Pharmacokinetic advantage of intra-arterial cyclosporin A delivery to vascularly isolated rabbit forelimb. I. Model development.

Effective antirejection therapy with minimal systemic morbidity is required if limb transplantation is to become a clinical reality. We investigated whether i.a. infusion of cyclosporin A (CSA) into the vascularly isolated rabbit forelimb will distribute drug homogeneously to the tissues and produce higher local drug levels than same-dose i.v. treatment, thereby improving the therapeutic index. CSA 4.0 mg/kg/day was infused continuously via osmotic minipump into either the right brachial artery (i.a. group) or jugular vein (i.v. group) of New Zealand rabbits. Ligation of all muscles at the rig

Correlation of forelimb malformation asymmetries with visceral organ situs in the transgenic mouse insertional mutation, legless.

We studied the relationship between the sidedness of visceral organs and the expression of limb abnormalities in the legless mutant. The control of asymmetry in visceral development appears to be random in the legless mutant; that is, 50% develop normally (situs solitus) and 50% develop with inverted viscera (situs inversus). We find that the sidedness of forelimb abnormalities expressed in the mutants is highly correlated with visceral sidedness. Abnormalities are more severe in the right forelimbs in situs solitus mutants, while the left forelimb is more severely affected in situs inversus m

Treatment of forelimb growth plate deformity in 11 dogs by radial dome osteotomy and external coaptation.

Radial dome osteotomy and external coaptation were used to correct forelimb growth deformities with carpal valgus angles of 15 to 43 degrees in 11 dogs (12 forelimbs). Osteotomy sites were clinically healed by week 4. Appearance and function in 10 limbs was good to excellent. The angular deformity recurred in one dog by week 4 due to partial closure of the distal radial growth plate. Carpal valgus recurred in one dog by week 2 because the ulnar osteotomy healed before radial growth ceased.

Unilateral damage to the entopeduncular nucleus causes dysfunction in skilled forelimb movement and hinders recovery in rats.

The entopeduncular nucleus (EPN) is an output nucleus of the basal ganglia and is suggested to play a critical role in motor control. This study aimed to clarify the impact of EPN damage on skilled motor function and its recovery. Unilateral EPN damage or intracerebral hemorrhage (ICH) was induced by stereotactic injection of kainic acid (KA) into the EPN or collagenase type Ⅳ into the internal capsule, respectively. Skilled motor function was assessed by the number of trials and success rates in a single-pellet reaching task from baseline to day 28. EPN damage was quantified by immunohistoche

Osteology of the pectoral girdle and forelimb of Siriusgnathus niemeyerorum, a Late Triassic traversodontid cynodont from Brazil.

Traversodontids are non-mammaliaform cynodonts, herbivorous to omnivorous, that lived during the Triassic. In the Late Triassic, some members of this group increased in body size relative to other cynodont lineages and evolved postcranial adaptations associated with locomotion. The Late Triassic Siriusgnathus niemeyerorum is one such traversodontid. Originally described in 2018 based on cranial material, the species has since been the focus of considerable advances in our understanding of its skull anatomy; however, its postcranial skeleton remains undescribed. The present study examined the p

Forelimb muscle activity during level and progressive incline and decline walking in dogs and implications for rehabilitation.

INTRODUCTION: Shoulder pathologies are a common reason for presentation to veterinary sports medicine and rehabilitation practices. Currently there are no standardized rehabilitation protocols for shoulder injuries but controlled walking, either on flat ground or on inclines/declines, is recommended in nearly every recovery rehabilitation program. The objective of this study was to evaluate the peak and average muscle activity of commonly targeted forelimb muscle groups using fine-wire and surface electromyography (EMG) during treadmill walking at five treadmill positions. Our hypothesis was t

Anterior-posterior gradient in the integrated processing of forelimb movement direction and distance in macaque parietal cortex.

A major issue in modern neuroscience is to understand how cell populations present multiple spatial and motor features during goal-directed movements. The direction and distance (depth) of arm movements often appear to be controlled independently during behavior, but it is unknown whether they share neural resources or not. Using information theory, singular value decomposition, and dimensionality reduction methods, we compare direction and depth effects and their convergence across three parietal areas during an arm movement task. All methods show a stronger direction effect during early move

A Whole-brain Map of Long-range Inputs to GABAergic Interneurons in the Mouse Caudal Forelimb Area.

The caudal forelimb area (CFA) of the mouse cortex is essential in many forelimb movements, and diverse types of GABAergic interneuron in the CFA are distinct in the mediation of cortical inhibition in motor information processing. However, their long-range inputs remain unclear. In the present study, we combined the monosynaptic rabies virus system with Cre driver mouse lines to generate a whole-brain map of the inputs to three major inhibitory interneuron types in the CFA. We discovered that each type was innervated by the same upstream areas, but there were quantitative differences in the i

Anatomical and functional changes in the organization of the cuneate nucleus of adult rats after fetal forelimb amputation.

A previous study has shown that fetal forelimb removal in the rat results in an increase in the size of the hindlimb representation in primary somatosensory cortex and suggested that this anomalous cortical organization may have resulted from alterations in the primary afferent innervation of the dorsal column nuclei (Killackey and Dawson, 1989). The present study used both anatomical and electrophysiological techniques to examine the effects of fetal forelimb amputation on the dorsal column nuclei. Rats sustained forelimb removals on embryonic day 16 and were used in terminal experiments when

Peripheral inputs and early unit activity in area 5 of the monkey during a trained forelimb movement.

Previous studies on area 5 in the monkey showed that an early neuronal activity up to 300 ms before the onset of movement was encountered in this cortical area after deafferentation of the trained forelimb (C1-T7). This present work indicates that none of the EMG activity recorded in other parts of the body may account for these early changes and lends weight to the hypothesis that their activation is purely central.

A docodont ulna from the Upper Jurassic in North America and its implications for the forelimb morphology in Docodonta (Mammaliaformes).

An isolated ulna referable to Docodon, from the Upper Jurassic Morrison Formation of Wyoming, resembles the ulna of Borealestes in the presence of a small facet for the radial condyle of the humerus that is confined to the middle of the lateral side of the facet for the ulnar condyle of the humerus and a prominent tubercle on the latter. These taxa and Haldanodon are characterized by the presence of a deep, slit-like lateral fossa for one of the ligaments of the elbow joint. These features are currently only known for Docodonta. However, the ulna of Docodon differs from those of Borealestes an

The Effect of Girth Design and Girth Tension on Saddle-Horse Pressures and Forelimb Stride Kinematics in Rising Trot.

The aim of this study was to investigate the effect of girth design and girth tension; six horses regularly ridden were used. Each horse underwent four experimental sessions in an unbalanced Latin-square design with two girth tensions (8 kg or 16 kg) and two girth designs (straight girth (S) or anatomical girth (A)). Pressure between the saddle and the horse was measured at 100 Hz with a pressure mat (0.5 sensels per cm2). Notably, 2D limb kinematics were determined from anatomical markers placed on the fore and hindlimbs. Video was collected at 240 fps. There was no significant effect of girt

Forelimb muscle activation patterns in American alligators: Insights into the evolution of limb posture and powered flight in archosaurs.

The evolution of archosaurs provides an important context for understanding the mechanisms behind major functional transformations in vertebrates, such as shifts from sprawling to erect limb posture and the acquisition of powered flight. While comparative anatomy and ichnology of extinct archosaurs have offered insights into musculoskeletal and gait changes associated with locomotor transitions, reconstructing the evolution of motor control requires data from extant species. However, the scarcity of electromyography (EMG) data from the forelimb, especially of crocodylians, has hindered underst

Spatial localization and projection densities of brainstem mossy fibre afferents to the forelimb C1 zone of the rat cerebellum.

The present study uses a double retrograde tracer technique in rats to examine the spatial localization and pattern of axonal branching in mossy fibres arising from three major sources in the medulla-the external cuneate nucleus, the sensory trigeminal nucleus and the reticular formation, to two electrophysiologically-identified parts of the cerebellar cortex that are linked by common climbing fibre input - the forelimb-receiving parts of the C1 zone in lobulus simplex and the paramedian lobule. In each experiment a small injection of rhodamine-tagged beads was injected into one cortical regio

Sensitivity to cadmium-chloride-induced forelimb ectrodactyly is independent of the p53 gene-dosage in the C57BL/6J mouse.

BACKGROUND: The p53 pathway plays an important role in the regulation of apoptosis, osteoblast differentiation, skeletal development, and teratogenic sensitivity. The administration of cadmium chloride (CdCl(2)) on gestational day 9 in susceptible mouse strains causes postaxial forelimb ectrodactyly in a percentage of fetuses through unknown mechanisms. In this study, the hypothesis that the p53 gene dosage might affect the incidence or severity of CdCl(2)-induced forelimb ectrodactyly was examined. METHODS: Heterozygous p53-null female mice, on the C57BL/6J background known to be sensitive t

Prospective evaluation of techniques for differentiating shoulder pathology as a source of forelimb lameness in medium and large breed dogs.

OBJECTIVE: To assess lameness evaluation, shoulder abduction angles, radiography, and ultrasonography for determining presence, location, and severity of forelimb pathology. STUDY DESIGN: Prospective cohort study. ANIMALS: Dogs >or=20 kg (n=30). METHODS: Each dog was assigned lameness scores. Shoulder abduction angles were determined. Radiographs of shoulders and elbows were subjectively graded for pathology. One investigator unaware of dog history (lameness, radiographic findings) performed ultrasonographic assessment of shoulders with subjective grading of pathology. Another investigator

Signal decomposition method of evaluating head movement to measure induced forelimb lameness in horses trotting on a treadmill.

In horses at a trot, the head moves up and down twice in one stride. In horses with unilateral forelimb lameness this movement is asymmetric. Computer-assisted kinematic analysis of vertical head movement can be used to quantify objectively lameness in horses in clinical trials. However, in mild lameness, absolute measurements of vertical head height may not be sensitive enough to detect small differences in lameness, and extraneous head movement by the horse due to curiosity, excitement or nervousness interferes with the accurate measurement of vertical head movement asymmetry. We describe a

A comparative molecular analysis of developing mouse forelimbs and hindlimbs using serial analysis of gene expression (SAGE).

The analysis of differentially expressed genes is a powerful approach to elucidate the genetic mechanisms underlying the morphological and evolutionary diversity among serially homologous structures, both within the same organism (e.g., hand vs. foot) and between different species (e.g., hand vs. wing). In the developing embryo, limb-specific expression of Pitx1, Tbx4, and Tbx5 regulates the determination of limb identity. However, numerous lines of evidence, including the fact that these three genes encode transcription factors, indicate that additional genes are involved in the Pitx1-Tbx hie

Sympathetic and afferent neurones projecting into forelimb and trunk nerves and the anatomical organization of the thoracic sympathetic outflow of the rat.

The anatomy of the cervicothoracic sympathetic nerves was studied in the rat. Details of the arrangements of white and grey rami communicantes and superior cervical, middle cervical and stellate ganglia are given. Dorsal root and sympathetic ganglion neurones projecting to skin and muscle of the forelimb and trunk were labelled retrogradely with horseradish peroxidase (HRP) in order to study their number, segmental distribution and location. HRP was applied to forelimb nerves supplying skeletal muscles (Ramus profundus of radial nerve, RP) or hairy skin (N. cutaneus brachii lateralis superior

Use of a type II external skeletal fixator for repair of delayed union in three calves with forelimb fractures.

Three calves, 3 to 6 months old, were admitted for evaluation and treatment of fractures of the forelimb that had resulted from use of forced extraction at birth. Each of these calves had clinical and radiographic evidence of delayed union of a fractured metacarpus. All calves were treated, using a type II external skeletal fixator consisting of 0.25-inch-diameter transcortical pins and acrylic connecting bars. Two calves required open reduction of the fracture to obtain anatomic alignment of the limb. On the basis of the 3 calves described here, it would appear that type II external skeletal
