Foodborne pathogens are one of the major cause of food-related diseases and food poisoning. Bacterial biofilms and quorum sensing (QS) mechanism of cell-cell communication have also been found to be associated with several outbreaks of foodborne diseases and are great threat to food safety. Therefore, In the present study, we investigated the activity of three tetrahedrally coordinated copper(I) complexes against quorum sensing and biofilms of foodborne bacteria. All the three complexes demonstrated similar antimicrobial properties against the selected pathogens. Concentration below the MIC i.

Biochemical confirmation of the identity of Burkholderia pseudomallei in Singapore previously relied on the API 20NE panel of tests. After introducing an alternative proprietary biochemical panel, the Microbact 24E (MedVet, Adelaide, Australia), we noted that the API panel identified some presumptive B. pseudomallei isolates as other species. We therefore compared the performance of the API 20NE against the Microbact 24E with 50 distinct clinical isolates of B. pseudomallei, after 24 hours and after five days incubation of primary cultures. The API panel correctly identified 40 isolates. Four

Two Streptomyces spp. were isolated from open ocean waters of the Gulf of Mannar. Of the two isolates, one was identified as Streptomyces coelicolor through conventional and molecular approaches, and it was found to produce a siderophore. Characterization revealed the siderophore to be of trihydroxamate type with hexadentate iron-binding capacity. FTIR analysis indicated the presence of aromatic rings with C-O and C = C stretching, while 1H and 13C NMR together with mass spectrometry confirmed the hydroxamate nature and identified the siderophore as ferrioxamine. A narrow shift in λ max indica

A total of 52 Pasteurella multocida strains of capsular serogroups (A, B and D) were screened for anti-quorum sensing activity against Chromobacterium violaceum. Of which, 12 strains of serogroups A were found to possess anti-quorum sensing activity. Inhibition activity was highest for strain NIVEDIPm9 and lowest for strain NIVEDIPm30 based on zone of pigment inhibition. Further, cell free extract of NIVEDIPm9 strain showed highest anti-biofilm activity in reference E. coli strain and concentration dependent degradation activity of C6-AHL molecule. In whole genome sequence annotation of NIVEDI

Resistant bacteria may kill more people than COVID-19, so the development of new antibacterials is essential, especially against microbial biofilms that are reservoirs of resistant cells. Silver nanoparticles (bioAgNP), biogenically synthesized using Fusarium oxysporum, combined with oregano derivatives, present a strategic antibacterial mechanism and prevent the emergence of resistance against planktonic microorganisms. Antibiofilm activity of four binary combinations was tested against enteroaggregative Escherichia coli (EAEC) and Klebsiella pneumoniae carbapenemase-producing K. pneumoniae (

Biofilms are aggregates of microorganisms anchored to a surface and embedded in a self-produced matrix of extracellular polymeric substances and have been associated with 80% of all bacterial infections in humans. Because bacteria in biofilms are less amenable to antibiotic treatment, biofilms have been associated with developing antibiotic resistance, a problem that urges developing new therapeutic options and approaches. Interfering with quorum-sensing (QS), an important process of cell-to-cell communication by bacteria in biofilms is a promising strategy to inhibit biofilm formation and dev

ETHNOPHARMACOLOGICAL RELEVANCE: Streptococcus pyogenes is a common cause of acute bacterial pharyngitis in children and adults. The global emergence of antibiotic resistance and increasing treatment failures have become serious clinical problems. Quercus infectoria galls have traditionally been used in Asian medicinal traditions as an astringent remedy for oral and pharyngeal conditions, including sore throat and related inflammations.
AIM OF THE STUDY: This study investigated the antibacterial activity and anti-virulence properties of Q. infectoria gall extract against S. pyogenes.

Multidrug resistance has also been accompanied by the prolonged use of antibiotics that makes complications in treatment. Biofilm in pathogenic bacteria is the most serious challenge linked with chronic illnesses and also contributes to virulence and drug resistance. Several bacterial pathogens employ the Quorum-sensing (QS) mechanism to coordinate their collective behaviors like bioluminescence, virulence, and biofilm formation. Therefore, agents that inhibit or interfere with bacterial QS and biofilm formation are emerging as a new class of next-generation antibacterial. Recently, nanopartic

Pseudomonas aeruginosa use small signaling molecules such as acyl homoserine lactones (AHLs), which play an important role in release virulence factors and toxin for further establishment of host infection. Thus, involving with the QS system would provide alternative ways of preventing the pathogenicity. In the present study, totally six medicinal plants (Terminalia bellerica, Celastrus paniculatus, Kingiodendron pinnatum, Schleichera oleosa, Melastoma malabathricum, Garcinia gummi-gutta) were screened for anti-QS activity using biomonitor strain of Chromobacterium violaceum CV12472. Gorgonian corals possess many novel natural products that could potentially mediate coral-bacterial interactions. Since many bacteria use quorum sensing (QS) signals to facilitate colonization of host organisms, regulation of prokaryotic cell-to-cell communication may represent an important bacterial control mechanism. In the present study, we examined extracts of twelve species of Caribbean gorgonian corals, for mechanisms that regulate microbial colonization, such as antibacterial activity and QS regulatory activity. Ethanol extracts of gorgonians collected from Puerto Rico and the Florida K

Listeria monocytogenes, a foodborne pathogen causing listeriosis, poses substantial societal, economic, and public health challenges due to its resistance, persistence, and biofilm formation in the food industry. Exploring subinhibitory concentrations of compounds to target virulence inhibition and increase susceptibility to adverse conditions presents a promising strategy to mitigate its impact of L. monocytogenes and unveils new potential applications. Thus, this study aims to explore the effect of linalool on virulence factors of L. monocytogenes and potential use in the reduction in its to

Gold nanoparticles (Au NPs) are often used to study the physiochemical behavior and distribution of nanomaterials in natural systems because they are assumed to be inert under environmental conditions, even though Au can be oxidized and dissolved by a common environmental compound: cyanide. We used the cyanogenic soil bacterium, Chromobacterium violaceum, to demonstrate that quorum-sensing-regulated cyanide production could lead to a high rate of oxidative dissolution of Au NPs in soil. After 7 days of incubation in a pH 7.0 soil inoculated with C. violaceum, labile Au concentration increased

A bacterium was isolated from the blood and empyema of a cirrhotic patient. The cells were facultatively anaerobic, nonsporulating, gram-negative, seagull shaped or spiral rods. The bacterium grows on sheep blood agar as nonhemolytic, gray colonies 1 mm in diameter after 24 h of incubation at 37 degrees C in ambient air. Growth also occurs on MacConkey agar and at 25 and 42 degrees C but not at 4, 44, and 50 degrees C. The bacterium can grow in 1 or 2% but not 3, 4, or 5% NaCl. No enhancement of growth is observed with 5% CO(2). The organism is aflagellated and nonmotile at both 25 and 37 degr

CONTEXT: Chromobacterium violaceum Bergonzini (Neisseriaceae), a Gram-negative bacterium, secretes a spectacular pigment called violacein. Violacein is a quorum-sensing metabolite and is also an active antimicrobial, anticancer agent. However, its efficiency as a potential drug, alone or in synergy with other active principles, has not being completely exploited. With the advent of different multi-drug resistant strains, it becomes essential to find a new natural product(s) that could be effectively used as a therapeutic agent.
OBJECTIVE: This work focused on the extraction of violacein from

Biofilms are implicated in roughly 80% of chronic and 65% of acute bacterial infections. Therefore, targeting QS-regulated virulence and biofilm presents a potential strategy for the development of novel antimicrobials. In this investigation, AgNPs-capped Guar Gum-co-Polyacrylonitrile hybrid (GPAG) nanocomposites were synthesized biogenically by modifying guar gum (GG) chains chemically with polyacrylonitrile (PAN) chains, resulting in a copolymer matrix with suitable capping sites preventing the aggregation of Ag NPs during the synthesis of GPAG. Chemical and structural properties of the synt

Oral bacterial infections are a great health concern worldwide especially in diabetic patients. Emergence of antimicrobial resistance with reference to biofilms in oral cavity is of great concern. We investigated antibiotics combination with proton pump inhibitors against oral clinical isolates. The strains were identified as Staphylococcus epidermidis and Staphylococcus aureus by the 16S rRNA gene sequencing. In molecular docking, ciprofloxacin, levofloxacin, and omeprazole best fit to active pockets of transcriptional regulators 4BXI and 3QP1. None of the proton pump inhibitors were active a

The catalytic domains of aromatic amino acid hydroxylases (AAAHs) contain a non-heme iron coordinated to a 2-His-1-carboxylate facial triad and two water molecules. Asp139 from Chromobacterium violaceum PAH (cPAH) resides within the second coordination sphere and contributes key hydrogen bonds with three active site waters that mediate its interaction with an oxidized form of the cofactor, 7,8-dihydro-l-biopterin, in crystal structures. To determine the catalytic role of this residue, various point mutants were prepared and characterized. Our isothermal titration calorimetry (ITC) analysis of
