Prediction of tau accumulation in prodromal Alzheimer's disease using an ensemble machine learning approach.

We developed machine learning (ML) algorithms to predict abnormal tau accumulation among patients with prodromal AD. We recruited 64 patients with prodromal AD using the Alzheimer's Disease Neuroimaging Initiative (ADNI) dataset. Supervised ML approaches based on the random forest (RF) and a gradient boosting machine (GBM) were used. The GBM resulted in an AUC of 0.61 (95% confidence interval [CI] 0.579-0.647) with clinical data (age, sex, years of education) and a higher AUC of 0.817 (95% CI 0.804-0.830) with clinical and neuropsychological data. The highest AUC was 0.86 (95% CI 0.839-0.885)

Computer-aided diagnosis of breast ultrasound images using ensemble learning from convolutional neural networks.

Breast ultrasound and computer aided diagnosis (CAD) has been used to classify tumors into benignancy or malignancy. However, conventional CAD software has some problems (such as handcrafted features are hard to design; conventional CAD systems are difficult to confirm overfitting problems, etc.). In our study, we propose a CAD system for tumor diagnosis using an image fusion method combined with different image content representations and ensemble different CNN architectures on US images. The CNN-based method proposed in this study includes VGGNet, ResNet, and DenseNet. In our private dataset

Ensemble machine learning prediction of posttraumatic stress disorder screening status after emergency room hospitalization.

Posttraumatic stress disorder (PTSD) develops in a substantial minority of emergency room admits. Inexpensive and accurate person-level assessment of PTSD risk after trauma exposure is a critical precursor to large-scale deployment of early interventions that may reduce individual suffering and societal costs. Toward this aim, we applied ensemble machine learning to predict PTSD screening status three months after severe injury using cost-effective and minimally invasive data. Participants (N = 271) were recruited at a Level 1 Trauma Center where they provided variables routinely collected at

Ensemble learning of inverse probability weights for marginal structural modeling in large observational datasets.

Inverse probability weights used to fit marginal structural models are typically estimated using logistic regression. However, a data-adaptive procedure may be able to better exploit information available in measured covariates. By combining predictions from multiple algorithms, ensemble learning offers an alternative to logistic regression modeling to further reduce bias in estimated marginal structural model parameters. We describe the application of two ensemble learning approaches to estimating stabilized weights: super learning (SL), an ensemble machine learning approach that relies on V-

An ensemble learning framework for protein stability prediction with enhanced recognition of stabilizing mutations.

Accurately predicting mutation-induced protein stability changes remains a central challenge in structural bioinformatics. Existing methods exhibit a strong bias toward destabilizing mutations, leading to limited performance for stabilizing mutations and constraining their utility in protein engineering. Here, we address this limitation through two complementary strategies: balanced dataset construction and integrative modeling. To mitigate the severe class imbalance in current stability datasets, we constructed undersampling-based balanced datasets and further evaluated reverse-mutation augme

Cultural validation of the RCADS and use of ensemble learning for symptom profiling of anxiety and depression.

INTRODUCTION: Depression and anxiety are the most prevalent global mental health concerns, especially among children and adolescents. Numerous screening tools are available to readily detect these issues. The cultural significance of these tools in specific communities should be validated, as socio-demographic factors can influence psychopathology. Moreover, screening tools are limited to the identification of a disorder and do not highlight critical symptoms that may be more dominant in disease progression. METHODS: In this study, a community sample of 237 Pakistani children and adolescents

ALEC: Active learning with ensemble of classifiers for clinical diagnosis of coronary artery disease.

Invasive angiography is the reference standard for coronary artery disease (CAD) diagnosis but is expensive and associated with certain risks. Machine learning (ML) using clinical and noninvasive imaging parameters can be used for CAD diagnosis to avoid the side effects and cost of angiography. However, ML methods require labeled samples for efficient training. The labeled data scarcity and high labeling costs can be mitigated by active learning. This is achieved through selective query of challenging samples for labeling. To the best of our knowledge, active learning has not been used for CAD

Automatic delineation of organ at risk in cervical cancer radiotherapy based on ensemble learning.

OBJECTIVES: The automatic delineation of organs at risk (OARs) can help doctors make radiotherapy plans efficiently and accurately, and effectively improve the accuracy of radiotherapy and the therapeutic effect. Therefore, this study aims to propose an automatic delineation method for OARs in cervical cancer scenarios of both after-loading and external irradiation. At the same time, the similarity of OARs structure between different scenes is used to improve the segmentation accuracy of OARs in difficult segmentations. METHODS: Our ensemble model adopted the strategy of ensemble learning. Th

Causal Uncertainty-Decomposed Ensemble Learning for Confounding-Aware Skin Cancer Detection in Dermoscopic Images.

Background/Objectives: Automated dermoscopic image analysis can assist research on early skin cancer detection, but deep learning models may learn acquisition-related shortcuts, including terminal hairs, illumination gradients, gel bubbles, shadows and measurement markings. This study evaluates a causal uncertainty-decomposed ensemble (CUDE) for confounding-aware skin-lesion classification within the ISIC 2019 benchmark setting. Methods: CUDE uses a structured variational autoencoder (SVAE) to impose separate lesion-relevant and acquisition-related nuisance latent heads. Three stochastic laten

EEG Ensemble Learning Framework for Prognostic Prediction of Post-rTMS Motor Recovery in Stroke.

Repetitive transcranial magnetic stimulation (rTMS) is a promising neuromodulatory therapy for post-stroke rehabilitation, offering the potential to modulate brain network dynamics. However, its clinical effectiveness remains debated due to the high inter-individual variability in patient responses, largely attributed to heterogeneous patterns of neural reorganization after stroke. In this study, we proposed an EEG ensemble learning framework (EEG-ELF) to predict motor recovery outcomes following rTMS therapy in stroke patients. The framework was evaluated on EEG recordings acquired during a m

Predicting 10-Year Diabetes Risk Through Physiological Acceleration: A Longitudinal Deep Learning Ensemble Approach.

Background/Objectives: Type 2 diabetes (T2D) develops gradually over many years through a prolonged preclinical phase, yet traditional static risk scores often fail to capture these dynamic metabolic trajectories. We propose a longitudinal deep learning framework to predict the 10-year risk of Type 2 diabetes onset defined by comprehensive ADA criteria by modeling the physiological acceleration of routine clinical biomarkers. Methods: Utilizing an 18-year longitudinal dataset from the community-based Korean Genome and Epidemiology Study (KoGES) cohort, we selected N=4354 participants with comp

An ensemble machine learning classifier for Parkinson's disease diagnosis using optical coherence tomography angiography.

Parkinson's disease (PD) is the fastest-growing neurodegenerative disorder worldwide, yet its early diagnosis remains a major challenge due to the absence of reliable biomarkers. Emerging evidence indicates that retinal microvascular alterations, detectable through Optical Coherence Tomography Angiography (OCTA), may serve as promising non-invasive biomarkers for PD. However, the lack of definitive diagnostic tests for early-stage PD underscores an urgent need for objective, non-invasive tools to facilitate timely detection and intervention. In this retrospective study, OCTA images were obtain

Harnessing Sequence Embedding and Ensemble Learning to Identify Antifungal Peptides with Low Hemolytic Risk.

The increasing prevalence of fungal infections represents a growing threat to human health, driven in part by the misuse of antibiotics and the rising incidence of resistance to conventional antifungal agents. Antifungal peptides (AFPs) have emerged as promising alternatives due to their diverse mechanisms of action and their relatively low propensity to develop resistance. To facilitate the systematic discovery of AFPs, we developed AI4AFP. This computational framework integrates curated antifungal peptide resources with advanced machine learning approaches to predict antifungal potential dir

When complexity does not pay: benchmarking deep learning and ensemble methods for biomarker discovery.

The integration of multi-omics data holds great promise for identifying robust and clinically relevant biomarkers, yet the increasing complexity of computational methods raises questions about their practical utility. In this study, we present a comprehensive benchmarking framework that evaluates 27 feature selection strategies and 11 predictive models across three real-world disease cohorts: Alzheimer's disease, progressive supranuclear palsy, and breast cancer. We compare traditional machine learning, ensemble-based methods, and state-of-the-art deep learning models in terms of predictive pe

A new paradigm for stratified cognitive assessment: fusing serum Raman spectroscopy and ensemble learning.

BACKGROUND: In China, the growing need for stratified cognitive assessment demands portable solutions. However, in community-based screening, neuropsychological assessments suffer from low acceptance and low efficiency. Thus, we aimed to develop a serum Raman spectroscopy-based ensemble learning approach for graded cognitive screening, and explore the Raman spectral features of cognitive impairment and their mapping relationship to brain function. METHODS: We recruited 220 subjects for modeling and 40 subjects for validation. Ensemble learning model was built using serum Raman spectra. High-we

Trustworthy Deep Feature Extraction and Ensemble-Based Machine Learning Approach for Breast Cancer Detections.

Breast cancer (BC) has become a major public health concern and is critically associated with the highest global death rate for cancer detection. The diagnosis process and the techniques remain complex and often influenced by the diagnostician's background, which makes it challenging. Despite advancements in BC detection, existing methods cannot often effectively combine interpretability and high accuracy in complex imaging data, limiting their clinical applicability. This study proposes a reliable strategy for detecting BC by combining deep learning (DL) strengths with ensemble-based machine
