Cinnamic Acid, Perillic Acid, and Tryptophan Metabolites Differentially Regulate Ion Transport and Serotonin Metabolism and Signaling in the Mouse Ileum In Vitro.

Phytochemicals and tryptophan (Trp) metabolites have been found to modulate gut function and health. However, whether these metabolites modulate gut ion transport and serotonin (5-HT) metabolism and signaling requires further investigation. The aim of this study was to investigate the effects of selected phytochemicals and Trp metabolites on the ion transport and 5-HT metabolism and signaling in the ileum of mice in vitro using the Ussing chamber technique. During the in vitro incubation, vanillylmandelic acid (VMA) reduced (p < 0.05) the short-circuit current, and 100 μM chlorogenic acid (CGA

Oxaziridinium salts as hydrophobic epoxidation reagents: remarkable hydrophobically-directed selectivity in olefin epoxidation.

Selective epoxidation of cinnamates versus crotonate was used to detect hydrophobic binding of the cinnamates in the transition states with hydrophobic oxidizing agents in water solution. With peracids as oxidants, no such effect was seen, in accord with the calculated geometries of epoxidation in which the hydrophobic groups of substrate and oxidant could not stack. However, with oxaziridinium salts carrying fused benzene rings there was significantly high selectivity for the cinnamates in water solution, which could be suppressed with added 2-propanol. The hydrophobically induced selectivity

Cinnamic-Hydroxamic-Acid Derivatives Exhibit Antibiotic, Anti-Biofilm, and Supercoiling Relaxation Properties by Targeting Bacterial Nucleoid-Associated Protein HU.

Finding novel compounds and drug targets is crucial for antibiotic development. The nucleoid-associated protein HU plays a significant role in bacterial DNA metabolism, supercoiling, and biofilm formation, making it a promising new target. In this work, structure-based screening and identified cinnamic-hydroxamic-acid derivatives (CHADs) are conducted as HU inhibitors, with a minimum inhibitory concentration (MIC) of as low as 12 µg mL-1 against a range of pathogenic bacteria. CHADs induce nucleoid deformation, preventing bacterial division and inhibiting growth. They exhibit low toxicity in m

Protective effect of (E)-(2,4-dihydroxy)-α-aminocinnamic acid, a hydroxy cinnamic acid derivative, in an ulcerative colitis model induced by TNBS.

Ulcerative colitis (UC) is a multifactorial disease that causes long-lasting inflammation and ulcers in the digestive tract. UC is the most common form of inflammatory bowel disease (IBD). The current treatment for mild-to-moderate UC involves the use of 5-aminosalicylates (5-ASA), but much of this compound is unabsorbed and metabolized by N-acetylation. Several efforts have since been made to evaluate new molecules from synthetic or natural sources. Recently, it was reported that (E)-(5-chloro-2-hydroxy)-α-aminocinnamic acid (2c) and (E)-(2,4-dihydroxy)-α-aminocinnamic acid (2f) are as good o

Biological evaluation of 1,3-benzodioxole acids points to 3,4-(methylenedioxy) cinnamic acid as a potential larvicide against Aedes aegypti (Diptera: Culicidae).

Aedes aegypti serves as the primary vector for viruses like dengue, Chikungunya, Zika, and yellow fever, posing a significant public health challenge in Brazil. Given the absence of approved vaccines for these diseases, effective mosquito control becomes paramount in preventing outbreaks. However, currently available chemical insecticides face issues related to toxicity and the emergence of resistance, necessitating the exploration of new active compounds. Drawing inspiration from natural products, we identified the 1,3-benzodioxole group as a key pharmacophore associated with insecticidal act

A Dose-Response Modeling Approach Shows That Effects From Mixture Exposure to the Skin Sensitizers Isoeugenol and Cinnamal Are in Line With Dose Addition and Not With Synergism.

Currently, hazard characterization of skin sensitizers is based on data obtained from studies examining single chemicals. Many consumer products, however, contain mixtures of sensitizers that might interact in such a way that the response induced by a substance is higher than predicted in the hazard assessment. To assess interaction of skin sensitizers in a mixture, a dose-response modeling approach is applied. With this approach, it is possible to assess whether or not responses from mixtures of sensitizers can be predicted from the dose-response information obtained from individual chemicals

The Effects of Several Natural Protoberberine Alkaloids and Cinnamic Acid Derivatives Used for Traditional Medicine on the Membrane Boundary Potential and Lipid Packing Stress.

Here we elucidated the effects of natural protoberberine alkaloids (rotundine, berberine, and nitidine) and cinnamic acid derivatives (ethyl-4-methoxycinnamate and osthole) found in Vietnamese medicinal plants, on the boundary potential of lipid bilayers and phase behavior of membrane lipids. Lipid bilayers were composed of neutral phosphatidylcholines (PC) and negatively charged phosphatidylserines (PS). Tested compounds did not produce any noticeable changes in the boundary potential with the exception of osthole, which caused a potential drop by about 30 mV independently of the membrane pho

Computational study and synthesis of multi-template molecularly imprinted polymer for isolating ethyl p-methoxycinnamate and ethyl cinnamate using bulk methods.

In recent years, research into molecularly imprinted polymers (MIPs) has expanded significantly, particularly for separation or isolation of analytes from extracts. MIPs designed for the simultaneous isolation of ethyl p-methoxycinnamate (EPMC) and ethyl cinnamate (EC) from Kaempferia galanga L. extract or multi-template molecularly imprinted polymer (MT-MIP) have been developed. However, the percentage recovery and purity have remained low. In this study, molecular modelling of MT-MIP for EPMC and EC was conducted using the semi-empirical Parametric Method 3 (PM3) to select functional monomer

Design, synthesis and bioactivity evaluation of cinnamic acid derivatives as potential anti-inflammatory agents against LPS-induced acute lung injury.

Acute lung injury (ALI), a common critical disease in clinical practice, has a mortality rate as high as 30-40 %, yet currently, no effective treatment methods are available. Research on ALI indicated that inhibition of overexpressed inflammatory factors might be a potential treatment for ALI. In this study, a series of cinnamic acid derivatives were obtained by introducing diverse aminothiazole fragments into the natural product cinnamic acid. Among these derivatives, compound 22 displayed excellent activity of inhibiting the release of IL-6 in J774A.1 cells. Moreover, it also ameliorated the

Thermodynamics of Formation of Biological Microemulsion (with Cinnamic Alcohol, Aerosol OT, Tween 20, and Water) and Kinetics of Alkaline Fading of Crystal Violet in Them.

The ternary phase diagrams for the formation of biological microemulsions of the combinations CA/AOT/water and CA/Tween 20/water have been presented. The thermodynamics of solution of water in AOT/CA as well as in Tween 20/CA forming w/o microemulsions and solution of oil in Tween 20/water forming o/w microemulsions have been calorimetrically studied. The solution processes are essentially exothermic (with a few exceptions) and have yielded negative entropies, i.e., producing an ordering effect. This has been supported by the measured specific heats of the resulting solution. The pseudo-first-
