Revisiting the recombinant history of HIV-1 group M with dynamic network community detection.

The prevailing abundance of full-length HIV type 1 (HIV-1) genome sequences provides an opportunity to revisit the standard model of HIV-1 group M (HIV-1/M) diversity that clusters genomes into largely nonrecombinant subtypes, which is not consistent with recent evidence of deep recombinant histories for simian immunodeficiency virus (SIV) and other HIV-1 groups. Here we develop an unsupervised nonparametric clustering approach, which does not rely on predefined nonrecombinant genomes, by adapting a community detection method developed for dynamic social network analysis. We show that this met

Memory Suppression Ability can be Robustly Predicted by the Internetwork Communication of Frontoparietal Control Network.

Memory suppression (MS) is essential for mental well-being. However, no studies have explored how intrinsic resting-state functional connectivity (rs-FC) predicts this ability. Here, we adopted the connectome-based predictive modeling (CPM) based on the resting-state fMRI data to investigate whether and how rs-FC profiles in predefined brain networks (the frontoparietal control networks or FPCN) can predict MS in healthy individuals with 497 participants. The MS ability was assessed by MS-induced forgetting during the think/no-think paradigm. The results showed that FPCN network was especially

Autophagic degradation of CCN2 (cellular communication network factor 2) causes cardiotoxicity of sunitinib.

Excessive macroautophagy/autophagy is one of the causes of cardiomyocyte death induced by cardiovascular diseases or cancer therapy, yet the underlying mechanism remains unknown. We and other groups previously reported that autophagy might contribute to cardiomyocyte death caused by sunitinib, a tumor angiogenesis inhibitor that is widely used in clinic, which may help to understand the mechanism of autophagy-induced cardiomyocyte death. Here, we found that sunitinib-induced autophagy leads to apoptosis of cardiomyocyte and cardiac dysfunction as the cardiomyocyte-specific Atg7-/+ heterozygous

Optimization: Molecular Communication Networks for Viral Disease Analysis Using Deep Leaning Autoencoder.

Developing new treatments for emerging infectious diseases in infectious and noninfectious diseases has attracted a particular attention. The emergence of viral diseases is expected to accelerate; these data indicate the need for a proactive approach to develop widely active family specific and cross family therapies for future disease outbreaks. Viral disease such as pneumonia, severe acute respiratory syndrome type 2, HIV infection, and Hepatitis-C virus can cause directly and indirectly cardiovascular disease (CVD). Emphasis should be placed not only on the development of broad-spectrum mol

Genetic Gastric Cancer Susceptibility in the International Clinical Cancer Genomics Community Research Network.

Few susceptibility genes for gastric cancer have been identified. We sought to identify germline susceptibility genes from participants with gastric cancer from an international hereditary cancer research network. Adults with gastric cancer of any histology, and with a germline DNA sample (n = 51), were retrospectively selected. For those without previously identified germline mutations (n = 43), sequencing was performed for 706 candidate genes. Twenty pathogenic or likely pathogenic variants were identified among 18 participants. Eight of the 18 participants had previous positive clinical tes

The Childhood Solid Tumor Network: A new resource for the developmental biology and oncology research communities.

Significant advances have been made over the past 25 years in our understanding of the most common adult solid tumors such as breast, colon, lung and prostate cancer. Much less is known about childhood solid tumors because they are rare and because they originate in developing organs during fetal development, childhood and adolescence. It can be very difficult to study the cellular origins of pediatric solid tumors in developing organs characterized by rapid proliferative expansion, growth factor signaling, developmental angiogenesis, programmed cell death, tissue reorganization and cell migra

Spontaneous coronary artery dissection: a disease-specific, social networking community-initiated study.

OBJECTIVE: To develop and assess the feasibility of a novel method for identification, recruitment, and retrospective and prospective evaluation of patients with rare conditions. PATIENTS AND METHODS: This pilot study is a novel example of "patient-initiated research." After being approached by several members of an international disease-specific support group on a social networking site, we used it to identify patients who had been diagnosed as having at least 1 episode of spontaneous coronary artery dissection and recruited them to participate in a clinical investigation of their condition.

The North Country on the Job Network: a unique role for occupational health nurses in a community coalition.

1. Through a community based program, nurses were hired by a rural medical center to facilitate care for injured workers in the community. This placed the nurses outside the industrial and insurance arenas. 2. Rapid access to care and expedited return to work were made possible largely through improved communication, facilitated by the nurse case coordinators, among all involved parties (i.e., workers, employers, health care providers, insurance carriers). 3. The program provides access to occupational health nurses, or case managers, to a large number of rural workers--many of whom work for s

Comparison of networks of loneliness, depressive symptoms, and anxiety symptoms in at-risk community-dwelling older adults before and during COVID-19.

Network analysis provides an innovative approach to examining symptom-to-symptom interactions in mental health, and adverse external conditions may change the network structures. This study compared the networks of common risk factors and mental health problems (loneliness, depressive symptoms, and anxiety symptoms) in community-dwelling older people before and during COVID-19. Older adults (aged ≥ 60) at risk for depression were recruited through non-governmental organizations. Loneliness, depressive symptoms and anxiety symptoms were measured using the three-item Loneliness Scale (UCLA-3), n

Unique microbial diversity, community composition, and networks among Pacific Islander endocervical and vaginal microbiomes with and without Chlamydia trachomatis infection in Fiji.

Chlamydia trachomatis (Ct) is the most common sexually transmitted bacterium globally. Endocervical and vaginal microbiome interactions are rarely examined within the context of Ct or among vulnerable populations. We evaluated 258 vaginal and 92 paired endocervical samples from Fijian women using metagenomic shotgun sequencing. Over 37% of the microbiomes could not be classified into sub-community state types (subCSTs). We, therefore, developed subCSTs IV-D0, IV-D1, IV-D2, and IV-E-dominated primarily by Gardnerella vaginalis-to improve classification. Among paired microbiomes, the endocervix

Women's social networks and use of facility delivery services for uncomplicated births in North West Ethiopia: a community-based case-control study.

BACKGROUND: High maternal mortality has remained an unmet public health challenge in the developing world. Maternal mortality in Ethiopia is among the highest in the world. Since most maternal deaths occur during labor, delivery, and the immediate postpartum period, facility delivery with skilled birth attendants is recommended to reduce maternal mortality. Nonetheless, the majority of women in Ethiopia give birth at home. Individual attributes and availability and accessibility of services deter service utilization. The role of social networks that may facilitate or constrain service use is n

Community structure analysis of transcriptional networks reveals distinct molecular pathways for early- and late-onset temporal lobe epilepsy with childhood febrile seizures.

Age at epilepsy onset has a broad impact on brain plasticity and epilepsy pathomechanisms. Prolonged febrile seizures in early childhood (FS) constitute an initial precipitating insult (IPI) commonly associated with mesial temporal lobe epilepsy (MTLE). FS-MTLE patients may have early disease onset, i.e. just after the IPI, in early childhood, or late-onset, ranging from mid-adolescence to early adult life. The mechanisms governing early (E) or late (L) disease onset are largely unknown. In order to unveil the molecular pathways underlying E and L subtypes of FS-MTLE we investigated global gen

Mitochondrial Dysfunction as a Central Hub of Aging: From Molecular Cascades to Inter-Organ Communication Networks.

: Background: Mitochondrial dysfunction serves as a fundamental driver of the aging process, precipitating progressive functional decline through complex molecular cascades. Summary: This review elucidates the core mechanisms underpinning this dysfunction, including reactive oxygen species (ROS)-induced redox imbalance, mitochondrial DNA (mtDNA) damage accumulation, impaired mitophagy, and metabolic reprogramming. Crucially, we examine how mitochondria act as signaling hubs for inter-organ crosstalk. Through the secretion of mitokines (e.g., FGF21, GDF15) and the release of damage-associated m

Exosomal microRNAs in colorectal cancer communication networks: implications for metastasis, therapy resistance, and precision medicine.

Exosome-derived microRNAs (miRNAs) have recently been recognised as important players in the process of intercellular communication in CRC, thus allowing tumour cells to affect not only each other but also the whole milieu of interactions occurring in the tumour microenvironment and metastatic niches. The growing body of evidence points to selective packaging of exosomal miRNAs via controlled biogenesis mechanisms and modulation by oncogenic signalling and microenvironmental stress. After being delivered to target cells, these miRNAs affect interrelated signalling pathways involved in the epit

Enhanced Security of Bidirectional Communication in IoT-Driven Utility Networks Using Sertainty UXP and LoRaWAN.

LoRaWAN holds immense potential in smart applications for its low-power, long-range communication capabilities and in-built AES-128 encryption for end-to-end security. However, prior research has identified critical security vulnerabilities, most notably its use of AES-128 encryption in ECB mode, which lacks semantic security. Sertainty UXP (Unbreakable Exchange Protocol) technology enhances AES by embedding intelligence directly into the data. Sertainty Corporation's UXP encryption employs AES-256-GCM, which offers authenticated encryption with integrated access control and policy enforcement

Bacterial and fungal communities and network dynamics in Otitis media patients upon 1,8-Cineol treatment.

The present study investigated the bacterial and fungal communities and cross-domain network interactions in patients with chronic Otitis media in response to the natural monoterpene medicinal 1,8-Cineol as a promising treatment option. While the bacterial community showed modest changes with preserved core genera, the fungal community underwent dramatic diversification and network expansion. This suggests that treatment may have disrupted competitive exclusion mechanisms in the latter, allowing for increased diversity and interactions, despite no significant changes in traditional diversity m
