Clinical laboratory evaluation of Invader® chemistry and hybrid capture for detection of high-risk human papillomavirus in liquid-based cytology specimens.

A laboratory-developed test for high-risk human papillomavirus (HPV) that utilizes the Invader technology (Hologic, Madison, WI, USA) was compared to hybrid capture (Digene, Gaithersburg, MD, USA). A total of 342 ThinPrep specimens were de-identified following cytology screening (10 high-grade squamous intraepithelial lesions, 14 low-grade squamous intraepithelial lesions, 199 atypical squamous cells of undetermined significance, 119 normal). DNA was manually extracted prior to interrogation with Invader. Hybrid capture/Invader discrepancies were subject to HPV sequencing. One sample (0.3%) wa

Clinical Chemistry and Hematology Values in Captive European Mink (Mustela lutreola): Reference Intervals and Evaluation of the Effects of Sex, Age, and Body Condition.

The European mink is a critically endangered mustelid. Species-specific hematological and biochemical reference intervals are lacking, complicating accurate health assessments in conservation programs. This study aimed to determine these reference intervals for European mink, following international guidelines, with 110 blood samples collected under isoflurane anesthesia, outside the breeding season, from 75 captive individuals (37 females, 38 males). Results were broadly comparable to the American mink and domestic ferret. Several parameters varied significantly according to sex, age, and bod

Updates on Intrinsic Medicinal Chemistry of 1,4-dihydropyridines, Perspectives on Synthesis and Pharmacokinetics of Novel 1,4-dihydropyrimidines as Calcium Channel Blockers: Clinical Pharmacology.

BACKGROUND: Several chemical studies described the physiological efficacy of 1,4- dihydropyridines (DHPs). DHPs bind to specific sites on the α1 subunit of L-type calcium channels, where they demonstrate a more pronounced inhibition of Ca2+ influx in vascular smooth muscle compared to myocardial tissue. This selective inhibition is the basis for their preferential vasodilatory action on peripheral and coronary arteries, a characteristic that underlies their therapeutic utility in managing hypertension and angina. Among the vascular-selective DHPs, nifedipine, felodipine, and isradipine are key

The 'mysterious' disease in Swedish moose. Concentrations of trace elements in liver and kidneys and clinical chemistry. Comparison with experimental molybdenosis and copper deficiency in the goat.

Moose (Alces alces L.) affected by a disease with unknown aetiology were compared with healthy moose regarding trace element concentrations in liver and kidneys, as well as certain clinical chemical parameters of blood plasma. The diseased moose showed decreased hepatic concentrations of Cu, Cd and Mg. Renal concentrations of Cd, Co, Mg and Mn were also decreased. Substantially increased concentrations in both liver and kidneys were seen for Al, Ca, Fe, Pb and Zn, while Se and Mo were increased in liver tissue. The hepatic levels of Mo in the affected moose were 36% higher and Cu was approxima

Assessment of Selected Clinical Chemistry Analytes Among Hypertensive Patients at Wallaga University Referral Hospital, Nekemte, Oromia, Western Ethiopia: A Comparative Cross-Sectional Study.

BACKGROUND AND AIMS: Hypertension (HTN) is a long-term condition marked by consistently high arterial blood pressure. It is a leading contributor to cardiovascular diseases and early mortality around the globe. It increases the risk of ischemic heart disease, stroke, chronic kidney disease (CKD), and dementia. So far, several studies have been conducted, but to the authors' knowledge, there is a scarcity of similar studies done on the selected analytes in our setting, which can be an important input for the early detection of disturbance in specific parameters. So, this study aims to assess se

Inactivation of Blood-Borne Enveloped Viruses with the Nonionic Detergent 2-[4-(2,4,4-Trimethylpentan-2-yl)Phenoxy]Ethanol Does Not Bias Clinical Chemistry Results.

BACKGROUND: Patients infected with virulent pathogens require the sophisticated diagnostic capabilities of a core laboratory for optimal care. This is especially true in outbreaks that strain healthcare system capacity. However, samples from such patients pose an infection risk for laboratory workers. We evaluated a strategy for mitigating this risk by preincubating specimens with 2-[4-(2,4,4-trimethylpentan-2-yl)phenoxy]ethanol, a non-ionic detergent commonly calledTriton X-100. METHODS: Lithium-heparinized plasma was mixed with the detergent Triton X-100 at 1%. Inactivation of Ebola virus (

Laboratory-based clinical audit as a tool for continual improvement: an example from CSF chemistry turnaround time audit in a South-African teaching hospital.

INTRODUCTION: Timeliness of laboratory results is crucial to patient care and outcome. Monitoring turnaround times (TAT), especially for emergency tests, is important to measure the effectiveness and efficiency of laboratory services. Laboratory-based clinical audits reveal opportunities for improving quality. Our aim was to identify the most critical steps causing a high TAT for cerebrospinal fluid (CSF) chemistry analysis in our laboratory. MATERIALS AND METHODS: A 6-month retrospective audit was performed. The duration of each operational phase across the laboratory work flow was examined.

Assessment of clinical chemistry and hematological parameters in female Sprague-Dawley rats following a 7-day oral exposure to three different species of Echinacea.

Echinacea has grown in popularity due to its broad therapeutic benefits. Despite its popularity, comprehensive safety evaluations for three medicinal species are limited. In this study, female Sprague-Dawley rats received oral doses (0, 25, 50, 100, 200 mg/kg/d) of 75% (v/v) ethanol extract from the aerial parts of 9 Echinacea samples of three species - Echinacea purpurea, Echinacea angustifolia, and Echinacea pallida - over a 7-day period. Blood and serum samples, collected twenty-four hours post the final dose, were analyzed for hematology and clinical chemistry parameters. The results revea

Protein precipitation as a possible important pitfall in the clinical chemistry analysis of blood samples containing monoclonal immunoglobulins: 2 case reports and a review of the literature.

Two case reports are presented, both illustrating an analytical interference caused by monoclonal immunoglobulins. Falsely low results were obtained in the routine analysis of glucose, CRP and HDL-cholesterol. When analysing samples containing paraproteins, various problems can be encountered in the clinical laboratory: next to the antibody effect, pseudohyponatraemia, hyperviscosity, cryoglobulinaemia and gel formation have to be taken into account. In our two cases the interference was caused by paraprotein precipitation, causing an increased turbidity and an apparent increase of light absor

Chromogenic and FRET peptide substrates, quantum-dot peptide conjugates and electrochemical peptide interfaces: A 25-year journey from combinatorial chemistry to clinical readouts-A viewpoint.

This Viewpoint synthesises 25 years of work from a single laboratory on activity-based protease sensing and offers a critical appraisal of where each of the three resulting platforms stands today. The activity of proteolytic enzymes-rather than their abundance-determines the outcome of inflammation, tumour progression, viral entry and barrier dysfunction. Measuring it in clinical samples is demanding: closely related proteases share substrate preferences, and biological matrices are crowded with endogenous inhibitors and competing hydrolases. Over the past 25 years, our group at the University

Assessment of lipemia interference on phenobarbital quantification using particle-enhanced turbidimetric inhibition immunoassay with Dimension clinical chemistry system and comparison to high-performance liquid chromatography.

Phenobarbital, an anti-seizure medication with a narrow therapeutic window, requires therapeutic drug monitoring (TDM) for safe and effective use. Lipemia is a well-known interferent in turbidimetric immunoassays, yet its impact on phenobarbital quantification compared with high-performance liquid chromatography (HPLC) has not been fully characterized. We evaluated the effect of lipemia on phenobarbital measurement using a particle-enhanced turbidimetric inhibition immunoassay (PETINIA; Dimension Xpand Plus clinical chemistry system, Siemens) versus HPLC. Simulated lipemic plasma was prepared

Development of a pregnancy-specific physiologically based pharmacokinetic model for cadmium and estimation of fetal internal dose using serial maternal biomarkers and clinical chemistry parameters.

Cadmium (Cd) poses developmental risks during pregnancy, yet single time-point biomarkers inadequately capture cumulative fetal internal dose. We developed a pregnancy-specific semi-mechanistic physiologically based pharmacokinetic (PBPK) model to estimate fetal Cd exposure trajectories in 2778 mother-infant pairs from the Korea Children's Environmental Health Study (Ko-CHENS). The ten-compartment maternal-fetal model incorporated serial blood Cd at early (T1; gestational weeks 12-16) and late pregnancy (T3; weeks 28-34) with paired cord blood Cd, and was individually parameterized using four

Preanalytical quality improvement. In pursuit of harmony, on behalf of European Federation for Clinical Chemistry and Laboratory Medicine (EFLM) Working group for Preanalytical Phase (WG-PRE).

Laboratory diagnostics develop through different phases that span from test ordering (pre-preanalytical phase), collection of diagnostic specimens (preanalytical phase), sample analysis (analytical phase), results reporting (postanalytical phase) and interpretation (post-postanalytical phase). Although laboratory medicine seems less vulnerable than other clinical and diagnostic areas, the chance of errors is not negligible and may adversely impact on quality of testing and patient safety. This article, which continues a biennial tradition of collective papers on preanalytical quality improveme

Experimental copper and chromium deficiency and additional molybdenum supplementation in goats. II. Concentrations of trace and minor elements in liver, kidneys and ribs: haematology and clinical chemistry.

Since the mid-1980s a previously undescribed disease has affected moose in south-western Sweden. Investigations made to reveal evidence of a viral aetiology have proved unsuccessful. Trace element studies in apparently healthy moose shot during regular hunting suggested a trace element imbalance, with excessive molybdenum uptake causing secondary copper deficiency. The results also indicated a possible chromium deficiency. To verify this hypothesis, an experimental study was performed in male goats fed a semi-synthetic diet for 1.5 years. The animals were kept and treated in four groups: Contr

Evaluation of an enzymatic one-step assay for Beta-Hydroxybutyrate blood testing on clinical chemistry platforms using HILIC-MS/MS as a non-enzymatic reference method.

BACKGROUND: β-Hydroxybutyrate (β-HB), the predominant circulating ketone body, serves as alternative energy source during periods of glucose deprivation. Accurate quantification of β-HB is essential for the clinical assessment and monitoring of ketosis and diabetic ketoacidosis (DKA). METHODS: We evaluated the analytical performance of the DiaSys β-Hydroxybutyrate 21 FS assay (BHB 21 FS), a one-step enzymatic assay based on β-hydroxybutyrate dehydrogenase-catalyzed oxidation of β-HB to generate NADH, which is photometrically measured at 340 nm. Physicochemical properties, biological chemistry and mechanisms of action of caries-arresting diammine-silver(I) fluoride and silver(I)-fluoride solutions for clinical use: a critical review.

This paper serves as a Part II follow-up of our research investigations performed on the molecular structures of silver(I)-fluoride (SF) and diammine-silver(I) fluoride (SDF) complexes in solution-based commercial products for clinical application, their precise chemical compositions, and their nature in aqueous solution, the latter including rapid fluoride-exchange processes at the silver(I) ion centre monitored by 19F NMR analysis (Part I). Part I of this series also explores the mechanisms of action (MoA) of these complexes, and is therefore largely focused on their chemical reactions with

Impact of hepatitis C virus co-infection on HIV patients before and after highly active antiretroviral therapy: an immunological and clinical chemistry observation, Addis Ababa, Ethiopia.

BACKGROUND: Hepatitis C virus (HCV) is an RNA virus which has been known to cause acute and chronic necro-inflammatory disease of the liver. It is the leading cause of end-stage liver disease and hepatocellular carcinoma. HIV is known to have a negative impact on the natural disease outcome and immune response of HCV infection, whereas the reverse remains unclear. We evaluated the impact of HCV co-infection on recovery of CD4+ and CD8+ T-cells and liver enzyme levels before and after initiation of highly active antiretroviral therapy (HAART) in HIV/HCV co-infected patients. METHODS: A hospita
