The activity of a complement-dependent cytotoxic antibody in the sera of 21 melanoma patients was investigated using a microcytotoxicity assay. Heat-inactivated sera were caused to react against mechanically dispersed fresh tumor cells in the presence of exogenous blood group AB complement. The frequency of 17 HLA antigens from locus A and locus B has been evaluated by microlymphocytotoxicity test in a population of 233 subjects (157 normal and 76 with deficiency of glucose-6-phosphate dehydrogenase [G-6PD]in red cells) from a village of Sardinia, a mediterranean island relatively close to continental Italy. It appears that G-6-PD-deficient people show a frequency of some HLA antigens (A2, A10, B12, BW35) significatively different from normal Sardinian subjects but close to (A10, BW35) or higher (A2, B12) than that of subjects from peninsular Italy.

Prospective monitoring of active Chronic Epstein-Barr virally infected patients receiving gammaglobulin therapy was performed using the antibody-dependent cell-mediated cytotoxicity assay (ADCC). Efforts were made to analyze and define with respect to clinical course the autologous immunological parameters (cells and sera). Our data demonstrate that the diagnosis and clinical course of this syndrome proved inconsistent with the assays usefulness in predicting suggestive benefits of gammaglobulin therapy.

The sera of three children with chronic benign neutropenia, due to anti-neutrophil antibodies, were studied with respect to their antibody specificity. This was done by screening the sera against a panel of leukocyte donors in the EDTA micro-agglutination test and in the indirect fluorescence test. Two of the sera contained antibodies against the known neutrophil-specific antigen NA2. The third serum was directed against a new neutrophil-specific antigen. Genetic analysis showed no correlation between this antigen and the already known neutrophil-specific antigens: 9A, NA1, NA2, NB1, and NC1.

Cellular immunology is rapidly becoming an important and clinically relevant area for the practitioner of clinical laboratory immunology. At present, most of the laboratory test procedures in this area depend on observations of biologic phenomena; however, the rapid developments in this area show promise that within the next decade we will be dealing with identification and quantitation of well defined chemical entities that form the basis of cellular immune interactions. These promising developments will hopefully resolve many of the quality control problems that plague us in the laboratory.

The preparation is given of an 111indium-oxine complex that is non toxic and can label reproductively various types of cells. They retain the label for long periods (24 hours and more). This method gives parallel results for cell-mediated cytotoxicity measurements to Chromium-51 labelling and is found to have wider uses.
