Targeting mitochondrial G-quadruplex DNA with a theragnostic small molecule for cervical Cancer imaging and therapy.

Mitochondria are emerging as critical hubs for cancer therapeutics, yet the development of agents that simultaneously enable real-time imaging and targeted therapy remains a formidable challenge. Here we report LN1, a smart theranostic small molecule designed to selectively target mitochondrial G-quadruplex (mtG4) DNA through a triphenylphosphonium (TPP) moiety integrated with a triphenylamine-derived fluorescent scaffold. LN1 exhibits high-affinity binding to mtG4 structures (Kd = 0.2 μM) with a 1:2 stoichiometry, leading to a pronounced fluorescence turn-on response via restriction of intram

G-quadruplex DNA reprograms the photochemistry of CX-5461 toward radical-driven anticancer activity.

CX-5461 (pidnarulex) is the most clinically advanced small-molecule ligand associated with G-quadruplex (G4) targeting and shows activity in DNA repair-deficient tumors, yet its clinical use is associated with dose-limiting phototoxicity. Here, we repurpose this intrinsic photoreactivity to investigate CX-5461 as a G4-associated, light-activated antitumor scaffold. In solution, CX-5461 produces both Type I and Type II reactive oxygen species (ROS), whereas complexation with G4 DNA markedly decreases detectable singlet oxygen (1O2) formation and favors radical-associated Type I oxidation. Selective Delivery of Anticancer Natural G-Quadruplex Ligands by the AT11 Aptamer for Gastric Cancer Treatment.

Searching for G-quadruplex-selective ligands as anticancer agents, we recently identified the natural compounds bulbocapnine, chelidonine, dicentrine, ibogaine, and rotenone as novel interactors of G-quadruplexes. Herein, to investigate their ability to interact with a specific carrier for selective delivery to cancer cells, the dimeric G-quadruplex-forming aptamer AT11 was used as a model. NMR spectroscopy, molecular modeling, circular dichroism, and fluorescence spectroscopy allowed the preferential interaction to be proven with the 3'-end G-quartet for bulbocapnine, chelidonine, dicentrine,

DHX36 modulates stress granule assembly independent of recruitment of mRNAs with G-quadruplex sequence motifs.

Stress granules are RNA-protein condensates that form in response to an increase in untranslating mRNPs (messenger ribonucleoproteins). Stress granules form by the condensation of mRNPs through a combination of protein-protein, protein-RNA, and RNA-RNA interactions. Several reports have suggested that G-rich RNA sequences capable of forming G-quadruplexes (rG4s) promote stress granule formation. Here, we provide three observations arguing that G-tracts do not promote messenger RNA (mRNA) accumulation in stress granules in human osteosarcoma cells. First, we observed no difference in the accumu

G-quadruplex stabilization induces DNA breaks in pericentromeric repetitive DNA sequences in B lymphocytes.

DNA secondary G-quadruplex (G4) structures can impair and even obstruct DNA replication. Defects in processing G4 structures are associated with replication stress, a common property of both B cell cancers and hyperproliferative premalignant cells. Genome instability arising from replication stress is a hallmark of cancer and strongly contributes to the chromosome rearrangements in B cell cancers. Here, we define the impact of G4-stabilizing ligands on generating genome instability in primary and malignant B cells. Treatment with the G4-stabilizing compound pyridostatin (PDS) causes breaks and

Exploring RNA G-quadruplex in the rabies virus genome and its potential against RABV infection.

Rabies virus (RABV) is a prototypical neurotropic RNA virus that can cause rabies with an almost 100% fatality rate. Despite great efforts having been made to identify an effective therapeutic target of rabies, the host factors involved in RABV infection and the underlying mechanisms remain poorly understood. There are no effective drug targets for human rabies to date. Therefore, a better understanding of rabies pathogenesis and exploring new targets for antiviral treatments are crucial and urgent. In this study, we identify the presence of G-quadruplex (G4) in the RABV genome and uncover a n

An ultrasensitive dual-mode aptasensor for patulin based on the upconversion particles and G-Quadruplex-hemin DNAzyme.

Patulin (PAT) is a mycotoxin-produced secondary metabolite that can contaminate foods, causing toxic effects on animal and human health. Therefore, for the first time, we have constructed a "turn-on" dual-mode aptamer sensor for PAT using oleic acid-coated upconversion nanomaterials (OA-UCNPs) and G-Quadruplex-hemin DNAzyme (G4-DNAzyme) as fluorescent and colorimetry probes. The sensor employs aptamers binding to PAT as recognition elements for specific molecule detection. Mxene-Au can be used as a biological inducer to assist OA-UCNPs in controlling fluorescence intensity. In addition, colori

Catalytic and Selective Red Light-Triggered Photodegradation of a G-Quadruplex DNA by a Zinc (II) Phthalocyanine.

Water-soluble phthalocyanine (Pc) derivatives have been regarded as potential G-quadruplex (G4) nucleic acid-targeting ligands for anticancer therapy and have been extensively studied as effective photosensitizers for photodynamic therapy (PDT). Understanding how photosensitizers interact with nucleic acids and the subsequent photolytic reactions is essential for deciphering the initial steps of PDT, thereby aiding in the development of new photosensitizing agents. In this study, we found that red-light irradiation of a mixture of a Zn(II) Pc derivative and an all-parallel G4 DNA leads to cata

Synthesis of 1,10-Phenanthroline-2,9-bistriazoles: Evaluation as G-Quadruplex Binders and Anti-Tumor Activity.

Novel 1,10-phenanthroline-2,9-bistriazoles derivatives have been synthesized by copper-catalyzed azide/alkyne cycloaddition reactions and assessed for their ability to bind and stabilize G-quadruplex (G4) structures. Ten novel compounds were evaluated using Förster resonance energy transfer (FRET) melting, circular dichroism (CD), and fluorescence spectroscopy on several G4 sequences. Biophysical characterization led to the identification of compounds 4 a, 4 b, and 5 b as good G4 ligands of KRAS G4 sequences. The impact on cell viability of all derivatives was also assessed, revealing weak eff

Genetic variations in G-quadruplex forming sequences affect the transcription of human disease-related genes.

Guanine-rich DNA strands can fold into non-canonical four-stranded secondary structures named G-quadruplexes (G4s). G4s folded in proximal promoter regions (PPR) are associated either with positive or negative transcriptional regulation. Given that single nucleotide variants (SNVs) affecting G4 folding (G4-Vars) may alter gene transcription, and that SNVs are associated with the human diseases' onset, we undertook a novel comprehensive study of the G4-Vars genome-wide (G4-variome) to find disease-associated G4-Vars located into PPRs. We developed a bioinformatics strategy to find disease-relat

G-quadruplex in the TMV Genome Regulates Viral Proliferation and Acts as Antiviral Target of Photodynamic Therapy.

Plant viruses seriously disrupt crop growth and development, and classic protein-targeted antiviral drugs could not provide complete protection against them. It is urgent to develop antiviral compounds with novel targets. Photodynamic therapy shows potential in controlling agricultural pests, but nonselective damage from reactive oxygen species (ROS) unexpectedly affects healthy tissues. A G-quadruplex (G4)-forming sequence in the tobacco mosaic virus (TMV) genome was identified to interfere the RNA replication in vitro, and affect the proliferation of TMV in tobacco. Fluorescent copper conjugated curcumin cysteine nanoprobe for selective determination of Fe3+ and G-quadruplex DNA.

The synthesis of fluorescent copper-curcumin-cysteine (Cu-CC) as a sensing platform is reported. The synthesized probe has been confirmed by UV-visible spectroscopy, FT-IR, XRD, SEM, and TEM characterization techniques, respectively. The mechanistic aspects of selective sensing of Fe3+ and detection of different G-quadruplex DNA have been illustrated based on the "turn-off-on" concept of a regeneratable fluorescence sensing probe at λex 450 nm. Interestingly, we have noticed a high selectivity to Fe3+ ion by the developed Cu-CC sensing probe in comparison with other metal ions. Furthermore, th

Targeting RNA G-Quadruplex in SARS-CoV-2: A Promising Therapeutic Target for COVID-19?

The COVID-19 pandemic caused by SARS-CoV-2 has become a global threat. Understanding the underlying mechanisms and developing innovative treatments are extremely urgent. G-quadruplexes (G4s) are important noncanonical nucleic acid structures with distinct biofunctions. Four putative G4-forming sequences (PQSs) in the SARS-CoV-2 genome were studied. One of them (RG-1), which locates in the coding sequence region of SARS-CoV-2 nucleocapsid phosphoprotein (N), has been verified to form a stable RNA G4 structure in live cells. G4-specific compounds, such as PDP (pyridostatin derivative), can stabi

Target-controlled in situ formation of G-quadruplex DNAzyme for a sensitive visual assay of telomerase activity.

It is of great importance to achieve facile and reliable detection of telomerase because it is an important cancer biomarker. The complex components of cell extracts and ultra-low concentration of telomerase makes it more difficult to realize a simple and sensitive visual detection of telomerase activity. Herein, a facile and sensitive visual strategy was developed for the detection of telomerase based on the telomerase-controlled in situ formation of a G-quadruplex-hemin DNAzyme. To avoid the influence of the complex components of cell extracts, a telomerase substrate (TS) was immobilized ont

CX-5461 is a DNA G-quadruplex stabilizer with selective lethality in BRCA1/2 deficient tumours.

G-quadruplex DNAs form four-stranded helical structures and are proposed to play key roles in different cellular processes. Targeting G-quadruplex DNAs for cancer treatment is a very promising prospect. Here, we show that CX-5461 is a G-quadruplex stabilizer, with specific toxicity against BRCA deficiencies in cancer cells and polyclonal patient-derived xenograft models, including tumours resistant to PARP inhibition. Exposure to CX-5461, and its related drug CX-3543, blocks replication forks and induces ssDNA gaps or breaks. The BRCA and NHEJ pathways are required for the repair of CX-5461 an

A Nitroxide-Tagged Platinum(II) Complex Enables the Identification of DNA G-Quadruplex Binding Mode.

We reported a novel strategy for investigating small molecule binding to G-quadruplexes (GQs). A newly synthesized dinuclear platinum(II) complex (Pt2L) containing a nitroxide radical was shown to selectively bind a GQ-forming sequence derived from human telomere (hTel). Using the nitroxide moiety as a spin label, electron paramagnetic resonance (EPR) spectroscopy was carried out to investigate binding between Pt2L and hTel GQ. Measurements indicated that two molecules of Pt2L bind with one molecule of hTel GQ. The inter-spin distance measured between the two bound Pt2L, together with molecula

Identification and characterization of a new G-quadruplex forming region within the kRAS promoter as a transcriptional regulator.

kRAS is one of the most prevalent oncogenic aberrations. It is either upregulated or mutationally activated in a multitude of cancers, including pancreatic, lung, and colon cancers. While a significant effort has been made to develop drugs that target kRAS, their clinical activity has been disappointing due to a variety of mechanistic hurdles. The presented works describe a novel mechanism and molecular target to downregulate kRAS expression--a previously undescribed G-quadruplex (G4) secondary structure within the proximal promoter acting as a transcriptional silencer. There are three distinc

CdSe/ZnS quantum dots-G-quadruplex/hemin hybrids as optical DNA sensors and aptasensors.

The luminescence of CdSe/ZnS QDs is quenched via electron transfer by hemin/G-quadruplex associated with the particles. This phenomenon is implemented to develop DNA sensors or aptasensors by tailoring hairpin-functionalized QDs that generate the hemin/G-quadruplex quenchers upon sensing of the respective analytes.

Multilevel ensemble for genome-wide prediction of cell-specific G-quadruplexes utilizing transformed sequences and enhanced chromatin accessibility with G4Beacon2.

G-quadruplexes (G4) are prevalent non-B DNA structures playing crucial biological roles in cells. Although experimental technologies for G4 identification in vitro and in vivo are advancing, computational prediction methods are increasingly preferred for their efficiency, convenience, and cost effectiveness. However, existing tools primarily perform in vitro G4 predictions that lack cell-specific information and are often non-genome-wide, with high-performance genome-wide in vivo cell-specific G4 prediction models still lacking. Here, we present G4Beacon2, a genome-wide cell-specific G4 predic

Stabilization of G-Quadruplexes Modulates the Expression of DNA Damage and Unfolded Protein Response Genes in Canine Lymphoma/Leukemia Cells.

G-quadruplexes have been identified as a promising anti-cancer target because of their ability to modulate the stability of mRNAs encoding oncogenes, tumor suppressor genes, and other potential therapeutic targets. Deregulation of DNA damage and Unfolded Protein Response pathways in cancer cells may create vulnerabilities that can be exploited therapeutically. Previous studies have shown variations in the relative expression of DDR and UPR components in canine lymphoma and leukemia cell lines CLBL-1, CLB70, and GL-1. In the present study, we report the presence of G-quadruplex structures in th
