Transgenic expression of dentin phosphoprotein inhibits skeletal development.

Dentin sialophosphoprotein (DSPP) is proteolytically processed into an NH2-terminal fragment called dentin sialoprotein (DSP) and a COOH-terminal fragment known as dentin phosphoprotein (DPP). These two fragments are believed to perform distinct roles in formation of bone and dentin. To investigate the functions of DPP in skeletal development, we generated transgenic mice to overexpress hemagglutinin (HA)-tagged DPP under the control of a 3.6 kb type I collagen (Col1a1) promoter (designated as Col1a1-HA-DPP). The Col1a1-HA-DPP transgenic mice were significantly smaller by weight, had smaller s

8DSS-promoted remineralization of demineralized dentin in vitro.

Dentin phosphoprotein (DPP) plays an extremely important role in the biomineralization of human tooth. The repetitive nucleotide sequence of aspartate-serine-serine (DSS) is the fundamental unit within DPP, and peptides containing 8 repeats of DSS (8DSS) have been shown to possess the ability to induce remineralization of the demineralized enamel. In this work, we coated the 8DSS peptide on the completely demineralized dentin to evaluate the effect of 8DSS peptide coating on dentin remineralization. Human dentin samples were demineralized with 37% phosphoric acid for 2 min, and then the 8DSS p

Dentin matrix protein-1 activates dental pulp fibroblasts.

INTRODUCTION: Dentin matrix protein-1 (DMP-1) is involved in the mineralization of hard dental tissues. DMP-1 is localized in several soft tissues, but its role is unclear. METHODS: Human inflamed dental pulps were collected from the endodontic clinic and human normal pulps from impacted teeth. Dental pulp cells from 8 subjects were explanted to test the effect of DMP-1 on interleukin-6 (IL-6) and IL-8 production by using enzyme-linked immunosorbent assay. RESULTS: DMP-1 was localized in pulp inflammation by using immunohistochemistry but was not present in impacted root pulps. Wherever foun

In vitro remineralization of severely compromised bonded dentin.

Biomimetic remineralization is potentially useful for the remineralization of incompletely resin-infiltrated collagen matrices created by etch-and-rinse adhesives. In this study, we tested the hypothesis that structurally altered dentin collagen cannot be remineralized to the same hierarchical order and dimension seen in structurally intact dentin collagen. The remineralization medium consisted of a set Portland cement/simulated body fluid system containing polycarboxylic acid and polyvinylphosphonic acid as biomimetic analogs. Remineralization of air-dried, collapsed hybrid layers was apparen

Hereditary opalescent dentin: a report of two cases.

AIM: The aim of this case report is to present the clinical and radiographic findings of hereditary opalescent dentin to facilitate an early diagnosis. BACKGROUND: Hereditary opalescent dentin (or dentinogenesis imperfecta) may manifest itself in three variations: i.e., Shields type I, Shields type II, and Shields type III. Dentinogenesis imperfecta occurs as an autosomal dominant trait with variable expressivity, either in presence with osteogenesis imperfecta or as a separate clinical entity in persons who have none of the features of osteogenesis imperfecta. CASE DESCRIPTION: A seven-year

Biomimetic remineralization of resin-bonded acid-etched dentin.

Degradation of denuded collagen within adhesive resin-infiltrated dentin is a pertinent problem in dentin bonding. A biomimetic remineralization scheme that incorporates non-classic crystallization pathways of fluidic amorphous nanoprecursors and mesoscopic transformation has been successful in remineralizing resin-free, acid-etched dentin, with evidence of intrafibrillar and interfibrillar remineralization. This study tested the hypothesis that biomimetic remineralization provides a means for remineralizing incompletely infiltrated resin-dentin interfaces created by etch-and-rinse adhesives.

Dentin in severe fluorosis: a quantitative histochemical study.

Dentin responds to different alterations in the enamel with hypermineralization, and is a biomarker of fluoride exposure. We hypothesized that severe fluorosis would lead to hypermineralization of the dentin when the enamel was severely affected. We used scanning electron microscopy and quantitative electron-probe microanalysis to compare dentin and enamel from healthy and fluorotic teeth. The dentin in fluorotic teeth was characterized by a highly mineralized sclerotic pattern, in comparison with control teeth (p < 0.001) and fluorotic enamel lesions (p < 0.001). Enamel near the lesions showe

Resin bonding to cervical sclerotic dentin: a review.

Several reports have indicated that resin bond strengths to noncarious sclerotic cervical dentine are lower than bonds made to normal dentine. This is thought to be due to tubule occlusion by mineral salts, preventing resin tag formation. The purpose of this review was to critically examine what is known about the structure of this type of dentine. Recent transmission electron microscopy revealed that in addition to occlusion of the tubules by mineral crystals, many parts of wedge-shaped cervical lesions contain a hypermineralised surface that resists the etching action of both self-etching pr

Remineralization of advanced root dentin lesions in vitro.

The possibility for remineralization of advanced root dentin lesions was investigated in vitro. Lesions were produced in acidic buffer solution with 0.1 and 0.5 ppm fluoride, for 3 and 5 weeks, respectively, using a single-section model. Next, the sections were immersed in a remineralization solution containing 0.1 ppm F (as NaF) for 20 weeks. Microradiographs were made after each 4-week-period. The lesions were around 500 micro m deep, with mineral loss values between 11,000 and 18000 vol%.microm. After 20 weeks of remineralization, the absolute amounts of deposited mineral ranged from 7500 t

Effect of different EGTA concentrations on dentin microhardness.

The effect of 1%, 3% and 5% EGTA (ethylene glycol-bis-(beta-amino-ethyl ether) N,N,N',N'-tetra-acetic acid) on the microhardness of root dentin of the cervical third of human teeth was studied. Five newly extracted maxillary incisors were sectioned transversely at the cementoenamel junction, and the crowns were discarded. The roots were embedded in blocks of high-speed polymerized acrylic resin and cut transversely into 1-mm sections. The second section of the cervical third of the root of each tooth was sectioned and divided into four parts. Each part was placed on an acrylic disc that was us

Type I dentin dysplasia: report of two cases.

Dentin dysplasia is a rare developmental disturbance of dentin affecting approximately 1:100,000 people. It has been classified as an autosomal dominant disease. Two distinct forms of dentin dysplasia have been described. As more cases of dentin dysplasia were reported these categories seemed inadequate; subclassification of type I dentin dysplasia were proposed based on root length and the presence or not of a pulpal remnant. This paper presents two cases demonstrating the classic features of type I dentin dysplasia in the mixed and permanent dentitions and discusses the suggested subclassifi

Further characterization of tetracycline's quantitative binding to dentin.

Unerupted human third molar crown segments were used to measure the change in the concentration of 3H-tetracycline that occurs when it moves across dentin from pulpal to occlusal surfaces. When 1.25, 2.5, and 5 x 10(-5) M 3H-tetracycline were filtered across dentin, binding increased by 21, 51, and 69%, respectively. When the same concentrations of 3H-tetracycline were preloaded onto crown segments and eluted with 1.1 x 10(-3) M nonradioactive tetracycline, the percentage of elution of 3H-tetracycline was not significantly different over time. When preloaded 3H-tetracycline was displaced by di

[Induced remineralization of carious dentin. Electronic microprobe analysis].

Our electron microprobe studies show the preferential incorporation in vitro of tin and fluorure into carious dentin. These ions do penetrate only into carious dentin through the body of the lesion. These ions do not progress through the sclerotic dentin barrier, when it exists which tend to demonstrate their affinity for demineralized tissues. Results of microprobe analysis may suggest the hypothesis of a crystalline arrangement as the product of reaction between residual apatite in carious dentin with stannous fluoride. A new crystalline phase could occur within the carious zone, up to the s

Dentin-predentin complex and its permeability: physiologic overview.

The major channels for solute diffusion across dentin are the dentinal tubules. Since dentin permeation is proportional to the product of tubule number and diameter, both of which increase as the tubules converge on the pulp, we find that dentin permeability increases rapidly as the pulp chamber is approached. The presence of a smear layer of cutting debris on top of cut dentin decreases dentin permeability, especially when permeability is measured by fluid filtration. Further, intratubular material--such as mineral deposits, collagen fibrils, proteoglycan linings, bacteria, etc.--can greatly

Periapical response to dentin filings. A pilot study.

Vital maxillary canines of mature cats were intentionally instrumented through the foramen with No. 50 K-files. Dentin chips were packed into the apical opening in one tooth of each animal. In the contralateral tooth a dentin plug was not formed. Both root canals were then filled with gutta-percha. The animals were sacrificed at 3, 5, and 8 months. The results showed quicker healing characterized by minimal inflammation and cementum deposition in the specimens where dentin fillings were packed.

Does Immediate Dentine Sealing Improve Bonding Effectiveness of Glass Ceramic Restorations Compared to Delayed Dentine Sealing?

A systematic review of the literature was conducted to determine whether immediate dentine sealing (IDS) or delayed dentine sealing (DDS) would lead to a more effective bond between dentine and indirect glass ceramic restorations. Bonding effectiveness was determined by assessing bond strength or force to fracture from the selected studies. This report followed the PRISMA Statement. A total of 10 articles were subsequently identified for inclusion. In vitro studies evaluating bond strength or force to fracture of etchable glass ceramic restorations, following a bonding protocol that incorporat

Dentine hypersensitivity: the effects of brushing desensitizing toothpastes, their solid and liquid phases, and detergents on dentine and acrylic: studies in vitro.

Dentine exhibiting symptoms of dentine hypersensitivity has tubules open at the dentine surface and patent to the pulp. The mechanisms whereby dentinal tubules are exposed is ill understood but probably involves a variety of abrasive and/or erosive agents. This study in vitro examined the quantitative and qualitative effects of toothpastes, their solid and liquid phases and detergents on dentine and acrylic. Abrasion of dentine and acrylic were measured by surfometry. Morphological changes to dentine were assessed by scanning electron microscopy. Abrasion of dentine and acrylic by toothpastes

Redistribution Effect of Demineralized Dentin on Hydrophobic Monomers.

Inadequate dentin bonding persists as a major cause of secondary caries and restoration failure, accounting for >60% of global dental restorative costs. Prevailing adhesion theory attributes this to challenges in displacing water from the demineralized dentin matrix (DDM) to infiltrate hydrophobic monomers. However, techniques such as ethanol-wet bonding, which eliminate water, yield only partial improvement, suggesting limitations in this water-centric view. Using chromatography equivalence modeling and Nile red molecular tracing, we identified a solvent-independent redistribution effect exer
