Pathogenomics of Culex quinquefasciatus and meta-analysis of infection responses to diverse pathogens.

The mosquito Culex quinquefasciatus poses a substantial threat to human and veterinary health as a primary vector of West Nile virus (WNV), the filarial worm Wuchereria bancrofti, and an avian malaria parasite. Comparative phylogenomics revealed an expanded canonical C. quinquefasciatus immune gene repertoire compared with those of Aedes aegypti and Anopheles gambiae. Transcriptomic analysis of C. quinquefasciatus genes responsive to WNV, W. bancrofti, and non-native bacteria facilitated an unprecedented meta-analysis of 25 vector-pathogen interactions involving arboviruses, filarial worms, ba

Efficacy of pirimiphos-methyl (50% EC), as a larvicide for the control of Culex quinquefasciatus.

BACKGROUND & OBJECTIVE: Pirimiphos-methyl, an organophosphorus insecticide is known to overcome resistance mechanisms of mosquitoes to other organophosphates. Information on the duration of efficacy of pirimiphos-methyl (50% emulsifiable concentrate) for the control of immatures of Culex quinquefasciatus, the vector of bancroftian filariasis, is scanty and hence site specific field trials are necessary to determine the field dosage. Pirimiphos-methyl (50% EC) was tested for its efficacy in cesspits and drains against the immatures of Cx. quinquefasciatus at Puducherry, an endemic area for fila

Vector competence of Culiseta incidens and Culex thriambus for West Nile virus.

The vector competence of Culiseta incidens (Thomson) and Culex thriambus Dyar for West Nile virus (WNV) were compared to Cx. quinquefasciatus Say or Cx. tarsalis Coquillett and Cx. stigmatasoma Dyar collected concurrently in California. Culiseta incidens were less susceptible to oral infection than Cx. quinquefasciatus, but transmitted virus at a significantly higher rate, thereby yielding comparable population transmission rates. Culex thriambus was equally susceptible to oral infection and transmitted virus at rates comparable to Cx. tarsalis or Cx. stigmatosoma. A mammalian host selection p

Role of some factors on the breeding of JE vector Culex vishnui group.

Vector of JE, Culex vishnui group, prefer to breed in rice field. During the present study, roles of some factors like irrigation, manuring, pesticide application, rainfall, temperature and pH of the breeding water, depth of breeding water and height of paddy plants on breeding of Culex vishnui group larvae have been studied. It is evident from the study that besides striking role of the factors like manuring and pesticide application, other factors have also some role on the breeding of JE vectors at their breeding sites.

The effect of diethylcarbamazine treatment on the development of bancroftian microfilariae in Culex p. fatigans.

Bancroftian microfilarie survived after one to three courses of diethylcarbamazine treatment in carriers and the larvae able to reach the infective stage in Culex p. fatigans. The infection rate and the development of infective larvae per infected mosquito from DEC-treated carriers was much lower than from the untreated carrier. The fact that surving MF can develop to infective stage in their vector indicates that such filarial carriers may be important sources for transmission of filariasis in the endemic areas after suspension of control measures.

[Insecticide tests for resistance on adults of Anopheles sinensis and Culex tritaeniorhynchus in Korea].

During the summer season(July-August) in 1967, the insecticide tests for resistance on adult A. sinensis and C. tritaeniorhynchus were carried out in Gobu-Myun Jeongup-Gun, Chollapuk-Do, where many Japanese encephalitis cases occur every year. 1)The mortality by 4.0% DDT in A. sinensis was 88.9% and by 4.0% dieldrin was 84.4%. Therefore the LC-50 of DDT for A. sinensis was 1.75%, while LC-50 of dieldrin was 1.1%. 2)The mortality by 4.0% DDT in C. tritaeniorhynchus was 36.7%, and by 4.0% dieldrin was 6.0% 3)The result indicates that A. sinensis in Korea is susceptible to DDT and resistant to di

Biochemical, molecular, and in silico insights into the effects of cyphenothrin on Culex quinquefasciatus mosquitoes.

BACKGROUND: Cyphenothrin, a type II synthetic pyrethroid, is widely recommended by the World Health Organization (WHO) for control programs for mosquitoes such as Culex quinquefasciatus. This cosmopolitan mosquito species plays a pivotal role in transmitting several neglected tropical diseases, including lymphatic filariasis, West Nile virus, Japanese encephalitis, and so on. Besides treatment, vector control programs heavily rely on insecticides, leading to varying resistance due to prolonged exposure. METHODS: The present study assessed the susceptibility status of field-collected Cx. quinq

Molecular detection of Usutu virus in pools of Culex pipiens mosquitoes in Greece.

Usutu virus (USUV) is a mosquito-borne flavivirus originating from Africa, that belongs to the Japanese encephalitis virus (JEV) complex. In nature, USUV involves Culex spp. mosquitoes acting as vectors and birds as amplifying hosts. The virus has recently spread in Europe and is considered an emerging human pathogen. This is the first research study performed in Greece revealing the presence and circulation of USUV in Culex spp. mosquito populations. Out of the 1,500 mosquito pools tested with real-time RT-PCR, four (Roesch et al., 2019) were positive for USUV. All four pools were collected f

Pathology of an iridescent virus in immature Culex pipiens L. (Diptera, Culicidae).

Iridovirus in Culex pipiens was reported for the first time in 2012. Later studies of horizontal transmission were performed, in which an interaction with the parasite Strelkovimermis spiculatus acting as viral vector was recognized. In the present study, we observed aspects of the pathology produced by an invertebrate iridescent virus in laboratory infected immature Cx. pipiens as well as in infected immature Cx. pipiens in the field. In the laboratory infected larvae, the infection and mortality were asynchronous. Signs of infection in larvae exposed to the virus were observed between the se

Susceptibility of Culex quinquefasciatus (Diptera: Culicidae) in Southern Louisiana to Larval Insecticides.

Mosquito control districts conduct rigorous insecticide treatments against both larval and adult Culex quinquefasciatus Say (Diptera: Culicidae), the primary vector of West Nile virus in the southern United States. However, the development of resistant populations of Cx. quinquefasciatus in response to extensive larvicide or adulticide applications has been demonstrated repeatedly across the world. Examining changes in insecticide susceptibility in treated field areas can help inform mosquito control districts as to whether or not their treatments remain effective. We hypothesized that frequen

Two cathepsins B are responsible for the yolk protein hydrolysis in Culex quinquefasciatus.

Despite the established role of Culex quinquefasciatus as a vector of various neurotropic viruses, such as the Rift Valley and West Nile viruses, as well as lymphatic filariasis, little is known regarding the organism's reproductive physiology. As in other oviparous animals, vitellogenin, the most important source of nutrients for the embryo development, is digested by intracellular proteases. Using mass spectrometry, we have identified two cathepsin B homologues partially purified by self-proteolysis of Cx. quinquefasciatus total egg extract. The transcriptional profile of these two cathepsin

Hexamerin a novel protein associated with Bacillus sphaericus resistance in Culex quinquefasciatus.

Bacterial insecticides like, Bacillus sphaericus and Bacillus thuringiensis serovar israelensis, have been used for the control of nuisance and vector mosquitoes for more than two decades. For many years, it was assumed that the use of microbial larvicides based on B. sphaericus would not lead to resistance in mosquitoes. However, recent reports have shown that B. sphaericus toxins are not free from this problem. Therefore, the resistance of mosquito populations to be will seriously threaten the sustainability of current mosquito control programme using these microbial insecticides. In the pre

Behavioral and electrophysiological responses of Culex quinquefasciatus to certain fatty acid esters.

Oviposition response of gravid Culex quinquefasciatus females to a series of synthetic fatty acid esters was evaluated at 10ppm under laboratory conditions. Octyl tridecanoate and propyl octadecanoate elicited 85% and 73% increased ovipositional responses respectively, compared to control, among the 16 esters tested. Other 14 esters showed highly significant repellency (67-96%) to gravid females. Standard 3-methyl indole received 69% increased egg deposition compared to control. In the Y-tube olfactometer, gravid C. quinquefasciatus females exhibited 78, 64% and 58% orientation respectively to

Overwintering biology of Culex (Diptera: Culicidae) mosquitoes in the Sacramento Valley of California.

At temperate latitudes, Culex (Diptera: Culicidae) mosquitoes typically overwinter as adult females in reproductive arrest and also may serve as reservoir hosts for arboviruses when cold temperatures arrest viral replication. To evaluate their role in the persistence of West Nile virus (WNV) in the Sacramento Valley of California, the induction and termination of diapause were investigated for members of the Culex pipiens (L.) complex, Culex tarsalis Coquillett, and Culex stigmatosoma Dyar under field, seminatural, and experimental conditions. All Culex spp. remained vagile throughout winter,

Insecticide susceptibility of field-collected populations of Culex tritaeniorhynchus in the Republic of Korea.

The toxicities of 10 insecticides were examined against late third instar Culex tritaeniorhynchus Giles (Diptera: Culicidae) using the direct-contact mortality bioassay. Six geospatially-distant field mosquitoes were collected from Chuncheon-si, Hwaseong, Seosan. Jeonju, Daegu, and Busan in the Republic of Korea. Marked regional variations of insecticide susceptibility were observed. Field populations of Seosan, Jeonju, and Daegu from agricultural areas showed higher to extremely higher insecticide susceptibility to pyrethroids than those of Chuncheon-si, Hwaseong, and Busan strains from non-a

Detection of Wolbachia endobacteria in Culex quinquefasciatus by Gimenez staining and confirmation by PCR.

BACKGROUND & OBJECTIVES: Wolbachia are common intracellular bacteria that are found in arthropods and nematodes. These endosymbionts are transmitted vertically through host eggs and alter host biology in diverse ways, including the induction of reproductive manipulations, such as feminization, parthenogenesis, male killing and sperm-egg incompatibility. Since they can also move horizontally across species boundaries, Wolbachia is gaining importance in recent days as it could be used as a biological control agent to control vector mosquitoes or for paratransgenic approaches. However, the study
