Fluoride is an environmentally prevalent inhibitor of fungi, plants, and other eukaryotes. The fluoride exporter (FEX) is the major resistance mechanism that prevents intracellular fluoride accumulation in eukaryotes. FEX activity is sodium-dependent, but the mechanism for Na+ activation and the impact of other cations on FEX function remain poorly understood. Here, we show that sodium and lithium have different effects on channel activity. We leverage these differences to understand how monovalent cations regulate FEX. Functional assays in a reconstituted system show that lithium acts as a co

This study aimed to investigate the effectiveness of the semi-annual application of 38% silver diamine fluoride (SDF) and that of 5% sodium fluoride (NaF) varnish in preventing occlusal cavitated caries in the primary molars of preschool children over a 30-mo period. This was a parallel 3-arm, double-blind, randomized controlled trial. Generally healthy children around 3 y old were randomly allocated to 1 of the 3 study groups: group 1: 38% SDF solution; group 2: 5% NaF varnish; and group 3: tonic water (placebo). The noncavitated occlusal surfaces of all primary molars received semi-annual to

BACKGROUND: Dental caries is the most common disease in childhood and has an impact on general health status. The topical application of fluoride varnishes has been used for the prevention and control of dental caries due to their high fluoride content, adhesion capacity, and safety. Silver has a varied application in medicine as well as in dentistry due to its anticaries, antimicrobial, and antirheumatic potentials. The introduction of nanosilver fluoride (NSF) was made with advancements in technology to overcome the drawbacks of silver diamine fluoride (SDF).
AIM: To compare and evaluate th

Fluorosis is a public health concern in 25 countries around the globe. The present study is about the mitigation of fluoride (F) toxicity by giving F-free water (FFW) and calcium (Ca). A study was conducted by taking 76 Wistar rats in two phases, phase I (6 months), where rats were randomly divided into four groups: normal-Ca diet (NCD) 0.5%; low-Ca diet (LCD) 0.25%; NCD + 100 ppm F and LCD + 100 ppm F in groups 1, 2, 3 and 4, respectively. F and Ca were given through water and diet respectively. Phase II is the reversal of fluorosis for 3 months, where LCD group 2 was treated with NCD. Groups

OBJECTIVE. Our in vitro study evaluated calcium fluoride formation in enamel and the anticaries effect of seven resin-based varnishes under cariogenic challenge. METHODS. Enamel blocks were subjected to pH cycling. The experimental groups received fluoride varnish application, the positive control received topical fluoride gel treatment, and the negative control did not receive any treatment. The pH cycling surface hardness (SH1 ) and integrated loss of subsurface hardness (ΔKHN) were then determined. We measured the amount of fluoride released into the demineralizing and remineralizing (DE-

Fluoride ion in drinking water is known for both beneficial and detrimental effects on health. The prevalence of fluorosis is mainly due to the intake of large quantities of fluoride through drinking water owing to more than 90% bioavailability. The objective of this study is to predict optimal fluoride level in drinking water for fluoride endemic regions by comprising the levels of fluoride and other water quality parameters in drinking water, prevalence of fluorosis, fluoride intake through water, food and beverages such as tea and coffee and also considering the progressive accumulation of

The key role of osteoblasts in skeletal fluorosis makes the exploration of the possible mechanisms of the fluoride-induced oxidative stress of osteoblasts of great importance. In this article, the in vitro effects of fluoride on the oxidative stress of osteoblasts are presented. To study the inhibitory effect of baicalein on the oxidative stress of osteoblasts, the antioxidant activity of baicalein was evaluated for osteoblasts exposed to fluoride. Calvarial osteoblasts were prepared and respectively treated with alpha-MEM (5% calf serum) containing 0.5, 1.0, 2.0, 4.0, 8.0, 12.0, and 20.0 mg/L

Our aim was to compare plaque fluoride and the level of mutans streptococci in saliva and plaque before and 1 and 2 years after discontinuation of water fluoridation in Kuopio, Finland. For comparison, a low-fluoride community was included in the study. Pooled plaque and saliva were collected from a random sample of 12-year-olds in both communities (n = 139). Enumeration of mutans streptococci in plaque was made on MSB agar and the level of salivary mutans streptococci was measured using the Strip mutans method. Fluoride was analyzed using a fluoride specific electrode. Caries, gingival status

Actin exhibits ATPase activity of unknown function that increases when monomers polymerize into filaments. Differences in the kinetics of ATP hydrolysis and the release of the hydrolysis products ADP and inorganic phosphate suggest that phosphate-rich domains exist in newly polymerized filaments. We examined whether the enrichment of phosphate on filamentous ADP-actin might modulate the severing activity of gelsolin, a protein previously shown to bind differently to ATP and ADP actin monomers. Binding of phosphate, or the phosphate analogs aluminum fluoride and beryllium fluoride, to actin fil

The aim was to determine the fluoride retention in plaque-covered and clean incipient enamel lesions after topical application of a CaF2/NaF varnish (Bifluorid 12). In 50 specimens of bovine enamel an incipient lesion was produced with acidic hydroxyethylcellulose (pH 4.8; 72 h). 40 specimens were fluoride varnished; the 10 remaining specimens were used for measuring baseline fluoride content. Each six of the fluoridated specimens were recessed in the buccal aspects of an intraoral appliance worn for 5 days. During the experimental period one side of the appliance was kept clean, and plaque gr

Healthy dental students received oral doses of 10.0 mg F daily in the form of compounds of widely varying solubilities. Parotid saliva F concentration and urinary F excretion were highest with NaF, Na2PO3F, and SnF2, lowest with CaF2, and intermediate with AlF3. Solubility of the ingested fluoride compound was a controlling factor in the amount of F excreted in the urine, the peak F concentration reached in parotid saliva, and the duration of a significant elevation in salivary F.

Dental caries remains as one of the most prevalent noncommunicable diseases even with advances in preventive dental care. This literature-based research examines how fluoride exposure influences enamel stability and caries formation. We hypothesized that fluoride reduces caries incidence by promoting remineralization and reinforcing the crystal lattice structure. A comprehensive search was done through PubMed, ScienceDirect, Karger, and Figshare to select peer reviewed studies that evaluated fluoride and enamel interactions. Quantitative data were extracted from open access in vitro and in viv

Fluorine is in the form of fluorides ubiquitous in nature and as such an inevitable part of our environment. In small amounts, it is known to have beneficial effects on dental health. On the other hand, excessive chronic intakes can result in adverse effects including the development of dental fluorosis in children and/or skeletal fluorosis in both children and adults. The adequate intake has been set, based on empirical observations, at 0.05 mg/day/kg body weight however, the threshold between beneficial and harmful effects is narrow. Despite numerous studies, knowledge on the fluoride toxici
