Genetic and molecular basis for low efficacy of vaccine strains against canine distemper virus in Brazil.

Canine distemper virus, CDV, is a worldwide distributed disease of the genus Morbillivirus that can affect dogs of all ages, breeds, and both sexes with varying degrees of morbidity and lethality. The virus consists of six structural proteins, of which the Hemagglutinin is responsible for the efficient fusion to the cell membrane, allowing the virus entrance and replication in susceptible animals. The Hemagglutinin protein is responsible for the virus binding to the SLAM and Nectin-4 proteins present on the host cell membrane to start the infection process. This biochemical mechanism is then u

Risk of canine distemper virus vaccination of domestic dogs in giant panda habitat to giant pandas.

The giant panda (Ailuropioda melanoleuca), a unique relic species in China and a global biodiversity conservation symbol, faces the threat of canine distemper virus (CDV). Vaccinating domestic dogs in panda habitats against CDV is crucial, yet the associated risks remain understudied. We investigated the safety of CDV vaccination in 69 domestic dogs within panda habitats, employing enzyme-linked immunosorbent assay for CDV antibodies and quantitative reverse transcription polymerase chain reaction for viral RNA, a marker of viral shedding. Results revealed that vaccinated dogs posed a risk thr

Meningoencephalitis caused by concurrent infection with canine distemper virus and a unique Sarcocystis sp. in a gray fox.

A deceased 9-wk-old male gray fox (Urocyon cinereoargenteus) with a history of decreased ambulation and diarrhea was submitted to the Texas A&M Veterinary Medical Diagnostic Laboratory. No significant gross findings were evident on postmortem examination. Histologically, the cerebrum and brainstem had mild necrotizing meningoencephalitis with protozoal schizonts and merozoites. Additionally, glial cells contained intracytoplasmic and intranuclear viral inclusion bodies. Sections of the cerebrum were positive for canine distemper virus (CDV) and negative for Sarcocystis neurona on immunohistoch

Coinfection of Canine Distemper Virus and Rabies Virus in Wildlife Samples Submitted for Routine Rabies Testing.

Canine distemper virus (also known as Canine morbillivirus), the etiologic agent of canine distemper, is a highly contagious pathogen causing a multisystemic infection in carnivores globally. Canine distemper may be clinically indistinguishable from rabies, and outbreaks of either disease are major concerns. In the US, both diseases are endemic and managed by parenteral vaccination in domestic animals. In wildlife, oral vaccination and trap-vaccinate-release programs are available for rabies prevention, but no such strategies exist for canine distemper. We evaluated the prevalence at which can

Pyogranulomatous pleuropneumonia caused by Nocardia asiatica in a dog coinfected with canine morbillivirus (canine distemper virus).

Nocardia asiatica causing pyogranulomatous pleuropneumonia is reported for the first time in a dog coinfected with canine morbillivirus (CM), diagnosed based on epidemiological, clinical, haematological, images, microbiological, histopathological, polymerase chain reaction and hsp65 gene sequencing findings. The immunosuppression of CM probably favoured the opportunistic behaviour of N. asiatica. Despite the therapeutic measures, the animal died, mainly due to respiratory distress. The association of methods to improve early diagnosis, therapy procedures and prognosis of canine nocardiosis is

Use of serologic tests to predict resistance to Canine distemper virus-induced disease in vaccinated dogs.

The objective of the current study was to determine whether detection of Canine distemper virus (CDV)-specific serum antibodies correlates with resistance to challenge with virulent virus. Virus neutralization (VN) assay results were compared with resistance to viral challenge in 2 unvaccinated Beagle puppies, 9 unvaccinated Beagle dogs (4.4-7.2 years of age), and 9 vaccinated Beagle dogs (3.7-4.7 years of age). Eight of 9 (89%) unvaccinated adult dogs exhibited clinical signs after virus challenge, and 1 (13%) dog died. As compared to adult dogs, the 2 unvaccinated puppies developed more seve

Zymographic patterns of MMP-2 and MMP-9 in the CSF and cerebellum of dogs with subacute distemper leukoencephalitis.

Distemper leukoencephalitis is a disease caused by the canine distemper virus (CDV) infection. It is a demyelinating disease affecting mainly the white matter of the cerebellum and areas adjacent to the fourth ventricle; the enzymes of the matrix metalloproteinases (MMPs) group, especially MMP-2 and MMP-9 have a key role in the myelin basic protein fragmentation and in demyelination, as well as in leukocyte traffic into the nervous milieu. To evaluate the involvement of MMPs during subacute distemper leukoencephalitis, we measured the levels of MMP-2 and MMP-9 by zymography in the cerebrospina

Rabies and canine distemper virus epidemics in the red fox population of northern Italy (2006-2010).

Since 2006 the red fox (Vulpes vulpes) population in north-eastern Italy has experienced an epidemic of canine distemper virus (CDV). Additionally, in 2008, after a thirteen-year absence from Italy, fox rabies was re-introduced in the Udine province at the national border with Slovenia. Disease intervention strategies are being developed and implemented to control rabies in this area and minimise risk to human health. Here we present empirical data and the epidemiological picture relating to these epidemics in the period 2006-2010. Of important significance for epidemiological studies of wild

Comparison of in vitro replication and cytopathology caused by strains of canine distemper virus of vaccine and field origin.

Three biological properties of canine distemper virus were examined to determine if any would consistently differentiate field from vaccine strains of the virus. The properties were the ability to (1) infect macrophages and epithelial cells, (2) produce distinct cytopathologic effect in alveolar and peritoneal macrophages and Vero cells, and (3) produce pocks on the chorioallantoic membrane of embryonated chicken eggs. Four vaccine strains and 5 field isolates were used in the study. The 5 field isolates were obtained directly from canine tissues. Of the 3 properties studied, only the comparis

A radioimmunoassay search for measles and distemper antigens in subacute sclerosing panencephalitis and multiple sclerosis brain tissues.

Solid-phase direct sandwich radioimmunoassays have been developed which will detect measles antigen in as little as 20 microgram of SSPE brain, measles antigen in 5 x 10(4) pfu/ml of measles virus, and canine distemper antigen in 5 x 10(5) pfu/ml of this virus. Using these assays, a search was carried out for measles and distemper antigens in CNS autopsy tissue from 3 multiple sclerosis (MS) patients. Even in 1000-fol higher concentrations of MS brain (20 mg), antigens of neither virus were detected. Additional studies were carried out using similar radioimmunoassays but employing IgG from 2 M

Molecular Evidence of Canine Distemper Virus Circulation in Red Foxes and Golden Jackals from Western Romania.

Canine distemper virus (CDV) is a highly contagious multi-host pathogen that poses a significant threat to both domestic and wild carnivore populations. The increasing overlap between wildlife habitats, peri-urban environments, and domestic animal populations has raised concerns regarding the epidemiological role of wild canids in maintaining and disseminating CDV. This study investigated the occurrence of CDV infection among free-ranging wild canids from western Romania. Brain tissue samples collected from 64 free-ranging wild canids, including 38 red foxes (Vulpes vulpes) and 26 golden jacka

Fusion protein and hemagglutinin of canine distemper virus co-induce apoptosis in canine mammary tumor cells.

BACKGROUND: Canine distemper virus (CDV) has been shown to have oncolytic activity against primary canine tumors. Previous studies from this laboratory had confirmed that CDV induces apoptosis in canine mammary tumor (CMT) cells, although the molecular mechanism remains unknown. METHODS: The CDV N, P, M, F, H, L, C, and V genes were identified in CDV-L and cloned separately. Mutants with deletions in the 5' region (pCMV-F L△60, pCMV-FL△107, and pCMV-FL△114) or with site-directed mutagenesis in the 3' region (pCMV-FLA602-610) of the F gene were generated. Comparison of immunity against canine distemper, adenovirus and parvovirus after vaccination with two multivalent canine vaccines.

BACKGROUND: Viral diseases are a major cause of morbidity and mortality in puppies. There is a belief among veterinary practitioners and even educational institutions that the vaccines made in Brazil against canine distemper virus (CDV), canine parvovirus (CPV) and canine adenovirus (CAV) are ineffective or only partially effective. OBJECTIVES: This study aimed at comparing the immunity of two multivalent vaccines in adult dogs in the city of Uberlândia, Minas Gerais state, Brazil. METHODS: The study was carried out at the Animal Protection Association and a total of 60 adult mongrel dogs we

Pathogenesis and phylogenetic analyses of canine distemper virus strain ZJ7 isolate from domestic dogs in China.

A new isolate of canine distemper virus (CDV), named ZJ7, was isolated from lung tissues of a dog suspected with CDV infection using MDCK cells. The ZJ7 isolate induced cytopathogenic effects of syncytia in MDCK cell after six passages. In order to evaluate pathogenesis of ZJ7 strain, three CDV sero-negative dogs were intranasally inoculated with its virus suspension. All infected dogs developed clinical signs of severe bloody diarrhea, conjunctivitis, ocular discharge, nasal discharge and coughing, fever and weight loss at 21 dpi, whereas the mock group infected with DMEM were normal. Persistence of Infectious Canine Distemper Virus in Murine Xenotransplants of Canine Histiocytic Sarcoma Cells after Intratumoral Application.

Oncolytic viruses and morbilliviruses in particular, represent an interesting therapeutic approach for tumors with a poor prognosis and frequent resistance to conventional therapies. Canine histiocytic sarcomas (HS) exemplify such a neoplasm in need for new curative approaches. Previous investigations demonstrated a limited success of an acute intratumoral application of canine distemper virus (CDV) on xenotransplanted canine histiocytic sarcoma cells (DH82 cells), while persistently CDV-infected DH82 cell transplants exhibited a complete spontaneous regression. Therefore, the present study fo

Canine distemper virus and canine adenovirus type-2 infections in neotropical otters (Lontra longicaudis) from Southern Brazil.

All descriptions of infectious diseases affecting otters were published in the Northern Hemisphere, with no occurrence identified in neotropical otters (Lontra longicaudis). Consequently, a retrospective histopathological study using archival tissue samples from six free-living neotropical otters was done to investigate the possible occurrence of disease patterns associated with common viral infectious disease agents of the domestic dogs. Immunohistochemical (IHC) assays were designed to identify intralesional tissue antigens of canine distemper virus (CDV), and canine adenovirus-1 (CAdV-1) an
