C26-Ceramide as highly sensitive biomarker for the diagnosis of Farber Disease.

Farber disease (FD) is a rare autosomal recessive disease caused by mutations in the acid ceramidase gene (ASAH1). Low ceramidase activity results in the accumulation of fatty substances, mainly ceramides. Hallmark symptoms at clinical level are periarticular nodules, lipogranulomas, swollen and painful joints and a hoarse voice. FD phenotypes are heterogeneous varying from mild to very severe cases, with the patients not surviving past their first year of life. The diagnostic aspects of FD are poorly developed due to the rarity of the disease. In the present study, the screening for ceramides

Sphingomyelinase D/ceramide 1-phosphate in cell survival and inflammation.

Sphingolipids are major constituents of biological membranes of eukaryotic cells. Many studies have shown that sphingomyelin (SM) is a major phospholipid in cell bilayers and is mainly localized to the plasma membrane of cells, where it serves both as a building block for cell architecture and as a precursor of bioactive sphingolipids. In particular, upregulation of (C-type) sphingomyelinases will produce ceramide, which regulates many physiological functions including apoptosis, senescence, or cell differentiation. Interestingly, the venom of some arthropodes including spiders of the genus Lo

Modulation of C16:0-ceramide in hypertrophied immature hearts by losartan.

BACKGROUND: The angiotensin type 2 receptor plays a unique role in growth inhibition in adult myocardium via modulation of ceramide synthesis. Angiotensin type 1 (AT1 )-receptor blockade results in increased angiotensin type 2 receptor activation by angiotensin II, and AT1 -receptor blockers are sometimes prescribed to children for the treatment of cardiac hypertrophy or heart failure. We investigated the changes of ceramide lipid components in hypertrophied immature rabbit hearts after chronic administration of the AT1 -receptor blocker, losartan. METHODS: One-week-old Japanese white rabbits

Fumonisins and fumonisin analogs as inhibitors of ceramide synthase and inducers of apoptosis.

Sphingoid bases are growth inhibitory and pro-apoptotic for many types of cells when added to cells exogenously, and can be elevated to toxic amounts endogenously when cells are exposed to inhibitors of ceramide synthase. An important category of naturally occurring inhibitors are the fumonisins, which inhibit ceramide synthase through structural similarities with both the sphingoid base and fatty acyl-CoA co-substrates. Fumonisins cause a wide spectrum of disease (liver and renal toxicity and carcinogenesis, neurotoxicity, induction of pulmonary edema, and others), and most-possibly all-of th

Ceramide stimulates prostaglandin production by human amnion and decidual cells.

OBJECTIVE: To determine whether ceramide regulates prostaglandin (PG) production by cultured human amnion cells and decidual cells independently of interleukin-1 beta (IL-1 beta). METHODS: Cells were grown in monolayer culture and then incubated with varying concentrations of ceramide, IL-1 beta, ceramide in the presence and absence of IL-1, and control media. Production of PGE2 was determined using a specific radioimmunoassay. RESULTS: Ceramide induced a significant concentration-dependent increase in PGE2 production by amnion cells and decidual cells. However, PGE2 production induced by IL

Elevated ceramide levels in GH4C1 cells treated with retinoic acid.

Ceramide is produced in HL 60 cells in response to 1 alpha,25-dihydroxy vitamin D3 (vitamin D3) (Okazaki et al. (1989) J. Biol. Chem. 264, 19076-19080). HL 60 cell differentiation can be mimicked by cell permeable ceramides (Okazaki et al. (1990) J. Biol. Chem. 265, 15823-15831). Vitamin D3 like other thyroid-steroid hormones binds to a member of the steroid hormone family. We sought to investigate whether agonists other than vitamin D3 which exert their effects through members of the steroid receptor family affect cellular ceramide levels. Treatment of GH4C1 cells with 10 microM all-trans ret

Complex fucolipids of hog gastric mucosa. Structure of the ceramide eikosahexoside.

The structure of a complex fucolipid from hog gastric mucosa containing twenty sugar residues and exhibiting blood-group (A + H) activity has been investigated. Based on the results of immunological assays, partial acid hydrolysis, sequential degradation with specific exoglycosidases, oxidation with periodate and chromium trioxide, and permethylation analysis, we suggest that the carbohydrate chain of this fucolipid contains four termini. One of the termini bears beta Gal1 leads to 4 beta GlcNAc disaccharide, two bear blood-group A determinant and one bears H determinant. Two of the branches,

Globoid cell leukodystrophy: deficiency of lactosyl ceramide beta-galactosidase.

Activity of lactosyl ceramide beta-galactosidase (beta-D-galactoside galactohydrolase, EC 3.2.1.23) was found to be extremely low in enzyme preparations from liver, brain, and cultured skin fibroblasts from patients with Krabbe's disease. Leukocytes from one set of parents had enzyme levels approximately half those measured in control leukocytes. The low activity observed for this galactolipid hydrolase is the fourth enzymatic deficiency noted for this genetic disease. Beta-galactosidase activity toward galactocerebroside, psychosine, and monogalactosyl diglyceride is also low in patients with

Hepatic FoxO1 loss leads to pancreatic β cell deficiency via liver-derived C18/22/24 ceramides.

Preserving pancreatic β cells is requisite for antihyperglycemic strategy. Hepatic FoxO1 represents a promising therapeutic target. However, the systemic consequences on β cell homeostasis upon hepatic FoxO1 loss and the underlying mechanism remain largely unexplored. Using liver-specific Foxo1 knockdown mouse and transgenic zebrafish models, we uncover that hepatic FoxO1 loss leads to β cell deficiency via triggering apoptosis. Lipidomic and pharmacological analyses identify that hepatic FoxO1 loss increases the liver-derived C18/22/24 ceramides, which contribute to apoptosis by inhibiting th

Cerebrospinal ceramides and cognition as a function of striatal asymmetry in early stage of Parkinson's disease.

BackgroundParkinson's disease (PD) is characterized by heterogeneous clinical phenotypes that may be influenced by the asymmetry of striatal denervation. The alpha-Synuclein Origin site and Connectome (SOC) model proposes that different disease onset patterns-"body first" versus "brain first"-affect symptom development and cognitive decline.ObjectiveThis study aims to explore the relationship between striatal denervation asymmetry, ceramide metabolism, and cognitive performance in early-stage PD.MethodsWe analyzed data from 329 patients with PD at baseline, categorized by the type of putamen d

Ceramides in Autoimmune Rheumatic Diseases: Existing Evidence and Therapeutic Considerations for Diet as an Anticeramide Treatment.

Autoimmune rheumatic diseases (AIRDs) constitute a set of connective tissue disorders and dysfunctions with akin clinical manifestations and autoantibody responses. AIRD treatment is based on a comprehensive approach, with the primary aim being achieving and attaining disease remission, through the control of inflammation. AIRD therapies have a low target specificity, and this usually propels metabolic disturbances, dyslipidemias and increased cardiovascular risk. Ceramides are implicated in inflammation through several different pathways, many of which sometimes intersect. They serve as signa

Identification of circulating plasma ceramides as a potential sexually dimorphic biomarker of pancreatic cancer-induced cachexia.

BACKGROUND: Cancer patients who exhibit cachexia lose weight and have low treatment tolerance and poor outcomes compared to cancer patients without weight loss. Despite the clear increased risk for patients, diagnosing cachexia still often relies on self-reported weight loss. A reliable biomarker to identify patients with cancer cachexia would be a valuable tool to improve clinical decision making and identification of patients at risk of adverse outcomes. METHODS: Targeted metabolomics, that included panels of amino acids, tricarboxylic acids, fatty acids, acylcarnitines, and sphingolipids,

Short-term magnesium deficiency upregulates ceramide synthase in cardiovascular tissues and cells: cross-talk among cytokines, Mg2+, NF-κB, and de novo ceramide.

The present study tested the hypotheses that 1) short-term dietary deficiency (MgD) of magnesium (21 days) would result in the upregulation of ceramide synthase (CS) in left ventricular (LV), right ventricular, atrial, and aortic smooth muscle, as well as induce a synthesis/release of select cytokines and chemokines into the LV and aortic smooth muscle and serum; 2) exposure of primary cultured vascular smooth muscle cells (VSMCs) to low extracellular Mg concentration would lead to the synthesis/release of select cytokines/chemokines, activation of N-SMase, and the de novo synthesis of ceramid

Identification of gangliosides and ceramides as biomarkers for mucopolysaccharidosis type II (hunter syndrome) through untargeted lipidomic analysis.

INTRODUCTION: Mucopolysaccharidosis type II (Hunter syndrome) is an X-linked recessive disorder caused by iduronate-2-sulfatase deficiency, affecting mainly male patients. The lack of its enzyme activity causes the accumulation of the glycosaminoglycans heparan sulfate and dermatan sulfate in all body tissues and leads to a secondary accumulation of gangliosides and ceramides. OBJECTIVE AND METHODS: We conducted a lipidomic study to investigate the dysregulation of lipid pathways in neuronopathic MPS type II. A modified liquid extraction was performed for untargeted lipid analysis. A reverse

Involvement of Ceramides in Non-Alcoholic Fatty Liver Disease (NAFLD) Atherosclerosis (ATS) Development: Mechanisms and Therapeutic Targets.

Non-alcoholic fatty liver disease (NAFLD) and atherosclerosis (ATS) are worldwide known diseases with increased incidence and prevalence. These two are driven and are interconnected by multiple oxidative and metabolic functions such as lipotoxicity. A gamut of evidence suggests that sphingolipids (SL), such as ceramides, account for much of the tissue damage. Although in humans they are proving to be accurate biomarkers of adverse cardiovascular disease outcomes and NAFLD progression, in rodents, pharmacological inhibition or depletion of enzymes driving de novo ceramide synthesis prevents the

Plasma Ceramides and Risk of Impaired Lower-Extremity Function in Older Adults: A Nested Case-Control Study.

BACKGROUND: Higher levels of ceramides have been linked to several chronic diseases; also there is emerging cross-sectional evidence that ceramides are associated with lower physical functioning. This research assessed for the first time the prospective relationship between ceramide species and impaired lower-extremity function (ILEF) in older adults. METHODS: Case-control study with 43 cases of ILEF and 86 age- and sex-matched controls, which was nested in the Seniors-ENRICA cohort of community-dwelling older adults. Association between increased plasma ceramides and chronic kidney disease in patients with and without ischemic heart disease.

AIM: Plasma levels of certain ceramides are increased in patients with ischemic heart disease (IHD). Many risk factors for IHD are also risk factors for chronic kidney disease (CKD), but it is currently uncertain whether plasma ceramide levels are increased in patients with CKD. METHODS: We measured six previously identified high-risk plasma ceramide concentrations [Cer(d18:1/16:0), Cer(d18:1/18:0), Cer(d18:1/20:0), Cer(d18:1/22:0), Cer(d18:1/24:0) and Cer(d18:1/24:1)] in 415 middle-aged individuals who attended our clinical Cardiology and Diabetes services over a period of 9 months.
