Nucleotide distribution analysis of 5'UTRs in genome-scale directs their redesign and expression regulation in yeast.

Non-conventional yeasts have emerged as important sources of valuable products in bioindustries. However, tools for the control of expression are limited in these hosts. In this study, we aimed to excavate the tools for the regulation of translation that are often overlooked. 5'UTR analysis of genome-scale annotated genes of four yeast species revealed a distinct decreasing 'G' frequency in -100 ∼ -1 region from 5040 5'UTRs in Komagataella phaffii. New 5'UTRs were regenerated by base substitutions in defined regions, and replacement of 'G' by 'A' or 'T' in the -50 ∼ -1 region highly facilitate

Hülle Cells of Aspergillus nidulans with Nuclear Storage and Developmental Backup Functions Are Reminiscent of Multipotent Stem Cells.

Some aspergilli are among the most cosmopolitan and ecologically dominant fungal species. One pillar of their success is their complex life cycle, which creates specialized cell types for versatile dispersal and regenesis. One of these cell types is unique to aspergilli-the Hülle cells. Despite being known for over a century, the biological and ecological roles of Hülle cells remain largely speculative. Previously reported data on in vivo Hülle cell formation and localization have been conflicting. Our quantification reveals that Hülle cells can occur at all locations on hyphae and that they s

Developmental regulators in Aspergillus fumigatus.

The filamentous fungus Aspergillus fumigatus is the most prevalent airborne fungal pathogen causing severe and usually fatal invasive aspergillosis in immunocompromised patients. This fungus produces a large number of small hydrophobic asexual spores called conidia as the primary means of reproduction, cell survival, propagation, and infectivity. The initiation, progression, and completion of asexual development (conidiation) is controlled by various regulators that govern expression of thousands of genes associated with formation of the asexual developmental structure conidiophore, and biogen

Transcriptomic responses of Saccharomyces cerevisiae and Hanseniaspora uvarum during separated co-fermentation.

Hanseniaspora uvarum is a predominant yeast species in vineyards and some strains demonstrate oenological potential by enhancing the aroma complexity and reducing the content of volatile acid and ethanol in wine. H. uvarum kept in our lab could improve the flavor and quality of wine when co-fermented with Saccharomyces cerevisiae, but its growth was inhibited by S. cerevisiae. To investigate the interaction mechanisms of S. cerevisiae and H. uvarum, double-compartment fermentor with dialysis tube of 1000 kDa MWCO was selected for separated co-fermentation (SCF) in synthetic grape juice at 25℃

Combined transcriptomics and cellular analyses reveal the molecular mechanism by which Candida tropicalis ZD-3 adapts to and degrades gossypol.

Microbial degradation techniques are often considered an environmentally friendly and cost-effective strategy for reducing gossypol toxicity. However, the mechanism by which Candida tropicalis degrades gossypol remains unclear. In the current study, we aimed to establish the mechanisms of biodegradation and adaptation mechanisms by C. tropicalis ZD-3. The toxicological evaluation results revealed that ZD-3 adapts to gossypol primarily by activating the antioxidant defense system to alleviate the oxidative stress response induced by gossypol. Transcriptomic analyses further suggested that ZD-3

Distinct cell morphotypes of Aureobasidium melanogenum ZN exhibit differential functional profiles in promoting maize growth.

Black yeast-like fungi of the genus Aureobasidium exhibit morphological plasticity, but whether distinct cellular states within the same genetic background are associated with different plant growth-promoting functions remains unclear. Here, yeast-like cells (YL), swollen cells (SC), and chlamydospores (CH) of Aureobasidium melanogenum ZN were characterized. YL was associated mainly with siderophore production and laccase activity, SC with extracellular polysaccharide accumulation, and CH with phosphate mobilization and higher ammonia and IAA production. Whole-genome and comparative genomic an

Metabolome and transcriptome unveil the mechanism of light on regulating beauvericin synthesis in Cordyceps chanhua.

Cordyceps chanhua, an important cordycipitoid medical mushroom with wide use in Asia, has gained attention for its bioactive component beauvericin (BEA), which is of medicinal value as a drug lead, but also of food safety risk. Recent observations by our group revealed a significant decrease of BEA content in C. chanhua when exposed to light, but the underlying regulatory mechanisms remain elusive. In this study, a comprehensive approach combining metabolomics and transcriptomics was employed to investigate the effects of white light on the secondary metabolism of C. chanhua for elucidation of

Cryptococcal nutrient acquisition and pathogenesis: dining on the host.

SUMMARYPathogens must acquire essential nutrients to successfully colonize and proliferate in host tissue. Additionally, nutrients provide signals that condition pathogen deployment of factors that promote disease. A series of transcriptomics experiments over the last 20 years, primarily with Cryptococcus neoformans and to a lesser extent with Cryptococcus gattii, provide insights into the nutritional requirements for proliferation in host tissues. Notably, the identified functions include a number of transporters for key nutrients including sugars, amino acids, metals, and phosphate. Here, we

Early transcriptional response of Paracoccidioides brasiliensis upon internalization by murine macrophages.

Paracoccidioides brasiliensis, a thermal dimorphic fungus, is the etiologic agent of the most common systemic mycosis in Latin America, paracoccidioidomycosis. The yeast form of P. brasiliensis acts as a facultative intracellular pathogen being able to survive and replicate within the phagosome of nonactivated murine and human macrophages. This ability has been proposed to be crucial to the development of disease. Thus, P. brasiliensis may have evolved mechanisms that counteract the constraints imposed by phagocytic cells. By using cDNA microarray technology we evaluated the early transcriptio

Regulation and compartmentalization of β-lactam biosynthesis.

Penicillins and cephalosporins are β-lactam antibiotics widely used in human medicine. The biosynthesis of these compounds starts by the condensation of the amino acids L-α-aminoadipic acid, L-cysteine and L-valine to form the tripeptide δ-L-α-aminoadipyl-l-cysteinyl-D-valine catalysed by the non-ribosomal peptide 'ACV synthetase'. Subsequently, this tripeptide is cyclized to isopenicillin N that in Penicillium is converted to hydrophobic penicillins, e.g. benzylpenicillin. In Acremonium and in streptomycetes, isopenicillin N is later isomerized to penicillin N and finally converted to cephalo

Iron regulatory pathways differentially expressed during Madurella mycetomatis grain development in Galleria mellonella.

Mycetoma is a chronic granulomatous infection of the subcutaneous tissue, most often caused by the fungal pathogen Madurella mycetomatis. Characteristic of the infection is the formation of grains. However, knowledge of the function and formation of the grain is limited. Here, we use a Galleria mellonella larvae infection model and transcriptomic profiling to identify processes associated with M. mycetomatis grain formation. Larvae were infected with M. mycetomatis and, after 4, 24, 72 and 168 h post-inoculation, RNA was extracted from larval content and sequenced. We found that 3498 G. mellon

The role of the veA gene in adjusting developmental balance and environmental stress response in Aspergillus cristatus.

veA belongs to the velvet regulatory system that regulates the development and secondary metabolism of many fungi. To identify the function of veA in Aspergillus cristatus, veA deletion mutants were constructed by homologous recombination via Agrobacterium tumefaciens-mediated transformation. Deletion of veA led to increased conidial production and reduced sexual sporulation. The regulatory role of veA in A. cristatus was not light-dependent, and this differed from its role in other Aspergilli. Furthermore, veA deletion mutants were more sensitive to environmental stressors, including salt, os

Selection and validation of reference genes for quantitative real-time PCR studies during Saccharomyces cerevisiae alcoholic fermentation in the presence of sulfite.

Sulfur dioxide is extensively used during industrial fermentations and contributes to determine the harsh conditions of winemaking together with low pH, high sugar content and increasing ethanol concentration. Therefore the presence of sulfite has to be considered in yeast gene expression studies to properly understand yeast behavior in technological environments such as winemaking. A reliable expression pattern can be obtained only using an appropriate reference gene set that is constitutively expressed regardless of perturbations linked to the experimental conditions. In this work we tested

CgrBrlA, CgrAbaA, and CgrWetA form a developmental regulatory cascade controlling conidiation, spore integrity, and virulence in Colletotrichum graminicola.

Colletotrichum graminicola, the causal agent of maize anthracnose, relies on asexual spores for disease initiation and spread, yet the transcriptional mechanisms controlling conidiation and infection-related development remain incompletely understood. Here, we functionally characterize three putative homologs of the conserved fungal developmental regulators BrlA, AbaA, and WetA in C. graminicola. Targeted deletion analyses showed that CgrbrlA is essential for conidiophore formation and conidium production, while CgrabaA is required for proper conidiophore differentiation and normal sporulation

Identification and characterization of antigenic proteins potentially expressed during the infectious process of Paracoccidioides brasiliensis.

Paracoccidioides brasiliensis causes paracoccidioidomycosis (PCM), a systemic mycosis presenting clinical manifestations ranging from mild to severe forms. A P. brasiliensis cDNA expression library was produced and screened with pooled sera from PCM patients adsorbed against antigens derived from in vitro-grown P. brasiliensis yeast cells. Sequencing DNA inserts from clones reactive with PCM patients sera indicated 35 open reading frames presenting homology to genes involved in metabolic pathways, transport, among other predicted functions. The complete cDNAs encoding aromatic-l-amino-acid dec

Phenotypic characterization of HAM1, a novel mating regulator of the fungal pathogen Cryptococcus neoformans.

UNLABELLED: Cryptococcus neoformans is a fungal pathogen responsible for >200,000 yearly cases with a mortality as high as 81%. This burden results, in part, from an incomplete understanding of its pathogenesis and ineffective antifungal treatments; hence, there is a pressing need to understand the biology and host interactions of this yeast to develop improved treatments. Protein palmitoylation is important for cryptococcal virulence, and we previously identified the substrates of its main palmitoyl transferase. One of them was encoded by the uncharacterized gene CNAG_02129. In the filamentou

PtaB, a lim-domain binding protein in Aspergillus fumigatus regulates biofilm formation and conidiation through distinct pathways.

The exopolysaccharide galactosaminogalactan (GAG) plays an important role in mediating adhesion, biofilm formation, and virulence in the pathogenic fungus Aspergillus fumigatus. The developmental modifiers MedA, StuA, and SomA regulate GAG biosynthesis, but the mechanisms underlying this regulation are poorly understood. PtaB is a lim-domain binding protein that interacts with the transcription factor SomA and is required for normal conidiation and biofilm formation. Disruption of ptaB resulted in impaired GAG production and conidiation in association with a markedly reduced expression of GAG

Exploring mechanisms of resistance to dimethachlone in Sclerotinia sclerotiorum.

BACKGROUND: The dicarboximide fungicide dimethachlone has been widely used in China for more than 12 years to control the Sclerotinia stem rot caused by Sclerotinia sclerotiorum disease. First signs of resistance in the field are reported at low frequency. In this study, four resistant isolate/mutants were used to explore still unknown mechanisms leading to dimethachlone resistance. RESULTS: The resistant isolate/mutants had significantly higher EC50 values compared with the sensitive control isolates. Cross-resistance was confirmed between dimethachlone and procymidone, iprodione and fludiox

Compensatory expression of multidrug-resistance genes encoding ABC transporters in dermatophytes.

Genomic sequencing of several dermatophyte species has revealed that they show small differences in genetic content and genome organization, although each fungus has adapted to specific niches. Thus, it seemed relevant to compare gene expression between species. Here, we examined the transcription modulation of three ATP-binding cassette (ABC) transporter genes (pdr1, mdr2 and mdr4), which code for membrane transporter proteins in four species of Trichophyton ; T. interdigitale, T. rubrum, T. tonsurans and T. equinum . These fungal species were challenged with sub-lethal doses of griseofulvin,

RTA-like proteins regulate azole susceptibility by affecting the expression of the oxidoreductase gene oxrA in Aspergillus fumigatus.

Aspergillus fumigatus, a saprophytic fungus and the principal etiologic agent of aspergillosis, is increasingly implicated in drug-resistant infections, posing challenges to clinical management. RTA-like proteins, a distinctive class of transmembrane fungal proteins, are involved in several stress responses. Nonetheless, their role in fungal azole resistance is yet to be determined. Transcriptomic studies performed in this investigation revealed that azole stimulation significantly upregulates the expression of RTA-like proteins in A. fumigatus. Phenotypic analyses indicated that relative to t
