Engineering a manganese superoxide dismutase with enhanced thermostability and activity via protein language Models: Toward antioxidant and anti-inflammatory applications in biomedicine and skincare.

Superoxide dismutases (SODs) are essential antioxidant enzymes that catalyze the dismutation of superoxide radicals, playing a crucial role in cellular defense against oxidative stress. However, most natural SODs lack the thermostability required for industrial and therapeutic applications. In this study, we identified a manganese-dependent SOD (PsSOD) from the extremophilic bacterium Pseudogracilibacillus sp., which exhibited high native activity but limited stability under high-temperature conditions. To enhance its robustness, we employed a protein language model-guided strategy that integr

Identification and Overexpression of Endogenous Transcription Factors to Enhance Lipid Accumulation in the Commercially Relevant Species Chlamydomonas pacifica.

Sustainable low-carbon energy solutions are critical to mitigating global carbon emissions. Algae-based platforms offer potential by converting carbon dioxide into valuable products while aiding carbon sequestration. However, scaling algae cultivation faces challenges like contamination in outdoor systems. Previously, our lab evolved Chlamydomonas pacifica, an extremophile green alga, which tolerates high temperature, pH, salinity, and light, making it ideal for large-scale bioproduct production, including biodiesel. Here, we enhanced lipid accumulation in evolved C. pacifica by identifying an

Biological treatment of methyl orange dye and textile wastewater using halo-alkaliphilic bacteria under highly alkaline conditions.

As the textile wastewater is highly saline and has high pH it is important to employ extremophilic microbes to survive in harsh conditions and provide effective bioremediation of textile dyes. This study aims to find a sustainable solution for dye removal by investigating the potential of an indigenously isolated bacterium, Nesterenkonia lacusekhoensis EMLA3 (halo-alkaliphilic) for treatment of an azo dye, methyl orange (MO) and textile effluent. MO dye decolorization studies were conducted using mineral salt media (MSM) by varying incubation time (0-120 h), initial dye concentration (50-350 m

Genome-scale data reveal that endemic Poecilia populations from small sulphidic springs display no evidence of inbreeding.

Populations with limited ranges can be highly vulnerable to changes in their environment and are, thus, of high conservation concern. Populations that experience human-induced range reductions are often highly inbred and lack genetic diversity, but it is unknown whether this is also the case for populations with naturally small ranges. The fishes Poecilia sulphuraria (listed as critically endangered) and Poecilia thermalis, which are endemic to small hydrogen sulphide-rich springs in southern Mexico, are examples of such populations with inherently small habitats. We used geometric morphometri

Proposed Missions to Collect Samples for Analyzing Evidence of Life in the Venusian Atmosphere.

The recent and still controversial claim of phosphine detection in the venusian atmosphere has reignited consideration of whether microbial life might reside in its cloud layers. If microbial life were to exist within Venus' cloud deck, these microorganisms would have to be multi-extremophiles enclosed within the cloud aerosol particles. The most straightforward approach for resolving the question of their existence is to obtain samples of the cloud particles and analyze their interior. While developing technology has made sophisticated in situ analysis possible, more detailed information coul

Assessment of Haloferax mediterranei Genome in Search of Copper-Molecular Machinery With Potential Applications for Bioremediation.

Heavy metals are essential micronutrients at low concentrations, serving as cofactors for relevant microbial enzymes (i.e., respiratory nitrate and nitrite reductases NADH dehydrogenase-2, amine oxidase, etc.), but they become harmful cellular intoxicants at significant low concentrations compared to other chemical compounds. The increasing need to incorporate bioremediation in the removal of heavy metals and other contaminants from wastewaters has led extremophiles to the spotlight of research. The haloarchaeon Haloferax mediterranei has promising physiological characteristics regarding biore

Experimental, Genomic, and Structural Evidence Supporting Putative PET-Hydrolases in Thermophilic Bacteria Isolated From Hot Springs in Cajamarca, Peru.

Polyethylene terephthalate (PET) waste represents a major environmental challenge due to limited recycling solutions. Thermophilic bacteria from geothermal environments harbor diverse enzymatic machinery adapted to extreme conditions, offering promising biocatalysts for plastic degradation; however, biological resources from Peru and other South American countries remain scarce. We characterized four bacterial strains isolated from two geothermal sites in Cajamarca, Peru, screened for PET hydrolysis at 50°C. Whole-genome sequencing using hybrid assembly achieved near-complete circular genomes.

Harnessing halophyte phytochemicals and biostimulants to enhance crop resilience under climate stress: a comprehensive review.

Global food security is increasingly compromised by the synergistic pressures of soil salinization and the climate-mediated proliferation of resilient agricultural pests and weeds. Halophytes, extremophile plants naturally adapted to high-salinity niches, represent an underexploited reservoir of bioactive compounds and microbial consortia with significant bioeconomic potential. This review synthesizes the multi-functional utility of halophyte-derived bioactive compounds in mitigating contemporary environmental stressors through two primary lenses. First, we discuss the biochemical efficacy of

Microbial communities and volatile profiles heterogeneity in a Mars-analogue volcanic lake.

Analyses of the Martian surface suggest the existence of sedimentary lacustrine deposits that may have once harboured life. Lake Bagno dell'Acqua (Pantelleria, Italy) represents an optimal planetary field analogue for investigating the evolution and adaptation of life in Mars-like environments and for validating potential life-detection strategies. Accordingly, we characterised microbial communities and volatile organic compound profiles in actively growing organosedimentary structures within lake sediments and in biofilm covering hot springs at the lake margins. The microbial communities exhi

Halophilic archaea produce wax esters and use an alternative fatty acyl-coenzyme A reductase for precursor synthesis.

Wax esters (WE) are fatty acid-based neutral lipids thought to be restricted to bacteria and eukaryotes, playing a key role in the functioning and maintenance of cells, especially under adverse conditions. Here, we show that several halophilic archaea (Halobacteriales) carry a homolog of the bacterial wax synthase gene. WE synthesis and accumulation are demonstrated in one of these (poly)extremophilic archaea, Natronomonas pharaonis, during growth on long-chain fatty acids. Our bioinformatic analysis also shows that the synthesis of fatty alcohols required for WE synthesis could be performed b

Bioengineering radioresistance by overproduction of RPA, a mammalian-type single-stranded DNA-binding protein, in a halophilic archaeon.

Halobacterium sp. NRC-1 is a wild-type extremophilic microbe that is naturally tolerant to high levels of ionizing radiation. Mutants of strain NRC-1 with even higher levels of resistance to ionizing radiation, named RAD, were previously isolated after selecting survival to extremely high doses of ionizing radiation. These RAD mutants displayed higher transcription levels for the rfa3 operon, coding two subunits of the RPA-like putative single-stranded binding protein, rfa3 and rfa8, and a third downstream gene, ral. In order to bioengineer cells with increased tolerance to ionizing radiation

Evolution of a new enzyme for carbon disulphide conversion by an acidothermophilic archaeon.

Extremophilic organisms require specialized enzymes for their exotic metabolisms. Acid-loving thermophilic Archaea that live in the mudpots of volcanic solfataras obtain their energy from reduced sulphur compounds such as hydrogen sulphide (H(2)S) and carbon disulphide (CS(2)). The oxidation of these compounds into sulphuric acid creates the extremely acidic environment that characterizes solfataras. The hyperthermophilic Acidianus strain A1-3, which was isolated from the fumarolic, ancient sauna building at the Solfatara volcano (Naples, Italy), was shown to rapidly convert CS(2) into H(2)S a

Structural and biochemical insights into the thermostable esterase Ta0887 from Thermoplasma acidophilum.

Microbial esterases are versatile and stable enzymes with a wide range of biotechnological applications. However, few esterases have been characterized from archaea, an important source of extremophilic enzymes. In this study, we report the biochemical characterization and crystal structure of Ta0887, a novel esterase from the thermoacidophilic archaeon Thermoplasma acidophilum. The protein was successfully cloned, expressed, and purified in Escherichia coli. Light scattering assays revealed that Ta0887 is a monomer in solution. Activity assays using p-nitrophenyl (p-NP) esters confirmed its e

Nitrate reduction and iron (II) oxidation by subsurface bacteria from the Iberian Pyritic Belt: Insights into the biogeochemical cycles in this environment.

Nitrogen and iron are central to Earth's biogeochemical cycles. The Iberian Pyrite Belt (IPB), a vast iron reservoir, hosts extremophilic microorganisms adapted to high metal and sulfur concentrations. This study investigates two isolates from the deep IPB subsurface, Citrobacter telavivensis T1.2D-1 and Stutzerimonas stutzeri T2.31D-1, evaluating their nitrate-reducing capacity and the role of iron in denitrification, with emphasis on nitrate-dependent ferrous iron oxidation (NDFO). Both species could reduce nitrate in sucrose-, lactate-, or acetate-fed medium, while their consortium enhanced

Enzymatic Deglycosylation and Lipophilization of Soy Glycosides into Value-Added Compounds for Food and Cosmetic Applications.

Soybean is one of the most important crops worldwide, being placed ninth on the chart of the most cultivated species. Its high level of production correlates with a huge amount of waste produced. These residues could be of great interest due to the presence of high-value-added molecules, including some glycosides (i.e., daidzin, genistin, glycitin) widely studied for their potent antioxidant properties. Due to their low bioavailability and limited solubility in lipidic media, lipophilization strategies have recently gained momentum to improve daidzin, genistin, and glycitin applications as mul

Exploring biosurfactant from Halobacterium jilantaiense as drug against HIV and zika virus: fabrication, characterization, cytosafety property, molecular docking, and molecular dynamics simulation.

Biosurfactants are surface-active molecules with unique qualities and various uses. Many microorganisms produce secondary metabolites with surface-active characteristics that serve various antiviral functions. The HIV and Zika viruses were chosen for this study because they can spread from mother to child and result in potentially fatal infections in infants. Halophilic bacteria from the Red Sea solar saltern in Egypt were screened using drop collapse, emulsification activity, and oil displacement assays to produce biosurfactants and emulsifiers. Halobacterium jilantaiense strain JBS1 was the

Thermostable Bacterial Laccase: Catalytic Properties and Its Application in Biotransformation of Emerging Pollutants.

Laccases have been predominantly reported in fungi, and primarily, fungal laccases are currently exploited in industrial applications. However, extremophilic bacterial laccases possess immense potential, as they can withstand extreme temperatures, pH, and salt concentrations. In addition, unlike fungal laccases, the production of bacterial laccases is cost-effective. Therefore, bacterial laccases are gaining significant attention for their large-scale applications. Previously, we reported a novel thermostable laccase (LacT) from Brevibacillus agri. Herein, we have confirmed that LacT shares a
