In this work, we experimentally investigate the potency of high pressure to drive a protein toward an excited state where an inhibitor targeted for this state can bind. Ras proteins are small GTPases cycling between active GTP-bound and inactive GDP-bound states. Various states of GTP-bound Ras in active conformation coexist in solution, amongst them, state 2 which binds to effectors, and state 1, weakly populated at ambient conditions, which has a low affinity for effectors. BACKGROUND: Autologous peripheral blood stem cell (PBSC) collection can be challenging in pediatric patients with solid tumors following intensive chemotherapy. Plerixafor, a hematopoietic stem cell mobilizing agent, has been increasingly used in such cases; however, reports from Japan remain limited.
METHODS: We conducted a single-center retrospective observational study to assess the efficacy and safety of plerixafor. Eight pediatric patients with solid tumors (nine mobilization attempts) who received plerixafor at our institution between 2015 and 2024 were included in the analysis.

OBJECTIVE: Overexpression of chemokine receptor subtype 4 (CXCR4) receptors is the root cause of the metastatic spread of various cancers. The use of CXCR4 antagonists as specific ligands to inhibit the CXCR4 interaction with its only natural ligand stromal cell derived factor-1α would benefit. Plerixafor is one such Food and Drug Administration approved CXCR4 antagonist used for hematopoietic stem cell mobilization in patients. Taking the specific advantage of low molecular weight, plerixafor conjugation with different bifunctional chelating agents, was optimized.
