An efficient in vitro propagation protocol of cocoyam [Xanthosoma sagittifolium (L) Schott].

Sprouted corm sections of "South Dade" white cocoyam were potted and maintained in a greenhouse for 8 weeks. Shoot tips of 3-5 mm comprising the apical meristem with 4-6 leaf primordial, and approximately 0.5 mm of corm tissue at the base. These explants were treated to be used into the culture medium. A modified Gamborg's B5 mineral salts supplemented with 0.05 μM 1-naphthaleneacetic acid (NAA) were used throughout the study. Thidiazuron (TDZ) solution containing 0.01% dimethyl sulfoxide (DMSO) was used. Erlenmeyer flasks and test tubes were used for growing cultures. The effect of different

Improving the efficiency of isolated microspore culture in six-row spring barley: II-exploring novel growth regulators to maximize embryogenesis and reduce albinism.

Two alternative cytokinins, thidiazuron and meta-topoline, were tested in isolated microspore culture on recalcitrant barley genotypes (six-row, spring), and green plant regeneration was improved substantially. Doubled-haploid (DH) plants are coveted in plant breeding and in genetic studies, since they are rapidly obtained and perfectly homozygous. In barley, DHs are produced mainly via androgenesis, and isolated microspore culture (IMC) constitutes the method offering the greatest potential efficiency. However, IMC can often be challenging in some genotypes because of low yield of microspores

Spatial Distribution Characterization of Hormonal Metabolites in Morphologically Diverse Jujube Fruits via MALDI-MS Imaging.

Ziziphus jujuba Mill. is an important economic tree species in China with nutritional, ecological and ornamental values, and its fruits show abundant variations in size and shape. Phytohormones regulate fruit morphogenesis, yet the spatial distribution characteristics of endogenous phytohormones in jujube fruits with different shapes remain unclear. In this study, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI) was applied to explore the spatial distribution patterns of phytohormones in round, constricted and long jujube fruits. The results indicated that phyt

In Vitro Propagation of Threatened Agave Species Using a Two-Stage System.

Agaves are plants with multiple possibilities of use and are naturally tolerant to low water availability conditions and high temperatures. This makes them species of great interest in the context of the necessary substitution of crops due to climate change. Unfortunately, the overexploitation of wild specimens has endangered many species of the genus that have not been domesticated or cultivated intensively. In vitro mass culture and propagation techniques have emerged as a very efficient option to produce agave plants that can be used without damage to the natural populations. A protocol is

Evolution of plant pathogenicity in Streptomyces.

Among the multitude of soil-inhabiting, saprophytic Streptomyces species are a growing number of plant pathogens that cause economically important diseases, including potato scab. Streptomyces scabies is the dominant pathogenic species worldwide, but is only one of many that cause very similar disease symptoms on plants. Molecular genetic analysis is beginning to identify the mechanisms used by plant pathogenic species to manipulate their hosts. The nitrated dipeptide phytotoxin, thaxtomin, inhibits cellulose biosynthesis in expanding plant tissues, stimulates Ca2+ spiking, and causes cell dea

Superior In Vitro Responses of a Native Rose Genotype to Driver Kuniyuki Walnut (DKW) Medium in a Comparative Study Using Natural and Synthetic Plant Growth Regulators.

Rosa canina is one of the precious native rose rootstocks with a high reputation among plant producers, which has potential horticultural and pharmacological properties related to the cosmetic values and the production of secondary metabolites. Due to high horticultural consumption, applying the plant tissue culture technique as a major tool for healthy and massive-scale production of R. canina plants is not unexpected. However, the response of R. canina in vitro plantlets to various plant tissue culture ingredients is not well understood to tender an efficient applied protocol for qualitative

The regulatory role of phytohormones in plant drought tolerance.

This paper highlights the role of various signaling hormones in drought stress tolerance. It explains how phytohormones act and interact under drought conditions. Drought stress significantly impairs plant growth, development and productivity. The likelihood of adverse impacts of drought will increase due to variations in global climate patterns. Phytohormones serve as key regulators of drought tolerance mechanisms in plants. The in-depth understanding of the role and signaling of such hormones is thus of great significance for plant stress management. In this review, we conducted a bibliometr

Characterization of a SUPERMAN-like Gene, MdSUP11, in apple (Malus × domestica Borkh.).

Arabidopsis SUPERMAN and its family members of its family play important roles in plant growth and floral organ development; yet much less is known about their functions expanding in apple tree development. Previous work has identified 12 SUP-like genes in the apple (Malus × domestica Borkh.) genome, and the MdSUP11 which is expressed in both vegetative and reproductive organs of apple. However, the function of MdSUP11 remains obscure. In this study, the β-glucuronidase expression driven by the MdSUP11 native promoter was detected in roots, young leaves, and floral organs of transgenic Arabido

Microbial production of plant hormones: Opportunities and challenges.

Plant hormones are a class of organic substances which are synthesized during the plant metabolism. They have obvious physiological effect on plant growth at very low concentrations. Generally, plant hormones are mainly divided into 5 categories: auxins, cytokinins, ethylene, gibberellins (GAs) and abscisic acid (ABA). With the deepening of research, some novel plant hormones such as brassinosteroid and salicylates have been found and identified. The plant hormone products are mainly obtained through plant extraction, chemical synthesis as well as microbial fermentation. However, the extremely

Regulation of nonsymbiotic and truncated hemoglobin genes of Lotus japonicus in plant organs and in response to nitric oxide and hormones.

• In legumes, symbiotic leghemoglobins facilitate oxygen diffusion to the bacteroids, but the roles of nonsymbiotic and truncated hemoglobins are largely unknown. Here the five hemoglobin genes of Lotus japonicus have been functionally characterized to gain insight into their regulatory mechanisms. • Plants were exposed to nitric oxide donors, stressful conditions, and hormones. Gene expression profiling was determined by quantitative PCR, and gene activities were localized using in situ hybridization and promoter-reporter gene fusions. • The LjGLB1-1, LjGLB2, and LjGLB3-1 mRNA expression leve

Effects of copper accumulation on growth and development of Scopelophila cataractae grown in vitro.

Scopelophila cataractae was cultured in vitro for 16 weeks to assess the contrasting effects of Cu on growth and reproduction, as well as gametophore stage. To induce buds and gametophores of S. cataractae, ten treatments (tr 1 to tr 10) of culture media were prepared using a combination of mineral salts, sugar, vitamin B complex, CuSO4, and exogenous hormones. Highest numbers of gametophores and buds were formed in media containing 500 µM CuSO4 in co-application with auxin and cytokinin, as shown in the modest Cu treatments (tr 6 and tr 7, 26 per cushion and 255 per 25 mm2, respectively). The dark septate endophyte S16 enhances drought tolerance in sweet cherry by coordinating metabolic reprogramming and the transcription factor PaHB12.

Drought severely limits the growth and productivity of sweet cherry (Prunus avium L.), a fruit crop highly sensitive to water deficit. Beneficial root-associated fungi, particularly dark septate endophytes (DSEs), have emerged as potential modulators of stress tolerance, yet their underlying mechanisms in perennial fruit trees remain poorly understood. In this study, we investigated the effects of the DSE fungus Helotiales sp. S16 on drought responses of sweet cherry rootstock Gisela 5 seedlings. Fungal symbiosis was established by homogenizing fungal suspension with sterile growth substrate,

Protein networks: integrating pathways for plant heat stress adaptation.

Plants' immobility renders them highly vulnerable to heat stress, which disrupts water relations, photosynthesis, respiration, and cellular homeostasis, ultimately reducing growth and yield. To survive, plants deploy a multifaceted heat stress response (HSR) that integrates calcium signaling, molecular chaperones, antioxidant enzymes, and phytohormonal networks. This review synthesizes recent advances in understanding the molecular crosstalk between phytohormones and protein synthesis during plant heat stress responses, with a particular focus on two key HSR modules: protein synthesis pathways

Evaluating the Phytohormone Proficiencies of Multifarious Bacillus rugosus for Growth Promotion in Arachis hypogaea (L.).

The application of Plant Growth-Promoting Rhizobacteria (PGPR) in agriculture is increasingly emphasized as a sustainable alternative to hazardous agrochemicals. This study aimed to isolate and characterize PGPR strains from the rhizospheric soil of Arachis hypogaea L., hypothesizing that the rhizosphere of a healthy plant harbors beneficial microbes with significant plant growth-promoting (PGP) attributes. The isolate AB1 demonstrated promising PGP traits, including phosphate solubilization (56.44 µg mL⁻¹), zinc solubilization (6.1 µg mL⁻¹), ammonia production (3.8 µM µg mL⁻¹), and the synthe

To Fight or to Grow: The Balancing Role of Ethylene in Plant Abiotic Stress Responses.

Plants often live in adverse environmental conditions and are exposed to various stresses, such as heat, cold, heavy metals, salt, radiation, poor lighting, nutrient deficiency, drought, or flooding. To adapt to unfavorable environments, plants have evolved specialized molecular mechanisms that serve to balance the trade-off between abiotic stress responses and growth. These mechanisms enable plants to continue to develop and reproduce even under adverse conditions. Ethylene, as a key growth regulator, is leveraged by plants to mitigate the negative effects of some of these stresses on plant d

Chemical composition of thermally processed coconut water evaluated by GC-MS, UPLC-HRMS, and NMR.

Thermally-processed coconut water often develop a commercially-undesirable pink color, thus, NMR, UPLC-HRMS, GC-MS analyses combined with chemometrics approach were applied to evaluate chemical variations in comparison to tender water (control) that could explain such color change. Chemometrics on negative ionization mode dataset showed trimeric and A-type dimeric procyanidins, and caffeoylshikimic acid as main identified secondary metabolites induced by processing, while, control water presented mainly cytokinin trans-zeatin riboside, procyanidin dimer, caffeoylshikimic acid and trihydroxy-oc

The potential signalling pathways which regulate surface changes induced by phytohormones in the potato cyst nematode (Globodera rostochiensis).

It has been demonstrated that the surface lipophilicity of the plant-parasitic nematode Globodera rostochiensis decreases when infective larvae are exposed to the phytohormones indole-3-acetic acid (auxin) or kinetin (cytokinin). In the present study, it was shown that inhibition of phospholipase C (PLC) or phosphatidylinositol 3 kinase (PI3-kinase) reversed the effect of phytohormones on surface lipophilicity. The signalling pathway(s) involved in surface modification were investigated using 'caged' signalling molecules and stimulators or inhibitors of different signalling enzymes. Photolysis

Integrated metabolomic-transcriptomic profiling of dwarf Camellia reticulata 'Hentiangao' reveals key gibberellin metabolism genes and multi-hormone crosstalk under GA3 treatment.

BACKGROUND: Camellia reticulata 'Hentiangao', a dwarf camellia cultivar, possesses distinctive ornamental traits and high value for urban landscaping. Dwarfism is an important agronomic and ornamental trait, and gibberellin (GA) plays a central role in regulating stem elongation and plant height. However, the molecular mechanisms underlying its dwarf phenotype and the interactions between stem development and endogenous hormone networks remain unclear, limiting the promotion and targeted breeding of this cultivar. We analyzed shoot tips of C. reticulata 'Hentiangao' treated with 800 mg·L-1 GA3

TaGAD2 is a potential downstream effector of Rht5 in controlling wheat plant height.

Plant height is a key determinant of wheat plant architecture that affect lodging resistance and grain yield. The GA-responsive (GAR) dwarfing gene Rht5 was previously shown to decrease plant height without compromise of wheat seedling vigor and considered as a promising candidate gene for breeding wheat varieties in water-limited conditions. However, the mechanisms underlying Rht5-mediated dwarfism are unclear. In this study, we investigated the genetic effects of Rht5 on wheat growth and development using recombinant inbred lines (RILs) and found that Rht5 reduces plant height through inhibi
