OBJECTIVE: A novel biocatalyst for Baeyer-Villiger oxidations is necessary for pharmaceutical and chemical industries, so this study aims to find a Baeyer-Villiger monooxygenase (BVMO) and to improve its stability by immobilization.
RESULTS: Acetone, the simplest ketone, was selected as the only carbon source for the screening of microorganisms with a BVMO. A eukaryote, Fusarium sp. NBRC 109816, with a BVMO (FBVMO), was isolated from a soil sample. FBVMO was overexpressed in E. coli and successfully immobilized by the organic-inorganic nanocrystal formation method. The immobilization improved

Mycetoma is a chronic granulomatous disease, classified into eumycetoma caused by fungi and actinomycetoma due to aerobic filamentous actinomycetes. Mycetoma can be found in geographic areas near the Tropic of Cancer. Mexico is one of the countries in which actinomycetoma is endemic. We report an extraordinary case of an adult male with double eumycetoma caused by Madurella mycetomatis and Fusarium verticillioides on both feet.

The objective of the presented study was to examine the influence of Fusarium mycotoxins (zearalenone--ZEN and deoxynivalenol--DON), administered separately and in combination, on the activity of cecal enzymes (β-glucosidase and β-glucuronidase) in gilts which were fed fodder con- taminated with these mycotoxins. The activity of β-glucosidase and β-glucuronidase varied in the range of 0.170-1.236 μmol · h(-1) · mg(-1) and 8.701-96.704 μmol · h(-1) · mg(-1), respectively. In the first two weeks, the toxins had no significant effect on the activity of β-glucosidase and β-glucuronidase in the asc

Redeemed interest in discovery and characterization of naturally occurring antimicrobials arises from the fact that these compounds possess diverse biological activities useful in the development of new therapeutics. In this study, 35 fungi from extreme environments of effluent treatment plants were screened for antimicrobial activity. An isolate HKF15 produced antimicrobial compound that showed activities against pathogenic and multidrug resistant bacteria; and was identified as Fusarium sp. The active principle was separated through activity-guided HPLC fractionation of 1:1 ethyl acetate cru

Fusarium species are common soil-inhabiting organisms and plant pathogens. Human infections are usually precipitated by local or systemic predisposing factors, and disseminated infection is associated with impaired immune responses. Skin infections caused by Fusarium spp. include keratitis, onychomycosis, mycetoma, painful discrete erythematous nodules. Hyperkeratotic skin lesions caused by Fusarium spp. are, however, rarely reported. We report a case of hyperkeratotic verrucous warty skin lesion in the foot of a 50-year-old immunocompetent male, farmer by occupation.

Two strains of soil-borne Fusarium solani, both characterized for their ability to produce cyclosporin A and C, were examined for their pathogenicity in severe combined immunodeficiency (SCID) and BALB/c male mice. Intravenous (i.v.) infections with F. solani conidia were performed. No mortality was observed after infection with 0.3-1.6 x 10(7) cfu per mouse in SCID and BALB/c mice. When mice were infected with 0.8-1.5 x 10(6) cfu per mouse and 2 days later with 1.2-1.9 x 10(6) cfu per mouse, 28.6-85.7% survival occurred over a 25-day period, depending on the F. solani strain and the inbred mo

Fungal infections of the skin and deeper tissues of the periorbital region are quite rare. We report a case of a localized, deep periorbital necrotizing Fusarium infection in an otherwise healthy, elderly lady. Since the clinical features and histopathological findings of Fusarium infection are by no means characteristic, the definitive diagnosis was achieved with the help of microbiological examination of cultured organisms. A combined medical and surgical therapy led to adequate control of infection. To conclude, localized, deep periorbital necrotizing soft tissue infection by Fusarium in an

PURPOSE: The authors report the first case of Fusarium solani keratitis that progressed to fungal endophthalmitis and was successfully treated with amphotericin B lipid complex (ABLC).
METHOD: The case of a 34-year-old immunocompetent woman who developed a contact lens-related F. solani keratitis requiring emergency penetrating keratoplasty (PKP) was analyzed. The immunocompetent patient developed fungal endophthalmitis (anterior chamber tap positive for F. solani three months after PKP) and was eventually treated with ABLC.
RESULTS: Systemic amphotericin B (total, 0.42 g) and ketoconazole i

In the European coastal dunes, marram grass (Ammophila arenaria) is planted in order to control sand erosion. In the years 1986 to 1991, workers on the Wadden islands in the Netherlands planting marram grass showed lesions of skin and mucous membranes, suggesting a toxic reaction. Fusarium culmorum dominated the mycoflora of those marram grass culms that were used for planting. This plant material had been cut and stored for more than one week in the open. The Fusarium toxin deoxynivalenol (DON) was detected in the suspect marram grass culms. Isolated F. culmorum strains were able to produce D

Fusarium species are saprophytic fungi that may colonize human skin and nails and may rarely cause invasive infections in traumatized tissue and in debilitated and immunocompromised patients. We report herein a case of invasive intranasal Fusarium oxysporum infection in a diabetic patient. This unusual presentation potentially can be confused with early rhinocerebral zygomycosis clinically and histologically. Distinguishing morphologic features and the possible role of diabetes in promoting this infection are discussed.

Disseminated infection caused by Fusarium moniliforme is described in a 32-year-old granulocytopenic man with malignant lymphoma being treated with cytotoxic drugs and corticosteroids. Infected skin denuded by antecedent severe varicella-zoster infection was the probable source of fungemia. F. moniliforme grows rapidly on common mycological media as a lavender- to violet-colored mold at 25 to 37 degrees C. Its aerial hyphae produce fusoid macroconidia and characteristic fusiform microconidia in chains. The morphology of hyphae in tissue closely resembles species of Aspergillus and is not diagn

Xylan, one of the sugar polymer components of biomass, is complex in the number and types of glycosidic bonds that comprise its structure. Due to this complexity, xylan hydrolysis requires many different enzymatic activities to fully release available sugars for subsequent fermentation to fuels and chemicals. As pretreatment severities are decreased to prevent degradation of biomass carbohydrates, the extent of hydrolysis of xylan bonds is also decreased. Due to the limited hydrolysis associated with lower severity treatments, esterases will be needed to hydrolyze acetyl-, feruloyl-, or coumar

We present the complete genome sequence of a novel alternavirus, tentatively named "Fusarium falciforme alternavirus 1 (FfAV1)", isolated from Fusarium falciforme. The host, F. falciforme strain Fod375, was isolated from a soil sample in Spain in 2012 and was found to be infected with a virus containing a tetra-segmented double-stranded (ds) RNA genome. The genome segments, designated as dsRNA1 (3529 bp), dsRNA2 (2641 bp), dsRNA3 (2459 bp), and dsRNA4 (1471 bp), each possess a single open reading frame (ORF). The protein predicted from dsRNA1 contains the typical domains of an RNA-dependent RN

Fusarium species are important opportunistic fungal pathogens. Diabetic foot infection (DFI) involves diverse causative pathogens; however, the potential for fungal etiologies is often overlooked. We describe a case of DFI following foot trauma. Despite prior broad-spectrum antimicrobial therapy, the wound failed to heal and presented persistent swelling and pain. Microbiological re-evaluation and molecular identification confirmed the causative agent as Fusarium keratoplasticum (F. keratoplasticum), a pathogenic species within Fusarium solani species complex (FSSC). The isolate exhibited high

Hairy cell leukaemia (HCL) is a B-cell malignancy whose induction chemotherapy consists of 5 days of cladribine, a purine nucleoside analogue. While there are consensus guidelines from the National Comprehensive Cancer Network (NCCN) on antifungal prophylaxis in the setting of acute myeloid leukaemia or acute lymphocytic leukaemia, there are no consensus guidelines for antifungal prophylaxis during induction treatment of HCL. We describe a case of angioinvasive Fusarium infection in a healthy, young patient who completed cladribine chemotherapy for HCL. While rare, fusariosis poses a significa

Modern fungicides have made significant contributions to crop disease management, but the development of resistant fungal strains has caused their failure in disease control. Therefore, developing fungicides with novel action mechanisms is the most effective measure to manage resistance. Quinofumelin, a novel quinoline fungicide, exhibits exceptional antifungal activity against phytopathogens. However, there is currently no available information on its mechanism of action. Here, we used transcriptome and metabolome analysis to observe a co-enrichment pattern of differentially expressed genes (
