Functional investigation of Zur in metal ion homeostasis, motility and multiple stresses resistance in cyanobacteria Synechocystis sp. PCC 6803.

Zur (zinc uptake regulator), a member of the Fur (ferric uptake regulator) family of transcriptional regulators, plays multifaceted roles by regulating the gene expressions, such as modulating zinc ion uptake by regulating the znuABC gene cluster and influencing bacterial motility by modulating genes associated with flagella or pili. The photosynthetic autotroph Synechocystis sp. PCC 6803 is frequently used as an indicator organism for water pollution and a cell factory for high-value biochemical production in synthetic biology. During its growth, this organism often encounters various abiotic

The three Bacillus anthracis toxin genes are coordinately regulated by bicarbonate and temperature.

The two Bacillus anthracis toxins are composed of three proteins, protective antigen, lethal factor, and edema factor. The structural genes for these three components are located on the virulence plasmid pXO1. We constructed transcriptional fusions between the regulatory region of each of these genes and lacZ. Each construct was then inserted as a single copy at the corresponding toxin gene locus on pXO1, resulting in three isogenic strains. Two environmental factors, bicarbonate and temperature, were found to induce beta-galactosidase synthesis in each recombinant strain. Furthermore, the tra

Role of FlbT in flagellin production in Brucella melitensis.

It was recently demonstrated that the pathogen Brucella melitensis produces a polar sheathed flagellum under the control of the master regulator FtcR. However, the regulatory mechanism controlling the flagellar assembly remains unknown. In this work, we investigate the flagellar hierarchy of B. melitensis as well as the flagellin FliC regulation. We show that a mutation in fliF or flgE (coding for the basal body structure and the hook, respectively) does not affect FliC synthesis, suggesting that production of FliC does not depend on the flagellar assembly. We demonstrate that FlbT is a FliC a

[Penicillin G acylase--synthesis, regulation, production].

Penicillin G acylase (PGA) is one of very important industrial enzymes used in the production of polysynthetic beta-lactam antibiotics. This enzyme catalyzes the hydrolysis of the amidic bond of penicillin G with the development of 6-aminopenicillanic acid which serves as the initial substance for the production of semisynthetic penicillins. In the strain Escherichia coli W ATCC 11105 and ATCC 9637, PGA is coded by the pga gene on the chromozome and synthesized as the pre-pro-PGA (pp PGA) precursor, which is transported, with probable participation of the chaperon system, to the periplasmatic

Calcium regulation of Pseudomonas aeruginosa metabolism.

UNLABELLED: Pseudomonas aeruginosa (Pa), a ubiquitous opportunistic pathogen, is notorious for causing diverse life-threatening infections, including lethal lung infections in patients suffering from cystic fibrosis (CF). Pa's ability to adapt to various environments is largely attributed to its metabolic flexibility and extensive regulatory networks. Understanding the mechanisms driving Pa adaptation during colonization of the airways of CF patients is of critical clinical importance. Previous studies by our group and others have shown that elevated Ca2+, a characteristic feature of CF airway

Temperature regulation of the streptococcal pyrogenic exotoxin A-encoding gene (speA).

The gene encoding the bacterial superantigen streptococcal pyrogenic exotoxin A is often found in streptococcal strains associated with the recently described streptococcal toxic shock syndrome. Here we demonstrate that this gene is expressed at approximate fourfold higher levels in cells grown at 37 degrees C when compared to cells grown at 26 degrees C. This suggests there is increased production of this toxin when Streptococcus pyogenes is found in infections of the soft tissues and bloodstream, as opposed to S. pyogenes that have not breached the epithelial layers and are living on the sur

The History of Bordetella pertussis Genome Evolution Includes Structural Rearrangement.

Despite high pertussis vaccine coverage, reported cases of whooping cough (pertussis) have increased over the last decade in the United States and other developed countries. Although Bordetella pertussis is well known for its limited gene sequence variation, recent advances in long-read sequencing technology have begun to reveal genomic structural heterogeneity among otherwise indistinguishable isolates, even within geographically or temporally defined epidemics. We have compared rearrangements among complete genome assemblies from 257 B. pertussis isolates to examine the potential evolution o

Breaking antimicrobial resistance by disrupting extracytoplasmic protein folding.

Antimicrobial resistance in Gram-negative bacteria is one of the greatest threats to global health. New antibacterial strategies are urgently needed, and the development of antibiotic adjuvants that either neutralize resistance proteins or compromise the integrity of the cell envelope is of ever-growing interest. Most available adjuvants are only effective against specific resistance proteins. Here, we demonstrate that disruption of cell envelope protein homeostasis simultaneously compromises several classes of resistance determinants. In particular, we find that impairing DsbA-mediated disulf

Overexpression of OsCM alleviates BLB stress via phytohormonal accumulation and transcriptional modulation of defense-related genes in Oryza sativa.

Xanthomonas oryzae is a serious pathogen causing bacterial leaf blight (BLB) disease in rice, markedly reducing its yield. In this study, the rice chorismate mutase (OsCM) gene was overexpressed in a bacterial leaf blight-susceptible rice line to investigate the functional role of OsCM in response to bacterial leaf blight stress. We reported that overexpression of OsCM altered the downstream pathway of aromatic amino acids, mitigating pathogen stress by altering stress-responsive genes and hormonal accumulation. Phenotypic evaluation showed that the lesion length in the transgenic line was sig

Identification, cloning and heterologous expression of biosynthetic gene cluster for desertomycin.

From our in-house microbial genome database of secondary metabolite producers, we identified a candidate biosynthetic gene cluster for desertomycin from Streptomyces nobilis JCM4274. We report herein the cloning of the 127-kb entire gene cluster for desertomycin biosynthesis using bacterial artificial chromosome vector. The entire biosynthetic gene cluster for desertomycin was introduced in the heterologous host, Streptomyces lividans TK23, with an average yield of more than 130 mg l-1.

LisRK is required for optimal fitness of Listeria monocytogenes in soil.

Listeria monocytogenes is a food-borne pathogen responsible for the disease listeriosis. It is ubiquitously found in the environment and soil is one of its natural habitats. Listeria monocytogenes is highly capable of coping with various stressful conditions. We hypothesized that stress-responsive two-component systems such as LisRK might contribute to the adaptation of L. monocytogenes to the soil environment. Indeed, investigations of the population dynamics of wild-type and mutant strains suggest an important role of LisRK for optimal fitness of L. monocytogenes in sterile soil. Results fro

Screening of genes encoding adhesion factors and biofilm formation in Staphylococcus aureus isolates from poultry.

Staphylococcus aureus isolates recovered from poultry between 1970 and 1972 (90 old isolates) and in 2006 (81 recent isolates) were screened for the presence of bap, icaA and icaD genes associated with biofilm formation, and for bbp, cna, ebpS, eno, fib, fnbA, fnbB, clfA and clfB genes that encode microbial surface components recognizing adhesive matrix molecules (MSCRAMMs). Most of the old isolates were collected from broiler breeders affected by staphylococcal tenosynovitis and arthritis, whilst nearly all recent isolates were derived from the nose and cloaca of healthy broilers. Ten recent

Control of virulence gene expression in Bacillus anthracis.

The atxA gene is an important regulator of virulence gene expression in Bacillus anthracis. atxA positively regulates expression of the three genes encoding the anthrax toxin proteins and at least one gene is required for capsule production. Here we report that an atxA-null mutant exhibits phenotypes unrelated to toxin and capsule synthesis. An atxA-null mutant grows poorly on minimal media and sporulates more efficiently than the parent strain. Numerous transposon-generated promoter-lacZ fusions at distinct loci on pXO1 exhibit CO2-enhanced atxA-dependent expression similar to that observed f

Regulation of Ansamitocin P-3 Biosynthesis by the Response Regulator CrsB in Actinosynnema pretiosum.

Ansamitocin P-3 (AP-3) is an ansamycin antibiotic produced by Actinosynnema pretiosum with high clinical and commercial value. Its biosynthesis is tightly controlled, yet the underlying transcriptional network is not fully understood. Here, we identified the two-component system CrsAB and characterized the response regulator CrsB. Deletion of crsB resulted in a 57% reduction in AP-3 yield without impairing morphological development or biomass accumulation. RNA‑Seq showed that CrsB acts as a global regulator, influencing energy metabolism, cell integrity, and biosynthetic capacity. At the ferme

Analysis of Chlamydia trachomatis Gene Function via Controlled Up- or Downregulation.

Due to genome streamlining, many genes in the ~1 Mb genome of Chlamydia trachomatis are essential and cannot be deleted. Ectopic overexpression and CRISPRi-based knockdown are complementary genetic approaches to study the function of genes in C. trachomatis, including essential genes. When used in combination with inducible promoters, these tools allow researchers to determine how precise regulation of gene expression contributes to proper progression of the chlamydial developmental cycle. This chapter includes protocols which detail ectopic overexpression of a protein or conversely, transcrip

MMP-3 expression and release by rheumatoid arthritis fibroblast-like synoviocytes induced with a bacterial ligand of integrin alpha5beta1.

Fibroblast-like synoviocytes (FLSs) play a major role in the pathogenesis of rheumatoid arthritis (RA) by secreting effector molecules that promote inflammation and joint destruction. How these cells become and remain activated is still elusive. Both genetic and environmental factors probably play a role in transforming FLSs into inflammatory matrix-degrading cells. As bacterial products have been detected in the joint and shown to trigger joint inflammation, this study was undertaken to investigate whether a bacterial ligand of integrin alpha5beta1, protein I/II, could contribute to the aggre

Developing cumate-inducible gene expression systems for application in Pseudomonas putida KT2440.

AIMS: Pseudomonas putida KT2440 possesses a versatile metabolic network and broad substrate utilization capacity, making it as an attractive chassis for synthetic biology applications. However, the limited availability of efficient gene expression tools for precise metabolic regulation has constrained its engineering potential. To address this limitation, we developed two novel cumate-inducible expression systems. METHODS AND RESULTS: In this study, we developed two novel cumate-inducible expression systems by engineering distinct cumate-responsive operators, CuO1 and CuO2, respectively deriv

Transcriptomic Analysis of Salmonella Typhimurium After Exposure to Blue Light.

Antimicrobial blue light (aBL) at 405 nm reduces the viability of foodborne pathogens, including Salmonella. Visible light, including blue light, can kill bacteria by a mechanism that involves reactive oxygen species (ROS) generated after excitation of endogenous photosensitizers. However, the molecular response of Salmonella to aBL is largely unknown. This study was undertaken to characterize differential gene expression in Salmonella Typhimurium strain 14028s following sublethal exposure to 405 nm aBL and to evaluate the phenotypic relevance of selected upregulated genes. Cell suspensions we

Comparative transcriptomic analysis of perfluoroalkyl substances-induced responses of exponential and stationary phase Escherichia coli.

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) are highly stable chemical contaminants of emerging concern for human and environmental health due to their non-natural chemistry, widespread use, and environmental persistence. Despite conventional metrology, mitigation strategies, and removal technologies, the complexity of this growing problem necessitates alternative approaches to tackle the immense challenges associated with complex environmental PFAS contamination. Recently, biology has emerged as an alternative approach to detect and mitigate PFAS and understand the molecular-level

High quality transcriptome profiling confirms the transcriptional landscape of Treponema pallidum subsp. pallidum.

Syphilis remains a critical global health challenge due to its potential for severe complications and the increase in its incidence rate over recent years. Until recently, the infectious agent of syphilis, Treponema pallidum subsp. pallidum (TPA), could not be cultured in vitro. Advances in co-culture techniques have enabled the effective long-term cultivation of TPA, providing a platform for studying its biology. Limited transcriptional data from TPA have been reported, and many genes in treponemal genomes are annotated based on in silico prediction of putative coding sequences without functi
