Groundwork for phage therapy: multi-faceted comparative analysis of lytic bacteriophages infecting uropathogenic Escherichia coli.

Phage therapy is a promising alternative for treating urinary tract infections (UTIs), particularly given the rapid emergence of antimicrobial resistance among uropathogenic Escherichia coli (UPEC). In this study, we review 98 reported lytic phages infecting UPEC, summarizing their morphology, host range, isolation sources, infection efficiency, progeny yield, adsorption rate, stability across pH and temperature ranges, biofilm-disrupting capacity, and other relevant characteristics. In addition, we conducted detailed bioinformatic and phylogenetic analyses of phage genomic sequences. This mul

Multisite binding of bacteriophages on lipopolysaccharides in Escherichia coli O157:H7 and the adaptive costs of phage resistance.

Escherichia coli (E. coli) O157:H7 is a major foodborne pathogen and a critical global food safety concern. Bacteriophage (phage) therapy offers a promising, highly specific biological alternative to antibiotic treatment. However, the clinical application of phage therapy is frequently limited by the rapid emergence of bacterial resistance. In this study, we examined the interaction between E. coli O157:H7 strain EDL933 and two T4-like phages, assessing the adaptive costs incurred by host strains in developing phage resistance. Our results indicate that phage PSD2001 utilizes capsular polysacc

UV disinfection of RBC-treated light greywater effluent: kinetics, survival and regrowth of selected microorganisms.

The microbial quality of raw greywater was found to be much better than that of municipal wastewater, with 1.6 x 10(7)cfu ml(-1) heterotrophic plate count (HPC), and 3.8 x 10(4), 9.9 x 10(3), 3.3 x 10(3) and 4.6 x 10(0)cfu 100 ml(-1) faecal coliforms (FC), Staphylococcus aureus sp., Pseudomonas aeruginosa sp. and Clostridium perfringes sp., respectively. Further, three viral indicators monitored (somatic phage, host: Escherichia coli CN(13) and F-RNA phages, hosts: E. coli F+(amp), E. coli K12) were not present in raw greywater. The greywater was treated by an RBC followed by sedimentation. Th

Applications of DNA probes for Vero cytotoxin-producing Escherichia coli.

Vero cytotoxin-producing Escherichia coli (VTEC) are now recognized as important aetiological agents in human disease. The symptoms of VTEC infection range from mild non-bloody diarrhoea to severe conditions such as haemorrhagic colitis (HC) and haemolytic uraemic syndrome (HUS). Two types of Vero cytotoxin (VT), VT1 and VT2, have been identified. The genes controlling production of VT1 and VT2 are phageencoded in several E. coli strains and DNA probes have been developed from cloned genes derived from these VT phages. Recently, synthetic oligonucleotide probes for VTEC have also been prepared

Second-site mutations in capR (lon) strains of Escherichia coli K-12 that prevent radiation sensitivity and allow bacteriophage lambda to lysogenize.

capR (lon) mutants of Escherichia coli K-12 are mucoid and sensitive to ultraviolet (UV) and X-ray radiation as well as to nitrofurantoin. The mutants form filaments after exposure to these agents. capR mutants are also conditionally lethal since they die when plated on complex medium even without UV treatment; this phenomenon is designated "complex medium-induced killing". Furthermore, capR mutants are poorly lysogenized by bacteriophage lambda. Second-site revertants were isolated by plating on media containing nitrofurantoin. All 17 of the independent revertants studied were still mucoid bu

R17 RNA replicase. V. Rifamycin sensitivity of R17 RNA synthesis in vivo.

The effect of rifamycin on R14 phage growth in vivo was examined using rifamycin sensitive and resistant strains of E. coli as host cells. The following conclusions were obtained: (1) There is a time lag between the addition of rifamycin to the culture and the initiation of inhibitory action by the drug. At 25 mug rifamycin per ml, this time lag is approximately 30 min. At 50 mug per ml, it takes 10 min to exert 90% inhibition of RNA synthesis in both infected and non-infected cultures. (2) The rifamycin sensitive stage for R17 growth is the first 20 min of its infectious cycle during which ti

The decisive period in the primary infection of muscle by Escherichia coli.

Escherichia coli superinfected with a non-replicating phage was used to study the course of infection in the adductor muscle of mice in terms of the content of viable bacilli and the in vivo multiplication rate. The infection was characterized by a steady decrease of bacterial numbers to 5% of the inoculum within 4 hours without any compensating multiplication. Within the next 3 hours there was slight multiplication (2 generation or less) followed by a slow decrease of numbers to nil in 72 hours. In terms of the viable count, the infection was temporarily enhanced between three- and eight-fold

"Navigating Healthy Waters": monitoring ship wastewater as a key defense against infectious diseases-a pilot study on a Mediterranean seaport.

Wastewater-based surveillance represents a non-invasive approach to monitor pathogen circulation, but data on cruise ships are scarce, although this enclosed environment is prone to infectious diseases spreading. We conducted a pilot study in a Mediterranean seaport (Italy) between July and October 2024, combining wastewater monitoring with onboard clinical surveillance. Twenty wastewater samples (10 untreated and 10 treated) were collected from 10 cruise ships. Viral detection targeted 10 human respiratory and gastrointestinal viruses. Metagenomic analysis of primary colorectal carcinomas and their metastases identifies potential microbial risk factors.

The paucity of microbiome studies at intestinal tissues has contributed to a yet limited understanding of potential viral and bacterial cofactors of colorectal cancer (CRC) carcinogenesis or progression. We analysed whole-genome sequences of CRC primary tumours, their corresponding metastases and matched normal tissue for sequences of viral, phage and bacterial species. Bacteriome analysis showed Fusobacterium nucleatum, Streptococcus sanguinis, F. Hwasookii, Anaerococcus mediterraneensis and further species enriched in primary CRCs. The primary CRC of one patient was enriched for F. alocis, S

Low voltage iron electrocoagulation as a tertiary treatment of municipal wastewater: removal of enteric pathogen indicators and antibiotic-resistant bacteria.

In this paper we analyse the feasibility of low voltage iron electrocoagulation as a means of municipal secondary effluent treatment with a focus on removal of microbial indicators, Antibiotic Resistant Bacteria (ARB) and nutrients. A laboratory scale batch unit equipped with iron electrodes was used on synthetic and real secondary effluent from a municipal wastewater treatment plant. Synthetic secondary effluent was separately assayed with spiked Escherichia coli WR1 and with bacteriophage ΦX174, while real effluent samples were screened before and after treatment for E. coli, Extended Spectr

TaqMan 5'-nuclease human immunodeficiency virus type 1 PCR assay with phage-packaged competitive internal control for high-throughput blood donor screening.

Screening of blood donors for human immunodeficiency virus type 1 (HIV-1) infection by PCR permits the earlier diagnosis of HIV-1 infection compared with that by serologic assays. We have established a high-throughput reverse transcription (RT)-PCR assay based on 5'-nuclease PCR. By in-tube detection of HIV-1 RNA with a fluorogenic probe, the 5'-nuclease PCR technology (TaqMan PCR) eliminates the risk of carryover contamination, a major problem in PCR testing. We outline the development and evaluation of the PCR assay from a technical point of view. A one-step RT-PCR that targets the gag genes

Variable fitness effects of bacteriophage resistance mutations in Escherichia coli: implications for phage therapy.

UNLABELLED: Bacteria exposed to bactericidal treatment, such as antibiotics or bacteriophages (phages), often develop resistance. While phage therapy is proposed as a solution to the antibiotic resistance crisis, the bacterial resistance emerging during phage therapy remains poorly characterized. In this study, we examined a large population of phage-resistant extra-intestinal pathogenic Escherichia coli 536 clones that emerged from both in vitro (non-limited liquid medium) and in vivo (murine pneumonia) conditions. Genome sequencing uncovered a convergent mutational pattern in phage resistanc

Expression of diagnostic 31/32 kilodalton proteins of Schistosoma mansoni as fusions with bacteriophage MS2 polymerase.

An expression plasmid pEx34b was used to synthesise parts of the 31/32 kDa Schistosoma mansoni antigens as fusions with the amino terminus of the phage MS2 polymerase. Purified MS2-schistosome fusion proteins reacted specifically in an enzyme-linked immunosorbent assay with sera from S. mansoni-infected patients. The observation that the majority of human sera tested recognised schistosome-specific epitopes, but not the MS2 polymerase fragment, suggests that the fusion proteins are useful for the immunodiagnosis of schistosomiasis and might be incorporated in a serological test system based on

[Study of spatial organization of the RNA of phage MSZ using nucleases specific for the secondary structure].

The limited hydrolisis of bacteriophage MS2 RNA by nuclease S1 and ds-specific snake venom RNase was studied in a wide range of ionic strength, at different pH, after heating and (slow and fast) cooling and at various enzyme-substrate relations. It was shown that the RNA has exposed hydrolisis sites for both nucleases. The localizations of these sites are very specific and are not altered in all conditions studied. The hydrolisis rate was changed in some conditions, at that the fragments patterns in denaturing electrophoresis did not move. It was supposed that the RNA has strongly predetermine

Retraction of F pili.

The disappearance of F pili on Escherichia coli cells in the presence of 10(-2) M NaCN was studied by electron microscopy and serum-blocking power. The pili which disappeared from the cell did not appear as free pili in the culture medium, suggesting that the pili had retracted into the cell. New pili were produced at a normal rate approximately 3 min after NaCN was removed. The adsorption of either F pili antibody or R17 bacteriophage to the sides of pili and temperatures below 24 C prevented retraction. The disappearance of pili was accompanied by a loss in the ability of cells to adsorb R17

Insights into Genome Ejection by a Therapeutic phiKMV-like Bacteriophage.

Ar-KM is a phiKMV-like therapeutic bacteriophage used in clinical candidate phage therapy cocktails to treat lung infections caused by P. aeruginosa. Here, we present an integrative structural atlas of Ar-KM proteins using cryo-EM, proteomics, and bioinformatics. From a single purified Ar-KM preparation, we identified three distinct populations: mature DNA-filled virions, open-nozzle particles with ejection proteins extending from the tail, and closed-nozzle empty particles. Near-atomic-resolution reconstructions of all three states enabled us to build atomic models for eleven structural prote

Site-specific oligonucleotide-directed mutagenesis using T4 DNA polymerase.

A simple and efficient mutagenesis procedure is described which uses both the 3'----5' exonuclease and 5'----3' polymerase activities of T4 DNA polymerase. Different types of mutation-deletion, insertion, and substitution-can be introduced into the DNA in a single reaction. The technique uses recombinant M13 single-stranded DNA and two complementary DNA oligonucleotides to target and control the extent of deletions catalyzed by T4 DNA polymerase. The second oligonucleotide not only directs ligation, but also serves as a template for insertion or substitution of nucleotides by T4 polymerase. Mu

Modified laminar flow biological safety cabinet.

Tests are reported on a modified laminar flow biological safety cabinet in which the return air plenum that conducts air from the work area to the high efficiency particulate air filters is under negative pressure. Freon gas released inside the cabinet could not be detected outside by a freon gas detection method capable of detecting 10(-6) cc/s. When T3 bacteriophage was aerosolized 5 cm outside the front opening in 11 tests, no phage could be detected inside the cabinet with the motor-filter unit in operation. Characterization of a novel phage vB_EcoP_P64441 and its potential role in controlling uropathogenic Escherichia coli (UPEC) and biofilms formation.

Urinary tract infections are one of the most common infectious diseases worldwide, with uropathogenic Escherichia coli (UPEC) being the leading cause. Phage therapy has emerged as a promising alternative due to the increasing prevalence of multidrug resistance UPEC strains. In this study, we isolated and characterized a novel phage, vB_EcoP_P64441, which lysed UPEC, including serogroup O75. The phage can survive from 4 to 50 °C and a pH range of 3-11. The optimal MOI was 1, latent period was 4 min, and burst size was 10 PFU/cell. Whole genome sequencing revealed that it consisted of 42,960 bas

Characterization and therapeutic potential of phage vB_Eco_ZCEC08 against multidrug-resistant uropathogenic Escherichia coli.

BACKGROUND: Urinary tract infections (UTIs) caused by antibiotic-resistant bacteria have become a significant public health concern. The increasing ineffectiveness of antibiotics has led to a renewed focus on investigating other strategies, such as bacteriophages, to target specific pathogenic bacteria and prevent future resistance. RESULTS: This study reports the isolation and characterization of bacteriophage vB_Eco_ZCEC08 targeting uropathogenic Escherichia coli (UPEC). Phage vB_Eco_ZCEC08 is morphologically a non-contractile tailed phage that exhibits strong lytic activity against UPEC wi
