Ultrasensitive Fluorescent Detection of Urinary Trypsin for Early Pancreatic Disease Diagnosis: Toward Smartphone-Assisted Analysis.

Early detection of pancreatic disease requires non-invasive, highly sensitive biomarker detection. Here, we report a fluorescence-based method for rapid, ultrasensitive detection of urinary trypsin without sample pretreatment. A π-extended pyrene fluorescent probe functionalized with four carboxylate groups was designed to facilitate selective interaction and micellar aggregation with protamine, a trypsin-cleavable substrate. Protamine-induced aggregation results in amplified fluorescence quenching, which is efficiently reversed by trypsin-mediated proteolysis. Trypsin cleavage reduces the arg

An ultrasensitive cyanine dye-based fluorescent probe for the determination of iron(III) and oxalate.

An ultrasensitive fluorescent probe based on the pentamethine indocyanine dye (Ind2) was developed for the quantitative determination of trace iron(III) and oxalate in aqueous solution. The analytical principle relies on fluorescence quenching of Ind2 by Fe3+ through radical oxidation under acidic conditions (pH 1.9 ± 0.05), whereas oxalate determination relies on a reduction of the effective oxidizing activity of Fe3+ upon complex formation. HRMS analysis confirmed the formation of a dye radical cation, followed by cleavage of the polymethine chain. Comparative studies revealed that pentameth

A nitrogen-rich Eu-MOF for ultrasensitive fluorescence detection of 6-mercaptopurine and 6-thioguanine.

The simultaneous detection of toxic antineoplastic drugs 6-mercaptopurine (6-MP) and 6-thioguanine (6-TG) remains challenging due to their low intrinsic luminescence and low quantum yield. To address this, a novel three-dimensional europium-based metal-organic framework, Eu2-tbia, was rationally constructed using a triazole-dicarboxylate ligand, 5-((4'-(triazol-1-yl)benzyl)oxy)isophthalic acid (H2tbia). The obtained Eu2-tbia exhibits strong luminescence via an efficient antenna effect and retains excellent stability in aqueous solutions over a wide pH range and in various organic solvents. The

Engineering a highly selective FRET-based genetically encoded Zn2+ sensor for real-time live-cell zinc imaging.

Genetically encoded Zn2+ probes (GEZPs) provide powerful tools for visualizing intracellular zinc dynamics with spatial and temporal resolution in living cells. Recently developed single-module ZnM-based scaffolds are derived from CaM-based metal-binding motifs and can serve as effective sensing elements. ZnM preferentially binds Zn2+, but their selectivity can be partially compromised by competing ions such as Ni2+, which share similar coordination properties. Here, we developed TV-ZnM1, a ratiometric FRET-based GEZP constructed through a stepwise engineering strategy centered on the ZnM sens

A ratiometric AIE probe for visual discrimination monitoring of hydrogen sulfide and biogenic amines by different sensing mechanism and its application in cells, tumor model and food.

Hydrogen sulfide (H2S) and biogenic amines (BAs) are naturally generated from foods that adversely affect human health. The very close structural similarities between cysteine and homocysteine present a great challenge to achieve their selective detection using regular fluorescent probes, limiting the biological and pathological studies of these two amino thiols. In this work, we have developed a novel ratiometric AIE probe named MTMC for the visual discrimination detection of H2S and BAs. Probe MTMC displayed a remarkable fluorescence color change after the addition of H2S (from red to yellow

A Novel Phthalimide-Based Turn-On Fluorescent Probe for Detection of Hydrogen Sulfide and Its Application in Real Samples.

In this paper, we rationally designed a turn-on fluorescent probe PSH for the selective and efficient detection of hydrogen sulfide (H2S) over other biologically relevant species, particularly glutathione (GSH), cysteine (Cys), and homocysteine (Hcy). Consisting of a phthalimide derivative linked with the 2,4-dinitrobenzenesulfonyl (DNBS) group through a sulfonyl ether bond, PSH exhibits a turn-on fluorescence response toward H2S with distinct advantages: a broad pH range (pH 5-9), rapid response kinetics (within 5 min), high selectivity against interfering substances (accompanied by an 8.5-fo

Ratiometric "on/off"-type fluorescence sensors based on nitrogen and silicon co-doped graphene quantum dots for identification and semi-quantitative visualized detection of dopamine, epinephrine, and norepinephrine.

Catecholamines (CAs) play pivotal physiological in the human body, and the aberrant levels of them are closely associated with various diseases. Therefore, developing a rapid, simple, and accurate method for CAs determination is of considerable clinical importance for precise disease diagnosis. In this work, a turn-off fluorescence sensor based on nitrogen and silicon co-doped graphene quantum dots (N,Si-GQDs) was first established for the detection of total CAs, leveraging the inner filter effect (IFE). To achieve the selective identification and separate quantification of dopamine (DA), epin

A Dual Responsive Fluorescent Probe Exhibiting ICT Based Colorimetric and Fluorimetric Detection of Cyanide.

2-(pyridin-4-yl)-8H-thieno[2,3-b] indole (ITP) fluorescent probe has been synthesized and utilized as a ratiometric sensor for cyanide anion. The color change upon the addition of cyanide anion is easily noticeable with naked eyes. The greater difference between the absorption and emission spectrum due to the inclusion of cyanide is attributed to the enhanced intramolecular charge transfer (CT) by hydrogen bonding of CN¯ with ITP, which greatly increased the electron density on the donor thieno[2,3-b] indole. The cyanide sensing mechanism is further confirmed by 1H NMR and DFT studies. Further

Tetrazine-based AIE probes for specific detection of superoxide anion.

Combining the single-electron recognition property of tetrazine with aggregation-induced emission (AIE), two fluorescent probes named SWJT-48 and SWJT-49 were rationally designed and synthesized. This molecular design relied on a specific reduction recognition mechanism to eliminate interference from various substances. Both probes exhibited rapid response, high sensitivity and excellent selectivity toward superoxide anion (O2•-). Both exogenous and endogenous O2•- in HeLa cells were successfully detected using SWJT-49. Furthermore, SWJT-49 achieved fluorescence imaging of O2•- in HT22 cells v

A Mini Review on Carbazole Fluoroscent Chemosensors for the Detection of Cu (II) Ion: A Comprehensive Review.

The first fluorescent chemosensor was introduced by F. Goppelsröder (1867) for the detection of Al (III) ion through the formation of strongly fluorescent chelate of morin ion. This mini review focuses on carbazole derivatives fluorescence for detecting Cu2+ ions in aqueous and living systems. Carbazole is an example of heterocyclic compound containing nitrogen atom. Carbazole and its derivatives have been used as fluorescent sensors for the detection of Cu2+ ions due to the some inherent features such as high solubility, presence of large conjugated system, active intramolecular charge transf

Red-emissive fluorescent probes based on PET mechanism for highly sensitive detection and multi-scenario imaging of thiophenol.

Thiophenol (PhSH) is a highly toxic aromatic thiol endangering ecosystems, food safety, and human health. Traditional detection methods are limited by instrument dependence and inability for real-time imaging. This study developed a red-emitting fluorescent probe, TTCP-DN, based on the PET mechanism. The probe employs 5-(4-diphenylamino)benzothiophene as the fluorophore and 2,4-dinitrophenoxy as the PhSH-specific recognition moiety, achieving mitochondrial targeting via a pyridine salt. Without PhSH, fluorescence is quenched; upon PhSH-induced reaction, fluorescence restores at 623 nm. The pro

Green Synthesis of a Reversible "ON-OFF-ON" Fluorescent Sensor for Fe3+ using Licorice-Derived N-Doped Carbon Dots.

The detection of ferric ions (Fe3+) is of crucial importance in environmental monitoring and biomedical diagnostics. However, developing highly selective, sensitive, and environmentally friendly detection methods remains an important challenge. In this study, nitrogen-doped carbon quantum dots (N-CQDs) were green-synthesized as a novel and sustainable fluorescent sensor for Fe3+ detection. The synthesis was achieved via a one-pot hydrothermal method using licorice powder as a renewable carbon source and p-phenylenediamine as a nitrogen dopant. The synthesized N-CQDs display bright blue fluores

A triphenylamine-boronic-acid-based theragnostic biomaterial for diabetes with concurrent fluorescence sensing of glucose and microviscosity.

Blood glucose and viscosity are two pivotal biomarkers associated with the chronic metabolic disorder, diabetes mellitus. Despite successful attempts at continuous monitoring of glucose, dual sensing of glucose and microviscosity with significant precision remains unexplored. To fill in this knowledge gap, a series of triphenylamine-boronic-acid-based fluorescence sensors were designed in this study. The triphenylamine-boronic acid derivatives (TPA-p-TBA, TPA-m-TBA, and TPA-o-TBA) were fabricated following the typical donor-π-acceptor arrangement, with triphenylamine as the electron-donating g

High-Fidelity NADH Imaging with a Mitochondria-Targeted Probe: Specific Interrogation of Oxidative Stress in Pneumonia and Tumors.

Abnormal accumulation of NADH in pneumonia and tumor microenvironments is a key pathological event driven by oxidative stress. It is very important to make a fluorescent probe that can accurately detect NADH and has high sensitivity and selectivity. Our research introduces a novel mitochondria-targeted probe, Mito-NQ, through a stepwise design strategy: screening N-methylquinoxaline salt as the optimal recognition group, extending the emission wavelength, and anchoring mitochondria with triphenylphosphine (TPP) for precise mitochondrial localization. Mechanistic studies revealed that the N-met

A Thiophilic Hg2+-Activated Fluorescent Probe for Multiscale Bioimaging: From Living Cells and Mice to Tomato Plants.

Mercury ions (Hg2+) have been demonstrated to disrupt key physiological and biochemical processes in plants, and therefore, early detection and real-time monitoring of Hg2+ concentration dynamics in plants, particularly in crops, are of critical importance. Currently, very few reported fluorescent probes have been applied for Hg2+ detection in crops, and in-depth investigations into Hg2+ accumulation behavior in plants remain scarce. To address this gap, we developed a novel "Turn-On" Hg2+-activated molecular probe, LBR, and systematically evaluated its performance. The probe exhibited high se

Glutathione-Triggered Nitrite Release From Organic Nitrites Enhances Nitric Oxide Bioavailability in Mammalian Cells.

The conversion of nitrite (NO2 -) to nitric oxide (NO) represents a key pathway for NO generation under ischemic conditions. Therefore, organic nitro compounds capable of releasing nitrite under physiological environments have attracted considerable attention. Here, we demonstrate that simple 4-nitro-1,8-naphthalimide derivatives efficiently and almost quantitatively liberate nitrite upon reaction with biological thiols, particularly glutathione (GSH). These compounds function as NO prodrugs, enhancing NO bioavailability in mammalian cells through intracellular nitrite-to-NO conversion. Notabl

A new fluorene-based Schiff-base as fluorescent chemosensor for selective detection of Cr3+ and Al3.

2-((9H-fluoren-2-ylimino) methyl)phenol (F3) was synthesized by condensation reaction of 9H-fluoren-2-amine and 2-hydroxybenzaldehyde in EtOH and characterized by its melting point, 1H-,13C NMR and molecular mass. F3 exhibits a high selectivity for detection of Cr3+ and Al3+ ions as a fluorescent chemosensor and showed a single emission band at 536nm upon excitation at 333nm according to fluorescence emission studies. The addition of Cr3+ and Al3+ make a significant increase in fluorescent intensity at 536nm in CH3CN, while other metal ions have almost no influence on the fluorescence. The flu
