Synthesis and estrogen receptor binding of (17 alpha, 20E)- and (17 alpha, 20Z)-21-phenylthio- and 21-phenylseleno-19-norpregna-1,3,5(10),20-tetraene-3,17 beta-diols.

Previous studies from our laboratory using 17 alpha-E- and 17 alpha-Z-halovinyl estradiols demonstrated a marked enhancement of receptor binding by the Z-isomers. This suggested tolerance at the 17 alpha-position was not previously observed by investigations using 16 alpha and 17 alpha-substituted estradiols. Because of the synthetic access provided by vinyl tin chemistry, we prepared the 17 alpha-E and Z-phenylthiovinyl and phenylselenovinyl estradiols and compared their binding characteristics to those of the previously reported 16 alpha/17 alpha-phenylseleno and methylseleno estradiols. Variations in maternal care alter corticosterone and 17beta-estradiol levels, estrous cycle and folliculogenesis and stimulate the expression of estrogen receptors alpha and beta in the ovaries of UCh rats.

BACKGROUND: Variations in maternal care are associated with neonatal stress, hormonal disturbances and reproductive injuries during adulthood. However, the effects of these variations on sex hormones and steroid receptors during ovary development remain undetermined. This study aimed to investigate whether variations in maternal care are able to influence the hormonal profile, follicular dynamics and expression of AR, ER-alpha and ER-beta in the ovaries of UCh rat offspring. METHODS: Twenty-four adult UCh rats, aged 120 days, were randomly divided into two groups (UChA and UChB) and mated. Ma

Effectiveness of papaya leaf extract (Carica papaya L.) as an infertility agent on estrus cycle length and expression of estrogen receptor beta on ovaries of female white rats (Rattus norvegicus).

BACKGROUND: Natural contraception is an affordable and accessible alternative to synthetic hormonal methods. One strategy for controlling overpopulation involves the use of plant-based agents that possess antifertility properties. Carica papaya (papaya) leaves have garnered scientific interest due to their rich content of bioactive compounds. Flavonoids, alkaloids, and saponins present in papaya leaves are known to influence reproductive physiology by modulating hormonal activity, disrupting follicular development, and altering uterine conditions. AIM: This study aimed to investigate the anti

The estrogen receptor family.

A significant flow of new data is currently being generated within the field of estrogen receptors and their mechanisms of action. This is primarily a result of the development of estrogen receptor knockout mice and the discovery of the second estrogen receptor, estrogen receptor beta. Both estrogen receptors appear to be involved in a multitude of regulatory events, the details of which will be worked out within the next few years. Estrogen receptor alpha appears to play a major role in the regulation of reproductive events and estrogen receptor alpha knockout female mice are completely infer

Effects of estrogen and progesterone on adrenoceptors and cyclic nucleotides in rat uterus.

The effects of estrogen and progesterone on adrenoceptors and cyclic nucleotides were studied in ovariectomized rat uterus. The effects on adrenoceptors were examined by measuring mechanical responses to noradrenaline and by binding site determinations with [3H]-dihydroergocryptine and [3H]-dihydroalprenolol. After acute administration of estradiol-17 beta, uterine cyclic GMP was progressively and significantly elevated, but cyclic AMP declined through it was not significant. Phentolamine suppressed this cyclic GMP elevation. Both acute and chronic treatments with estrogen increased the number

Characterization of the cytosolic estrogen receptor in rat skeletal muscle.

A charcoal assay and the synthetic estrogen [6,7-3H]R 2858 (17 alpha-ethynyl-11 beta-methoxyestradiol-17 beta) were used to show that rat skeletal muscle cytosol contains an estrogen receptor, which was characterized with regard to association and dissociation rate-constants at several temperatures. The degradation kinetics was also studied, and was more rapid in the absence than in the presence of ligand. Association, dissociation and degradation were all temperature-dependent. The apparent equilibrium dissociation constant (Kd), however, was not temperature-dependent, and was about 0.1-1.0 n

Estrogen and the inner ear: megalin knockout mice suffer progressive hearing loss.

Megalin, the largest member of the low-density lipoprotein receptor-related protein family, functions as an endocytic receptor for a variety of essential lipophilic metabolites, including the steroid hormone estrogen. In the cochlea, megalin is strongly expressed within the marginal cells of the stria vascularis, and previous studies demonstrated that beta-estrogen receptors are also expressed in megalin-expressing marginal cells. In the present study, we demonstrate that homozygous megalin mutant mice exhibit profound hearing loss at 3 months of age associated with features of presbycusis, en

Transforming growth factors beta 1, beta 2, and beta 3 messenger RNA and protein expression in mouse uterus and vagina during estrogen-induced growth: a comparison to other estrogen-regulated genes.

Increasing evidence suggests that the differential regulation of multiple peptide growth factors by steroid hormones contributes significantly to the pleiotropic effects elicited in target tissues. We report here an evaluation of the effects of the potent estrogen, diethylstilbestrol, on the expression of the three mammalian transforming growth factor beta (TGF beta) isoforms, TGF beta 1, TGF beta 2, and TGF beta 3, in both the uterus and the vagina of the prepubescent mouse. Immunohistochemical protein detection, in situ hybridization, and Northern RNA analyses demonstrate overlapping but dis

Differential expression of estrogen receptors in women with systemic lupus erythematosus.

OBJECTIVE: Systemic lupus erythematosus (SLE) is an autoimmune disease primarily affecting women. T cell activation markers (calcineurin, CD154) increase in SLE T cells cultured with estradiol 17-beta. Biological effects of estradiol are mediated through 2 receptor proteins, estrogen receptor-alpha (ER-alpha) and estrogen receptor-beta (ER-beta). We compared the amount of estrogen receptor subtypes in T cells and measured the ability of receptor agonist-specific ligands to activate marker gene expression. METHODS: T cells were isolated from 22 female patients with SLE and 17 control women. Th

A yeast estrogen screen for examining the relative exposure of cells to natural and xenoestrogens.

Xenoestrogens, such as o,p'-DDT and octyl phenol (OP), have been associated with reproductive abnormalities in various wildlife species. Xenoestrogens mimic the natural estrogen 17 beta-estradiol and compete for binding to the estrogen receptor. Even though the affinity of o,p'-DDT and OP for the estrogen receptor is approximately 1000-fold lower than 17 beta-estradiol, the actions of xenoestrogens could be enhanced if their bioavailability in serum were greater than 17 beta-estradiol. To test this hypothesis, the yeast estrogen screen (YES) was created by expressing human estrogen receptor (h

Failure of rotenone to interfere with 17 beta-estradiol action in the rat uterus.

The involvement of rotenone in rat mammary carcinogenesis has been suggested to occur through estrogenic effects. This hypothesis was tested by determining the extent of rotenone inhibition of 17 beta-estradiol binding to the estrogen receptor and of the 17 beta-estradiol-induced uterotrophic response in ovariectomized Sprague-Dawley rats. Estradiol binding to the uterine estrogen receptor in the presence of rotenone was determined by charcoal assay and Scatchard analysis. Additionally, 17 beta-estradiol-receptor interactions were assessed on sucrose density gradients. No inhibition of binding

Estrogen modulation of adrenoceptor responsiveness in the female rat pineal gland: differential expression of intracellular estrogen receptors.

The effect of different doses of 17beta-estradiol (E2) on the pineal response to beta-adrenoceptor stimulation in female rats was examined. Pinealocytes from 21-day-old ovariectomized rats were exposed to different estrogen doses and treated with beta-adrenergic agonists. Estrogen treatment produced a dose-dependent, biphasic response to beta-adrenoceptor-induced accumulation of cAMP. This effect was inhibitory at estrogen doses up to 0.1 nM and fitted to a negative exponential curve, while at doses from 0.1 to 100 nM the effect was stimulatory and fitted to a standard positive hyperbola. [The effect of estrogens on male reproduction].

One third of estrogens in the male are from the testis and the others from outside the testis. Aromatase P450 (CYP19) is an enzyme responsible for the conversion of androgens to estrogens. Estrogens regulate cell function via specific receptors--estrogen receptors (ER) which include ER alpha and ER beta. It has been found that the role of estrogens in male reproduction is complex and important, particularly during the neonatal life. Males lacking ER alpha are completely infertile because ER alpha-induced estrogens regulate the reabsorption of luminal fluid in the head of the epididymis and dis

Specific binding of estradiol to rat coronary artery smooth muscle cells.

We report the expression and characteristics of the estrogen receptor in rat coronary artery-derived smooth muscle cells. Polymerase chain reaction analyses of total and poly(A) + mRNA from rat coronary artery-derived smooth muscle cells indicate the presence of estrogen receptor mRNA. Binding analyses reveal the presence of high affinity binding sites for 17 beta-estradiol, with a Kd equivalent to that observed for authentic estrogen receptors in other estrogen responsive tissues. Scatchard and Hill plot analyses of the properties of receptor-ligand binding indicate the presence of a single s

An epigenetic basis for autism spectrum disorder risk and oral contraceptive use.

In the United States 1 in 68 children are diagnosed with autism spectrum disorder (ASD). Although the etiology is unknown, many scientists believe ASD is caused by a combination of genetic and environmental factors and/or epigenetic factors. The widespread use of oral contraceptives is one environmental risk factor that has been greatly overlooked in the biomedical literature. Oral contraceptives, synthetic hormones created to imitate natural human hormones and disrupt endogenous endocrine function to inhibit pregnancy, may be causing the harmful neurodevelopmental effects that result in the i

The pancreatic beta-cell as a target of estrogens and xenoestrogens: Implications for blood glucose homeostasis and diabetes.

The estrogen receptor ERalpha is emerging as a key molecule involved in glucose and lipid metabolism. The main functions of pancreatic beta-cells are the biosynthesis and release of insulin, the only hormone that can directly decrease blood glucose levels. Estrogen receptors ERalpha and ERbeta exist in beta-cells. The role of ERbeta is still unknown, yet ERalpha plays an important role in the regulation of insulin biosynthesis, insulin secretion and beta-cell survival. Activation of ERalpha by 17beta-estradiol (E2) and the environmental estrogen bisphenol-A (BPA) promotes an increase of insuli

Evaluation of estrogenic receptors in the nasal mucosa of women taking oral contraceptives.

BACKGROUND: There is evidence that the nasal mucosa is affected by estrogen influence. Some authors have already detected estrogen receptors (ER) in the nasal mucosa. However, there doesn't seem to be a consensus about the concentration and distribution of the ER or the possible influence of hormonal contraceptives in the nasal mucosa. OBJECTIVE: The study was conducted to evaluate the influence of oral contraceptives on the distribution and concentration of estrogenic receptors in nasal mucosa. STUDY DESIGN: Two groups of 32 women with regular menstrual cycles were selected. One group of wo

Identification and characterization of estrogen and progesterone receptors from the oviduct of the American alligator (Alligator mississippiensis).

A protein extract prepared from the egg shell fiber-secreting region of the oviduct of Alligator mississippiensis was assayed for the presence of receptors for estrogen and progesterone. In the presence of [3H]-estradiol-17 beta, the extract contained an estrogen-binding activity which reached equilibrium at 25 degrees C in 1 h. Scatchard analysis demonstrated that a single estrogen-binding activity was present in the extract with a Kd of 0.5 nM for [3H]estradiol-17 beta. A steroid specificity competition assay showed the estrogen binding activity strongly recognized estradiol-17 beta and diet
