Genetic reprogramming of transcription factor ap-2gamma in bovine somatic cell nuclear transfer preimplantation embryos and placentomes.

Bovine somatic cell nuclear transfer (SCNT) efficiency remains very low despite a tremendous amount of research devoted to its improvement over the past decade. Frequent early and mid-gestational losses are commonly accompanied by placental abnormalities. A transcription factor, activating protein AP-2gamma, has been shown to be necessary for proper placental development in the mouse. We first evaluated the expression of the gene coding for AP-2gamma (Tfap2c) in several bovine fibroblast donor cell lines and found it was not expressed. Subsequently we determined the expression profile of Tfap2

Postnatal transient renal insufficiency in the feto-fetal transfusion syndrome.

Twinning and higher-order multiple-gestation pregnancies have become relatively frequent in the current era of assisted reproductive techniques. Vascular interconnections are present in nearly all monochorionic twin placentae, yet hemodynamically significant arteriovenous anastomoses resulting in the feto-fetal transfusion syndrome occur in only 5%-18% of these. When arteriovenous connections through a shared placental cotyledon are present, variable amounts of blood may be transfused from one fetus to the other, and feto-fetal transfusion syndrome may result. While reports of renal failure du

High-Throughput Identification of Resistance to Pseudomonas syringae pv. Tomato in Tomato using Seedling Flood Assay.

Tomato is an agronomically important crop that can be infected by Pseudomonas syringae, a Gram-negative bacterium, resulting in bacterial speck disease. The tomato-P. syringae pv. tomato pathosystem is widely used to dissect the genetic basis of plant innate responses and disease resistance. While disease was successfully managed for many decades through the introduction of the Pto/Prf gene cluster from Solanum pimpinellifolium into cultivated tomato, race 1 strains of P. syringae have evolved to overcome resistance conferred by the Pto/Prf gene cluster and occur worldwide. Wild tomato species

SMI for imaging of placental infarction.

Since superb Micro-vascular Imaging (SMI) is a new blood flow imaging technique that employs a unique algorithm to minimize motion artifacts, it can visualize low-velocity blood flow in small vessels. We demonstrate SMI imaging of the placental infarction in a case with fetal growth restriction, comparing a normal placenta on the same gestation. While SMI image in normal placenta clearly shows structures from the branching vessels to the peripheral villous trees in the cotyledons, they could not be seen in the case of fetal growth restriction. We think that the clinical value and future potent

The content of fagopyrin and polyphenols in common and tartary buckwheat sprouts.

Dried buckwheat herb is used in medicinal products whereas fresh green plant parts, especially sprouts, are consumed as a vegetable. The herb contains fagopyrins, which cause sensitivity to light after ingestion. The aim of this study was to investigate the impact of different growing conditions and the development phase on the content of fagopyrin and phenolic compounds in buckwheat sprouts. Total flavonoid and total phenol contents, fagopyrin content and antioxidant activity were determined spectrophotometrically. Fagopyrin and flavonoids were located almost exclusively in cotyledons. Based

Ultrastructural changes in the placenta of the ewe following foetal infusion of cortisol.

The placental changes which followed continuous infusion of cortisol into the sheep foetus in the later stages of gestation were, like the hormonal changes, broadly similar to those of spontaneous parturition. There was, however, a premature separation of foetal and maternal tissues in certain areas of the placental cotyledons, and this separation appeared to protect the foetal epithelium from the degenerative changes which normally take place in the short space of time between the birth of the lamb and the delivery of the foetal membranes. The results suggest that an experimental model in whi

Developmental and light effects on the accumulation of FtsH protease in Arabidopsis chloroplasts--implications for thylakoid formation and photosystem II maintenance.

The chloroplast ATP-dependent metalloprotease FtsH is involved in the degradation of unassembled proteins, the repair of photosystem II (PSII) from photoinhibition, and, apparently, the formation of thylakoids. In Arabidopsis, it is encoded by a family of 12 genes. However, the products of only four of them, FtsH1, 2, 5 and 8, have been found in chloroplasts to date. Mutations in two of these, FtsH2 and 5, demonstrate a visible phenotype of variegated leaves, with the phenotype of the FtsH2 mutant being more pronounced. Moreover, the degree of variegation appears to be dependent on development

Expression of prolactin-related hormones in the early bovine conceptus, and potential for paracrine effect on the endometrium.

Hormones related to the pituitary hormones PRL and GH are produced by the utero-placental unit of many species. In the cow, these include bovine placental lactogen (bPL) and a distantly related subfamily including the protein encoded by bovine PRL-related complementary DNA I (bPRCI). In the present studies, we defined the onset of expression of these genes in order to begin to study the regulation of their expression and function before implantation. Messenger RNA levels of both bPL and bPRCI were assessed by dot blot analysis of total RNAs prepared from conceptuses on days 15-25. Specific tra

Antioxidant transport by the human placenta.

We investigated the transfer of three antioxidants - melatonin, S-adenosyl methionine (SAM) and various forms of vitamin E - across the term, normal human placenta. The transport technique involved the single, isolated placental cotyledon system in vitro. Melatonin crossed the placental rapidly, equally to the freely diffusible marker, antipyrine. There was no biotransformation of the agent. SAM was transferred slowly, similarly to passively transported L-glucose as a marker. There was a breakdown of SAM to at least one other derivative; the process appeared to be nonenzymatic. Vitamin E was t

Maternal ethanol use and selective fetal malnutrition.

Maternal ethanol ingestion may cause fetal injury, particularly impaired somatic and brain growth, by at least two mechanisms: (1) directly, by fetotoxicity from ethanol and/or acetaldehyde; (2) indirectly, by ethanol-induced placental injury and selective fatal malnutrition. There is ample evidence that ethanol and/or acetaldehyde is fetotoxic. In addition, alcoholics are frequently in a state of poor nutrition. However, regardless of maternal nutritional status, ethanol can be placentotoxic, impairing the normal transfer of essential fetal nutrients. Ethanol and/or acetaldehyde has been show

Plant regeneration from single-nodal-stem explants of legume tree Prosopis alba Griseb.

Seeds of Prosopis alba were scarified with abrasive paper and placed to germinate on MS (Murashige and Skoog 1962) nutrient medium. After 7 days of culture, the basal part of cotyledons was removed and pieces of 4 mm" from distal parts were cultured on Murashige and Skoog (1962) mineral salts and vitamins (MS) (3% sucrose) supplemented with growth regulators. Callus proliferation took place in the majority of the media tested. A low percentage of calluses with green buds that developed on MS basal medium containing 0.1 mg.L-1 2,4-D alone or supplemented with BAP at 0.1 mg.L-1 was observed. Nei

Maternal nutrient restriction alters uterine artery hemodynamics and placentome vascular density in Bos indicus and Bos taurus.

The objective was to examine uterine artery blood flow (UBF) as well as macroscopic and microscopic placentome vascular density in nutrient-restricted Angus and Brahman heifers. Angus (n = 6) and Brahman (n = 6) heifers were bred to a single sire and pregnancy confirmed at 30-d postbreeding. Heifers were randomly assigned to 1 of 2 dietary treatments consisting of 100% (control-fed; CON; n = 6) or 60% (total nutrient-restricted; RES; n = 6) based from net energy requirements for gestating heifers. Nutritional treatments were imposed from days 50 to 180 of gestation. On day 175 of gestation, UB

Placental Health Score (PHS) for Early Prediction of Placental Insufficiency and Preterm Birth: A Cross-Sectional Analytical Study.

Background Placental insufficiency is a pathological state in which the placenta fails to provide adequate oxygen and nutrients to the fetus, leading to impaired growth, preterm birth, and adverse neonatal outcomes. Despite its clinical relevance, diagnostic approaches remain limited, and there is a need for a quantifiable tool to assess placental health. The objective of this study was to develop a Placental Health Score (PHS) by integrating key morphometric placental parameters into a multiple linear regression model after normalization and to validate the score by assessing its correlation

Seedling diversity, development and evolution in Bromeliaceae Juss. (Poales).

BACKGROUND AND AIMS: Understanding post-seminal development is essential for species conservation and provides key insights into reproductive biology and macroevolution. Despite their remarkable structural diversity among monocots, seedlings remain an understudied developmental stage, although they are critical for juvenile sporophyte survival and establishment. In Bromeliaceae, one of the major adaptive radiations of monocots, knowledge of seedling development remains fragmented and taxonomically biased. This study comparatively characterizes the ontogeny and morphoanatomy of bromeliad seedli

Maternal Rumen-Protected Leucine Supplementation Enhances Placental Nutrient Transport Capacity and Increases Birth Weight in Hu Sheep.

Leucine, an essential branched-chain amino acid, serves not only as a substrate for protein synthesis but also as a key regulator of placental function and fetal development. This study investigated the effects of dietary supplementation with RP-Leu during late gestation on placental development and offspring performance in Hu sheep. Sixty twin-pregnant ewes at day 80 of pregnancy were randomly assigned to either a control group (fed a basal diet) or an RP-Leu group (fed a basal diet supplemented with 19 g/day RP-Leu). The feeding trial lasted for 60 d. The ewes were slaughtered at day 140 of

A novel Ca2+-activated protease from germinating Vigna radiata seeds and its role in storage protein mobilization.

Calcium (Ca(2+))-dependent/activated proteases make decisive cleavages in proteins, affecting their further degradation/activation. Few such Ca(2+)-dependent proteases have been reported from plants, and none during germination-related events. Seeds are woken up from their quiescent state upon imbibition of water. The subsequent process of germination is strongly influenced by hormones (mainly gibberellins) and light, with both resulting in change in intracellular Ca(2+). We have investigated the effect of Ca(2+) on protease activity in extracts prepared from dry Vigna radiata (L.) Wilczec see

Ectopic expression of BABY BOOM triggers a conversion from vegetative to embryonic growth.

The molecular mechanisms underlying the initiation and maintenance of the embryonic pathway in plants are largely unknown. To obtain more insight into these processes, we used subtractive hybridization to identify genes that are upregulated during the in vitro induction of embryo development from immature pollen grains of Brassica napus (microspore embryogenesis). One of the genes identified, BABY BOOM (BBM), shows similarity to the AP2/ERF family of transcription factors and is expressed preferentially in developing embryos and seeds. Ectopic expression of BBM in Arabidopsis and Brassica led

Comparison of the pathogenicity for pregnant sheep of Rift Valley fever virus and a live attenuated vaccine.

A live attenuated mutant of Rift Valley fever virus, MV P12, was previously shown to be non-pathogenic in young lambs, but capable of producing protective immunity. The studies reported here show that the abortion in sheep caused by an infection with virulent virus is the result of necrosis of the maternal villi and cotyledons arising from an acute inflammation of the maternal caruncles. Pregnant ewes infected with the attenuated mutant virus MV P12 showed none of these lesions in the placenta and gave birth to healthy lambs. Colostrum from ewes infected with MV P12 virus was able to induce pr

Plastic and Placenta: Identification of Polyethylene Glycol (PEG) Compounds in the Human Placenta by HPLC-MS/MS System.

The placenta is a crucial interface between the fetus and the maternal environment. It allows for nutrient absorption, thermal regulation, waste elimination, and gas exchange through the mother's blood supply. Furthermore, the placenta determines important adjustments and epigenetic modifications that can change the phenotypic expression of the individual even long after birth. Polyethylene glycol (PEG) is a polyether compound derived from petroleum with many applications, from medicine to industrial manufacturing. In this study, for the first time, an integration of ultra-high-performance liq

[PLACENTAL BED RESEARCH - UNDERSTANDING THE PATHOPHYSIOLOGY OF THE GREAT OBSTETRICAL SYNDROMES].

Research on the placenta bed - the interface between the mother and the fetus, substantially contributed to the understanding of the pathophysiology of the so-called great obstetrical syndromes (preeclampsia, fetal growth restriction, preterm labor, placental abruption). The physiological modifications of the utero-placental spiral arteries was the key finding, thereafter, many other unique physiological and pathophysiological changes were discovered, specifically related to these clinical syndromes. In the current issue, a group of researchers from the Edith Wolfson's Obstetrics and Gynecolog
