-motor symptom during "off" periods in Parkinson's disease (PD), often associated with end-of-dose wearing-off (EODWO). However, severe abdominal manifestations such as painful abdominal contraction (PAC) are rare, and their pathogenesis remains unclear. We report a unique case in which both PAC and belly dancer's dyskinesia (BDD) emerged following subthalamic nucleus deep brain stimulation (STN-DBS).
CASE PRESENTATION: A 74-year-old woman with PD underwent bilateral STN-DBS. Following a postoperative reduction in her levodopa dosage

Swallowing is regulated by cortical networks and the caudal brainstem. Damage to central pattern generators, commonly seen after stroke, can result in severe dysphagia with limited treatment options. Pharyngeal electrical stimulation (PES) presents a promising therapy as it may enhance sensory input to impaired brainstem networks by targeting pharyngeal sensory afferents, the primary site of dysfunction in brainstem infarction. This single-case study reports on a 78-year-old woman with a confirmed left-sided infarction in the dorsolateral medulla oblongata, resulting in severe dysphagia. PES w
