Background:Cymbidium goeringii, one of China's traditional and valuable orchids, possesses significant ornamental and economic value. However, it is relatively sensitive to low temperature and other abiotic stresses, which severely restrict its application in landscaping and industrial development. WRKY transcription factors play important roles in plant responses to abiotic stresses, yet related research in C. goeringii remains limited. Methods: In this study, based on transcriptome data of C. goeringii under four different stresses, we identified and cloned the WRKY transcription factor gene

Cold stress poses a significant challenge to the poultry industry, leading to impaired health and economic losses. However, the specific mechanisms by which cold stress induces injury in the chicken spleen, a critical immune organ, remain poorly defined. This study aimed to investigate the protective effects of the natural flavonoid galangin against cold stress-induced splenic injury and to elucidate the underlying mechanisms, with a focus on the NLRP3 inflammasome-mediated pyroptosis pathway. Using an in vivo chicken model, we demonstrated that acute cold exposure (10±1°C for 48 h) significan

Despite a high sucrose accumulation in its taproot vacuoles, sugar beet (Beta vulgaris subsp. vulgaris) is sensitive to freezing. Earlier, a taproot-specific accumulation of raffinose was shown to have beneficial effects on the freezing tolerance of the plant. However, synthesis of raffinose and other oligosaccharides of the raffinose family depends on the availability of myo-inositol. Since inositol and inositol-metabolising enzymes reside in different organelles, functional inositol metabolism and raffinose synthesis depend on inositol transporters. We identified five homologues of putative

Cold stress is a widely distributed abiotic stress that severely limits the yield and quality of tea plants. Tryptophan (Trp) and downstream indole compounds play an important role in plant growth and stress response. However, beyond its involvement in indole compounds synthesis, the other physiological functions of Trp in tea plants remain unknown. In this study, anthranilate synthetase (ASA2) was a positive regulator to enhance cold stress tolerance in Camellia sinensis. The expression of CsASA2 was strongly induced by cold stress. Suppression of CsASA2 expression in C. sinensis reduced the

Recent reports indicate that Heat Shock Proteins (HSPs) are induced in mammalian tissues as part of a homeostatic response to environmental stressors. Administration of sympathomimetic drugs and neuroendocrine stress hormones has been shown to evoke an HSP response in unstressed animals indicating that cell signaling events exists that couple specific neurotransmitter/hormone-receptor interactions with HSP expression in mammalian tissues. Herein, we demonstrate that exposure of rats to a cold ambient temperature (6 degrees C) results in increased expression of constitutive and inducible member

Cold stress severely restricts pepper (Capsicum annuum L.) growth and productivity by disrupting cellular homeostasis and inducing oxidative damage. Although HD-ZIP transcription factors play important roles in plant abiotic stress responses, the molecular mechanisms regulating their function during cold stress remain largely unknown. Here, we characterized the pepper HD-ZIP transcription factor CaHD-ZIP and demonstrated that it is a nuclear-localized, cold-responsive protein that positively contributes to cold stress tolerance. To further investigate the regulatory mechanism underlying CaHD-Z

Sigma factors (SIGs) are nuclear-encoded regulators of chloroplast gene transcription. We conducted a genome-wide analysis in Brassica napus, identifying 23 SIG genes that were phylogenetically classified into six distinct subfamilies. Characterization of gene structure, conserved motifs, and chromosomal locations indicated family expansion primarily through segmental duplication under purifying selection. Promoter analysis identified cold-responsive elements enriched in BnSIG5A. Expression profiling showed that BnSIG5 subfamily members, particularly BnSIG5A, are strongly induced by cold stres

Grapevine (Vitis vinifera) is a high-value fruit crop frequently challenged by cold stress. Here, we identify the E3 ubiquitin ligase Plant U-box 26 (VvPUB26) as a pivotal regulator of the grapevine cold signaling pathway. We demonstrated that the VvPUB26-VvPIF4-VvCBF3 module fine-tunes cold tolerance by integrating light and temperature signals. Overexpression of VvPUB26 (VvPUB26-OE) significantly enhances cold tolerance in transgenic plants, whereas RNA interference (VvPUB26-RNAi) lines exhibit increased cold sensitivity. Biochemical analysis reveals that VvPUB26 modulates the CBF signaling

Combined saline-alkali and cold stresses severely constrain plant growth at middle and high latitudes. Plants have evolved cross-adaptation mechanisms wherein exposure to one stress enhances resistance to another. However, the specific mechanisms driving cross-adaptation between saline-alkali and cold stresses in alfalfa (Medicago sativa L.) remain to be elucidated. Here, we performed integrated transcriptomic and metabolomic analyses coupled with functional validation to elucidate these mechanisms. We found that saline-alkali pretreatment significantly enhanced the tolerance of alfalfa to sub

Cyclophilins are ubiquitous proteins that are essential for cis-trans isomerisation of peptide bonds preceding the proline residue. Apart from catalysing the cis-trans isomerization, these proteins are also implicated in diverse cellular functions. We investigated the role of a cyclophilin gene PoxCYP62-2, isolated from a halotolerant strain of Penicillium oxalicum, in abiotic stress tolerance of plants. Ectopic expression of PoxCYP62-2 gene in transgenic Arabidopsis thaliana plants conferred tolerance to cold stress. The transgenic plants exhibited higher rosette fresh and dry mass, lower MDA

NADPH oxidases (NOXs) are crucial enzymes for reactive oxygen species (ROS) generation in plants and play vital roles in growth, development, and stress responses. To elucidate the sequence characteristics of the NOX gene family and its low-temperature response patterns in melon (Cucumis melo L.), this study conducted genome-wide identification and expression profiling of NOX family members using bioinformatics analysis, RNA-seq transcriptome sequencing, and real-time quantitative PCR (RT-qPCR). The results revealed that eight NOX members were identified in the melon genome, distributed across

BACKGROUND: Early-life environmental stressors can program immune development, but how the frequency of cold stress shapes thymic programming remains unknown.
METHODS: Developmentally matched (day 4, poikilothermic, yolk sac or milk dependent) zebrafish larvae and neonatal mice were exposed to two or four cycles of cold stress (termed 2× or 4× stress). Thymic development (zebrafish) and thymic T cell subsets (mice) were assessed, followed by influenza A virus (IAV) challenge in mice. Mechanisms were explored by RT-qPCR, western/dot blotting, and irf4/5 transgenic or irf5 knockout zebrafish.

Campylobacter jejuni is a foodborne pathogen with limited ability to thrive at low temperatures. This study investigated the survival and proteomic responses of C. jejuni NCTC 11168 during prolonged storage at 4 °C, with specific focus on its behavior in chicken juice. An initial rapid decrease of 80-90 % in viable cell counts across various broth media was observed within the first 10 days; however, chicken juice provided the most conducive environment for slowing the decline in cell viability compared to Brain Heart Infusion Broth (BHI) and Mueller Hinton (MH) broths. Cold stress can affect ventricular repolarization (VR), which may serve as a potential trigger for malignant arrhythmias. Compared to conventional electrocardiogram (ECG) devices, wearable technology is more suitable for out-of-hospital monitoring due to their portability. However, outdoor cold environments can limit their application. We aimed to develop a wearable ECG capable of functioning in a cold environment. Moreover, to explore the impact of cold stress on VR parameters in a real-world cold environment and identify associated cardiovascular risks. A self-developed handheld 12-lead wear

Bacteria use post-transcriptional regulatory mechanisms to rapidly adjust gene expression during environmental change. In the gut-associated genus Bacteroides, these mechanisms remain poorly defined as these organisms lack canonical RNA chaperones like Hfq and CsrA that coordinate post-transcriptional stress responses in many well-studied model bacteria. Most Bacteroides possess conserved RNA recognition motif-1 (RRM-1) domain-containing RNA-binding proteins (more common in eukaryotes than bacteria) that have been proposed to act as global RNA chaperones. Here, we show that these RNA binding p

Acclimation can alleviate the damage caused by adverse environmental factors. To investigate the effects of cold stimulation on immunity in tracheal of broilers, 360 one-day-old chicks were raised at normal temperatures during 1-7 days. From 8 day, G1 (control) continued to be raised at normal temperatures, whereas G2 and G3 (treatment groups) were cold-stimulated at 3°C and 12°C below the temperature of G1, respectively. At 42 day, all the groups were subjected to a 24-hr acute cold stress, designated as S1, S2, and S3. Tracheal tissues were collected to detect gene levels of immunoglobulins,

PURPOSE: Cold seawater immersion aggravates hemorrhagic shock-induced homeostasis imbalance and organ dysfunction, leading to increased mortality. Previous studies have shown that treatments targeting oxidative stress and mitochondrial dysfunction have limited efficacy for cold seawater immersion combined with hemorrhagic shock (SIHS). Thus, the mechanisms responsible for SIHS need further investigation.
METHODS AND RESULTS: Data from the hemorrhagic shock transcriptome and cold seawater immersion targets used for bioinformatics analysis revealed the involvement of endoplasmic reticulum stres
