Serotonergic psychedelics as epigenetic modulators: A paradigm shift in Alzheimer's disease therapeutics.

Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder characterized by amyloid-β (Aβ) accumulation, tau pathology, synaptic dysfunction, and neuroinflammation, which collectively drive progressive memory loss, cognitive decline, and behavioral changes. Increasing evidence implicates epigenetic dysregulation as a key contributor to these pathological processes by altering gene expression programs. Serotonergic psychedelics, which primarily act as agonists of the serotonin 2 A receptor (5-HT₂AR), have recently attracted attention for their ability to robustly promote neuroplast

Environmental Transgenerational Epigenetics: Effects on Fertility and Offspring Health.

The effects of environmentally induced epigenetic transgenerational inheritance on the fertility and health of organisms' offspring are reviewed. Previously, researchers have demonstrated a wide variety of environmental toxicants can promote an increase in disease and pathology in future generations through epigenetic transgenerational inheritance. These epigenetic alterations are transmitted through the germ line to future generations, influencing subsequent generations fertility and health. These epigenetic alterations provide a molecular mechanism for the decrease in fertility rates of subs

Intergenerational Effects of Paternal Diets on Offspring Health and Reproduction.

Fat, protein, and micronutrients serve as essential components of human nutrition, and their balanced intake is critical for maintaining health. Chronic nutritional imbalances may adversely affect not only individual health but that of subsequent generations. Accumulating evidence indicates that paternal nutritional status before conception contributes to offspring health through diet-induced alterations in sperm quality and epigenetic programming. The genetic integrity and physiological condition of sperm are susceptible to long-term dietary exposures, providing a potential biological basis f

Multimodal Peripheral Blood Immunophenotyping and Single-Cell Epigenetic Analysis of Knee Osteoarthritis Reveals Expansion and Epigenetic Dysregulation of Treg Cell and Nonclassical Monocyte Populations.

OBJECTIVE: Osteoarthritis (OA) involves systemic inflammation, yet peripheral blood immunophenotypes and their underlying epigenetic landscapes remain poorly defined. We characterized cellular and chromatin accessibility profiles in knee OA and radiographic progressors (RPs). METHODS: We performed multimodal immunophenotyping using cytometry by time-of-flight mass spectrometry (CyTOF) on peripheral blood samples from 21 patients with knee OA and 11 healthy controls (HCs). To confirm findings, we developed DNA methylation-based imputation models and applied them to two independent validation c

Epigenetically programmed identity crisis to combat diffuse large B cell lymphoma.

Germinal center B cell-like diffuse large B cell lymphoma (GCB-DLBCL) originates from the malignant transformation of germinal center B cells. This process is driven by transcriptional and epigenetic dysregulations, frequently caused by recurrent mutations and chromosomal translocations. These changes lead to a differentiation arrest associated with unchecked proliferation and survival. This review highlights key transcriptional and epigenetic dependencies that sustain the GCB-DLBCL phenotype and identifies therapeutic vulnerabilities. Epigenetic targeting of these vulnerabilities unlocks tumo

Histone Deacetylase Inhibitor Entinostat (MS-275) Restores Anesthesia-induced Alteration of Inhibitory Synaptic Transmission in the Developing Rat Hippocampus.

Recent evidence strongly supports the idea that common general anesthetics (GAs) such as isoflurane (Iso) and nitrous oxide (N2O; laughing gas), as well as sedative drugs such as midazolam are neurotoxic for the developing mammalian brain having deleterious effects on neural circuits involved in cognition, learning and memory. However, to date, very little is known about epigenetic mechanisms involved in GA-induced plasticity of synaptic transmission in the hippocampus, the main memory-processing region in the brain. Here, we used patch-clamp recordings of miniature inhibitory post-synaptic cu

Metabolic-epigenetic crosstalk in innate immune cell plasticity within the tumor microenvironment.

The tumor microenvironment exerts profound metabolic and epigenetic pressures that shape the plasticity of innate immune cells, influencing their capacity to promote or suppress tumor progression. Emerging evidence highlights the intricate interplay between metabolic reprogramming and epigenetic modifications in macrophages, neutrophils, and other innate immune populations within the tumor microenvironment. Tumor-derived metabolites, hypoxia, and nutrient availability dynamically regulate chromatin accessibility, histone modifications, and DNA methylation patterns, thereby driving context-depe

Collective Mechanical Memory Encoded by Long-Lasting Supracellular Cytoskeletal Structures in Multicellular Spheroids.

Animal cells can sense and "remember" the stiffness of their extracellular environment, resulting in sustained changes in form and function. Such "mechanical memory" has been previously explored using individual cells and attributed to epigenetic changes and transcriptional activity. However, it is unclear whether such memory is retained across collective cells. Here, we report that collective cells sustain mechanical memory through self-organized actin-CK18 networks spanning multiple cell lengths, even under dramatically changing mechanical environments, such as those encountered during cance

Activity-dependent Signaling and Epigenetic Abnormalities in Mice Exposed to Postnatal Ethanol.

Postnatal ethanol exposure has been shown to cause persistent defects in hippocampal synaptic plasticity and disrupt learning and memory processes. However, the mechanisms responsible for these abnormalities are less well studied. We evaluated the influence of postnatal ethanol exposure on several signaling and epigenetic changes and on expression of the activity-regulated cytoskeletal (Arc) protein in the hippocampus of adult offspring under baseline conditions and after a Y-maze spatial memory (SP) behavior (activity). Postnatal ethanol treatment impaired pCaMKIV and pCREB in naïve mice with

Disc-Derived Induced Pluripotent Stem Cells and Environmental Cues for Nucleus Pulposus Regeneration.

Notochordal cells (NCs), abundantly found in the developing nucleus pulposus (NP), show potential for intervertebral disc regeneration because of their unique instructive and healthy matrix-producing capacity. However, NCs are lost early in life, and attempts at in vitro expansion have failed because they lose their specific phenotype. Therefore, much effort is focused on the generation of cells resembling the properties of healthy matrix-producing NP-like cells from human induced pluripotent stem cells (hiPSCs). They are considered a promising alternative for employing native NCs. Given the o

Neuroprotective Epigenetic Changes Induced by Maternal Treatment with an Inhibitor of Soluble Epoxide Hydrolase Prevents Early Alzheimer's Disease Neurodegeneration.

Modulation of Alzheimer's disease (AD) risk begins early in life. During embryo development and postnatal maturation, the brain receives maternal physiological influences and establishes epigenetic patterns that build its level of resilience to late-life diseases. The soluble epoxide hydrolase inhibitor N-[1-(1-oxopropyl)-4-piperidinyl]-N'-[4-(trifluoromethoxy)phenyl] urea (TPPU), reported as ant-inflammatory and neuroprotective against AD pathology in the adult 5XFAD mouse model of AD, was administered to wild-type (WT) female mice mated to heterozygous 5XFAD males during gestation and lactat

Epigenetic mechanisms involved in methamphetamine addiction.

Methamphetamine (METH) is an illicit psychostimulant that is widely abused. The molecular mechanism of METH addiction is complicated and still unknown. METH causes the release of the neurotransmitters including dopamine, glutamate, norepinephrine and serotonin, which activate various brain areas in the central nervous system. METH also induces synaptic plasticity and pathological memory enhancement. Epigenetics plays the important roles in regulating METH addiction. This review will briefly summarize the studies on epigenetics involved in METH addiction.

Advances in the study of the role of epigenetic mechanisms in plant growth and response to stress.

Plants face the challenge of balancing dynamic environmental stresses with internal developmental demands during growth, necessitating the precise regulation of gene expression networks to optimize resource allocation. Recent studies have shown that epigenetic regulatory systems, which serve as bridges between genotype and phenotype, enable plants to respond rapidly to environmental changes and facilitate adaptive evolution by establishing heritable epigenetic memory. This article systematically elucidates the molecular mechanisms by which core epigenetic processes coordinate plant growth and

Targeting the NAD+-SIRT3 axis to mitigate metabolic memory in diabetic kidney disease.

Metabolic memory-the persistent risk of diabetic complications after early hyperglycemia-drives progressive renal injury in diabetic kidney disease (DKD) via sustained oxidative stress, inflammation, and epigenetic reprogramming. We synthesize clinical and experimental evidence showing the nicotinamide adenine dinucleotide (NAD+)-SIRT3 (sirtuin 3) axis as a central mechanistic hub linking mitochondrial dysfunction to epigenetic and inflammatory programs in DKD metabolic memory, while evaluating restoration strategies. Integrating data from preclinical, cellular, and human studies, we review SI

HDAC inhibition prevents hypobaric hypoxia-induced spatial memory impairment through PΙ3K/GSK3β/CREB pathway.

Hypobaric hypoxia at higher altitudes usually impairs cognitive function. Previous studies suggested that epigenetic modifications are the culprits for this condition. Here, we set out to determine how hypobaric hypoxia mediates epigenetic modifications and how this condition worsens neurodegeneration and memory loss in rats. In the current study, different duration of hypobaric hypoxia exposure showed a discrete pattern of histone acetyltransferases and histone deacetylases (HDACs) gene expression in the hippocampus when compared with control rat brains. The level of acetylation sites in hist

Mechanical memory as a metastatic compass: a comparative framework for understanding organotropism through biophysical priming.

The seed and soil hypothesis has long explained organotropic metastasis through biochemical compatibility between tumor cells and distant tissues. However, accumulating evidence points to the role of mechanobiology in this process. This review proposes a framework in which cellular mechanical memory, the persistent adaptation to physical cues, functions as a mechanical compass that influences metastatic destination. We analyze how cancer cells, conditioned by the physical properties of the primary tumor (such as stiffness and viscosity), are epigenetically programmed to colonize distant organs

HDAC inhibitor treatment restores transcriptome, chromatin accessibility, and memory deficits in a mouse model for Down syndrome.

Down syndrome (DS) is associated with widespread molecular alterations, including aberrant gene expression, which not only affects chromosome 21 genes but perturbs the whole transcriptome. The genome-wide nature of this alteration suggests that it might be epigenetically mediated, which may also offer therapeutic opportunities given the reversibility of epigenetic modifications. Here, we show that decreased global acetylation level in the hippocampus of the trisomic mouse model Ts65Dn is associated with decreased chromatin accessibility at gene promoters in hippocampal neurons. Strikingly, pha

[Epigenetics in Parkinson's Disease].

The genetic information encoded in the DNA sequence provides a blueprint of the entire organism. The epigenetic modifications, in particular DNA methylation and histone modifications, determine how and when this information is made available and define the specific gene transcription pattern of a given cell. Epigenetic modifications determine the functional differences of genetically identical cells in multicellular organisms and are important factors in various processes from embryonic development to learning and memory consolidation. DNA methylation patterns are altered by environmental cond
