Canine cataplexy is preferentially controlled by adrenergic mechanisms: evidence using monoamine selective uptake inhibitors and release enhancers.

Narcolepsy is currently treated with anti-depressants to control REM-related symptoms such as cataplexy and with amphetamine-like stimulants for the management of sleepiness. Both stimulant and antidepressant drugs presynaptically enhance monoaminergic transmission but both classes of compounds lack pharmacological specificity. In order to determine which monoamine is selectively involved in the therapeutic effect of these compounds, we examined the effects of selective monoamine uptake inhibitors and release enhancers on cataplexy using a canine model of the human disorder. A total of 14 comp

Lisdexamfetamine dimesylate-exposition in male rats during the peripubertal period impairs inflammatory mechanisms, antioxidant activity, and apoptosis process in kidneys of male pubertal rats.

Lisdexamfetamine dimesylate (LDX) is a prodrug of dextroamphetamine, which has been widely recommended for the treatment of Attention-Deficit/Hyperactivity Disorder (ADHD). There are still no data in the literature relating the possible toxic effects of LDX in the kidney. Therefore, the present study aims to evaluate the effects of LDX exposure on morphological, oxidative stress, cell death and inflammation parameters in the kidneys of male pubertal Wistar rats, since the kidneys are organs related to the excretion of most drugs. For this, twenty male Wistar rats were distributed randomly into

Coadministration of lithium and celecoxib reverses manic-like behavior and decreases oxidative stress in a dopaminergic model of mania induced in rats.

The present study intends to investigate the effect of lithium (Li) and celecoxib (Cel) coadministration on the behavioral status and oxidative stress parameters in a rat model of mania induced by dextroamphetamine (d-AMPH). Male Wistar rats were treated with d-AMPH or saline (Sal) for 14 days; on the 8th day of treatment, rats received lithium (Li), celecoxib (Cel), Li plus Cel, or water until day 14. Levels of oxidative stress parameters were evaluated in the serum, frontal cortex, and hippocampus. d-AMPH administration induced hyperlocomotion in rats, which was significantly reversed by Li

Characterization of the Neurochemical and Behavioral Effects of Solriamfetol (JZP-110), a Selective Dopamine and Norepinephrine Reuptake Inhibitor.

Excessive sleepiness (ES) is associated with several sleep disorders, including narcolepsy and obstructive sleep apnea (OSA). A role for monoaminergic systems in treating these conditions is highlighted by the clinical use of US Food and Drug Administration-approved drugs that act on these systems, such as dextroamphetamine, methylphenidate, modafinil, and armodafinil. Solriamfetol (JZP-110) is a wake-promoting agent that is currently being evaluated to treat ES in patients with narcolepsy or OSA. Clinical and preclinical data suggest that the wake-promoting effects of solriamfetol differ from

Clinical correlates of oppositional defiant disorder and attention-deficit/hyperactivity disorder in adults.

OBJECTIVE: Oppositional defiant disorder (ODD) is a common comorbidity of attention-deficit/hyperactivity disorder (ADHD) in both children and adolescents. Although there is research demonstrating that ADHD persists into adulthood, less is known about the frequency of its persistence, clinical characteristics, and impairment when associated with comorbid ODD in adults with ADHD. METHOD: Data from a randomized clinical trial of adults with ADHD were analyzed to determine the prevalence and clinical correlates of comorbid ODD. As per the Diagnostic and Statistical Manual of Mental Disorders, Fo

Attention deficit disorder and epilepsy.

Countless studies have demonstrated that patients with epilepsy have a significant increase in behavioral disturbances of all kinds, including hyperactivity and inattention. This finding has been demonstrated in studies utilizing observer questionnaires and behavior rating scales, neuropsychological test batteries, and standardized tests of attention such as continuous performance tests. Multiple factors must be considered in the evaluation of a child with epilepsy and hyperactivity or inattention. For instance, inattention could be due to subclinical seizures, undiagnosed learning disabilitie

Attention-deficit hyperactivity disorder in adults.

It has been estimated that 30% to 70% of children who are diagnosed as having attention-deficit hyperactivity disorder (ADHD) will continue to show symptoms of the condition as adults. Since the prevalence of ADHD among school children may be 3% or more, its prevalence among adults may be 1% or 2%. The third revised edition of the Diagnostic and Statistical Manual (1987) of the American Psychiatric Association lists three essential features for the diagnosis of ADHD: "developmentally inappropriate inattention, impulsiveness, and hyperactivity." Other conditions associated with ADHD in adults i

Drug effects on orthostatic intolerance induced by bedrest.

Effective and practical preventive procedures for postflight orthostatic intolerance are highly desirable. The current practice of attempts to expand plasma volume by ingestion of salt and fluids before reentry has proven benefits. This study evaluated alternative options using fludrocortisone (F) to expand plasma volume (PV), dextroamphetamine (Dex) to enhance norepinephrine (NE) release and atropine (A) to reduce the effects of vagal stimulation. Seven subjects with proven post-bedrest orthostatic intolerance returned for a 7-day 6 degrees head-down bedrest study. F (0.2 mg) was given at 8:0

Development and validation of an automated microfluidic perfusion platform for parallelized screening of compounds in vitro.

Monoamine transporters are of great interest for their role in the physiological activity of the body and their link to mental and behavioural disorders. Currently, static well-plate assays or manual perfusion systems are used to characterize the interaction of psychostimulants, antidepressants and drugs of abuse with the transporters but still suffer from significant drawbacks caused by lack of automation, for example, low reproducibility, non-comparability of results. An automated microfluidic platform was developed to address the need for more standardized procedures for cell-based assays.

Contrasting baseline-dependent effects of amphetamine, chlorpromazine and scopolamine on response switching in the pigeon.

Both amphetamine and scopolamine increase low rates and reduce high rates of responding. It has been suggested that the dependence of the effects of these drugs on control rate may be due to their non-specific disruptive cue properties rather than to specific pharmacological actions. To examine whether this possible non-specific disruption also applied to response-choice, pigeons were trained under a schedule in which 30 key-peck responses were required. This fixed-ratio could be completed by responding in any order on either or both of two keys, and then the first switch between the two keys

Hypothermia-induced anterograde amnesia and its reversal in rats trained on a T-maze escape task.

Previous studies have demonstrated anterograde amnesia (AA) and its reversal in rats trained on passive avoidance tasks. The present investigation was conducted to determine whether induction and/or reversal of AA is limited to inhibitory learning tasks or whether these phenomena can be illustrated in behavioral situations involving choice. Accordingly, in Experiment 1, rats were trained on a T-maze escape task as either hypothermic (28 degrees C) or normothermic. Twenty-four hours later half of each acquisition group was tested as either hypothermic or normothermic. Results indicated a stern

Failure of naloxone to modify the effects of chlorpromazine and d-amphetamine on avoidance behavior in the squirrel monkey.

Lever-pressing by squirrel monkeys was maintained under a continuous avoidance schedule in which each response postponed for 30 s the delivery of an electric shock to the tail. Dose-response curves were determined for chlorpromazine (0.03--0.3 mg/kg) and d-amphetamine (0.03--1.0 mg/kg) administered alone and administered concomitantly with 1.0 or 10 mg/kg of naloxone. The dose-response curves for chlorpromazine and d-amphetamine were similar to those previously reported for monkeys under other schedules of shock-maintained behavior: Chlorpromazine decreased responding in a dose-related manner

Tyramine-binding by synaptosomes from rat brain: effect of centrally active drugs.

Incubation of p-tyramine (TRM) with rat midbrain plus corpus striatum (MB+ CS) crude synaptosomal preparations, under conditions which reduce to a minimum amine uptake, results in appreciable binding of this amine by synaptosomes. This process is inhibited by preincubation with a number of drugs active in the CNS, e.g., chlorpromazine, desipramine, d-amphetamine, and diphenhydramine. Morphine, however, does not affect this binding. The Ki for each one of these compounds, as well as the K association constant and concentration of the binding sites of TRM, were determined. These results suggest

Comparison of the time course of the anorectic effect of fenfluramine and amphetamine with drug levels in blood.

Time courses of the suppressive effects on food intake of (+)-amphetamine and (plus or minus)-fenfluramine in deprived rats were found to be different. Amphetamine displayed a potent initial action which rapidly decayed, and this behavioural effect was consistent with the measured blood concentration of amphetamine which showed a peak at 1 h followed by rapid clearance. For fenfluramine, the initial suppression of eating was maintained over several hours and was, for the first hour, related to the blood concentration of fenfluramine but later to an active metabolite, norfenfluramine. The study

Morphine enhancement of shuttle avoidance prevented by alpha-methyltyrosine.

Rats were given 6 days of training (180 trials) to make a two-way (shuttle) avoidance response. "Poor performers" making less than 50% avoidances in the 6th block of 30 tials were given drug treatments on the next 2 days. Both morphine sulfate (15 mg/kg) and d-amphetamine sulfate (1 mg/kg) increased the avoidance level by 70-100%. Prior treatment with alpha-methyltyrosine (50 mg/kg) prevented the effects of both drugs. Nalorphine hydrochloride (5 mg/kg) also blocked the effect of morphine. The action of alpha-methyltyrosine to block enhancement of avoidance is taken to indicate that this effec

Effects of d-amphetamine and chlordiazepoxide on spaced responding in pigeons.

The effects of d-amphetamine and chlordiazepoxide were studied in pigeons on performance (1) under a schedule that reinforced responses on a key (food key) if they were more than 20 sec apart, (2) under the same schedule when responses also were required on a collateral key during the interresponse time on the food key, and (3) under the same schedule when responses were required on a collateral key during the interresponse time on the food key and collateral-key responses could produce a stimulus correlated with the availability of food. Under all three spaced-responding schedules, d-amphetam

Using a controlled stimulant to treat cocaine use disorder in the setting of attention deficit/hyperactivity disorder: a case study.

Cocaine Use Disorder (CUD), like all other stimulant use disorders, remains a challenging condition to treat, particularly in the absence of FDA-approved pharmacotherapies. While contingency management (CM) is considered first-line treatment and has demonstrated strong evidence for efficacy, it remains underutilized due to financial, philosophical, and logistical barriers. Off-label use of psychostimulants has emerged as a potential therapeutic strategy, especially in patients with comorbid attention-deficit/hyperactivity disorder (ADHD). We present a case of a middle-aged male with a long-sta

Sleep- and Wake-Promoting Drugs: Where Are They Being Sourced, and What Is Their Impact?

Background: Recent decades have seen both an increased number of shift workers in order to deliver services 24/7, and increased potential for social interactions at all hours of the day. People have sought to engage in strategies, which either promote vigilance or facilitate sleep, with the use of sleep- and wake-promoting drugs representing one strategy. Methods: We investigated use of sleep- and wake-promoting drugs in participants (n = 377) who completed a survey investigating the type and source of sleep- and wake-promoting drugs, and their impact on sleep and performance outcomes.
