Upstream open reading frames (uORFs) are intrinsic regulatory elements that fine-tune gene expression at the translational level without altering genomic sequences or protein structures. Here, we targeted the conserved uORF that resides in the 5'-untranslated region (5'-UTR) of a GDP-galactose phosphorylase gene (GhGGP1) in cotton (Gossypium hirsutum). Regulation of GGP1 is a key control point of plant L-ascorbic acid biosynthesis. Using a dual-luciferase reporter system, we demonstrate that this uORF represses translation of the downstream GGP1 coding sequence in a feedback manner in response

Microbial growth is central to soil carbon cycling. However, how microbial communities grow under climate change is still largely unexplored. Here we use a unique field experiment simulating future climate conditions (increased atmospheric CO2 and temperature) and drought concomitantly and investigate impacts on soil microbial activity. We trace 2H or 18O applied via water-vapor exchange into membrane (and storage) fatty acids or DNA, respectively, to assess community- and group-level adjustments in soil microbial physiology (replication, storage product synthesis, and carbon use efficiency).

Drought stress is a major constraint for tomato production, affecting plant water status, growth, and physiological performance. Grafting onto genetically diverse rootstocks offers a promising strategy to improve drought adaptation. Based on previous evaluations, interspecific tomato MAGIC-derived lines (ToMAGIC) showing contrasting drought responses were selected as candidate tolerant and susceptible genotypes. We hypothesized that these contrasting genotypes would differentially modulate the drought response of a grafted Moneymaker scion when used as rootstock, and that the selected tolerant

Tetraploidization was induced in the drought-tolerant tomato landrace 'de Ramellet' to evaluate its physiological and anatomical response under well-watered (WW) and water-deficient (WD) conditions. Under WW, tetraploid plants exhibited approximately 40% lower stomatal density and approximately 80% larger stomata than diploids. Net photosynthetic rate (P N), intrinsic water-use efficiency, and intercellular CO2 concentration remained unchanged between diploids and tetraploids. Under WD, both genotypes reduced P N and stomatal conductance by similar proportions; however, only diploids decreased

Drought is one of the most detrimental factors that causes significant effects on crop development and yield. However, the negative effects of drought stress may be alleviated with the aid of exogenous melatonin (MET) and the use of plant-growth-promoting bacteria (PGPB). The present investigation aimed to validate the effects of co-inoculation of MET and Lysinibacillus fusiformis on hormonal, antioxidant, and physio-molecular regulation in soybean plants to reduce the effects of drought stress. Therefore, ten randomly selected isolates were subjected to various plant-growth-promoting rhizobac

Drought alters both plant growth and biomass accumulation and allocation, but how molecular responses and functional traits jointly reflect these changes in woody plants remains insufficiently understood. We investigated whether plant ecological strategies, quantified using the competitive-stress tolerant-ruderal (CSR) framework, could provide an integrative description of drought-associated biomass responses in mulberry (Morus atropurpurea) seedlings. In this study, mulberry seedlings were maintained under well-watered, mild-drought, or moderate-drought conditions for 30 days, after which bio

Drought stress is a primary abiotic constraint limiting potato productivity. While polyphenol oxidase (PPO) is known to participate in stress responses, the specific role of StuPPO9 in drought tolerance remains poorly understood. In this study, we generated StuPPO9-overexpressing (OE) and CRISPR/Cas9-mediated knockout (C4) lines in the potato cultivar 'Atlantic'. Under sustained drought stress, OE lines exhibited significantly superior growth phenotypes compared to wild-type (WT) and C4 plants, characterized by increased leaf and root relative water content, root number and enhanced photosynth

The ECERIFERUM (CER) gene family is essential for the biosynthesis of plant cuticular wax. In this study, 32 BoCER genes were identified in cabbage through genome-wide analysis. We found that the BoCER genes are highly conserved with their homologous counterparts in Arabidopsis thaliana. However, there was a significant divergence in the expression pattern among the BoCER paralogs, which suggests the occurrence of functional specialization during evolution. The expression analysis also showed that most of the BoCER genes are expressed in the aboveground part. Cis-regulatory element analysis su

Drought stress is a major constraint in jujube production. Although abscisic acid (ABA) and cytokinin exert opposing effects on plant growth, recent findings revealed that their combined application enhanced drought tolerance in jujube; however, the underlying mechanism remains elusive. In this study, three exogenous treatments were applied to sour jujube seedlings under drought stress: 5 mg L-1 ABA, a cytokinin mixture of 50 mg L-1 6-benzylaminopurine (6-BA) and 100 mg L-1 kinetin (KT), and a combined ABA and cytokinins solution. Transcriptomic and metabolomic analyses were integrated to eluc

Increased evidence has shown that hydrogen sulfide (H2S), a novel gasotransmitter, could enhance drought resistance in plants by inducing stomatal closure, with concurrent enhancement of photosynthetic efficiency, but little is known about the mechanism behind this contradictory phenomenon. This study examined the regulating mechanism of H2S in response to drought stress from stomatal and non-stomatal factors in Chinese cabbage. The results showed that exogenous H2S could increase the accumulation of photosynthetic pigments and alleviate the damage caused by drought stress. It also regulated t

INTRODUCTION: Soybean is an indispensable crop supplying protein and oil for humans and animals, and playing an essential role in global food security. Drought represses soybean seed germination, reducing biomass accumulation and even inhibiting yield.
METHODS: In order to dissect the genetic components underlying soybean drought tolerance during different development stage, a natural population containing 140 accessions was employed to evaluate seven drought tolerance-related traits under water-welled and drought stress conditions. Subsequently, genome-wide association study (GWAS) was condu

INTRODUCTION: Drought is one of the most severe abiotic stresses limiting crop productivity, and abscisic acid (ABA)-induced stomatal closure is a key mechanism underlying plant drought resistance. Although ABA signaling has been extensively characterized in Arabidopsis thaliana, the role and regulatory mechanisms of microfilaments (F-actin) in this process remain poorly understood. Previously, we identified the actin-binding protein CAP1 as an interactor of the ABA receptor RCAR12 and a phosphorylation target of OPEN STOMATA 1 (OST1), suggesting a potential link between actin dynamics and ABA

To evaluate the effect of drought stress on some physiological characteristics of six endophyte-infected (E+) genotypes and four endophyte-free (E-) genotypes of Lolium perenne, an experiment was carried out within a completely randomized design in three replications. In pot culture, one- month stress conditions for E+ and E- genotypes of L. perenne were handled by limiting irrigation to fulfill 20% field capacity of the soil. The physiological characteristics of E+ and E- genotypes of L. perenne indicated a better resistance of E + genotypes under drought stress in terms of higher biomass and

Global phosphorus scarcity and the environmental impacts of chemical fertilizers necessitate sustainable microbial alternatives for agriculture. We characterized Burkholderia gladioli ST3M-39a, a maize rhizosphere isolate, as a multifunctional plant growth-promoting rhizobacterium with exceptional climate resilience. The strain achieved rapid phosphate solubilization (177.96 ± 5.26 µg/mL within 24 h; molybdenum-antimony assay), zinc solubilization, and ammonia production, EPS production, produced stress-alleviating enzymes (cellulase and protease), and biosurfactant production. Crucially, it m

Drought stress is a major environmental constraint limiting growth, water relations, and metabolic stability in pepper (Capsicum annuum). This study evaluated the individual and combined effects of melatonin (MEL) and salicylic acid (SA) on drought tolerance by assessing growth, water status, photosynthetic pigments, oxidative stress markers, osmotic adjustment, and antioxidant defence systems. Plants were grown under non-stressed and drought-stressed conditions and treated with MEL, SA, or their combination. Drought stress significantly reduced fresh weight, relative water content, and photos

Global cotton production often experiences significant obstacles from climatic variability, particularly due to drought stress. The present study evaluated the efficacy of seed priming with chitosan nanoparticles (CNPs) as a sustainable strategy to mitigate drought stress in cotton under rainfed ecology of cotton cultivation in India. The laboratory synthesized chitosan nanoparticles using the ionic gelation method with trisodium polyphosphate (TTP) as a crosslinker were characterized through transmission electron microscopy (Joel JEM 2100 Plus) and particle size analyser (Malvern Zetasizer Na

Mangrove species mainly inhabit tropical and subtropical intertidal zones, where they suffer periodic tidal inundation. They display diverse perforation-plate types and pit structures; however, the extent to which these anatomical features contribute to their adaptation to coastal environments remains unclear. In this study, we quantified 27 traits, representing hydraulics, anatomy and pit characteristics, in eight woody mangrove species from Hainan, Southern China, including four simple-perforation (SI) and four scalariform-perforation (SC) plate species. All species exhibited high xylem embo

Drought stress exacerbates non-stomatal water loss, which hinders agricultural growth and global food security. The cuticle, a layer of cutin, waxes, and other polymers, protects plants from the negative impact of drought. One crucial adaptation that has enabled terrestrial plants to survive and flourish in drought-prone regions is the development of cuticle structures. In this review, we (1) provide an overview of the molecular and enzymatic pathways involved in the biosynthesis of cutin and wax; (2) analyze important regulatory networks that control the formation of cuticles, such as abscisi

Common bean productivity in arid and semi-arid regions is frequently constrained by water deficit and micronutrient limitations. This study aimed to (i) quantify the effects of deficit irrigation and foliar micronutrient application on growth, yield, and water productivity, and (ii) elucidate the physiological and biochemical mechanisms underlying genotype-specific drought tolerance. Field experiments were conducted over two growing seasons on sandy soil using three common bean varieties (Nebraska, Giza 3, and Giza 6). Treatments were arranged in a split-split-plot randomized complete block de

The first T2T reference genome of Guazuma ulmifolia is reported, which serves as a core genomic resource for stress adaptation research and stress-tolerant breeding in cacao wild relatives. Climate change, particularly increased incidence of drought, poses a major threat to food security. Understanding the genomic basis of environmental adaptation in crop wild relatives can provide valuable resources for improving stress resilience. Guazuma ulmifolia, a wild relative of Theobroma cacao with important ecological and medicinal value, lacks high-quality reference genomic resources. Here, we repor
