[Chemical composition of lipoid anticoagulant and mechanism of its action].

A lipid anticoagulant (ether extract of brain tissue) after repeated treatment with alcohol and acetone contained, mainly, ethanolamine phosphatides and sphingomyelin. The product inhibited the conversion of prothrombin into thrombin due to interaction of the lipid anticoagulant with the substrate (prothrombin). The anticoagulant inhibited also the reaction of thrombin with fibrinogen, producing a kind of "coupling inhibition" as shown by graphical analysis of the inhibition kinetics. A fraction of the anticoagulant, enriched in ethanolamine phosphatides and a fraction, enriched in sphingomyel

Discovery of novel diagnostic biomarkers for Sjögren-Larsson syndrome by untargeted lipidomics.

AIM: Sjögren-Larsson syndrome (SLS) is a rare neurometabolic disorder that mainly affects brain, eye and skin and is caused by deficiency of fatty aldehyde dehydrogenase. Our recent finding of a profoundly disturbed brain tissue lipidome in SLS prompted us to search for similar biomarkers in plasma as no functional test in blood is available for SLS. METHODS AND RESULTS: We performed plasma lipidomics and used a newly developed bioinformatics tool to mine the untargeted part of the SLS plasma and brain lipidome to search for SLS biomarkers. Plasma lipidomics showed disturbed ether lipid metab

EPT1 (selenoprotein I) is critical for the neural development and maintenance of plasmalogen in humans.

Ethanolamine phosphotransferase (EPT)1, also known as selenoprotein 1 (SELENOI), is an enzyme that transfers phosphoethanolamine from cytidine diphosphate-ethanolamine to lipid acceptors to produce ethanolamine glycerophospholipids, such as diacyl-linked phosphatidylethanolamine (PE) and ether-linked plasmalogen [1-alkenyl-2-acyl-glycerophosphoethanolamine (plasmenyl-PE)]. However, to date there has been no analysis of the metabolomic consequences of the mutation of EPT1 on the concentration of ethanolamine glycerophospholipids in mammalian cells. We studied a patient with severe complicated h

Efficacy and evaluation of safety of sclerosants for intramuscular venous malformations: clinical and experimental studies.

Excision of intramuscular venous malformations may damage intact functional muscles, and sclerotherapy is an alternative way of relieving symptoms. Several sclerosants are available, but selection of the optimal one is controversial. We report our clinical experiences of sclerotherapy, and experimental studies in rats that investigated muscular damage after injection of various sclerosants. For the clinical study, 10 patients with intramuscular venous malformations were reviewed who had been treated by sclerotherapy using ethanolamine oleate. The rate by which the volume reduced was assessed q

The biochemical role of platelet-activating factor in reproduction.

The presence of Co-A independent transacylase activity in amnion cells and the preferential transfer of arachidonic acid to acceptor-ethanolamine plasmalogen provide a satisfactory explanation to the questions raised by the observation that arachidonate-enriched ethanolamine plasmalogen increases in amnion late in gestation without alteration in the total amount of ethanolamine glycerophospholipids. The proposed mechanism also serves as a link between the observed changes in glycerophospholipid composition and the generation of PAF. We have emphasized a role for PAF in fetal lung maturation, t

Stimulatory effect of calmodulin antagonists on phospholipid base-exchange reactions in rabbit platelet membranes.

The properties of Ca2+-dependent incorporation of [3H]serine, [3H]ethanolamine and [3H]choline into the corresponding phospholipids mediated by base-exchange enzymes in rabbit platelet membranes were studied in the presence or absence of the calmodulin antagonists chlorpromazine, trifluoperazine and N-(6-aminohexyl)-5-chloro-1-naphthalenesulfonamide (W-7), all of which markedly activate three base-exchange reactions. The base-exchange activities were dependent on Ca2+ both in the presence and absence of the drugs. Other metal ions tested did not stimulate the base-exchange reactions, even in t

Ethanolaminephosphate cytidylyltransferase is essential for survival, lipid homeostasis and stress tolerance in Leishmania major.

Glycerophospholipids including phosphatidylethanolamine (PE) and phosphatidylcholine (PC) are vital components of biological membranes. Trypanosomatid parasites of the genus Leishmania can acquire PE and PC via de novo synthesis and the uptake/remodeling of host lipids. In this study, we investigated the ethanolaminephosphate cytidylyltransferase (EPCT) in Leishmania major, which is the causative agent for cutaneous leishmaniasis. EPCT is a key enzyme in the ethanolamine branch of the Kennedy pathway which is responsible for the de novo synthesis of PE. Our results demonstrate that L. major EP

High-Resolution Color Transparent Display Using Superimposed Quantum Dots.

In this paper, a high-resolution full-color transparent monitor is designed and fabricated using the synthesized quantum dots for the first time. For this purpose, about 100 compounds that had the potential to emit blue, green, and red lights were selected, and simulation was performed using the discrete dipole approximation (DDA) method, in which the shell layer was selected to be SiO2 or TiO2 in the first step. Among the simulated compounds with SiO2 or TiO2 shells, Se/SiO2 and BTiO3/SiO2 were selected as blue light emitters with high intensity and narrow bandwidth. Accordingly, CdSe/SiO2 na

Hemolytic anemia in hereditary pyrimidine 5'-nucleotidase deficiency. II. Effect of pyrimidine nucleotides and their derivatives on glycolytic and pentose phosphate shunt enzyme activity.

We evaluated the glycolytic intermediate concentrations from the erythrocytes of a patient with hereditary pyrimidine 5'-nucleotidase (P5'N) deficiency. Conclusive evidence for a metabolic block was not found. We evaluated the effects of the pyrimidine (cytidine and uridine) tri- and diphosphate nucleotides (CTP, CDP, UTP, UDP) and the choline and ethanolamine derivatives of CDP (CDP-choline, CDP-ethanolamine) on the activities of key enzymes of the Embden-Meyerhof pathway. Structural basis for catalysis and selectivity of phospholipid synthesis by eukaryotic choline-phosphotransferase.

Phospholipids are the most abundant component in lipid membranes and are essential for the structural and functional integrity of the cell. In eukaryotic cells, phospholipids are primarily synthesized de novo through the Kennedy pathway that involves multiple enzymatic processes. The terminal reaction is mediated by a group of cytidine-5'-diphosphate (CDP)-choline /CDP-ethanolamine-phosphotransferases (CPT/EPT) that use 1,2-diacylglycerol (DAG) and CDP-choline or CDP-ethanolamine to produce phosphatidylcholine (PC) or phosphatidylethanolamine (PE) that are the main phospholipids in eukaryotic

Contact dermatitis caused by diphenhydramine hydrochloride.

Three cases of contact dermatitis caused by an ethanolamine antihistamine, diphenhydramine hydrochloride (Benadryl), are described. Patients who develop contact dermatitis from it should avoid its oral or parenteral administration because they may develop a localized or generalized flare of dermatitis. Dimenhydrinate (Dramamine), clemastine fumarate (Tavist), carbinoxamine maleate (Clistin-D), and doxylamine succinate (Decapryn) are also ethanolamine antihistamines. Since they may cross-react with diphenhydramine hydrochloride, they should be avoided in sensitive patients.

Dysfunctional plasmalogen dynamics in the plasma and platelets of patients with schizophrenia.

BACKGROUND: Based upon the concept of a prodromal risk syndrome for first psychosis, intense efforts are being applied to define potential biomarkers of disease onset in schizophrenia. One such tentative biomarker may be plasmalogens, complex structural glycerophospholipids that serve multiple functions in all cells. METHODS: To further investigate this possibility, we undertook a lipidomics analysis of choline and ethanolamine plasmalogens in the plasma and platelets of 23 patients with schizophrenia and 27 age-matched controls. RESULTS: Plasma levels of both choline and ethanolamine plasma

Apropos: critical analysis of molluscicide application in schistosomiasis control programs in Brazil.

Schistosomiasis is a snail-transmitted infectious disease affecting over 200 million people worldwide. Snail control has been recognized as an effective approach to interrupt the transmission of schistosomiasis, since the geographic distribution of this neglected tropical disease is determined by the presence of the intermediate host snails. In a recent Scoping Review published in Infectious Diseases of Poverty, Coelho and Caldeira performed a critical review of using molluscicides in the national schistosomiasis control programs in Brazil. They also described some chemical and plant-derived m

Variable Substrate Preference among Phospholipase D Toxins from Sicariid Spiders.

Venoms of the sicariid spiders contain phospholipase D enzyme toxins that can cause severe dermonecrosis and even death in humans. These enzymes convert sphingolipid and lysolipid substrates to cyclic phosphates by activating a hydroxyl nucleophile present in both classes of lipid. The most medically relevant substrates are thought to be sphingomyelin and/or lysophosphatidylcholine. To better understand the substrate preference of these toxins, we used (31)P NMR to compare the activity of three related but phylogenetically diverse sicariid toxins against a diverse panel of sphingolipid and lys

Insights from selenoprotein I mouse models for understanding biological roles of this enzyme.

Selenoprotein I (selenoi) is a metabolic enzyme expressed in a wide variety of tissues that catalyzes the transfer of the ethanolamine phosphate group from CDP-ethanolamine to lipid acceptors to generate ethanolamine phospholipids. It is a member of the selenoprotein family, a class of proteins that mostly play fundamental roles in redox homeostasis and are defined by the co-translational incorporation of selenium in the form of selenocysteine. Loss-of-function mutations in the human SELENOI gene have been found in rare cases leading to a complex form of hereditary spastic paraplegia. Understa

p-hydroxyphenylacetaldehyde, an aldehyde generated by myeloperoxidase, modifies phospholipid amino groups of low density lipoprotein in human atherosclerotic intima.

Oxidation of low density lipoprotein (LDL) may be of critical importance in the pathogenesis of atherosclerosis. Recent studies suggest that oxidized phospholipids render LDL atherogenic. However, both the structures and the physiologically relevant pathways for the formation of modified phospholipids in oxidized LDL remain poorly understood. We previously showed that p-hydroxyphenylacetaldehyde (pHA) is the major product of L-tyrosine oxidation by the myeloperoxidase/hydrogen peroxide/chloride system of phagocytes. In the current studies, we demonstrate that this reactive aldehyde targets the

Phosphatidylethanolamine in Trypanosoma brucei is organized in two separate pools and is synthesized exclusively by the Kennedy pathway.

Phosphatidylethanolamine is a major phospholipid class of all eukaryotic cells. It can be synthesized via the CDP-ethanolamine branch of the Kennedy pathway, by decarboxylation of phosphatidylserine, or by base exchange with phosphatidylserine. The contributions of these pathways to total phosphatidylethanolamine synthesis have remained unclear. Although Trypanosoma brucei, the causative agent of human and animal trypanosomiasis, has served as a model organism to elucidate the entire reaction sequence for glycosylphosphatidylinositol biosynthesis, the pathways for the synthesis of the major ph

Polydactylous subungual squamous cell carcinoma caused by chemical contact.

Polydactylous squamous cell carcinoma (SCC) is rare and has been associated with human papillomavirus (HPV). Our recent case was HPV negative and provides greater evidence for chemical irritants being an alternative cause of subungual SCC. Our patient had spent a number of years with her hands in direct contact with undiluted cleaning chemicals including one containing ethanolamine. Ethanolamine has been shown to have carcinogen sensitizing role. Although HPV has a strong association with subungual SCCs, the accumulation and concentration of noxious substances around and under the nails must a

Brain lipid changes after organic solvent exposure.

Animals (rats, gerbils and guinea pigs) were exposed to various common solvents. The brain lipids were extracted and analysed. In particular the fatty acid pattern of a major phospholipid, ethanolamine, phosphoglyceride was studied. A major finding was that some chlorinated solvents, trichloroethylene, perchloroethylene and 1,1,1-trichloroethane, had effects on the ethanolamine phosphoglyceride fatty acid pattern, while most other solvents, like toluene, xylene, white spirit and Freon 11 lacked this effect. Relatively small, but significant, changes were observed in the proportions of the poly

Homeostatic response of phospholipid pathways to PCYT2 deficiency and impaired de Novo synthesis of phosphatidylethanolamine.

PCYT2 is the key regulatory enzyme in the biosynthesis of phosphatidylethanolamine (PE) via the CDP-ethanolamine Kennedy pathway. Deficiencies in this gene have been linked to metabolic, neurological, and cardiac disorders; however, most studies report that PE levels remain unchanged. This study aimed to identify the metabolic mechanisms that preserve PE levels when its synthesis is impaired in PCYT2-knockdown human fibroblasts. We investigated alternative pathways that could compensate for reduced PE synthesis, including phosphatidylcholine (PC) and PE base-exchange to phosphatidylserine (PS)
