Induction of a polarized Th1 response by insertion of multiple copies of a viral T-cell epitope into adenylate cyclase of Bordetella pertussis.

The adenylate cyclase (CyaA) of Bordetella pertussis delivers the N-terminal catalytic domain into the cytosol of a large number of eukaryotic cells, in particular, professional antigen-presenting cells. This allows the delivery of CD8(+) T-cell epitopes to the major histocompatibility complex class I presentation pathway. We have previously shown that immunization of mice with CyaA carrying a single CD8(+) T-cell epitope leads to antiviral protection as well as to protective and therapeutic antitumor immunity associated with the induction of specific cytotoxic T-lymphocyte (CTL) responses. Immunoinformatics Design of a Broad-Spectrum Multi-Epitope Vaccine Targeting HA2 and M1 of H9N2 AIV.

H9N2 avian influenza virus (AIV) continues to mutate, leading to immunosuppression and secondary infections in poultry. Traditional inactivated vaccines mainly induce humoral immunity and have limited cross-protection efficacy against various subtypes of virus strains. In this study, we targeted the HA2 and M1 proteins of H9N2 as antigens and used immunoinformatics methods to design a broad-spectrum multi-epitope vaccine (MEV) that can simultaneously activate humoral and cellular immunity. Firstly, through systematic evolutionary analysis and sequence comparison, highly conserved amino acid se

Development of a multi-epitope vaccine candidate to combat SARS-CoV-2 and dengue virus co-infection through an immunoinformatic approach.

BACKGROUND: Although the SARS-CoV-2 and dengue viruses seriously endanger human health, there is presently no vaccine that can stop a person from contracting both viruses at the same time. In this study, four antigens from SARS-CoV-2 and dengue virus were tested for immunogenicity, antigenicity, allergenicity, and toxicity and chosen to predict dominant T- and B-cell epitopes. METHODS: For designing a multi-epitope vaccine, the sequences were retrieved, and using bioinformatics and immunoinformatics, the physicochemical and immunological properties, as well as secondary structures, of the vac

Design of a multi-epitope recombinant BCG vaccine targeting Brucella OMP31, LptE and VirB2 in immunoinformatics approaches.

Brucellosis, caused by the intracellular pathogen Brucella, remains a significant health challenge, alongside substantial economic impacts on livestock industries. Despite antibiotic treatments, the absence of licensed human vaccines necessitates innovative preventive strategies. In this study, we employed reverse vaccinology to design a novel multi-epitope vaccine (MEV) targeting Brucella melitensis. Three immunogenic proteins-outer membrane protein OMP31, LPS assembly protein LptE, and the type IV secretion system protein VirB2-were selected as vaccine candidates. Comprehensive bioinformatic

Brucellosis novel multi-epitope vaccine design based on in silico analysis focusing on Brucella abortus.

Brucella is a common kind of bacteria that has the ability to live within cells and may cause diseases that can be transmitted between animals and humans. Current medical therapy struggles to effectively eradicate Brucella. Thus, it is necessary to develop a multi-epitope vaccine (MEV) in order to effectively prevent Brucella infection. To achieve this objective, we used the reverse vaccinology methodology based on omp19 and Bacterial surface antigen (D15). After conducting our research, we successfully identified 2 cytotoxic T lymphocyte (CTL) epitopes, 2 helper T lymphocyte (HTL) epitopes, a

One action and multiple results: Influenza A virus and SARS-CoV-2 poly-epitope-based hybrid vaccine exploration by reverse vaccinology and immunoinformatic methods.

INTRODUCTION: Influenza A virus (IAV) and severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) coinfection have been shown to intensify symptoms and, therefore, could be considered a significant threat to susceptible populations. Although vaccination is an effective and safe strategy to be used against viruses, large amounts of mutations in these two viruses remain an issue for vaccine design, as some strains of the IAV and SARS-CoV-2 are still present worldwide, regardless of the introduction of various vaccines to protect against the viruses. METHODS: We employed a bioinformatic app

Development of a novel multi-epitope vaccine for brucellosis prevention.

Brucellosis, a zoonotic disease caused by Brucella, presents a significant threat to both animal and human health. In animals, the disease can lead to infertility, miscarriage, and high fever, while in humans, symptoms may include recurrent fever, fatigue, sweating, hepatosplenomegaly, and joint and muscle pain following infection. Treatment often involves long-term antibiotic therapy, placing a substantial psychological and financial burden on patients. While vaccination is crucial for prevention, current animal vaccines have drawbacks such as residual virulence, and a safe and effective huma

Reverse vaccinology and immunoinformatics approach to design a chimeric epitope vaccine against Orientia tsutsugamushi.

Scrub typhus is a vector-borne infectious disease caused by Orientia tsutsugamushi and it is reportedly associated with up to 20 % of hospitalized cases of febrile illnesses. The major challenge of vaccine development is the lack of identified antigens that can induce both heterotypic and homotypic immunity including the production of antibodies, cytotoxic T lymphocyte, and helper T lymphocytes. We employed a comprehensive immunoinformatic prediction algorithm to identify immunogenic epitopes of the 56-kDa type-specific cell membrane surface antigen and surface cell antigen A of O. tsutsugamus

Prediction and identification of novel IBV S1 protein derived CTL epitopes in chicken.

Infectious bronchitis virus (IBV) is a major pathogen common in the poultry industry. Broad cytotoxic T lymphocyte (CTL) response against IBV is one of the crucial factors that help to control viral replication. Spike glycoproteins on the surface of the IBV virion harbor major T cell epitopes. In this study, based on the peptide-binding motifs of chicken MHC I molecules for the BF2*4, BF2*12, BF2*15, and BF2*19 haplotypes, potential CTL epitopes were predicted using S1 proteins from different IBV strains. Twenty-one peptides were predicted to be potential CTL epitopes; they were manually synth

Modeling TCR-Epitope Recognition Specificity: What We Should Learn to Succeed.

T-cell recognition of infected or malignant cells is central to both spontaneous and therapy-induced cellular immune responses against pathogens and cancer. This recognition is elicited by the interaction between T-Cell Receptors (TCRs) and epitopes, which consist of antigenic peptides displayed on major histocompatibility complex molecules. TCR-epitope interactions are characterized by high diversity in TCR and epitope sequences and high structural flexibility in TCR loops. As a result, deciphering the rules of TCR-epitope recognition specificity and accurately predicting these interactions r

Broad and Efficient Activation of Memory CD4+ T Cells by Novel HAdV- and HCMV-Derived Peptide Pools.

Reactivation of Human Cytomegalovirus (HCMV) and Human Adenovirus (HAdV) in immunocompromised patients following stem cell transplantation (SCT) or solid organ transplantation (SOT) is associated with high morbidity and mortality. The adoptive transfer of virus-specific CD8+ and CD4+ T cells has been shown to re-establish the antiviral T-cell response and improve clinical outcome. The viral load in immunocompromised patients can efficiently be reduced solely by the infusion of virus-specific CD4+ T cells. The identification of CD4+ T-cell epitopes has mainly focused on a limited number of vira

In vitro peptide immunization of target tax protein human T-cell leukemia virus type 1-specific CD4+ helper T lymphocytes.

PURPOSE: Adult T-cell leukemia/lymphoma induced by human T-cell leukemia virus type 1 (HTLV-1) is usually a fatal lymphoproliferative malignant disease. HTLV-1 Tax protein plays a critical role in HTLV-1-associated leukemogenesis and is an attractive target for vaccine development. Although HTLV-1 Tax is the most dominant antigen for HTLV-1-specific CD8(+) CTLs in HTLV-1-infected individuals, few epitopes recognized by CD4(+) helper T lymphocytes in HTLV-1 Tax protein have been described. The aim of the present study was to study T-helper-cell responses to HTLV-1 Tax and to identify naturally
