Genetics, cross-resistance and mechanism of resistance to spinosad in a field strain of Musca domestica L. (Diptera: Muscidae).

The house fly, Musca domestica L., is a cosmopolitan insect with the ability to develop resistance to insecticides used for their management. In the present study, we investigated the genetics of spinosad resistance, and cross-resistance potential to other insecticides by selecting a field strain with a commercial spinosad formulation. Bioassays with the field strain, before selection with spinosad, gave resistance ratios (RRs) of 4, 5, 66, 21 and 5 fold for spinosad, indoxacarb, abamectin, imidacloprid and deltamethrin, respectively, in comparison to a laboratory susceptible (Lab-susceptible)

Dramatic genotypic difference in, and effect of genetic crossing on, tissue culture-induced mobility of retrotransposon Tos17 in rice.

KEY MESSAGE : We show for the first time that intraspecific crossing may impact mobility of the prominent endogenous retrotransposon Tos17 under tissue culture conditions in rice. Tos17, an endogenous copia retrotransposon of rice, is transpositionally active in tissue culture. To study whether there exists fundamental genotypic difference in the tissue culture-induced mobility of Tos17, and if so, whether the difference is under genetic and/or epigenetic control, we conducted this investigation. We show that dramatic difference in tissue culture-induced Tos17 mobility exists among different r

Genome-wide cross-trait analysis to compare genetic architecture between Parkinson's disease and kidney-related traits.

Epidemiological studies link kidney function to Parkinson's disease risk, but the genetic basis of this relationship remains unclear. Here, we leverage genome-wide association data for Parkinson's disease and five UK Biobank-derived kidney-related traits in individuals of European ancestry, combining genetic correlation, pleiotropy mapping, colocalization and enrichment across sex-combined and sex-stratified analyses. Genome-wide genetic correlations with Parkinson's are modest, with only a significant correlation with urinary potassium-to-creatinine ratio significant, yet we identify shared l

Awareness, uptake, and determinants of germline genetic testing in Chinese epithelial ovarian cancer patients: a cross-sectional study.

OBJECTIVE: Various guidelines recommend offering germline genetic testing to all patients diagnosed with epithelial ovarian cancer, as it helps guide treatment decisions, identify cancer risks in other organs, and empowers patients to notify families for timely screening and risk-reducing surgery. Our study aimed to explore the awareness and uptake rate of germline genetic testing among Chinese women with epithelial ovarian cancer, and to analyze the determinants influencing their decision. METHODS: A cross sectional study was conducted at Obstetrics and Gynecology Hospital of Fudan Universit

Pharmacotherapy Risks in Rare Genetic Diseases: Cross-Referencing ACMG Secondary Findings v3.2 List With Clinical Databases.

Clinical genomics and pharmacogenomics have largely remained separate fields, though some genetic variants have overlapping disease risk and drug implications. However, the extent of this overlap is not well studied. To explore this gap, we cross-referenced genes from the American College of Medical Genetics Secondary Findings v3.2 list with genomic databases and drug labeling to identify gene-phenotype pairs with overlapping clinical genomics and pharmacogenomic implications. We searched GeneReviews and PharmGKB (now called ClinPGx) for each gene-phenotype pair and reviewed the FDALabel datab

A genome-wide cross-trait analysis characterizes the shared genetic architecture between rheumatoid arthritis and psychiatric disorders.

OBJECTIVES: Patients with RA have a 2- to 3-fold elevated risk of psychiatric disorders, suggesting an underlying genetic link between these phenotypes. However, the shared genetic architectures and pathological mechanisms driving RA-psychiatric disorder comorbidity remain to be fully elucidated. Herein, we performed cross-trait analysis to investigate the shared genetic architecture between RA and psychiatric disorders. METHODS: Leveraging European-ancestry genome-wide association studies (GWASs) datasets of RA (n = 1 026 690) and 10 major psychiatric disorders (n = 14 307-1 222 882), we per

Identification of genetic variants associated with idiopathic inflammatory myopathies via cross-trait analysis with B cell lymphomas.

The genetic architecture of idiopathic inflammatory myopathies (IIMs) remains incompletely defined. When increasing sample size is not feasible, cross-trait analysis of genetically correlated diseases offers an effective strategy for discovering risk loci. Using summary statistics of IIM and B cell lymphoma subtypes, we applied conditional false discovery rate (condFDR) and multi-trait analysis of genome-wide association studies (GWASs) (MTAG) to detect genetic associations with IIM risk. Single-nucleotide polymorphisms (SNPs) outside the human leukocyte antigen (HLA) region meeting significan

Cross-family and phage-specific gene requirements for Klebsiella infection revealed by scalable RB-TnSeq genetic screens.

Bacteriophages are being cataloged at an accelerating pace and are recognized as key players in nutrient and energy cycling across ecosystems. Yet the bacterial genetic determinants that govern phage-host specificity and infection success remain poorly understood, particularly in clinically and ecologically important genera such as Klebsiella where prior receptor characterization has been almost entirely limited to capsulated strains. Here we used a randomly barcoded, genome-wide, loss-of-function transposon mutant library (RB-TnSeq) of Klebsiella sp. M5al, a naturally acapsular, nitrogen-fixi

System genetic analysis of intestinal cancer and periodontitis development as influenced by aging and diabesity using Collaborative Cross mice.

It is increasingly recognized that young, chow-fed inbred mice poorly model the complexity of human carcinogenesis. In humans, age and adiposity are major risk factors for malignancies, but most genetically engineered mouse models (GEMM) induce carcinogenesis too rapidly to study these influences. Standard strains, such as C57BL/6, commonly used in GEMMs, further limit the exploration of aging and metabolic health effects. A similar challenge arises in modeling periodontitis, a disease influenced by aging, diabesity, and genetic architecture. We propose using diverse mouse populations with hyb

Novel insights into the relationship between glaucoma and brain diseases from the genetic to diseases levels: A cross-sectional study.

Glaucoma is a heterogeneous group of diseases which is one of the leading causes of irreversible blindness worldwide. Although the eye-brain axis has been proposed, its functional connectivity remains poorly defined. This study aimed to explore the mechanisms and causal relationship between glaucoma and brain cortical structure, focusing on the eye-brain axis. A Mendelian randomization (MR) study was conducted using inverse variance weighting as the primary estimator, alongside MR-PRESSO, MR-Egger, and weighted median methods to assess sensitivity, heterogeneity, and pleiotropy. Pathway analys

No association between anti-cytomegalovirus seropositivity and arthritis: evidence from the cross-sectional epidemiology and genetic association analyses.

Human cytomegalovirus (CMV), a β-herpesvirus associated with chronic inflammation and lifelong latency, has been implicated in the pathogenesis of arthritis. However, the nature of this relationship remains controversial. In this study, we integrate cross-sectional epidemiology analyses, genetic correlation assessments, and Mendelian randomization (MR) approaches to elucidate the potential association between CMV infection and arthritis-related conditions. Observational analysis of 5133 participants from the NHANES database revealed a positive association between CMV IgG seropositivity and art

Fear of cancer recurrence in breast cancer survivors carrying a BRCA1 or 2 genetic mutation : a cross-sectional study.

BACKGROUND: Fear of cancer recurrence (FCR) affects virtually all patients who have been treated for cancer, to varying degrees. Breast cancer survivors who carry a BRCA1 or BRCA2 gene mutation are at high risk of cancer recurrence. No study has yet assessed FCR specifically in this population. OBJECTIVES: This cross-sectional study, conducted in women who were treated for breast cancer and carrying a BRCA1/2 mutation, aimed to: (1) assess the mean level of FCR and estimate the proportion of patients with clinical levels of FCR; (2) examine the relationships between FCR and selected psycholog

Diversity is key for cross-ancestry transferability of glaucoma genetic risk scores in Hispanic Veterans in the Million Veteran Program.

A major goal of precision medicine is to stratify patients based on their genetic risk for a disease to inform future screening and intervention strategies. For conditions like primary open-angle glaucoma (POAG), the genetic risk architecture is complicated with multiple variants contributing small effects on risk. Following the tepid success of genome-wide association studies for high-effect disease risk variant discovery, genetic risk scores (GRS), which collate effects from multiple genetic variants into a single measure, have shown promise for disease risk stratification. We assessed the a
