A new microbial gluten-degrading prolyl endopeptidase: Potential application in celiac disease to reduce gluten immunogenic peptides.

Gluten is a complex of proteins present in barley, wheat, rye and several varieties of oats that triggers celiac disease in genetically predisposed subjects. Gluten is notoriously difficult to digest by mammalian proteolytic enzymes and therefore, proline-rich digestion-resistant peptides contain multiple immunogenic epitopes. Prolyl endopeptidases (PEP) hydrolyse internal proline residues on the carboxyl side of peptides and have been proposed for food gluten detoxification and as oral enzyme supplementation for celiacs. The aim of this study was to identify new gluten-degrading microbial enz

Effect of Ichthyophthirius multifiliis infection on host immunity and microbiota shifts of Takifugu fasciatus.

Ichthyophthirius multifiliis (Ich) is a major pathogen responsible for high mortality rates in Takifugu fasciatus aquaculture. Despite its significance, the local and systemic immune responses of pufferfish to Ich infection remain poorly understood. Additionally, the interaction between ectoparasite infection and gut microbiota has not been thoroughly investigated. To address these gaps, we established an Ich infection model in pufferfish through bath administration. RT-PCR confirmed that Ich invades the skin, gill, and fin at 10 days post-infection (dpi) using 5000 theronts per fish. RT-qPCR

Theoretical Analysis of Selectivity Differences in Ketoreductases toward Aldehyde and Ketone Carbonyl Groups.

Lactobacillus kefir alcohol dehydrogenase (LkADH) and ketoreductase from Chryseobacterium sp. CA49 (ChKRED12) exhibit different chemoselectivity and stereoselectivity toward a substrate with both keto and aldehyde carbonyl groups. LkADH selectively reduces the keto carbonyl group while retaining the aldehyde carbonyl group, producing optically pure R-alcohols. In contrast, ChKRED12 selectively reduces the aldehyde group and exhibits low reactivity toward ketone carbonyls. This study investigated the structural basis for these differences and the role of specific residues in the active site. Mo

Effect of Thermostable Enzymes Produced by Psychrotrophic Bacteria in Raw Milk on the Quality of Ultra-High Temperature Sterilized Milk.

Ultra-high temperature sterilized milk (UHT) is a popular dairy product known for its long shelf life and convenience. However, protein gel aging and fat quality defects like creaming and flavor deterioration may arise during storage. These problems are primarily caused by thermostable enzymes produced by psychrotrophic bacteria. In this study, four representative psychrotrophic bacteria strains which can produce thermostable enzymes were selected to contaminate UHT milk artificially. After 11, 11, 13, and 17 weeks of storage, the milk samples, which were contaminated with Pseudomonas fluoresc

Bambara Nut Root-Nodules Bacteria from a Semi-Arid Region of South Africa and Their Plant Growth-Promoting Traits.

Rhizobial nitrogen-fixing bacteria are the main inhabitants of the root nodules of legume plants. Studying the bacterial community of legume nodules is important in understanding plant growth and nutrient requirements. Culture-based technique was used to examine the bacterial community of these underground organs from Vigna subterranea L. Verdc (Bambara nut), an underutilized legume in Africa, for plant growth-promoting traits. In this study, Bambara nuts were planted to trap root-nodule bacteria, and the bacteria were morphologically, biochemically, and molecularly characterized. Five selecte

Field-based evidence for enrichment of antibiotic resistance genes and mobile genetic elements in manure-amended vegetable soils.

The increasing prevalence of antibiotic resistance genes (ARGs) and antibiotic resistant bacteria (ARB) in the soil environment represents a serious threat to public health. In this study, the diversity and abundance of ARGs and mobile genetic elements (MGEs) in different years of manure-amended vegetable soils were investigated. A total of eight genes, including four tetracycline resistance genes: tetW, tetM, tetO and tetT; two sulfonamide resistance genes: sul1 and sul2; and two MGEs: intI1 and intI2; were quantified in ten vegetable soils. The relative abundance of ARGs in soils amended wit

Microbial Diversity of Source and Point-of-Use Water in Rural Haiti - A Pyrosequencing-Based Metagenomic Survey.

Haiti endures the poorest water and sanitation infrastructure in the Western Hemisphere, where waterborne diseases cause significant morbidity and mortality. Most of these diseases are reported to be caused by waterborne pathogens. In this study, we examined the overall bacterial diversity of selected source and point-of-use water from rural areas in Central Plateau, Haiti using pyrosequencing of 16s rRNA genes. Taxonomic composition of water samples revealed an abundance of Firmicutes phyla, followed by Proteobacteria and Bacteroidetes. A total of 38 bacterial families and 60 genera were iden

Occupational exposure to printer toner-emitted nanoparticles at printing facilities influences air and airway microbiomes.

Workplace exposure to printer toner-emitted nanoparticles at commercial printing facilities poses respiratory health risks to workers on the printing floor, however, its impact on environmental and airway microbiomes and how this relates to worker health remains unknown. To investigate this, we prospectively evaluated five printing centres in Singapore, collecting air samples from office areas and printing floors and airway specimens from workers stationed in office or printing floor areas. All specimens were subjected to targeted amplicon sequencing to determine bacteriome and mycobiome profi

"Unmasking the Uncommon": A case series of multi-drug resistant Elizabethkingia meningoseptica causing late-onset sepsis and meningitis in preterm neonates.

Elizabethkingia meningoseptica is an uncommon nosocomial pathogen that causes meningitis, pneumonia, and sepsis in neonates and in immunocompromised individuals. It exhibits resistance to many commonly employed first-line antibiotics used to treat gram-negative pathogens. Herein, we present three cases of late-onset sepsis with multi-drug resistant (MDR) Elizabethkingia meningoseptica in high-risk neonates. Case 1 was a one-day-old preterm low-birth-weight infant who presented with respiratory distress syndrome and septic shock. The patient was intubated and administered empirical broad-spectr

Thallium exposure disturbs gut microbiota composition, metabolic function, and host gut energy metabolism in aged mice.

Gut microbiota regulates the development of environmental exposure-induced diseases. Although previous studies have reported the effect of thallium exposure on the gut microbiota of adult male mice, its effects on the aging gut microbiome, particularly in females, remain largely unexplored. Herein, we showed that thallium exposure for 4 weeks altered fecal microbial β diversity in aged female and male mice, whereas exposure for 2 weeks did not. Combined LEfSe and indicator analyses revealed that in aged female mice, thallium exposure for 2 weeks increased Oscillibacter abundance, while exposur

Helicobacter pylori infection affects the human gastric microbiome, as revealed by metagenomic sequencing.

Helicobacter pylori infection is a prevalent infectious disease, associated with many gastric diseases, including gastritis, gastric ulcer, and gastric cancer. To reveal the characteristics of the gastric microbiome in patients infected with H. pylori, we performed metagenomic shotgun sequencing of stomach swab samples from 96 patients and then conducted metagenomic association analyses between alterations in the gastric microbiome and H. pylori infection status. The overall composition of the gastric microbiota in H. pylori-infected individuals was distinctly different from the negative contr

In vivo fluorescence imaging of exogenous enzyme activity in the gastrointestinal tract.

Exogenous enzymes are administered orally to treat several diseases, such as pancreatic insufficiency and lactose intolerance. Due to the proteinaceous nature of enzymes, they are subject to inactivation and/or digestion in the gastrointestinal (GI) tract. Here we describe a convenient fluorescence-based assay to monitor the activity of therapeutic enzymes in real time in vivo in the GI tract. To establish the proof of principle, the assay was applied to proline-specific endopeptidases (PEPs), a group of enzymes recently proposed as adjuvant therapy for celiac disease (a highly prevalent immun

Structural Characterization of an α-D-glucan from Bellamya purificata and Its Protective Effects on Non-Alcoholic Fatty Liver Disease in Zebrafish.

Non-alcoholic fatty liver disease (NAFLD) is a primary metabolic disorder that threatens adolescent health globally, with no effective therapeutic agents currently available. Bellamya purificata is a traditional Chinese medicine categorized as "medicinal food", and polysaccharides are among its active components. However, its physicochemical structure remains poorly characterized, and no study has evaluated its effects on NAFLD. In this study, a homogeneous neutral polysaccharide, α-D-glucan (Mw = 6412.704 kDa), was isolated from B. purificata. The structure of the polysaccharide was character

Microbial co-reduction of selenate and selenite in zeolite-packed columns in the presence of sulfate and nitrate: effects on removal efficiency and transformation of microbial community structure.

The escalating global concern over the presence of selenium species in aqueous systems, necessitates effective remediation strategies. Selenium, existing as selenite and selenate oxyanions, poses challenges due to its mobility in aqueous systems. Various technologies, particularly biological wastewater treatment systems, leverage microorganisms' unique abilities for waste removal. Challenges in selenium bioremediation, including interference from other anions and reoxidation, can be addressed with appropriate microorganisms. This study explores the responses of zeolite-packed (natural and modi

Microbe associated molecular patterns from rhizosphere bacteria trigger germination and Papaver somniferum metabolism under greenhouse conditions.

Ten PGPR from different backgrounds were assayed on Papaver somniferum var. Madrigal to evaluate their potential as biotic elicitors to increase alkaloid content under the rationale that some microbe associated molecular patterns (MAMPs) are able to trigger plant metabolism. First, the 10 strains and their culture media at two different concentrations were tested for their ability to trigger seed germination. Then, the best three strains were tested for their ability to increase seedling growth and alkaloid levels under greenhouse conditions. Only three strains and their culture media enhanced

Systemic disease protection elicited by plant growth promoting rhizobacteria strains: relationship between metabolic responses, systemic disease protection, and biotic elicitors.

A study of plant defensive systemic responses induced by three plant growth promoting rhizobacteria (PGPR) on Arabidopsis thaliana Col 0 against Pseudomonas syringae pv. tomato DC3000 at the biochemical and transcriptional levels is reported in this paper. All three strains decreased disease severity when applied to A. thaliana prior to pathogen inoculation. At the biochemical level, each of the three strains induced ethylene (ET) when incubated with 1-amino-cyclopropane-1-carboxylic acid, and salicylic acid (SA) production in the plant. Plants treated with each of the three strains were also

Cloning and characterization of blaVIM, a new integron-borne metallo-beta-lactamase gene from a Pseudomonas aeruginosa clinical isolate.

Production of a metallo-beta-lactamase activity was detected in a carbapenem-resistant Pseudomonas aeruginosa clinical isolate (isolate VR-143/97) from an Italian inpatient at the Verona University Hospital (northern Italy). The metallo-beta-lactamase determinant was isolated from a genomic library of VR-143/97, constructed in an Escherichia coli plasmid vector, by screening for clones with reduced susceptibility to imipenem. Sequencing of the cloned gene revealed that it encoded a new class B beta-lactamase that was named VIM-1. At the sequence level VIM-1 was rather divergent from the other
