Hypoxia initiates the neurosecretory response of the carotid body (CB) by inhibiting one or more potassium channels in the chemoreceptor cells. Oxygen-sensitive K(+) channels were first described in rabbit CB chemoreceptor cells, in which a transient outward K(+) current was reported to be reversibly inhibited by hypoxia. Although progress has been made to characterize this current with electrophysiological and pharmacological tools, no attempts have been made to identify which Kv channel proteins are expressed in rabbit CB chemoreceptor cells and to determine their contribution to the native

Nuclear receptors (NRs) encompass a superfamily of cytoplasmic/nuclear localized receptors that on ligand binding (or by phosphorylation) directly regulate the transcription of target genes. NRs are involved in many developmental processes, including moulting in insects and dauer larva formation in Caenorhabditis elegans. Here we report the isolation of two genes related to NRs from the filarial nematode Brugia pahangi. Bp-nhr-1 is a member of the NGF1-B subfamily of NRs and is expressed at very low levels in post-infective larval stage 3 (L3) after their transmission to the mammalian host. Th

Female mosquitoes are vectors of many devastating human diseases because they require blood feeding to initiate reproduction. Thus, elucidation of molecular mechanisms managing female mosquito reproduction is essential. Although the regulation of gene expression during the mosquito gonadotrophic cycle has been studied in detail, how this process is controlled at the chromatin level remains unclear. Chromatin must be accessible for transcription factors (TFs) governing gene expression. A specialized class of TFs, called pioneer factors (PFs), binds and remodels closed chromatin, permitting othe

BLTY is a pepper cultivar that has resistance to young larvae of Spodoptera litura Fabricius, whereas FXBX is a pepper cultivar suitable for the growth of S. litura larvae. The obtained microbiomics data suggest that the BLTY leaves fed S. litura larval guts inhabited more Brucella. In this study, two Brucella spp. (2FC-4 and 2FC-15) and one Microbacterium sp. (2FC-22) were isolated from the guts of BLTY and FXBX leaf-fed S. litura, respectively. The loading of 2FC-4 and 2FC-15 reduced the survival rate, average larval weight, diet consumption, pupation rate, and emergence rate of S. litura, w

The arthropod-specific hormone family of ecdysteroids plays an important role in regulating diverse physiological processes, such as moulting and metamorphosis, reproduction, diapause and innate immunity. Ecdysteroids mediate their response by binding to a heterodimeric complex of two nuclear receptors, the ecdysone receptor (EcR) and the retinoid-X-receptor/ultraspiracle (RXR/USP). In this study we investigated the role of EcR and RXR in metamorphosis and development of the desert locust, Schistocerca gregaria. The desert locust is a voracious, phytophagous, swarming pest that can ruin crops

The effect of xenobiotics (phenobarbital and atrazine) on the expression of Drosophila melanogaster CYP genes encoding cytochromes P450, a gene family generally associated with detoxification, was analyzed by DNA microarray hybridization and verified by real-time RT-PCR in adults of both sexes. Only a small subset of the 86 CYP genes was significantly induced by the xenobiotics. Eleven CYP genes and three glutathione S-transferases (GST) genes were significantly induced by phenobarbital, seven CYP and one GST gene were induced by atrazine. Cyp6d5, Cyp6w1, Cyp12d1 and the ecdysone-inducible Cyp

FTZ-F1, a member of the nuclear receptor superfamily, has been implicated in the activation of the segmentation gene fushi tarazu during early embryogenesis of Drosophila melanogaster. We found that an isoform of FTZ-F1, betaFTZ-F1, is expressed in the nuclei of almost all tissues slightly before the first and second larval ecdysis and before pupation. Severely affected ftz-f1 mutants display an embryonic lethal phenotype, but can be rescued by ectopic expression of betaFTZ-F1 during the period of endogenous betaFTZ-F1 expression in the wild type. The resulting larvae are not able to molt, but

The insulin superfamily constitutes a phylogenetically conserved hormonal system that critically regulates growth, metabolism, and reproduction in Metazoa. However, the functional diversification and the architecture of its regulatory networks of this superfamily remains poorly characterized in crustaceans. Here, we identify and characterize two little-studied insulin superfamily genes in the Pacific white shrimp Litopenaeus vannamei, gonadulin (LvGon) and insulin-related peptide (LvIRP). LvGon is predominantly expressed in neural tissues (brain and thoracic ganglia), while LvIRP is specifical

BACKGROUND: Natural products are important sources of insecticidal lead compounds and are central to the development of green pesticides. Arecoline, the primary alkaloid in betel nut, shows insecticidal activity against Aedes aegypti. This study aimed to synthesize arecoline benzyl ester derivatives with improved activity and to investigate how they act by interfering with hormone signaling pathways and chitin metabolism in Ae. aegypti.
RESULTS: This study synthesized 22 arecoline benzyl ester derivatives and evaluated for insecticidal activity against fourth-instar Ae. aegypti larvae. Among

Maintenance of somatic sex identity is essential for adult tissue function. In the Drosophila testis, adult somatic stem cells known as cyst stem cells (CySCs) require the transcription factor Chinmo to preserve male identity. Loss of Chinmo leads to reprogramming of CySCs into their ovarian counterparts through induction of the female-specific RNA-binding protein TransformerF (TraF), though the underlying mechanism has remained unclear. Here, we identify the pioneer transcription factor Zelda (Zld) as a critical mediator of this sex reversal. In wild-type CySCs, zld mRNA is repressed by micro

The dried rhizomes of Paris yunnanensis Franch. have been extensively utilized in traditional Chinese medicine as hemostatic, antitumor, and antimicrobial agents. An examination of classical texts and renowned Chinese medical formulations showcased its efficacy in acne treatment. Presently, there is a significant scarcity of Paris resources. Consider directing attention towards the non-medicinal parts of Paris to mitigate the strain on medicinal resources within this realm. To address these resource limitations, this study investigated the bioactivity and pharmacodynamics of the above-ground p

Chitin synthase is the key regulatory enzyme for chitin synthesis and excretion in insects, as well as a specific target of insecticides. The chitin synthase A gene (BmChsA) cloned from Bombyx mori, the model species of lepidopteran, is an epidermis-specific expressed gene during the molting stage. Knockdown BmChsA gene in 3rd instar larvae increased the number of non-molting and abnormal molting larvae. Exposure to nikkomycin Z, a chitin synthase inhibitor downregulated the expression of BmChsA and decreased the amount of epidermis chitin during the molting process. The thickness of the new e

The crustacean neuropeptide, molt-inhibiting hormone (MIH), directly inhibits Y-organ ecdysteroidogenesis, an effect mediated by cyclic AMP (cAMP) and antagonized by calcium-calmodulin. We investigated regulation of Y-organ protein. RNA, and DNA syntheses by MIH, cAMP, and calcium in relation to steroidogenesis in vitro. Ecdysteroid production and [3H]leucine incorporation into protein were inhibited 50-60 and 80-90%, respectively, by MIH activity in eyestalk extracts (4 eyestalk equivalents), 10(-6) M forskolin, or a combination of 10(-2) M dibutyryl cAMP and 10(-4) M 3-isobutyl-1-methylxanth

INTRODUCTION: Pulsatillae radix (PR), a medicinal root plant and a well-known Chinese herbal remedy, is primarily used for its heat-clearing, detoxifying, blood-cooling, and antiinflammatory properties.
METHODS: The constituents of PR were systematically analyzed using UHPLC-Q-TOFMS/ MS. Potential targets of active components were identified via the SwissTargetPrediction and PharmMapper databases, while (ulcerative colitis) UC-related disease targets were retrieved from GeneCard and other relevant databases. Overlapping targets between PR and UC were determined using Venn analysis. The STRING

Developmental transcriptional programs establish neuronal diversity and circuit assembly, but how these regulators continue to shape and preserve neuronal identity remains unresolved. The Drosophila central complex comprises precisely wired circuits of diverse neuronal types that coordinate complex behaviors. Within this structure, dorsal fan-shaped body (dFB) neurons integrate internal-state signals to regulate sleep, feeding, and energy homeostasis, yet how distinct dFB subtypes are specified and maintained remains unknown. More broadly, whether developmental transcription factors continue t

The transforming growth factor-β (TGF-β) superfamily encodes a large group of proteins, including TGF-β isoforms, bone morphogenetic proteins and activins that act through conserved cell-surface receptors and signaling co-receptors. TGF-β signaling in insects controls physiological events, including growth, development, diapause, caste determination and metamorphosis. In this study, we used the red flour beetle, Tribolium castaneum, as a model species to investigate the role of the type I TGF-β receptor, saxophone (Sax), in mediating development. The yellow fever mosquito Aedes aegypti is an obligatory blood feeder and a major arboviral disease vector, evoking severe public health concerns worldwide. In adult female mosquitoes, the gut is critical for blood digestion and pathogen entry. We aimed for a systematic exploration of microRNA expression dynamics in the gut during the gonadotrophic cycle. Small RNA libraries were constructed from female mosquito gut tissues at five time points. Unsupervised hierarchical clustering revealed three expression clusters (early, mid and late) peaking at sequential time points - 24, 48 and 72 h poste

Somatic reproductive tissues, such as the male accessory glands in insects, are critical for insect fertility. However, the signaling pathways that maintain accessory gland homeostasis remain poorly understood. Using tissue-specific knockdown or overexpression in Drosophila, we modulated insulin signaling pathway genes in male accessory glands and examined their effects on male fertility. The impact of FOXO overexpression on gland structure and function was studied by analyzing cellular integrity, expression of genes encoding accessory gland proteins, and autophagy markers. Additionally, we ex

Rising seawater temperature poses a severe threat to crustaceans and marine ecosystem stability by perturbing reproductive physiology, whereas the underlying molecular mechanisms remain largely elusive. To address this knowledge gap, the present study conducted transcriptomic and histopathological analyses on gonadal tissues (ovary and testis) of Scylla paramamosain sampled at 0, 6, 24, and 72 h following exposure to heat stress (HS, 35 °C). Differential expression analysis revealed progressive transcriptomic reprogramming in both gonads, with the number of DEGs increasing over time. HS profou

Antibiotics are widely detected in the environment due to their extensive use in medicine, aquaculture, and animal husbandry. Among these, tetracycline (TC) is recognized as a significant environmental contaminant with considerable ecological and public health implications. Continuous exposure to TC through contaminated food, water, and soil can lead to the accumulation of TC residues in the body, potentially exerting adverse effects on human health. To evaluate these effects, we used Drosophila melanogaster as a model organism. Our findings demonstrated that chronic exposure to TC at 1 mg/ml
