Development of fast and sensitive protocols for the detection of viral pathogens using a small portable convection PCR platform.

One of the most crucial steps for preventing viral pandemics is the early detection of the causative virus on site. Various molecular and immunological approaches have been developed for virus detection. In this study, we investigated the utility of the recently introduced convection polymerase chain reaction (cPCR) platform for the rapid and sensitive detection of various animal viruses in the field, including the foot-and-mouth disease virus (FMDV) and avian influenza viruses (AIVs). Primer sets were designed to simultaneously detect two highly conserved regions of the FMDV, including the 5'

Sample convection in liquid-state NMR: why it is always with us, and what we can do about it.

Many NMR experiments on liquids suffer if the sample convects. This is particularly true for applications, such as the measurement of diffusion, that rely on spatial labelling of spins. It is widely assumed that, in most well-conducted experiments with stable temperature regulation, samples do not convect. Unfortunately this is not the case. It is shown here that typical NMR samples show measurable convective flow for all but a very narrow range of temperatures; convection is seen both above and below this range, which can be as small as a degree or so for a mobile solvent such as chloroform.

Convection-enhanced delivery improves distribution and efficacy of tumor-selective retroviral replicating vectors in a rodent brain tumor model.

In the present study, we compared the therapeutic effect of tumor-selective retroviral replicating vectors (RRV) expressing the yeast cytosine deaminase (CD) delivered by convection-enhanced delivery (CED) or simple injection, followed by systemic administration of the pro-drug, 5-fluorocytosine (5-FC). Treatment with RRV-CD and systemic 5-FC significantly increased survival in rodent U87MG glioma model in comparison with controls (P<0.01). Interestingly, CED of RRV-CD followed by 5-FC further enhanced survival in this animal model in comparison with intra-tumoral injection of RRV-CD, followed

Synergistic combination of convective self-assembly and hollow core fiber for sensitive SERS detection of glucose molecules.

This study demonstrates a highly sensitive surface-enhanced Raman scattering (SERS) platform developed by integrating convective self-assembly (CSA) with hollow core fibers (HCFs). Traditional solid-core-based fiber-optic SERS probes, such as multi-mode fibers (MMFs), often suffer from limited light-matter interaction area and poor hot-spot generation. To overcome these limitations, we utilized the CSA to deposit plasmonic nanoparticles onto the long interior walls of the HCF. This architecture confines both the excitation laser and the target analyte within the micro-capillary core, dramatica

Non-invasive convective head cooling during stroke thrombectomy: A prospective multi-center feasibility trial.

INTRODUCTION: Non-invasive convective head cooling is a promising putative neuroprotective therapy for ischemic stroke patients as it may portably, non-invasively, and selectively cool the ischemic penumbra. We aimed to investigate the feasibility of utilizing non-invasive convective head cooling in ischemic stroke patients before and during endovascular thrombectomy (EVT). PATIENTS AND METHODS: We conducted a multi-center, prospective, non-randomized, open-label trial at two comprehensive stroke centers in ischemic stroke patients where EVT was planned. Patients were assessed for eligibility

Improving lightning representation in global model through convective cloud based Price and Rind 1992 scheme.

Accurate representation of lightning activity in coarse-resolution medium-range weather forecasting models remains a major challenge due to the fine scale nature of electrification processes. In the National Centre for Medium-Range Weather Forecasting Global Unified Model (NCUM-G), lightning flash rates are computed following the widely adopted Price and Rind (1992) (PR92) formulation. However, the current parameterization relies on storm detection criterion solely a cloud-thickness threshold, resulting in a large number of false alarms over India. To address the limitations of the current par

A Climatology of Extreme Convective Storms in Tropical and Subtropical East Asia and Their Ingredients for Heavy Rainfall as Seen by TRMM.

Heavy rainfall is a challenge to forecast due to the variety of rainfall intensities and durations across a wide spectrum of high-impact storm types. In this study, we analyze extreme storms in Tropical and Subtropical East Asia, a moisture-rich environment with complex terrain and oceanic regions. The Tropical Rainfall Measuring Mission's Precipitation Radar is utilized to characterize the frequency and rainfall intensity of four extreme storm types. Extreme storms producing heavy precipitation are categorized into four types: deep convective cores (DCCs), deepwide convective cores (DWCCs), w

Performance and application analysis of ANFIS artificial intelligence for pressure prediction of nanofluid convective flow in a heated pipe.

Heat transfer augmentation of the nanofluids is still an attractive concept for researchers due to rising demands for designing efficient heat transfer fluids. However, the pressure loss arisen from the suspension of nanoparticles in liquid is known as a drawback for developing such novel fluids. Therefore, prediction of the nanofluid pressure, especially in internal flows, has been focused on studies. Computational fluid dynamics (CFD) is a commonly used approach for such a prediction of fluid flow. The CFD tools are perfect and precise in prediction of the fluid flow parameters. But they mig

Convective burn from use of hairdryer for heel warming prior to the heel prick test - a case report.

BACKGROUND: Blood sampling through heel lancing is the most common invasive painful procedure performed on newborn infants. CASE PRESENTATION: We report the case of a five day old infant who sustained burns to the left foot and leg after the mother's hairdryer was used by the midwife to warm the baby's heel prior to capillary blood sampling (CBS) with an automated device. CONCLUSION: Heel warming is not recommended for routine CBS although it is often practiced. If pre-warming is to be practiced, standardised devices should be used rather than improvised techniques. This will reduce the risk

Therapeutic hypothermia with rapid thin liquid convection is safe and feasible in acute ischemic stroke patients: the SISCO pilot study.

BACKGROUND: Laboratory studies have shown that rapid therapeutic hypothermia (TH) of <34 °C can reduce stroke infarct volume by over 50%. The EuroHyp-1 and ICTUS 2/3 trials found no benefits in the slow cooling of ischemic stroke patients, while extensive shivering was observed. More powerful cooling methods are required to improve outcomes. METHODS: In a feasibility study approved to include up to 30 patients, the ThermoSuit® System (TSS) was used to cool sedated ischemic stroke patients to 32-34 °C. Patients were cooled after reperfusion, and TH was maintained for 24 h. Cooling speeds, adve

Acute and chronic convection-enhanced muscimol delivery into the rat subthalamic nucleus induces antiseizure effects associated with high responder rates.

Intracerebral drug delivery is an emerging treatment strategy aiming to manage seizures in patients with systemic drug-resistant epilepsies. In rat seizure and epilepsy models, the GABAA receptor agonist muscimol has shown powerful antiseizure potential when injected acutely into the subthalamic nucleus (STN), known for its capacity to provide remote control of different seizure types. However, chronic intrasubthalamic muscimol delivery required for long-term seizure suppression has not yet been investigated. We tested the hypothesis that chronic convection-enhanced delivery (CED) of muscimol

Novel 2D Layered MXene Nanofluids for Enhancing the Convective Heat Transfer Performance of Double-Pipe Heat Exchangers.

This paper proposes a new class of novel 2D layered structured materials, such as MXene (MX), for the synthesis of innovative nanofluids as coolants to evaluate the convective heat transfer performance of a double-pipe heat exchanger (DPHE). Convective heat transfer experiments were successfully conducted in lab-scale fabricated DPHE using low-concentration MXene nanofluids by varying the volume concentration of MXene nanoparticles (0.01-0.05 vol %) in different base fluids. The influence of the MXene nanofluids on various convective heat transfer parameters, such as LMTD, Nusselt number, heat

A novel point-of-care test of respiratory syncytial viral RNA based on cellulose-based purification and convective PCR.

BACKGROUND: Respiratory syncytial virus (RSV) infection is a global public-health problem. Timely diagnostics are needed for high-risk patients. Several methods have been used for RSV detection but not suitable for on-site detection due to the requirement of specialized laboratories and expensive equipment. METHODS: We developed a convenient, rapid and low-cost method of nucleic acids (NA) extraction based on cellulose paper, which could extract NA from nasopharyngeal swabs (NPSs) within 1 min. This extraction method was integrated with fluorescence convection polymerase chain reaction (CPCR)

Numerical study of high-frequency oscillatory air flow and convective mixing in a CT-based human airway model.

High-frequency oscillatory ventilation (HFOV) is considered an efficient and safe respiratory technique to ventilate neonates and patients with acute respiratory distress syndrome. HFOV has very different characteristics from normal breathing physiology, with a much smaller tidal volume and a higher breathing frequency. In this study, the high-frequency oscillatory flow is studied using a computational fluid dynamics analysis in three different geometrical models with increasing complexity: a straight tube, a single-bifurcation tube model, and a computed tomography (CT)-based human airway mode

A semi-analytical physics-informed neural network framework with hessian analysis for singularly perturbed fractal convection-dominated anomalous diffusion problems.

The present study introduces the concept of physics-informed neural networks to develop a numerical approach for solving singularly perturbed convection-dominated anomalous diffusion problems. A key focus of this research is to analyse the boundary layers that arise in convection-dominated anomalous diffusion problems using an artificial neural network, namely a physics-informed neural network. We explore solutions to convection-dominated anomalous diffusion problems using neural-network corrector functions derived from underlying auxiliary conditions. The thin layers represent regions with ra

Segmentation and structural connectivity of the putamen for targeted convection enhanced drug delivery in Parkinson's disease: a tractography-based approach.

BACKGROUND: Convection-enhanced drug delivery (CED) for Parkinson's disease (PD) is costly, and current methods lack precision, often targeting the entire putamen, leading to inefficient use of resources. Our simulation study explores a more targeted approach, which could reduce treatment costs by focusing therapy on specific putaminal regions, aiming to optimize delivery without compromising efficacy. METHODS: Twenty PD patients underwent diffusion-weighted imaging (DWI) to visualize the structural connectivity. A commercial subcortical auto-segmentation tool was used to define the putamen a
