Decoding dexmedetomidine's multiple mechanisms in atherosclerosis: effects on lipid metabolism, inflammation, cholesterol efflux, and foam cell formation.

Atherosclerosis is a leading cause of cardiovascular disease and is characterized by dysregulated lipid metabolism and chronic vascular inflammation. Dexmedetomidine (DEX) has been reported to exert anti-inflammatory, antioxidant and lipid-lowering effects across several disease settings, yet its potential therapeutic impact on atherosclerosis remains unclear. In this study, we investigated the anti-atherosclerotic efficacy of DEX and explored the underlying mechanisms in vivo and in vitro. An atherosclerosis model was established by feeding ApoE−/− mice a Western diet for 13 weeks, followed b

Biodegradable cell-laden starch foams for the rapid fabrication of 3D tissue constructs and the application in neural tissue engineering.

Cells encapsulation by biomaterials has been widely studied as a strategy of building tissue construct in tissue engineering. Conventional encapsulation of cells using hydrogels often needs the polymerization process or relatively complex molding process. In this study, we developed a facile strategy for the in situ fabrication of biodegradable cell-laden starch foams. By utilizing the unique gelatinization property of starch, cell-laden starch foams with tunable architecture were rapidly prepared in a green and biological-friendly process. The bubble size and stiffness of starch foams could b

Hematopoietic arginase 1 deficiency results in decreased leukocytosis and increased foam cell formation but does not affect atherosclerosis.

BACKGROUND AND AIMS: Arginase1 (Arg1), an M2 macrophage marker, plays a critical role in a number of immunological functions in macrophages, which are the main cell type facilitating atherosclerotic lesion development. Arg1 uses the substrate l-arginine to create l-ornithine, a precursor molecule required for collagen formation and vascular smooth muscle cell differentiation. By reducing l-arginine availability, Arg1 limits the production of nitric oxide (NO), a pro-atherogenic factor in macrophages. In endothelial cells, conversely, NO is strongly anti-atherogenic. However, until now, the rol

A Novel PAMAM G3 Dendrimer-Based Foam with Polyether Polyol and Castor Oil Components as Drug Delivery System into Cancer and Normal Cells.

One of the intensively developed tools for cancer therapy is drug-releasing matrices. Polyamidoamine dendrimers (PAMAM) are commonly used as nanoparticles to increase the solubility, stability and retention of drugs in the human body. Most often, drugs are encapsulated in PAMAM cavities or covalently attached to their surface. However, there are no data on the use of PAMAM dendrimers as a component of porous matrices based on polyurethane foams for the controlled release of drugs and biologically active substances. Therefore, in this work, porous materials based on polyurethane foam with incor

Peptonics: Shear protection and foam prevention in mammalian cell cultures producing monoclonal antibodies and viral vectors for gene therapy.

Biologics produced by cell culture are central to modern pharmaceutical production. In this context, the surfactants added to culture media for protecting cells from shear are essential for ensuring product yield and quality. Commercial surfactants exhibit lot-to-lot variability, which has caused failures in production campaigns. Addressing this issue, our team introduced Peptonics, a family of peptidic surfactants that offer comparable tensioactivity to Pluronics while improving cell productivity and removing foaming issues. Peptonics were originally demonstrated on model Chinese Hamster Ovar

Identification of foam cell like M2 macrophages, AEBP1 biomarkers, and resveratrol as potential therapeutic in MASLD using Ecotyper and WGCNA.

The immune cell landscapes in metabolic dysfunction-associated fatty liver disease (MAFLD) and their clinical relevance have not been explored. We used Ecotyper to identify immune cell states based on gene expression and examined their roles in metabolic dysfunction-associated steatotic liver disease (MASLD) progression. Limma was applied to identify differentially expressed genes (DEGs). Weighted gene co-expression network analysis (WGCNA) was used for module identification. Bidirectional Mendelian randomization (MR) analysis was used to validate the causal effect of AEBP1 on metabolic dysfun

Intracellular Galectin-3 as a Crucial Regulator of Foam Cell Formation and Apoptosis Progression Through the Modulation of Membrane Lipid Rafts.

BACKGROUND: Atherosclerosis, a leading cause of cardiovascular disease (CVD) mortality worldwide, is characterized by dysregulated lipid metabolism and unresolved inflammation. Macrophage-derived foam cell formation and apoptosis contribute to plaque formation and vulnerability. Elevated serum galectin-3 (Gal-3) levels are associated with increased CVD risk, and Gal-3 in plaques is strongly associated with macrophages. However, its impact on inflammation remains controversial. METHODS: An in vitro model using human THP-1-derived macrophages was established to simulate inflammatory foam cells.

Modulatory Effects of Mdivi-1 on OxLDL-Induced Metabolic Alterations, Inflammatory Responses, and Foam Cell Formation in Human Monocytes.

Atherosclerosis, a major contributor to cardiovascular disease, involves lipid accumulation and inflammatory processes in arterial walls, with oxidized low-density lipoprotein (OxLDL) playing a central role. OxLDL is increased during aging and stimulates monocyte transformation into foam cells and induces metabolic reprogramming and pro-inflammatory responses, accelerating atherosclerosis progression and contributing to other age-related diseases. This study investigated the effects of Mdivi-1, a mitochondrial fission inhibitor, and S1QEL, a selective complex I-associated reactive oxygen speci

Sulforaphane Inhibits Foam Cell Formation and Atherosclerosis via Mechanisms Involving the Modulation of Macrophage Cholesterol Transport and the Related Phenotype.

Sulforaphane (SFN), an isothiocyanate, is one of the major dietary phytochemicals found in cruciferous vegetables. Many studies suggest that SFN can protect against cancer and cardiometabolic diseases. Despite the proposed systemic and local vascular protective mechanisms, SFN's potential to inhibit atherogenesis by targeting macrophages remains unknown. In this study, in high fat diet fed ApoE-deficient (ApoE-/-) mice, oral SFN treatment improved dyslipidemia and inhibited atherosclerotic plaque formation and the unstable phenotype, as demonstrated by reductions in the lesion areas in both th

A Snake Venom-Secreted Phospholipase A2 Induces Foam Cell Formation Depending on the Activation of Factors Involved in Lipid Homeostasis.

MT-III, a snake venom GIIA sPLA2, which shares structural and functional features with mammalian GIIA sPLA2s, activates macrophage defense functions including lipid droplet (LDs) formation, organelle involved in both lipid metabolism and inflammatory processes. Macrophages (MΦs) loaded with LDs, termed foam cells, characterize early blood vessel fatty-streak lesions during atherosclerosis. However, the factors involved in foam cell formation induced by a GIIA sPLA2 are still unknown. Here, we investigated the participation of lipid homeostasis-related factors in LD formation induced by MT-III

Feeding Angptl4-/- mice trans fat promotes foam cell formation in mesenteric lymph nodes without leading to ascites.

Angiopoietin-like 4 (ANGPTL4) regulates plasma triglyceride levels by inhibiting LPL. Inactivation of ANGPTL4 decreases plasma triglycerides and reduces the risk of coronary artery disease. Unfortunately, targeting ANGPTL4 for the therapeutic management of dyslipidemia and atherosclerosis is hampered by the observation that mice and monkeys in which ANGPTL4 is inactivated exhibit lipid accumulation in the mesenteric lymph nodes (MLNs). In mice these pathological events exclusively unfold upon feeding a high saturated FA diet and are followed by an ultimately lethal pro-inflammatory response an

High density lipoprotein deficiency and foam cell accumulation in mice with targeted disruption of ATP-binding cassette transporter-1.

Recently, the human ATP-binding cassette transporter-1 (ABC1) gene has been demonstrated to be mutated in patients with Tangier disease. To investigate the role of the ABC1 protein in an experimental in vivo model, we used gene targeting in DBA-1J embryonic stem cells to produce an ABC1-deficient mouse. Expression of the murine Abc1 gene was ablated by using a nonisogenic targeting construct that deletes six exons coding for the first nucleotide-binding fold. Lipid profiles from Abc1 knockout (-/-) mice revealed an approximately 70% reduction in cholesterol, markedly reduced plasma phospholipi

Structure-based drug repurposing and in vitro evaluation of VAV2-associated candidates for suppression of macrophage foam-cell formation in atherosclerosis.

Cardiovascular disease remains a leading cause of mortality worldwide, and macrophage-derived foam-cell formation represents an important event in atherosclerosis. VAV2, a Rho-family guanine nucleotide-exchange factor, participates in signalling pathways associated with oxidized low-density lipoprotein uptake and macrophage lipid accumulation. In this study, a structure-based drug-repurposing workflow was used to identify clinically relevant compounds with predicted affinity for a full-length VAV2 structural model. Tanshinone IIA inhibits foam cell formation and alleviates atherosclerosis by regulating the Hedgehog signaling pathway through TRPM2 suppression.

The development of foam cells is crucial in the advancement of atherosclerosis (AS). Tanshinone IIA (Tan IIA), the primary lipophilic component of Salvia miltiorrhiza, has various pharmacological effects. Despite this, the precise role of Tan IIA in AS has not been fully elucidated. In this research, we employed ApoE-/- mice to establish an AS model. Oil Red O and HE staining indicated that Tan IIA obviously reduced plaque areas in both the aorta and aortic arch. Additionally, serum analysis revealed that Tan IIA notably decreased lipid and inflammatory factor levels in AS mice. In vitro studi

Tailoring the cell morphology and characteristics of poly(lactic acid) foams via a stereocomplex crystallization-assisted phase separation strategy.

Stereocomplex (SC) crystallization between poly(l-lactide) (PLLA) and its enantiomer poly(d-lactide) (PDLA) provides an effective approach to enhancing the physicochemical properties of poly(lactic acid) (PLA). In this study, we systematically investigate the effects of SC crystallization on the thermal-induced phase separation (TIPS) behavior of PLA enantiomers/1, 4-dioxane solutions, as well as the microstructure, pore morphology, and properties of the resulting PLA foams. The results show that increasing PDLA content leads to a higher relative fraction of SC crystallites and alters the cell

Paeonol inhibits NETs-mediated foam cell inflammation through the CitH3/NLRP3/caspase-1 signaling pathway in atherosclerosis.

Atherosclerosis is a chronic inflammatory disease characterized by lipid streaks, which are produced by aggregates of lipid-rich foam cells. Foam cells intensify atherosclerosis by secreting a range of inflammatory mediators. Neutrophil extracellular traps produced by activated neutrophils, which are abundantly present in lipid-accumulating plaques. However, the relationship between neutrophil extracellular traps and foam cells inflammation is still unclear. Paeonol is well known for its anti-inflammatory effects in atherosclerosis. Nevertheless, the exact pharmacological mechanisms by which p
