Self-Assembly of the Second Transmembrane Domain of hCtr1 in Micelles and Interaction with Silver Ion.

Human copper transporter 1 (hCtr1) transports copper and silver by a homotrimer. The protein contains three transmembrane domains in which the second transmembrane domain (TMD2) is a key component lining the central pore of the trimer. The MXXXM motif in the C-terminal end of TMD2 plays a significant role in the function of hCtr1. In this study, we characterized the structure and assembly of isolated TMD2 of hCtr1 in sodium dodecyl sulfate (SDS) micelles and the interaction of the micelle-bound peptide with silver ion using nuclear magnetic resonance, circular dichroism, isothermal titration c

Regulation of heme synthesis and proteasomal activity by copper: possible implications for Wilson's disease.

Wilson's disease (Wd) is a genetic disorder resulting in Cu2+ accumulation, and is caused by mutations in the ATP7B gene, the copper transporter. In vivo studies show a correlation between Cu2+ accumulation and malfunction of the heme biosynthesis pathway. In this study, we describe multiple effects of Cu2+ accumulation on heme synthesis, which, in turn, affect proteasomal activity. Cu2+ toxicity was examined in two hepatocellular carcinoma cell lines, HepG2 and Hep3B, with Hep3B cells containing an integrated hepatitis B virus genome. Exposure of HepG2 and Hep3B cells to Cu2+ inhibited the en

Cuproptosis involvement in neonatal ischemic-hypoxic encephalopathy through the COMMD1/ATP7A signaling axis.

Neonatal hypoxic-ischemic encephalopathy (HIE) is a severe brain lesion caused by perinatal asphyxia that often results in neonatal death or long-term neurological deficits. The in-depth study of cell death mechanisms has recently led to the discovery of cuproptosis, a new type of cell death. Although understanding the relationship between HIE and cuproptosis is still in its preliminary stages, there is evidence that copper ions and their associated metabolic pathways play an important role in the pathophysiology of HIE. We used the Rice-Vannucci method to construct a mouse model of HIE. Cereb

Quantitative relationship between mutated amino-acid sequence of human copper-transporting ATPases and their related diseases.

Copper-transporting ATPase 1 and 2 (ATP7A and ATP7B) are two highly homologous P-type copper ATPase exporters. Mutations in ATP7A can lead to Menkes disease which is an X-linked disorder of copper deficiency. Mutations in ATP7B can cause Wilson disease which is an autosomal recessive disorder of copper toxicity. In this study, we attempt to build a quantitative relationship between mutated ATPase and Menkes/Wilson disease. First, we use the amino-acid distribution probability as a measure to quantify the difference in ATPase before and after mutation. Second, we use the cross-impact analysis t

Taxifolin regulates SLC31A1-mediated cuproptosis and tumor progression in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) is a primary malignant neoplasm exhibiting a high mortality rate. Taxifolin is a naturally occurring flavonoid compound that exhibits a range of pharmacological properties. The effects of taxifolin on HCC remain largely unexplored. Therefore, the aim of this study was to examine the potential roles of taxifolin in the development and progression of HCC. In this study, CCK-8 assay was utilized to examine the impact of taxifolin on the cell viability. The copper ions level and the activity of mitochondrial respiratory chain were determined by the correspondent kits

Wilson's disease: does a neuropsychiatric phenotype without liver involvement truly exist?

INTRODUCTION: Wilson’s disease (WD) is an inherited disorder of copper metabolism resulting in pathological copper accumulation in multiple organs, predominantly the liver, kidneys, and brain, with subsequent organ damage due to copper toxicity. Although the underlying metabolic defect originates in impaired hepatic copper transport with early hepatocellular injury, the currently accepted clinical phenotypic classification of WD distinguishes two major forms based on presenting symptoms: (1) hepatic and (2) neuropsychiatric, including cases without clinically overt liver disease. The aim of th

Nephrotoxicity induced by PLA NPs and CuSO4: Crosstalk between Cu metabolism and ferroptosis via ATP7B autophagy.

Polylactic acid (PLA) plastics exhibit greater environmental degradability than petroleum-based plastics, potentially exacerbating nanoplastic (NPs) formation that enters the food chain. PLA NPs can accumulate in kidneys and transport heavy metals like copper (Cu) due to their high surface area and charge properties. Ferroptosis, an iron-dependent cell death pathway, is implicated in renal diseases. While Cu homeostasis disruption may interact with ferroptosis. This study explored PLA NPs and Cu-induced kidney injury, examining Cu-ferroptosis crosstalk and autophagy's regulatory role. Our find

MiR-495 Inhibits Cisplatin Resistance and Angiogenesis in Esophageal Cancer by Targeting ATP7A.

Background: Cancer resistance to chemotherapy is closely associated with changes in transporter systems. In this study, we investigated the possible regulation of 1 copper ion transporter (ATP7A; ATPase copper transporting alpha) by microRNA miR-495 and its implications in cisplatin resistance and angiogenesis in esophageal cancer. Methods: MiR-495 and ATP7A mRNA expression in clinical tissue samples and 2 cancer cell lines (Eca-109 and TE1) were detected by quantitative real-time polymerase chain reaction. The levels of miR-495 and ATP7A expression in Eca-109 and TE1 cells were increased by t

[Hereditary hemochromatosis, alpha-1-antitrypsin deficiency and Wilson's disease. Pathogenesis, clinical findings and pathways to diagnosis].

Primary hemochromatosis, alpha-1-antitrypsin (AAT) deficiency, and Wilson's disease are the most common hereditary causes of unclear hepatopathy. Classical primary hemochromatosis (type I) on the basis of a homozygous mutation of the HFE gene, usually presents in adults with increasing hepatocellular siderosis and chronic progressive necroinflammatory liver disease. Homozygous AAT deficiency type PiZZ becomes manifest in newborns as a giant cell hepatitis or findings similar to bile duct atresia, in adults as chronic hepatitis or "cryptogenic cirrhosis". The heterozygous PiZ mutation can lead

Diagnosis, management and genetic analysis of a cat with primary copper hepatopathy.

CASE SUMMARY: A 2-year-old spayed female domestic longhair cat was presented for evaluation of chronic ocular discharge and occasional vomiting. While physical examination findings were consistent with an upper respiratory infection (URI), serum chemistry results revealed increased liver enzyme activities. Histopathologic examination of a liver biopsy identified substantial centrilobular accumulation of copper in hepatocytes - strongly suggestive of primary copper hepatopathy (PCH). Retrospective cytologic examination of a liver aspirate also identified copper aggregates in hepatocytes. After

Copper and ectopic expression of the Arabidopsis transport protein COPT1 alter iron homeostasis in rice (Oryza sativa L.).

Copper deficiency and excess differentially affect iron homeostasis in rice and overexpression of the Arabidopsis high-affinity copper transporter COPT1 slightly increases endogenous iron concentration in rice grains. Higher plants have developed sophisticated mechanisms to efficiently acquire and use micronutrients such as copper and iron. However, the molecular mechanisms underlying the interaction between both metals remain poorly understood. In the present work, we study the effects produced on iron homeostasis by a wide range of copper concentrations in the growth media and by altered cop

Increased frequency of congenital heart defects in Menkes disease.

ATP7A is a copper-transporting ATPase critical for central and peripheral nervous system function. Mutations in ATP7A cause Menkes disease and occipital horn syndrome (OHS), allelic X-linked recessive conditions that feature vascular abnormalities ascribed to low activity of lysyl oxidase, a copper-dependent enzyme. From a recently created Menkes disease/OHS patient registry, we identified four of 95 patients with major congenital heart defects (4.2%), a proportion exceeding the general population prevalence (≈1%). In conjunction with mouse models of Menkes disease, OHS, and lysyl oxidase defi

Melatonin inhibits liver ferroptosis in copper-laden rats: a potential therapy mechanism underlying Wilson's disease.

Wilson's disease (WD) is an autosomal recessive disorder associated with impaired copper metabolism that results in hepatic manifestations. However, as a rare disease, the underlying pathogenic mechanism and drug development have lagged behind. Studies have reported that copper accumulation is associated with potential increases in iron levels, which can lead to further exacerbation of oxidative damage and has been observed in WD patients. Therefore, removing excess copper from the body and enhancing antioxidant capacity are crucial in treatment. Melatonin (MLT) is an endogenous hormone with a

Liver Expression of a MiniATP7B Gene Results in Long-Term Restoration of Copper Homeostasis in a Wilson Disease Model in Mice.

Gene therapy with an adeno-associated vector (AAV) serotype 8 encoding the human ATPase copper-transporting beta polypeptide (ATP7B) complementary DNA (cDNA; AAV8-ATP7B) is able to provide long-term copper metabolism correction in 6-week-old male Wilson disease (WD) mice. However, the size of the genome (5.2 kilobases [kb]) surpasses the optimal packaging capacity of the vector, which resulted in low-yield production; in addition, further analyses in WD female mice and in animals with a more advanced disease revealed reduced therapeutic efficacy, as compared to younger males. To improve effica

Role of copper in regression of cardiac hypertrophy.

Pressure overload causes an accumulation of homocysteine in the heart, which is accompanied by copper depletion through the formation of copper-homocysteine complexes and the excretion of the complexes. Copper supplementation recovers cytochrome c oxidase (CCO) activity and promotes myocardial angiogenesis, along with the regression of cardiac hypertrophy and the recovery of cardiac contractile function. Increased copper availability is responsible for the recovery of CCO activity. Copper promoted expression of angiogenesis factors including vascular endothelial growth factor (VEGF) in endothe

Neurological Wilson's Disease Signs-Hepatic Encephalopathy or Copper Toxicosis?

Wilson's disease (WD) is a rare autosomal recessive (AR) disorder resulting from mutations in the ATP7B gene, which is responsible for the encryption of transmembrane copper transporting ATPase. The symptomatic presentation of the disease is estimated to be about 1 in 30,000. The impairment of ATP7B function results in a copper overload in hepatocytes, which further leads to liver pathology. This copper overload also occurs in other organs, most particularly in the brain. This could then cause the occurrence of neurological and psychiatric disorders. Symptoms differ substantially and most ofte

Nitroxyl relieves acute kidney injury by suppressing SLC31A1-mediated cuproptosis in renal tubular epithelial cells.

CONTEXT: Cuproptosis, a copper-dependent form of programmed cell death, has been implicated in the pathogenesis of acute kidney injury (AKI). Nitroxyl (HNO), the one-electron reduced and protonated form of nitric oxide (NO), is an emerging regulator of cellular function. However, the role of HNO in modulating cuproptosis during AKI remains largely unexplored. OBJECTIVE: This study aims to investigate the effect of HNO on cuproptosis in a murine model of renal ischemia-reperfusion (IR) injury. MATERIALS AND METHODS: An in vitro hypoxia/reoxygenation (HR) model using human kidney-2 (HK-2) cell

Altered copper transport in oxidative stress-dependent brain endothelial barrier dysfunction associated with Alzheimer's disease.

Oxidative stress and blood-brain barrier (BBB) disruption due to brain endothelial barrier dysfunction contribute to Alzheimer's Disease (AD), which is characterized by beta-amyloid (Aβ) accumulation in senile plaques. Copper (Cu) is implicated in AD pathology and its levels are tightly controlled by several Cu transport proteins. However, their expression and role in AD, particularly in relation to brain endothelial barrier function remains unclear. In this study, we examined the expression of Cu transport proteins in the brains of AD mouse models as well as their involvement in Aβ42-induced

ATF3/SPI1/SLC31A1 Signaling Promotes Cuproptosis Induced by Advanced Glycosylation End Products in Diabetic Myocardial Injury.

Cuproptosis resulting from copper (Cu) overload has not yet been investigated in diabetic cardiomyopathy (DCM). Advanced glycosylation end products (AGEs) induced by persistent hyperglycemia play an essential role in cardiotoxicity. To clarify whether cuproptosis was involved in AGEs-induced cardiotoxicity, we analyzed the toxicity of AGEs and copper in AC16 cardiomyocytes and in STZ-induced or db/db-diabetic mouse models. The results showed that copper ionophore elesclomol induced cuproptosis in cardiomyocytes. It was only rescued by copper chelator tetrathiomolybdate rather than by other cel

Copper nanoregulator with organelle-level precision reprograms COMMD1-Mediated copper homeostasis for myocardial infarction repair.

Copper (Cu) homeostasis dysregulation is one of the key factor exacerbating mitochondrial dysfunction and impairing cardiac repair in myocardial infarction (MI). Herein, by integrating single-cell transcriptomics, clinical specimens, and animal models, we first identified COMMD1 as a key negative regulator of Cu homeostasis in MI. We then constructed a Cu nanoregulator, Qu@Cu-SS31, that selectively accumulate in the mitochondria of ischemic cardiomyocytes and release bioactive Cu2+ in a pH-responsive manner, enhancing mitochondrial function and promoting myocardial recovery.
