Autosomal-dominant sensorineural hearing loss is genetically heterogeneous, with a phenotype closely resembling presbycusis, the most common sensory defect associated with aging in humans. We have identified SLC17A8, which encodes the vesicular glutamate transporter-3 (VGLUT3), as the gene responsible for DFNA25, an autosomal-dominant form of progressive, high-frequency nonsyndromic deafness. In two unrelated families, a heterozygous missense mutation, c.632C-->T (p.A211V), was found to segregate with DFNA25 deafness and was not present in 267 controls. Linkage-disequilibrium analysis suggeste

Medial olivocochlear (MOC) efferent neurons in the brainstem comprise the final stage of descending control of the mammalian peripheral auditory system through axon projections to the cochlea. MOC activity adjusts cochlear gain and frequency tuning, and protects the ear from acoustic trauma. The neuronal pathways that activate and modulate the MOC somata in the brainstem to drive these cochlear effects are poorly understood. Evidence suggests that MOC neurons are primarily excited by sound stimuli in a three-neuron activation loop from the auditory nerve via an intermediate neuron in the cochl

The six receptors of the inner ear (cochlea, two otolith organs and three semicircular canals) share a common transduction unit made up of a sensory hair cell, a first order sensory neuron and the synapse between them. Displacement of the stereocilia in a particular direction leads to excitation of the hair cell and activation of the neuron. Electrical and mechanical reflections of these stages of transduction can be recorded non-invasively in humans and in animals. These include cochlear microphonic potentials, otoacoustic emissions, auditory and vestibular evoked potentials. The ability to r

The effect of various Ca(2+)-channel blockers on exocytosis has been studied at the level of single presynaptic terminals in rat hippocampal cell cultures. The fluorescence change of the styryl dye FM 1-43 has been used as a measure of exocytosis during electrical stimulation. omega-Conotoxin GVIA (2-10 microM) completely inhibited exocytosis in approximately 45% of the boutons in the field of view, while in approximately 55% exocytosis was inhibited incompletely (by 38%). This heterogeneity in response of presynaptic boutons was not seen with isradipine (5 microM) or omega-agatoxin IVA (80 nM

We investigated the influence of degenerated retinal networks on the efficacy of subretinal prosthetic devices in eliciting retinal neural responses. We present a computational model that incorporates intricate descriptions of retinal connectivity spanning neural layers, conductance-based cellular and synaptic parameters, and analytical formulas governing the electrical field. Our results suggest the possibility of selective modulation of functionally-distinct retinal pathways through subretinal stimulation, even in the absence of all photoreceptors. However, we observed a decreasing level of

Magnetic tunnel junctions (MTJs) in nanoscale have emerged as promising candidates for energy-efficient neuromorphic computing. As a pioneering demonstration, the radiofrequency (RF) neural network based on the intrinsic RF-to-DC conversion capability of MTJs features multilayer interconnectivity and native processing of RF inputs. However, most existing devices rely on magnetic field lines to modulate their behavior in neural networks, resulting in high energy consumption and increased area overhead. Moreover, the limited tunable bandwidth of the MTJs constrains the number of synapses per lay

To determine which part of the parasympathetic bronchoconstrictor pathway is most sensitive to depression by general anesthetics, we stimulated different parts of the pathway in dogs after initial anesthesia with chloralose and urethan and then after additional anesthetic drugs. We stimulated the entire reflex pathway by producing apnea or hypoventilation, the sensory pathway by electrically stimulating the proximal end of cut superior laryngeal nerves, and the motor pathway by stimulating the distal end of a cut cervical vagus nerve. Bronchoconstrictor responses to all stimuli were assessed w

Ciliary ganglionectomy and/or postganglionic ciliary neurectomy in the cynomolgus monkey was followed by supersensitivity to intramuscular (i.m.) pilocarpine and lack of response to topical eserine and to electrical stimulation of the Edinger-Westphal nucleus. Normal responsiveness to pilocarpine and eserine returned in most instances after about 6 months. An accommodative response to stimulus of the Edinger-Westphal nucleus was also present and, as in control eyes, could be blocked by hexamethonium. Collectively, these findings demonstrate that parasympathetic innervation to the ciliary muscl

When activated at low frequencies (0.1-1 Hz), second postnatal week synapses onto the most distal part of the apical dendritic tree (stratum lacunosum-moleculare) of rat hippocampal CA1 pyramidal cells display a frequency-dependent synaptic depression not observed for the more proximal (stratum radiatum) synapses. Depression in this frequency range is thought of as a possible contributor to behavioural habituation. In fact, in contrast to the proximal synapses, the distal synapses provide more direct sensory information from the entorhinal cortex as well as from thalamic nuclei. The use of ant

Distribution of the binding sites of Joro spider toxin (JSTX), a specific inhibitor of the glutamate receptors in the crustacean neuromuscular synapse, was studied by using autoradiography. JSTX was synthesized and made radioactive by conjugation with iodine-125. 125I-JSTX irreversibly blocked the excitatory postsynaptic potentials of the lobster neuromuscular synapse in a similar manner as the natural spider toxin. Light microscopic autoradiography of 125I-JSTX treated muscle showed sporadic aggregates of reduced silver grains on the surface of muscles. Electron microscopy of adjoining ultrat

Neurons of the visual cortex of the cat were penetrated with intracellular electrodes and postsynaptic potentials evoked by visual stimuli recorded. By alternately polarizing the cell with steady current injected through the recording electrode, IPSPs and EPSPs could be recorded and analyzed independently. Hyperpolarizing current suppressed IPSPs and enhanced EPSPs by moving the membrane potential toward the IPSP equilibrium potential. Depolarizing the cell toward the EPSP equilibrium potential enhanced IPSP. The responses to electrical stimulation of the LGN, where EPSPs and IPSPs could be di

The axonal branching pattern of the two cerebral giant neurones (CGCs) of Lymnaea stagnalis was studied with intrasomatically applied horseradish peroxidase. The cells are symmetrical. Each CGC projects to the ipsilateral n. labialis medius and n. arteriae labialis, the subcerebral commissure, and to all ipsi- and contralateral buccal nerves. The contralateral buccal nerves are reached via the ipsilateral cerebro-buccal connective and the buccal commissure. The CGC fire action potentials 1:1 in a driver-follower relationship. Each cell is capable of both driving and following. The relationship

At the crustacean neuromuscular junction, facilitation elicited by a repetitive stimulation reaches a plateau level that is proportional to the stimulation frequency. In the present study we demonstrated that plateau facilitation (F(plateau)) does not depend on Ca(2+) manipulations. We manipulated Ca(2+) concentration in the following ways: (1) applying cell permeable chelators BAPTA-AM or EGTA-AM; (2) decreasing Ca(2+) concentration in the extracellular media; (3) enhancing Ca(2+) influx by 4-aminipyridin. We found that neither F(plateau) is decreased by lowering Ca(2+) nor it is increased by

Synaptic ribbons (SRs) of mammalian pinealocytes exhibit day/night changes in number and size, changes that are apparently regulated by the suprachiasmatic nucleus via postganglionic sympathetic nerve fibres. Since the neural control of SR changes is far from clear and as pinealocytes produce action potentials, we undertook to investigate whether electrical stimulation affects SR changes. Isolated rat pineal glands removed during the daytime were kept in vitro for 0, 30, 60, 90 or 120 min, with or without continuous electrical stimulation (1 mA, 1 Hz), followed by the quantification of SR prof

The purpose of this study was to develop neurobiologically plausible models to account for the response properties of several types of cochlear nucleus neurons. Three cell types--the bushy cells, stellate cells, and fusiform cells--were selected because useful data from intracellular recordings were available for these cell types, and because these three cell types exhibit distinct contrasts in their neuronal signal coding strategies. Stellate cells have primarily linear current-voltage (I-V) characteristics, but both bushy and fusiform cells have highly non-linear I-V characteristics. In ligh

1. In the lobster Palinurus vulgaris a sensory input in the lateral posterolateral nerve (lpln) of the stomatogastric nervous system (STS) is able to turn on the cardiac sac (CS) network and to induce dramatic long-lasting alterations in the output of the pyloric network. This long-lasting alteration of pyloric network output consists primarily of changes in the activity of the two neurons that innervate the muscles of the cardiopyloric valve of the stomach, with the dilator neuron (the ventricular dilator, VD) transferring from the pyloric network to the CS network and the constrictor neuron

Paired corticospinal motoneuronal stimulation (PCMS) applying transcranial magnetic stimulation (TMS) over the primary motor cortex paired with peripheral electrical nerve stimulation (PNS) can induce bidirectional associative plasticity at the corticospinal motoneuronal synapses. Direction of the induced plasticity depends on the interstimulus interval (ISI) between TMS and PNS, and the voluntary motor output is modulated in the same manner. Here we investigated the effects of PCMS on the motor learning of finger movements. Excitability at the corticospinal motoneuronal synapses was evaluated

BACKGROUND: Prenatal alcohol exposure (PAE) induces a wide range of neurodevelopmental disabilities that are grouped under the term 'fetal alcohol spectrum disorders' (FASD). The effects of PAE on brain development are dependent on complex neurochemical events, including modification of AMPA receptors (AMPARs). We have recently found that chronic ethanol (EtOH) exposure decreases AMPA-mediated neurotransmission and expression through the overexpression of the specific microRNA (miR)137 and 501-3p, which target GluA1 AMPA subunit, in the developing hippocampus in vitro. Here, we explored how PA

Excitatory and inhibitory postsynaptic potentials are the two fundamental categories of synaptic responses underlying the diverse functionalities of the mammalian nervous system. Recent advances in neuroscience have revealed the co-release of both glutamate and GABA neurotransmitters from a single axon terminal in neurons at the ventral tegmental area that can result in the reconfiguration of the postsynaptic potentials between excitatory and inhibitory effects. The ability to mimic such features of the biological synapses in semiconductor devices, which is lacking in the conventional field ef
