Diosgenin suppresses COX-2 and mPGES-1 via GR and improves LPS-induced liver injury in mouse.

Using a wild yam (Dioscorea japonica), we previously found novel anti-inflammatory and anti-carcinogenic effects via the downregulation of cyclooxygenase (COX)-2 and microsomal prostaglandin E synthase (mPGES)-1. One of the substances in wild yam is a steroidal saponin, diosgenin. We demonstrated that diosgenin suppressed COX-2 in human non-small-cell lung carcinoma A549 cells via nuclear factor-kappa B (NF-κB) translocation and the effects were reversed by a glucocorticoid receptor antagonist, RU486. In lipopolysaccharide (LPS)-induced mouse liver injury, COX-2 and mPGES-1 were induced and lo

Diosgenin attenuates hepatic stellate cell activation through transforming growth factor-β/Smad signaling pathway.

Activation of hepatic stellate cells (HSC) plays a pivotal role in the development of hepatic fibrosis. Transforming growth factor-β1 (TGF-β1) is considered to be the main stimuli factor responsible for the activation of HSC. Diosgenin is a steroidal saponin found in several plants including Solanum and Dioscorea species, and it inhibited high glucose-induced renal tubular fibrosis. However, the effects of diosgenin against hepatic fibrosis remain elusive. Therefore, in this study, we investigated the effects of diosgenin on TGF-β1-induced HSCs and elucidate the possible mechanism of its anti-

Evidence for Polyamine, Biogenic Amine, and Amino Acid Adduction Resulting from Metabolic Activation of Diosbulbin B.

Dioscorea bulbifera L. (DBL), a traditional Chinese medicine, is a well-known herb with hepatotoxicity, and the biochemical mechanisms of the toxic action remain unknown. Diosbulbin B (DSB), a major component of DBL, can induce severer liver injury which requires cytochrome P450-catalyzed oxidation of the furan ring. It is reported that a cis-enedial reactive intermediate resulting from metabolic activation of DSB can react with thiols and amines to form pyrrole or pyrroline derivatives. In this study, we investigated the interaction of the reactive intermediate with polyamines, biogenic amine

Prophylactic oral diosgenin attenuates acute sleep deprivation-induced liver injury in mice via modulation of the gut microbiota and vascular barrier.

Acute sleep deprivation (SD) is an increasingly prevalent health risk, yet the early inflammatory processes associated with acute hepatic injury following SD remain largely elusive. Following a two-step screening of 265 natural products and bioactive small molecules, diosgenin (DG), a sapogenin found in Dioscorea species, was prioritized for subsequent in vivo evaluation. Under the rotating-platform paradigm in mice, 24-hour SD (SD24h) was associated with acute hepatic inflammation, increased overall intestinal permeability, gut vascular barrier (GVB)-associated alterations, gut microbiota dys

An integrated approach to safer plant production on metal contaminated soils using species selection and chemical immobilization.

In order to examine the species specific accumulation of heavy metals in medicinal crops, seven different common medicinal plants were cultivated on a Cd (55mgkg(-1)) and Pb (1283mgkg(-1)) contaminated soil. Subsequently, the effect of various immobilizing agents, applied in isolation and in combination, on Cd and Pb uptake by two medicinal plant species was examined. Cadmium and Pb root concentrations in medicinal plants grown in the control soil varied between 0.5 and 2.6mgkg(-1) for Cd and 3.2 and 36.4mgkg(-1) for Pb. The highest accumulation occurred in Osterici Radix (Ostericum koreanum)

Effects of Heat-Moisture Treatment on the Physicochemical Properties, Structural Characteristics, and Anti-Ulcerative Colitis Activity of Yam Starch.

Yam starch (YS) alleviates ulcerative colitis (UC), but whether heat-moisture treatment (HMT) enhances this effect remains unclear. This study examined the impact of HMT on the structural features of yam starch and yam flour (YF) and their UC-mitigating capacity. The results showed that HMT increased the resistant starch (RS) content in YF by forming starch-lipid complexes and, in YS, it enhanced the RS content through recrystallization. Animal experiments demonstrated that YF, HMT-modified YF, YS, and HMT-modified YS all alleviate UC symptoms, with HMTYS being the most effective. Specifically

Methyl Protodioscin Promotes Ferroptosis of Prostate Cancer Cells by Facilitating Dissociation of RB1CC1 from the Detergent-Resistant Membranes and Its Nuclear Translocation.

Methyl protodioscin (MPD), a furostanol saponin found in the rhizomes of Dioscorea plants, has been shown to effectively inhibit proliferation of prostate cancer cells in vitro and in vivo. However, the mechanism underlying this inhibitory action remains unclear. To elucidate the mechanism, we used mass spectrometry to analyze protein rearrangements in detergent-resistant membranes (DRMs). Ferroptosis-related factors were identified in cells in vitro and in vivo. MPD induced the expression of acyl-CoA synthetase long chain family member 4 and reduced expression levels of glutathione peroxidase

DNA damage by reactive oxygen species resulting from metabolic activation of 8-epidiosbulbin E acetate in vitro and in vivo.

8-Epidiosbulbin E acetate (EEA), a furan-containing diterpenoid lactone, is one of main component of Dioscorea bulbifera L. (DBL). It has been reported that EEA induces severe hepatotoxicity in mice and that its hepatotoxicity is associated with metabolic activation. The present study demonstrated that exposure to EEA (50, 100 or 200 μM) induced DNA damage, including significant DNA fragmentation, increases of tail DNA and olive tail moment, H2AX phosphorylation and PARP-1 activation, in cultured mouse primary hepatocytes. Similar observation was obtained in mice administered EEA at 50, 100 or
